1. 维持 IMOP 细胞的自我更新
<ol>
	<li>配制 iMOP 培养基：DMEM/F12、1× B27 添加剂、25 µg/ml 羧苄青霉素和 20 ng/ml bFGF。使用无菌试剂配制 50 ml iMOP 培养基。将 49 ml DMEM/F12 加入 50 ml 锥形管中，并在 37 °C 水浴中预热。

		<li>在 37 °C 水浴中解冻 50× B27 添加剂和经滤膜除菌的 100 mg/ml 羧苄青霉素分装液，持续约 5 分钟。在室温下解冻 100 µg/ml bFGF 分装液。向 DMEM/F12 中加入 1 ml 50× B27、10 µl 100 µg/ml bFGF 和 12.5 µl 100 mg/ml 羧苄青霉素。</li>
	</ol>
	</li>
	<li>在 60 mm 组织培养皿中使用 3 ml 培养基培养 iMOP 细胞。每隔一天通过加倍培养基体积的方式更换新鲜培养基。确保 bFGF 浓度不低于 5 ng/ml。
	<ol>
		<li>将 iMOP 细胞在 37 °C、5% CO2 条件下培养。培养 5–7 天后按以下步骤进行传代。使用 10 ml 移液枪头将培养物转移至 15 ml 锥形管中。</li>
	</ol>
	</li>
	<li>通过将 0.1 ml 0.5 M EDTA（pH 8.0）稀释至 50 ml HBSS 中，配制 1 mM EDTA 的 HBSS 溶液。将配制好的 1 mM EDTA HBSS 溶液在 37 °C 水浴中预热。</li>
	<li>通过重力沉降法或在室温下以 200 × g 离心 5 分钟收集细胞。对于重力沉降法，将含有培养物的 15 ml 锥形管置于 37 °C 培养箱中 5–10 分钟。之后观察耳球体是否已沉降至 15 ml 锥形管底部。 
	注意：过高的离心力可能导致细胞损伤。</li>
	<li>使用 2 ml 吸液枪头小心吸除废弃培养基，避免扰动细胞沉淀。向细胞沉淀中加入 0.5 ml 预热的 1 mM EDTA HBSS 溶液。使用 P1000 移液器轻轻吹打 2–3 次。将锥形管置于 37 °C 条件下孵育 &#60;5 分钟，以促进细胞解离为单细胞悬液。</li>
	<li>轻轻摇动细胞悬液，判断耳球体是否已完全解离。若无耳球体沉降至锥形管底部，则表明细胞已解离。孵育时间可能因情况而异。 
	注意：EDTA 处理时间过长会导致细胞死亡率显著升高。</li>
	<li>将细胞从 37 °C 取出，加入 2 ml 培养基以中和并稀释 EDTA。在室温下以 200 × g 离心 5 分钟收集细胞。使用 2 ml 吸液枪头吸除稀释后的 EDTA 溶液，并加入 5 ml 1× PBS 洗涤细胞。</li>
	<li>在室温下以 200 × g 离心 5 分钟，使用 2 ml 吸液枪头吸除 1× PBS。使用 P1000 移液器将细胞沉淀重悬于 0.5 ml iMOP 培养基中，轻轻吹打 2–3 次。</li>
	<li>使用配备适当微流控计数板的微流控颗粒计数仪进行细胞计数28。一个汇合的 iMOP 细胞培养皿约含有 ~6 × 106 个细胞。将细胞悬液用培养基按 1:100 稀释，取 75 µl 加入计数板中进行计数。将 1 × 106 个细胞接种至 6 cm 培养皿中（iMOP 培养基中约 1:10 稀释）。每 5–7 天传代一次 iMOP 细胞。 
	注意：形成较大耳球体或贴附于组织培养皿底部的细胞会发生分化。若培养物过度汇合，细胞也会发生死亡。</li>
</ol>
2. IMOP 细胞的冻存与复苏
<ol>
	<li>在冻存细胞前，将合成冻存液解冻并平衡至 4 °C，以制备合成冻存液。</li>
	<li>从一个汇合的 6 cm 培养皿中收集 iMOP 细胞。使用 10 ml 移液器将细胞（约 6 - 8 × 106 个细胞）转移至 15 ml 锥形离心管中。</li>
