干扰素-γ（IFN-γ）是一种细胞因子，在介导针对胞内病原体的免疫反应以及控制肿瘤生长中起关键作用5。该细胞因子在抗细菌免疫中的重要性体现在：携带IFN-γ信号通路突变的人类个体对分枝杆菌和沙门氏菌感染高度易感6。类似地，缺乏IFN-γ或IFN-γ受体的小鼠在抵抗分枝杆菌7-9及其他胞内病原体（包括单核细胞增生性李斯特菌（L. monocytogenes）10,11、主要利什曼原虫（Leishmania major）12、鼠伤寒沙门氏菌（Salmonella typhimurium）13以及某些病毒11）方面均表现出防御缺陷。除了抗病原体作用外，IFN-γ在宿主抗肿瘤防御中也发挥着关键作用14。尽管在感染或癌症背景下高水平的IFN-γ产生是有益的，但该细胞因子的持续产生已被证实与系统性自身免疫性疾病的发生15-17以及非肥胖型糖尿病小鼠模型中1型糖尿病的加速进展相关18。

IFN-γ 通过以下方式增强固有免疫和适应性免疫：（1）上调主要组织相容性复合体（MHC）I 类和 II 类分子的表达，（2）增加抗原呈递细胞上共刺激分子的表达，（3）增强巨噬细胞的吞噬作用以及促炎性细胞因子和杀菌因子的产生例如，一氧化氮和活性氧），（4）促进初始CD4+ 将T细胞分化为Th1效应细胞，（5）促进抗体类别转换为免疫球蛋白（Ig）2a和IgG3（小鼠），（6）诱导趋化因子产生以募集免疫细胞至感染部位，以及（7）增强自然杀伤（NK）细胞和细胞毒性T淋巴细胞（CTL）应答5,19鉴于IFN-γ在宿主对抗病原体和肿瘤反应中的关键作用，重组IFN-γ已被用于治疗多种感染和恶性肿瘤（详见综述于19然而，由于全身性给予干扰素-γ（IFN-γ）或促进Th1型免疫反应的细胞因子白细胞介素-12（IL-12）会伴随副作用及剂量相关的毒性19,21，人们正致力于开发替代策略以增强免疫细胞产生干扰素-γ。新型生物制剂和小分子的开发需要 体内 用于检测这些制剂是否在免疫反应过程中增加干扰素-γ（IFN-γ）的产生，以及这种增加是否转化为有意义的生物学效应，例如提高动物存活率的筛选工具。
利用革兰氏阳性菌L. monocytogenes对小鼠进行实验性感染，已成为解析IFN-γ在宿主免疫应答中对抗胞内病原体作用的关键模型1,22。通过静脉或腹腔（i.p.）途径感染小鼠后，病原菌迅速扩散至脾脏和肝脏，并被组织驻留的巨噬细胞和肝细胞吞噬，脾脏中的细菌载量通常在感染后3至4天达到峰值1,3,22。自然杀伤细胞（NK细胞）产生的IFN-γ对于激活巨噬细胞以及早期抵抗病原体具有重要作用3；然而，在高感染剂量下，IFN-γ的产生也可能不利于病原体的清除23。在病原体早期控制阶段，NKT细胞也是脾脏和肝脏中IFN-γ的重要来源2,24，并且已有研究表明，其IFN-γ的产生可增强其他细胞类型（包括NK细胞）的IFN-γ分泌2。另一方面，后期发挥作用的适应性T淋巴细胞，尤其是CD8+ T细胞，在介导病原体清除以及提供再感染保护方面具有关键作用1,4,22。
该感染模型因其多种优势而受到研究人员的广泛关注（详见文献1综述）。首先，病原体感染具有高度可重复性，并能诱导强烈的Th1型和细胞免疫反应。其次，在亚致死性感染过程中，细菌载量主要集中于肝脏和脾脏，便于检测。第三，该病原体可在生物安全二级（BSL2）条件下安全操作。第四，该生物体及其引发的免疫反应已被广泛表征。最后，目前已开发出多种突变株和基因修饰菌株，可供研究使用。

本实验方案的总体目标是培养C57 blacksix小鼠并用单核细胞增生性李斯特菌（listeria monocytogenes）EGG菌株进行感染，检测细菌载量、干扰素γ反应以及动物对该病原体感染的存活情况和应答反应。该方案可帮助研究人员筛选影响干扰素γ反应及细菌感染后动物存活的药物或基因。该技术的主要优势在于其在实验室中建立相对简便。即使对于细菌培养经验较少的人而言也是如此。本实验操作将由我实验室的研究生 Jennifer Ahn 演示。首先，将一个无菌的移液器吸头浸入室温融化的单核细胞增生李斯特菌 EGG 菌株的甘油菌种中。立即用移液器吸头在脑心浸液（BHI）琼脂平板的一个区域上来回划线，此为初次划线。将平板旋转90度，用新的移液器吸头穿过第一次划线区域，将菌液涂布至平板的下一个四分之一区域。这是第二次划线。然后，转动平板并将第二次划线区域延伸至平板的第三个象限，进行第三次划线。现在，将平板倒置于37°C条件下，过夜培养。在16至24小时之间，最后一组划线区域应可见单个均匀的菌落。