1. 供体手术
<ol>
	<li>在手术前，根据每种药物制剂的特点，在适当的时间点给予镇痛处理。按照批准的动物护理与使用方案，在皮肤切口前1小时，皮下注射0.1 mg/kg体重的丁丙诺啡。</li>
	<li>通过连接异氟烷挥发器的麻醉舱以4%异氟烷对供体动物进行镇静诱导，并通过鼻罩以2%异氟烷维持镇静与麻醉状态。在手术开始前，通过脚趾夹捏回缩反射评估麻醉深度。</li>
	<li>佩戴口罩、一次性隔离衣和手套。</li>
	<li>剃除手术区域毛发，特别是后肢和腹股沟区域，并使用10%聚维酮-碘进行皮肤消毒。</li>
	<li>使用无菌手术巾、经高压灭菌的器械以及高倍显微镜（40倍）。</li>
	<li>使用剪刀从腹股沟区域向大腿中部做皮肤切口，并环绕切口连接，以将后肢与小鼠其余身体部分分离。</li>
	<li>识别并分离股动脉、股静脉和神经。使用镊子和显微剪刀将这三种结构完全分开。</li>
	<li>在完成血管蒂的解剖后，使用显微剪刀在腹股沟韧带水平切断血管。</li>
	<li>接着，继续使用剪刀在大腿中段水平向上切断个体腹侧（缝匠肌和大腿内侧肌群）和背侧肌群20，以将移植物从供体动物上完全分离。</li>
	<li>横断股骨，并使用剪刀在股骨干中点处切断。</li>
	<li>通过过量异氟烷麻醉后行颈椎脱位法处死动物，确认心跳和呼吸停止。</li>
	<li>使用安装在注射器上的33G灌注针（见材料表），以2 ml&#160;含肝素（30 I.E.）的冷（4 °C）生理盐水灌注肢体。</li>

2. 受体操作

	</li>
	<li>移植植入
	<ol>
		<li>通过使用温热生理盐水（37 °C）冲洗手术区域，并在术前和术后各注射0.3 ml温热生理盐水，以尽量减少体液流失。</li>
		<li>将移植物放置于准确反映原生后肢解剖位置的方向，对齐受体与移植物的股骨，并使用20 G脊髓穿刺针作为髓内钉连接两者。</li>
		<li>使用可吸收缝线材料（6-0 Polysorb）吻合腹侧和背侧肌群。</li>
		<li>采用无缝合套环技术连接股血管；具体操作为将受体侧血管拉过先前安装在移植物血管末端的套环上。使用10-0尼龙缝线行环形结扎，将受体血管固定于套环上。</li>
		<li>随后松开血管夹。此时应目视确认套环旋转情况及位置是否理想，以避免血管扭转或成角。</li>
		<li>使用电凝设备仔细止血，特别关注肌肉供体-受体交界面及骨断端区域。</li>
		<li>使用不可吸收尼龙缝线（6-0 Ethilon）缝合皮肤。</li>
		<li>通过将动物置于加热灯下的笼中恢复，建立正常体温环境。在送回饲养设施前，持续常规监测至少4小时。</li>
		<li>术后给予丁丙诺啡镇痛，剂量为0.1 mg/kg，皮下注射，每6–8小时一次&#160;，持续3天。</li>

20世纪90年代末标志着重建性移植的开创性时代，1998年在法国成功完成了首例手部移植手术。自此以后，血管化复合组织移植（VCA）已被成功应用于广泛患者群体中，用于修复严重的组织缺损。截至目前，全球已有76名患者接受了112例上肢移植，以及31例面部移植1-3。此外，还开展了其他多种类型的VCA移植，包括腹壁4、喉5、气管6、血管化关节7，甚至阴茎8移植。最近，子宫移植后成功实现活产婴儿的案例也已有报道9。全球日益积累的临床经验表明，对于那些因严重功能组织缺损而无法通过传统重建和修复手术及治疗手段改善病情的患者而言，重建性移植已成为一种有效的治疗选择。