	<li>通过重力沉降或在室温下以 200 × g 离心 5 分钟收集细胞。如果耳球体较小，细胞将无法通过重力沉降沉淀。可选：使用步骤 1.9 计数细胞，以确定总细胞数量。</li>
	<li>使用 2 ml 吸液移液器吸除废弃培养基，同时保留松散的细胞沉淀。</li>
	<li>加入 0.25 ml 合成冻存液，将细胞重悬至密度约为 5 × 105 至 3 × 106 个细胞/ml。使用 P1000 移液器轻轻吹打混匀。使用带 1 ml 滤芯吸头的 P1000 移液器将细胞悬液转移至冻存管中。</li>
	<li>将冻存管放入无酒精的冻存盒中，并将冻存盒置于 -80 °C 环境中，使温度以每分钟 1 °C 的速率下降，直至达到 -80 °C。</li>
	<li>如需长期保存细胞，应将冻存管转移至液氮储存罐的气相中。复苏细胞时，先将含有冻存细胞的冻存管在 -80 °C 下平衡。</li>
	<li>将 iMOP 培养基在 37 °C 水浴中预热。</li>
	<li>将冻存管在 37 °C 水浴中轻轻旋转管底，快速解冻。当最后一块冰晶消失后，立即向解冻的细胞中加入 1 ml 预热的 iMOP 培养基。将细胞转移至 15 ml 锥形离心管中，并额外加入 4 ml iMOP 培养基。</li>
	<li>在 200 × g 条件下离心 15 ml 锥形管 5 分钟，吸除上清培养基。用 2 ml iMOP 培养基重悬细胞，并将细胞接种至 6 cm 培养皿中。将培养物置于 37 °C、5% CO2 条件下孵育以进行扩增。</li>
</ol>
3. 将 IMOP 细胞分化为感觉上皮
<ol>
	<li>制备50 ml iMOP感觉上皮分化培养基：DMEM/F12，1X B27补充剂，25 µg/ml羧苄西林。在50 ml锥形管中将49 ml DMEM/F12置于37 °C水浴中预热。在37 °C水浴中解冻50X B27补充剂和100 mg/ml羧苄西林分装液5分钟。向DMEM/F12中加入1 ml 50X B27和12.5 µl 100 mg/ml羧苄西林，配制成50 ml iMOP感觉上皮分化培养基。</li>
	<li>收获、解离、重悬并计数细胞，请重复步骤1.4–1.9。</li>
	<li>在第-3天，使用iMOP培养基将1 × 106个细胞接种至6 cm培养皿中。在第0天，用10 ml移液器将培养物转移至15 ml锥形管中。按照步骤1.6所述方法，通过重力沉降收集耳球体。使用2 ml吸液移液器吸除废弃培养基，保留耳球体在锥形管底部。</li>
	<li>轻柔加入2 ml感觉上皮分化培养基。使用大孔径10 ml移液器将耳球体转移至6 cm培养皿。 
	注意：剧烈吹打可能因机械剪切作用导致耳球体细胞解离。</li>
	<li>每隔一天向培养物中添加2 ml新鲜的感觉上皮分化培养基。如有需要，可通过重力沉降收集细胞，并更换培养基。</li>
	<li>在第10天，将培养物转移至15 ml锥形管，按照步骤1.6所述让耳球体沉降，收集耳球体。使用2 ml吸液移液器吸除废弃培养基，保持耳球体不受扰动。</li>
	<li>在室温下用含4%多聚甲醛的1X PBS固定耳球体15分钟。移除多聚甲醛溶液，用洗涤缓冲液（含0.1% Triton X-100的1X PBS）洗涤耳球体，随后在封闭缓冲液（含10%正常山羊血清和0.1% Triton X-100的1X PBS）中孵育1小时。
	<ol>
		<li>更换缓冲液，加入稀释的一抗，在4 °C孵育。用含0.1% Triton X-100的1X PBS洗涤样本，对耳球体进行免疫染色27。将耳球体转移至1X PBS中，使用P1000移液器将其置于载玻片的封片剂中，盖上盖玻片，并在4 °C下让封片剂干燥。</li>
	</ol>
	</li>
