将样品盘和基底熔化到样品台上，并向样品中加入一滴成像液。然后将悬臂支架连接到 AFM。将悬臂和样品靠近，以便在悬臂尖端上的液体和样品上的液体之间形成毛细管桥。
使用 AFM 软件将测量激光对准悬臂尖端。接下来，从悬臂的热谱中的主峰找到悬臂的驻留频率。如果校准了悬臂的挠度，则使用简单的谐振子模型拟合驻留峰值，可得到悬臂的弹簧常数。
然后，通过找到悬臂在接近热谱中确定的谐振频率的频率范围内进行外部驱动时的振幅响应来调整悬臂。调整驱动振幅，使自由振荡振幅约为 5 纳米。这通常对应于大多数 AFM 上的 0.2-0.8 伏特。
然后，将振幅设定点调整到自由振幅的 80% 左右。接下来，将 feedback gains 设置得相对较高。确保没有出现不稳定或振铃后，将初始扫描速率设置为大约 1 赫兹，并将扫描大小设置为 10 纳米。
使用 AFM 控制软件启动针尖接近表面。通过稍微改变设定点值来评估尖端是否已到达表面而不开始成像。如果尖端位于表面，则对 ZPA 区扩展的影响应该可以忽略
不计。尖端到达表面后，缩回 ZPA 区域并重新调整悬臂。由于针尖采样的相互作用，谐振频率可能已经转移到较低的值。现在，将设定点更改为新调谐自由振幅的 80% 左右，并接合悬臂，以调幅模式对表面进行 10 x 10 纳米平方的扫描，以验证成像参数是否合适。
检查 trace 和 retrace 配置文件是否叠加。如果没有，请进一步降低设定点，并尝试增加增益。如果图像变得杂色，请降低增益。
如果可能的话，对样品的 1 到 5 平方微米的大区域重复该作。对于软样品或生物样品，这可能会导致吸头污染。将扫描大小减小到适合可视化感兴趣特征的值。
这可以低至 20 x 20 纳米。接下来，减小悬臂的驱动振幅，使反馈回路自动缩回 ZPA 区域，从表面增强尖端。当悬臂远离表面时，调整驱动振幅，使悬臂振幅为 1 到 2 纳米峰到峰。
以小步骤逐渐降低设定点，直到 ZPA 区域再次向表面延伸，并恢复原始图像。将设定点振幅保持在新自由振幅的 75% 和 95% 之间。然后，重新调整增益，因为可以在较低的幅度下使用较高的增益，而不会引入明显的噪声。
优化系统以找到自由幅度、设定点和增益的最佳组合，以实现高分辨率。最佳系统条件取决于样品、液体的润湿特性以及所使用的悬臂。对于亲溶剂界面，使用弹簧常数为每米 0.5 到 2 牛顿的悬臂。