当菌落出现后，将10毫升无菌BHI肉汤加入无菌通气50毫升试管中，并使用无菌移液器吸头从平板上转移一个单核细胞增生李斯特菌（L monocytogenes）菌落至肉汤中。将接种后的肉汤置于37摄氏度、225转/分钟的振荡培养箱中过夜培养，直至OD600达到1。在生物安全柜中，取100微升过夜培养物接种于一管BHI培养基中，以制备实验用单核细胞增生李斯特菌感染接种物。在37摄氏度振荡培养至达到所需OD600值后，将培养物转移至无菌离心管中进行离心。使用连接含漂白剂的捕集瓶的真空装置吸取上清液。在相同的离心条件下，用 PBS 清洗沉淀物两次。第二次清洗后，将细菌重悬于 PBS 中，并稀释至适当浓度，以在 200 微升体积中向每只小鼠递送目标 CFU。接下来，使用无菌移液器吸头充分混匀悬浮液，以确保细菌均匀分布。然后，用带有25号针头的1毫升安全型注射器吸取200微升接种物。现在，用非优势手抓住小鼠肩部周围的松弛皮肤使其背部皮肤绷紧。使用新的针头和注射器，将全部200微升接种物注射到动物腹部下象限、腹中线外侧区域，以避开膀胱。为测定感染高峰期的细菌载量，在感染后第3天，将实验动物仰卧固定于解剖板上，使其四肢固定。随后用70%乙醇对皮肤进行消毒。使用无菌的硬质剪刀，从胸部向腹股沟做一切口，然后从腹股沟中点分别向两侧膝盖、从胸部中点分别向两侧肘部做切口。钝性分离并翻起皮肤，用25号针头将其固定张开。用70%乙醇再次消毒肌层。然后使用无菌精细剪刀在腹膜壁上做正中切口。用镊子夹住剑突。从剑突开始，在腹膜壁两侧分别做横向切口，沿肋骨延伸至膈肌下方。这将暴露出肝脏。接下来，使用无菌剪刀从一个叶中切下约100毫克的组织块。将组织放入已预称重的含有0.2至0.3克经酸洗的无菌1.5至2毫米玻璃珠的1.5毫升微量离心管中。然后使用镊子轻轻拨开腹腔左侧的器官，以暴露脾脏。用镊子轻轻夹住脾脏，剪除周围结缔组织，将器官从腹腔中分离。将脾脏置于与肝脏不同的试管中，并将组织样本放入含有冰块的防漏容器内送至实验室。使用珠磨匀浆器以30赫兹频率振荡试管3分钟。然后，将匀浆液在含0.1% Triton X-100的PBS中进行十倍系列稀释。接着，用无菌涂布棒将每种稀释后的匀浆液各100 µL涂布于BHI琼脂平板上。将平板置于37°C培养箱中，过夜培养。保留每个平板上菌落数不超过300个的平板。计数每个平板上的菌落。为检测CT8 T细胞干扰素γ反应中的CD4，按照上述方法，从感染了病原体2×10⁻⁴ CFU的小鼠中分离脾脏。将脾脏处理成单个红细胞悬液。用含胎牛血清的完全RPMI培养基重悬细胞沉淀，调整细胞浓度至每毫升4×10⁶个细胞。随后，将1毫升细胞悬液接种至24孔板的每个孔中，并加入等体积已解冻且经热灭活的单核细胞增生性李斯特菌（L. monocytogenes），然后将培养物置于37°C培养箱中培养。20 小时后，向各孔中加入每毫升 0.66 微升的蛋白质转运抑制剂，并将培养板放回培养箱。4 小时后，在生物安全柜中收集 500 微升培养上清液，并将样品于 -80 摄氏度冷冻，用于后续通过 ELISA 检测干扰素 γ。然后将细胞转移至一个无菌的15毫升离心管中，用PBS洗涤孔板，并将洗涤液与收集的细胞合并，用于流式细胞术染色和分析。脾脏中的细菌负荷峰值出现在感染后2至3天。在感染前添加PPAR Alpha拮抗剂可显著降低该峰值。感染后第7天，使用热灭活病原体对脾脏CD4阳性及CD8阳性T细胞进行体外再刺激，可通过流式细胞术检测到CD4阳性与CD8阳性细胞产生干扰素γ的阳性反应。在感染前使用PPARα拮抗剂处理，可增强CD4和CD8细胞的干扰素γ反应。此外，PPARα拮抗剂处理可显著提高动物在感染LD50剂量病原体后的存活率，表明该模型适用于研究调控干扰素γ反应的新药对感染后动物存活的影响。一旦掌握，本方案中的所有步骤若正确操作，可在数周内获得所需数据。该技术还可进一步改进，例如用表达模型抗原的单核细胞增生性李斯特菌（*Listeria monocytogenes*）感染小鼠，并使用针对这些抗原的MAC I类和II类四聚体试剂，从而可在感染后检测T细胞的扩增情况。该技术建立后，成为解析干扰素γ在宿主对胞内病原体免疫反应中作用的关键模型。观看本视频后，您应能够充分掌握如何培养C57BL/6小鼠并用单核细胞增生性李斯特菌进行感染，以及如何测定感染后的细菌载量和干扰素γ反应。请注意，单核细胞增生李斯特菌对免疫功能低下者（如孕妇、婴幼儿或老年人）可能具有危险性。因此，该病原体只能由免疫功能健全的人员在生物安全二级条件下操作。