	<li>使用配备16位CCD相机以及20X 0.75空气物镜或40X 1.3 NA油浸物镜的倒置显微镜系统采集落射荧光图像。使用以下波长（激发和发射）采集不同荧光通道的信号：蓝色（377 ± 25 nm / 447 ± 30 nm），绿色（475 ± 25 nm / 540 ± 25 nm），红色（562 ± 20 nm / 625 ± 20 nm）和红外（628 ± 20 nm / 692 ± 20 nm）。</li>
</ol>
4. EdU 掺入检测
<ol>
	<li>在第 -3 天，将 1 × 106 个 iMOP 细胞接种于一个 6 cm 培养皿中。三天后的第 0 天，收获细胞，消化解离，重悬并计数，重复步骤 1.4–1.9。</li>
	<li>将 2.5 × 105 个细胞接种于 iMOP 培养基中，另将 5 × 105 个细胞接种于感觉上皮分化培养基中，分别置于 6 孔板的不同孔中。</li>
	<li>在第 3 天通过 EdU 掺入实验测定处于 S 期的细胞百分比
	<ol>
		<li>将 EdU 储存液直接加入 iMOP 培养物中，使培养基中 EdU 的终浓度为 1 µM。</li>
		<li>在 37 °C、5% CO2 条件下，将 EdU 与 iMOP 培养物共同孵育 2 小时&#160;。收获细胞，消化解离并收集细胞，重复步骤 1.4–1.9。加入含 4% 多聚甲醛的 1× PBS，室温固定 15 分钟。在室温下以 200 × g 离心 5 分钟收集细胞。移除多聚甲醛溶液并妥善处理。</li>
		<li>根据制造商说明书，使用 Hoechst 对细胞核进行染色，并使用绿色荧光染料-叠氮化物标记掺入的 EdU。 
		注意：所有洗涤步骤均使用含 1× PBS、3% BSA 和 0.1% Tween 20 的溶液进行，用 1× PBS 洗涤细胞两次。</li>
		<li>使用抗荧光淬灭剂将细胞封片，盖上 1.5 盖玻片。采用落射荧光显微镜获取标记细胞的荧光图像。</li>
	</ol>

</ol>
5. 分化IMOP来源的神经元
<ol>
	<li>配制50 ml神经元分化培养基：Neurobasal培养基、1X B27添加剂、2 mM L-谷氨酰胺。将50X B27添加剂和200 mM L-谷氨酰胺溶液在37 °C水浴中解冻5分钟。向48.5 ml Neurobasal培养基中加入1 ml 50X B27添加剂和0.5 ml 200 mM L-谷氨酰胺。</li>
	<li>将12 mm圆形1.5级玻璃盖玻片放入无菌10 cm培养皿中，并向皿内加入70%乙醇，以对盖玻片进行灭菌和清洁。</li>
	<li>轻轻振荡盖玻片，确保其完全被乙醇覆盖，在室温下静置10分钟。用无菌1X PBS清洗盖玻片3次，以去除残留乙醇。再用无菌H2O清洗盖玻片1次，以去除残留的1X PBS。</li>
	<li>使用2 ml移液器吸除H2O，让盖玻片自然干燥。将盖玻片置于生物安全柜中用紫外光照射15分钟。若不立即使用，应将盖玻片保存在无菌环境中。</li>
	<li>将一片12 mm圆形盖玻片放入24孔板的每一孔中。轻轻摇动培养板，确保盖玻片平整地贴附于孔底。</li>
	<li>在室温下解冻2 mg/ml聚-D-赖氨酸储备液，并用1X PBS稀释至10 µg/ml的工作浓度。向每孔中加入0.25 ml 10 µg/ml聚-D-赖氨酸溶液。将培养板置于37 °C培养箱中孵育1小时。</li>
	<li>用无菌1X PBS清洗各孔3次。在室温下解冻10 mg/ml层粘连蛋白储备液，并用1X PBS稀释至10 µg/ml。向24孔板的单个孔中加入0.25 ml 10 µg/ml层粘连蛋白工作液。将培养板置于37 °C培养箱中孵育。使用2 ml移液器吸除层粘连蛋白溶液。</li>
	<li>使用P1000移液器向每孔加入1 ml 1X PBS，再用2 ml移液器吸除，重复此洗涤步骤共3次。最后一次洗涤后保留1X PBS于孔中，直至细胞准备接种。通过执行步骤1.4–1.9进行细胞收获、解离和计数，启动神经元分化。在第-3天，将1 × 106个细胞接种至6 cm培养皿中。</li>
