1. 新生仔猪的手术麻醉与手术准备
<ol>
	<li>在独立的前室中，于颈部基部对25 kg的猪进行肌肉注射镇静-麻醉混合药物，药物成分为：阿扎哌隆（0.3 mg/kg）、盐酸氯胺酮（10 mg/kg）、右美托咪定（15 µg/kg）。 
	注意：在麻醉前用药混合物中加入布托啡诺（0.005–0.02 mg/kg），以增强术中镇痛效果。</li>
	<li>使用吸入式异氟烷气体麻醉猪（4-5% 异氟烷，氧气流速为 500 ml-1,000 ml/min）2) 使用面罩。使用兽用喉镜（17–25 cm 长直型镜片）观察声带，并对声带局部喷洒10%利多卡因喷雾。在插管前30–60秒喷洒利多卡因，使药物与声带充分接触，以降低声带痉挛和气道阻塞的风险。</li>
	<li>通过声门插入带套囊的气管插管（5.0–7.0 mm 内径（ID））对猪进行插管，并使用异氟烷气体（0.5–2.0% 异氟烷，氧气流速 1,000–2,000 ml/min）维持麻醉2使用密闭循环呼吸麻醉系统在整个手术过程中进行麻醉。通过下颌张力以及足底和眼睑反射反应来评估麻醉深度。呼出的麻醉气体经收集后排出手术室外。</li>
	<li>用2.0%氯己定外科洗手液清洁耳部外表面，随后用70%异丙醇冲洗。使用20 G静脉导管穿刺耳静脉，术中给予静脉输液（乳酸林格液；5–10 ml/kg/hr）。用医用胶带将脉搏血氧仪固定于舌体黏膜表面，以监测心率及外周血氧饱和度（SpO₂）。2).</li>
	<li>将麻醉后的猪置于仰卧位，从胸部中段向尾侧至耻骨腹面区域剃除腹部腹侧的毛发。使用两种交替的2.0%氯己定外科擦洗液与无菌水清洗对该区域进行清洁。</li>
	<li>将麻醉后的猪转移至手术室，用70%异丙醇溶液进行最终手术擦洗，待其自然干燥后，为动物铺巾。</li>
	<li>将直肠温度探针插入直肠约2-4 cm以监测体温。手术过程中将猪置于循环水加热垫上，以维持正常体温（38-40 °C）。</li>
	<li>用四块毛巾以重叠的象限方式覆盖猪的腹部。第一块毛巾横跨剑突处，第二块毛巾沿腹部侧面放置，位于腹正中线外侧约5 cm处。第三块毛巾横跨骨盆的髂嵴，第四块毛巾与第二块毛巾类似（但在对侧），沿腹部侧面放置，位于腹正中线外侧约5 cm处。</li>
	<li>在下方的毛巾铺巾上放置一张带有切口开口以便接触手术部位的大型手术台铺巾，覆盖猪体及整个手术台。最后的铺巾是在大型手术台铺巾上覆盖一张一次性无菌铺巾。</li>
</ol>
2. 腹部手术与肠淋巴干插管
<ol>
	<li>使用手术刀片做一条20 cm的皮肤切口，以暴露下方的腹肌。使用单极电凝（设定为20瓦）切开腹肌各层，暴露壁层腹膜。用梅岑鲍姆剪刀切开一段20 cm长的线性壁层腹膜，以进入腹腔脏器并暴露淋巴管。</li>
	<li>在手术切口的头侧放置牵开器，以在整个手术过程中保持腹腔开放。</li>
	<li>在整个手术过程中，用温热（37 °C）的无菌生理盐水润湿所有组织。轻柔地提起包含结肠、盲肠、回肠和空肠的大段肠管，将其从腹腔移出并置于猪的左侧 flank 区，以暴露上腹部、肝脏和淋巴管。使用额外的布巾单固定外置的肠管，形成吊带以轻柔支撑肠管。</li>
	<li>定位淋巴管：其位置大约在右肾静脉头侧内侧约4 cm处，腔静脉孔尾侧内侧腹侧约6 cm处，并位于右肝叶脏面下方靠近胰腺的位置22,36,37。将淋巴管识别为与门静脉右腹侧段相邻的透明结构36,37。</li>
	<li>使用棉签轻轻分离附着的组织，将淋巴管从周围筋膜中游离出来。当血管的两侧已与周围组织分离后，使用精细钝头镊子在血管下方创建一个&#34;隧道&#34;开口。</li>
