通常使用对采样滤膜的“离线”分析来研究大气气溶胶（见图2）。32 对采样物质的化学分析包括有机成分（例如 蛋白质、碳氢化合物分子、糖类）和无机成分（例如 金属、盐类）。生物学分析包括可存活与不可存活微生物的含量、利用DNA方法或显微镜进行物种鉴定，以及基于DNA的定量分析。

空气采样滤膜中内毒素与DNA含量的分析

本实验的总体目标是描述如何准确分析采集在滤膜上的空气颗粒物中内毒素和DNA含量。该方法有助于回答室内外生物气溶胶研究中的关键问题，例如理解与健康相关的成因。滤膜采样过程将由我实验室的研究生Yinon Mazar进行演示。对于高体积采样，使用石英纤维制成的滤膜。或者根据研究需求，选择特定类型的滤膜以及最合适的滤膜截留粒径。将用于有机物和生物化合物采样的滤膜分别用铝箔单独包裹后进行预烘烤。然后在实验室马弗炉中于450摄氏度下烘烤至少5小时，以彻底去除任何预先存在的有机残留物。将烘烤后的滤膜储存于20摄氏度，直至用于采样。接下来，清洁并消毒高流量采样器，例如随附文本方案中所述的采样器。然后将预烘烤的滤膜放入滤膜盒中，并按照制造商的说明运行采样器以收集适当的样品。在二级生物安全柜中，使用洁净的直径为1.12厘米的打孔器，将洁净的预烘烤滤膜材料切割成22个圆形小片。使用这些滤膜制备一系列含内毒素加标滤膜，作为对照使用。然后，使用洁净的直径为1.12厘米的打孔器切割含有采集样品的滤膜，并将每份样品放入各自的无热原2毫升离心管中。同时，将干燥的内毒素加标滤膜转移至单独的无热原2毫升离心管中。向每支试管中加入一毫升无热原水，并使用实验室摇床在室温下振荡60分钟。接着，将样品管在微量离心机中以375 ×g离心10分钟。然后，将含有内毒素的上清液转移至新的无热原的 2 毫升离心管中。为内毒素检测准备一个带盖的无热原 96 孔平底微孔板，并首先规划好样品的位置。接下来，打开酶标仪，并将其程序设置为在37摄氏度下进行动力学反应，每5分钟振荡一次，随后在405纳米波长下进行检测。重复5分钟振荡和读板操作共18次。将之前用于加标滤膜的标准内毒素溶液各取 50 微升，分别加入 A1 至 A10 和 B1 至 B10 孔中。将空白样品加入 A11、A12、B11 和 B12 孔。接着，将经离心处理的加标滤膜提取液及其空白样品的上清液加入 C 和 D 行的孔中。然后，根据需要将实验样品及其空白样品的上清液加入其余孔中。为启动检测，迅速向每个孔中加入 50 微升鲎变形细胞甘氨酸溶液。轻轻水平摇动培养板。在将其放入酶标仪并开始实验运行之前完成上述操作。完成后，从酶标仪中取出微孔板。利用标准曲线确定每个样本中的内毒素水平，并计算从样本滤膜中提取内毒素的效率。首先，按照随附的文字实验方案制备DNA加标滤膜。对于每个滤膜样品，准备酸洗玻璃珠混合物：在0.5毫升无菌离心管中加入0.1克直径为425至600微米的玻璃珠和0.3克直径小于或等于106微米的玻璃珠。接着，使用经消毒的直径为1.12厘米的打孔器，从每张滤膜样品的随机位置切下三个圆形小片。将滤膜样品放入独立的螺口2毫升离心管中。然后，将玻璃珠混合物加入每个试管中，并加入60 µL DNA提取试剂盒提供的细胞裂解缓冲液。盖紧试管，将样品放入珠磨机中。使用珠磨机对过滤后的细胞进行机械破碎，持续1分钟，然后将样品置于冰上冷却1分钟。重复此循环五次。随后按照试剂盒供应商的说明书操作，从裂解的细胞中提取DNA。洗脱后，将洗脱液再次加载到提取柱中进行第二次过柱，以提高DNA得率。提前开启定量 PCR 仪，使其有足够时间预热。在新建的程序文件中，于仪器操作软件内输入定量 PCR 运行的相关参数。随后，使用表面 DNA 去污染剂对工作台面进行消毒，并全程使用无菌离心管、枪头和试剂进行操作。此外，在工作台旁放置一桶冰，并将 tac polemoraris 混合物放入冰桶中。按照随附文本方案中的表四所述配制定量 PCR 反应混合液。将反应混合液避光并置于冰上保存，直至使用。接下来，将一块洁净的微孔板放入板架中，以防止其接触工作台面并保持清洁。在定量PCR微孔板的各孔中，分别加入1 µL的标准DNA、样本DNA或无核酸酶水，每个样本设三个复孔。然后，向每个反应孔中加入9 µL的主反应混合液。完成后，用光学粘合膜覆盖板子，并在所有边缘紧密封好。在放入热循环仪之前，将板子放入配备板桶的离心机中，以1000 × g离心1分钟。最后，启动并运行已准备好的定量PCR程序。在内毒素提取过程中，将滤膜样品置于离心机中旋转，以从滤膜上提取内毒素。不同的离心速度会导致提取效率的差异，尤其是在较高内毒素浓度时更为明显。应针对所选择的特定滤膜和执行的方案进行提取效率的初步测试。与采样后的石英滤膜相比，向洁净滤膜中加标可获得更高的提取效率，但这两者的提取效率均低于直接检测标准溶液所达到的效率。一旦掌握该方法，内毒素分析可在三个半小时内完成，前提是操作得当。完成此操作后，可进行其他方法（如DNA测序），以进一步回答有关生物体在大气中传播的更多问题。观看本视频后，您应能充分理解如何高效地从空气样本滤膜中提取内毒素和DNA，并对其进行准确分析。