	<li>在第0天，重复步骤1.4–1.9进行细胞收获、解离和计数。将1 × 105 至 1.5 × 105个iMOP细胞接种至24孔板的每孔中，每孔加入0.5 ml预热的神经元分化培养基。每隔一天吸除旧培养基并更换为预热的神经元分化培养基。</li>
	<li>在第7天，用4%多聚甲醛在室温下固定iMOP来源的神经元15分钟。吸除多聚甲醛溶液，用洗涤缓冲液（含0.1% Triton X-100的1X PBS）清洗iMOP来源的神经元，然后在封闭缓冲液（含10%正常山羊血清和0.1% Triton X-100的1X PBS）中于室温孵育1小时。

		<li>更换缓冲液，将样品在含稀释抗体的封闭缓冲液中孵育，4 °C孵育过夜。使用含0.1% Triton X-100的1X PBS清洗样品，进行免疫染色27。在将盖玻片置于封片剂上之前，先用1X PBS清洗。将封片后的样品在4 °C下干燥，然后按照步骤3.8所述条件采集落射荧光图像。</li>

bFGF 撤除降低 IMOP 细胞的增殖

本实验的总体目标是将永生化的多能耳源性祖细胞分化为内耳细胞类型。因此，该方法可帮助我们解决听觉再生领域的一些关键问题，例如耳源性祖细胞如何转变为耳蜗毛细胞或听觉神经元。重要的是，该技术提供了一种体外细胞系统，可与体内哺乳动物动物模型系统平行使用。今天由我实验室的博士后研究员 Azadeh Jadali，以及实验室的两名研究生 Zhichao Song 和 Alejandra Laureano 演示该实验步骤。按照文本方案中所述配制培养基后，取三毫升该液体，将一乘以十的六次方个永生化多能祖细胞（IMOP 细胞）接种至一个 60 毫米的组织培养皿中。将细胞在 37 摄氏度、5% 二氧化碳条件下孵育 5 至 7 天。每隔一天，通过添加新鲜培养基使培养液的体积增加一倍。传代细胞时，首先将细胞从培养皿转移至15毫升锥形管中，然后将锥形管置于37摄氏度环境中5到10分钟。在此期间，预计耳球样克隆将通过重力沉降聚集在锥形管底部。接下来，吸除上清液，注意不要扰动沉淀。通过加入0.5毫升预先配制并预热的、含1毫摩尔EDTA的平衡盐溶液（HBSS），轻轻吹打2至3次，重新悬浮耳球样克隆。为了促进耳球体解离为单个细胞，将锥形管置于37摄氏度的培养箱中。定期轻轻摇动溶液，当观察到管底无沉淀的耳球体时，将其转移至室温。随后向细胞中加入2毫升培养基以中和EDTA。在室温下以200 ×g离心5分钟。然后吸除稀释的EDTA溶液，并用5 mL的1×PBS洗涤细胞。使用相同条件离心细胞后，吸除1×PBS。接下来，向锥形管中加入 0.5 毫升培养基，并重悬细胞沉淀。进行细胞计数时，将 IMOP 细胞用培养基按 1:100 比例稀释。然后取75 µL该溶液加入微流控颗粒计数器中的小盒内，测定细胞数量。随后，按先前所述方法，将1×10⁶个IMOP细胞接种于60 mm组织培养皿中，并每5至7天重复一次传代操作。接种后3天，利用重力沉降法收集耳球体，操作方法同前。继续吸除上清液，然后在2 mL感觉上皮分化培养基中轻轻重悬细胞。接着使用10 mL大孔移液管将耳球转移至新的60 mm培养皿中。将耳球培养物分化十天，每隔一天向培养物中添加两毫升新鲜分化培养基。随后，再次通过重力沉降法收集耳球培养物。吸除用过的培养基。接着，在室温下用含4%多聚甲醛的1×PBS固定耳球15分钟。然后去除甲醛溶液，并用含0.1% Triton X-100的1×PBS洗涤细胞。用含10%山羊血清和0.1% Triton X-100的1×PBS对耳球进行封闭。封闭1小时后，弃去封闭液，加入含有适当稀释比例的Cdkn1b或CDH1抗体的封闭液。随后将细胞在4°C条件下与一抗孵育过夜。第二天，按照其他部分所述方法，使用荧光标记的二抗、鬼笔环肽（Phalloidin）和 Hoechst 继续进行免疫染色。