	<li>使用精细镊子将三根2-0丝线缝合线穿过淋巴管下方。首先结扎最尾侧的缝线，以闭塞、扩张并使淋巴管充满淋巴液。有意将此缝线的末端留得相对较长（4 cm），以便将导管固定在淋巴管上。另外两根缝线彼此相距1.0 cm，位于已固定的尾侧缝线头侧约1.0–1.5 cm处。将这两根缝线以“单个松结”方式留置，以便更快地将导管固定于血管内。 
	注意：位于最尾侧结扎缝线与上方两根缝线中段缝线之间的淋巴管节段，即为导管插入部位。如需要，可用2-0聚乳酸羟基乙酸共聚物（polyglactin）缝线替代2-0丝线。</li>
	<li>使用虹膜剪在血管上剪开一个小孔，并用精细钝头镊子扩张血管。将一根带有斜面端的特制导管（外径4.06 mm × 内径2.31 mm）插入血管约1.0–1.5 cm，然后结扎上方两根缝线以固定导管位置。使用尾侧缝线的长端将导管牢固固定在血管上。</li>
	<li>用大量温热生理盐水冲洗外置的肠管，并轻柔地将其还纳回腹腔，确保肠道处于正确的解剖位置。</li>
	<li>将导管从左侧中 flank 区（腰 fossa 腹侧5–10 cm处）引出体外。使用手术刀做皮肤切口，并将套管针从腹腔穿至皮肤表面，以创建导管引出通道。使用大型凯利钳通过套管针开口将导管从腹腔内引出体外。</li>
	<li>使用带圆针（锥形针）的2-0聚乳酸羟基乙酸共聚物缝线，以连续单纯缝合法关闭壁层腹膜。使用带圆针的2-0聚乳酸羟基乙酸共聚物缝线，以间断单纯缝合法关闭腹肌各层。</li>
	<li>使用带角针的2-0聚乳酸羟基乙酸共聚物缝线，以皮下连续缝合法关闭皮肤。使用带角针的2-0尼龙缝线，以荷包缝合法将外置的导管固定于皮肤上。</li>
	<li>在猪仍处于麻醉状态时为其穿上特制背心，以便于穿戴，并减少其在恢复期间的应激反应。</li>
</ol>
3. 术后恢复与淋巴液采集
<ol>
	<li>在停止吸入麻醉前约10分钟，肌肉注射布托啡诺（0.1 mg/kg）以提供术后即时镇痛。术后继续每12小时给予布托啡诺（0.1 mg/kg），持续24-48小时，以维持术后镇痛效果。</li>
	<li>术后7天内，每8-12小时监测一次猪的术后并发症。</li>
	<li>使用涂有乙二胺四乙酸（EDTA）并添加抗生素（青霉素6,000 IU、链霉素6 mg和两性霉素B 3 mg）的500 ml聚丙烯冲洗瓶，每12小时收集一次淋巴液，持续7天。</li>
</ol>
4. 淋巴液中载脂蛋白B48、甘油三酯、胆固醇和总蛋白的定量分析
<ol>
	<li>将淋巴液样品在 4 °C 条件下以 1,800 × g 离心 5 分钟。收集上清液，用于甘油三酯、胆固醇和总蛋白的定量分析。</li>
	<li>将上清液分为三个样品：未稀释样品、用蒸馏水 1:20 稀释的样品，以及用蒸馏水 1:100 稀释的最终样品。</li>
	<li>使用未稀释的上清液，采用市售试剂盒测定胆固醇水平。</li>
	<li>使用 1:20 稀释的样品通过市售试剂盒测定甘油三酯水平，使用 1:100 稀释的样品通过二辛可宁酸-总蛋白测定法（bicinchoninic acid-total protein assay）测定总蛋白水平。</li>
</ol>
5. 来自淋巴液收集的载脂蛋白B48脂蛋白的定量38
<ol>
	<li>采用改良的免疫印迹法测定载脂蛋白ApoB48的浓度38。使用3-8% Tris-乙酸-十二烷基硫酸钠聚丙烯酰胺凝胶电泳（SDS-PAGE）分离总淋巴液。</li>