为成像做准备时，先在玻璃载玻片上滴加封片剂，然后将染色后的耳球体放入封片介质中。接下来，将盖玻片覆盖在样品上。在4°C条件下让封片剂过夜干燥后，将玻片置于配备16位CCD相机的倒置显微镜载物台上。使用20倍0.75数值孔径的平场物镜，通过采集蓝色、绿色、红色和红外通道的信号，获取落射荧光图像。首先，将圆形玻璃盖玻片放入无菌的100毫米培养皿中，并用70%乙醇浸没，以进行清洁。轻轻振荡盖玻片，确保其充分接触乙醇，并在室温下放置10分钟。随后，用无菌1×PBS冲洗盖玻片3次，再用无菌水冲洗1次。然后将盖玻片置于生物安全柜中，暴露于紫外光下15分钟使其干燥。如需立即使用，将一片无菌盖玻片放入24孔板的每个孔中，并轻轻振荡培养板，确保所有盖玻片平铺于孔底。然后通过向每个孔中加入含有每毫升10微克聚D-赖氨酸的0.25毫升1×PBS溶液来包被盖玻片。将培养板在37摄氏度孵育1小时后，移除聚D-赖氨酸溶液，并用无菌1×PBS洗涤各孔3次。最后一次洗涤液吸除后，向24个孔中的每个孔加入含有每毫升10微克层粘连蛋白的0.25毫升1×PBS溶液。在37摄氏度下孵育平板过夜后，吸除Laminin溶液，然后用1毫升1×PBS洗涤每个孔三次，但最后一次洗涤后将PBS保留在孔中，直至细胞准备接种。开始神经元分化时，按照先前所述方法计数解离的耳球细胞，并将其重悬于预热的神经元分化培养基中。将每孔含有包被盖玻片的培养板接种一至一点五乘以十的五次方个IMOP细胞。在37摄氏度、5%二氧化碳条件下孵育培养板，每隔一天更换一次神经元分化培养基。七天后，吸去培养基，向每孔中加入4%多聚甲醛，固定已贴附在盖玻片上的细胞，室温孵育15分钟。然后去除多聚甲醛，用含0.1% Triton X-100的1×PBS洗涤细胞一次。接下来，移除洗涤缓冲液，并向每孔中加入封闭缓冲液，封闭缓冲液由含10%正常山羊血清和0.1% Triton X-100的1×PBS组成。将细胞在室温下孵育1小时。然后吸去液体，并加入含有适当稀释比例的 Cdkn1b 或 Tubb3 抗体的封闭缓冲液。在 4°C 下孵育过夜后，用含 1% Triton X-100 的 1× PBS 洗涤细胞一次。随后按照先前所述方法进行免疫染色。染色完成后，通过向孔中加入1× PBS进行最后一次洗涤。为准备分析，将附着有染色细胞的盖玻片从24孔板中取出，并将其放置在载玻片的封片剂上。在4°C条件下让封片剂过夜干燥后，按照先前所述方法采集落射荧光图像。在耳上皮分化培养基中培养10天的耳球体相较于未分化细胞，显示出Cdkn1b表达增加，Cdkn1b是一种细胞周期依赖性激酶抑制剂，也是内耳分化的标志物。图中Hoechst染色标记了细胞核。这些细胞还表现出肌动蛋白丝的形态学变化以及细胞间黏附区域的变化，这些特征与上皮细胞相似。这一现象可通过Phalloidin染色以及Cdh1在细胞周边的标记显现。耳球还可被诱导分化为神经元。这幅明场图像显示，在神经元分化培养基中培养七天后，IMOP 细胞出现神经突。使用神经元标记物 Tubb3 对这些细胞进行免疫染色，证实了其分化特征。此外，这些细胞还强烈表达 Cdkn1b。这表明 Tubb3 和 Cdkn1b 的双重信号可作为 IMOP 细胞神经元分化的指标。通过计算 Cdkn1b 荧光信号与 Hoechst 信号的比值，对已分化和未分化细胞的 Cdkn1b 荧光进行归一化处理，从而获得定量结果。此处展示了该数据的直方图。这些图表展示了归一化 Cdkn1b 水平超过预设阈值的未分化和已分化 IMOP 细胞所占的百分比。两种已分化的细胞群体中表达 Cdkn1b 的细胞百分比均较未分化的对照组显著增加，证实了免疫荧光所提示的分化结果。一旦掌握操作要点，若操作得当，IMOP 细胞的常规培养与分化可在 30 分钟内完成。观看本视频后，您应该能够充分了解如何启动 IMOP 细胞的分化。感谢观看本视频，祝您的实验顺利成功。
