目前，hiPSC疾病建模领域的主流方法是将细胞在单层培养中分化为特定的神经元亚型，或使用一种称为类器官的三维培养系统来模拟大脑发育的某些方面1,2,3。这些系统在研究和揭示人类发育及疾病独特特征方面已显示出极高的价值4,5,6,7。然而，无论是神经元培养还是类器官，通常都需要数周至数月的培养时间才能进入可研究状态。这些实验方案耗时较长，且维持培养体系所需的资源量较大，常常限制了可开展的实验数量以及可测试的变量（如生长因子或药物）数量。此外，许多利用终末分化神经元和类器官开展的研究主要聚焦于树突发育或突触形成等发生在发育后期的过程。尽管这些过程已被证实与自闭症和精神分裂症等发育性疾病的病理机制相关，但在明确神经元分化之前发生的早期发育事件同样对疾病发生至关重要8,9,10,11,12,13。事实上，近期的基因组学研究显示，包含细胞增殖、突起生长和细胞迁移的胎儿中期在自闭症的致病过程中尤为关键11,14。因此，研究神经干细胞和前体细胞群体对于深入理解这些早期发育过程具有重要意义。类器官系统因其三维结构和组织化的特征被认为更能真实模拟人类大脑发育，其中确实包含可用于研究部分早期事件的前体细胞池。然而，类器官中的前体细胞群体通常较为稀疏，且更接近放射状胶质细胞，而非典型的神经干细胞或前体细胞5,15。因此，建立一种高通量的方法，在具有活跃增殖能力的细胞群体中研究神经发育的早期阶段将具有重要意义。
在实验室中，我们建立了一种利用人诱导多能干细胞（hiPSC）来源的神经前体细胞（NPCs）的研究方案。NPCs 是一类具有高度增殖能力的神经干细胞与祖细胞混合群体，可用于研究神经发育过程，如细胞增殖、细胞迁移以及初始突起（神经突）的延伸。这些检测方法源于我们实验室数十年来成功用于大鼠和小鼠皮层培养物中神经发育研究的技术16,17,18,19,20,21,22,23。重要的是，已有研究表明，在大鼠和小鼠培养系统中定义的表型及调控信号能够高度预测体内（in vivo）活跃的分子机制，这体现了这些技术的重要价值16,17,18,19,24。在 hiPSCs 初始分化为 NPCs 后，这些方法使我们能够在数天内研究关键的发育过程。该方法具有多项优势：（1）所需仪器设备简单，易于实施；（2）可在短时间内开展大量实验重复，从而快速验证结果的可重复性；（3）可快速且经济高效地测试诸如包被基质、生长因子作用以及药物活性等培养变量。此外，我们充分利用了细胞外生长因子作为多种发育过程关键调控因子的明确作用。通过将 NPCs 暴露于特定的发育信号分子，直接刺激细胞增殖、神经突生长和细胞迁移等事件，我们发现这些处理可增强对在对照条件下不明显的缺陷的识别能力19,25,26,27,28。同样，药物效应的易检测性为采用精准医学策略测试不同治疗干预措施的有效性提供了有力途径。因此，本方案提供了一种高通量、可重复且简便的方法，用于研究早期脑发育、疾病发病机制，以及生长因子和药物对神经发育表型的潜在有益影响。

Rapid Detection of Neurodevelopmental Phenotypes in Human Neural Precursor Cells (NPCs)

这些研究的一个目标是确定神经前体细胞（NPCs）的增殖活性，即细胞数量的增加。这可通过评估整个细胞群体的DNA合成来实现，该方法是一种高通量技术，通过检测放射性示踪剂氚标记的胸苷掺入细胞提取物的情况，反映所有处于S期的细胞，无论其DNA合成持续5分钟或长达两小时。此外，这些检测还可用于确定进入S期的细胞比例及总细胞数量，这是一种更为耗时的单细胞分析方法。细胞在S期合成DNA，此阶段发生在有丝分裂和细胞分裂之前，而细胞分裂必须发生才能实现细胞数量的增加。由于这些过程需要一定时间，因此在48小时测得的DNA合成变化可能与此时间点的细胞数量变化无直接关联。尽管如此，研究发现，48小时时DNA合成的变化能够可靠地预测第4天和第6天细胞数量的增加。
在评估DNA合成时，将细胞以每孔100,000个细胞（约50%汇合度）的密度接种于24孔板中，并培养48小时后进行检测。采用该细胞密度可确保细胞保持其单层生长环境的特性，同时避免在48小时内因过度增殖导致培养基过酸。...

增殖、迁移和神经突生长等神经发育过程在神经精神疾病中常受到干扰。因此，我们提供了快速且可重复地评估人诱导多能干细胞来源的神经前体细胞（NPCs）中这些神经发育过程的实验方案。这些方案还可用于评估相关生长因子和治疗药物对NPC发育的影响。

本方案的总体目标是以快速且可重复的方式表征人神经前体细胞的早期脑发育过程，并明确重要细胞外因子在调控这些过程中的作用。该方法有助于回答神经精神疾病和神经发育障碍领域的重要问题，例如：所有受影响个体是否具有相同的细胞异常，或者相反，是否每个人表现出特定于个体的功能障碍？本方案的主要优势在于其快速、高度可重复且易于实施。尽管该方法可用于研究神经发育障碍，也可用于研究神经退行性疾病和神经精神疾病，如阿尔茨海默病和抑郁症。由于某些技术涉及将细胞从一个培养皿转移至另一个培养皿，或需要进行形态学分析，因此有必要通过视觉演示来展示其中部分方法，以达到最佳传达效果。要分离人神经前体细胞，需吸除培养基，并用1×磷酸盐缓冲液洗涤细胞一次。吸去磷酸盐缓冲液，向含有神经前体细胞的孔中加入500微升1倍细胞解离溶液。然后在37摄氏度下孵育培养板10分钟。接着使用P1000移液器加入500微升室温保存的磷酸盐缓冲液，以洗涤并去除细胞。然后，将磷酸盐缓冲液中的细胞转移至一个15毫升的锥形管中。再用1毫升磷酸盐缓冲液洗涤一次培养孔，并将洗涤液转移至该管中。接着，以300 ×g离心5分钟，使细胞形成沉淀。然后，弃去上清液，将沉淀物重悬于1至5毫升预热的DMEM/F12培养基中。每孔接种100,000个细胞，每个样本设三个复孔，使用24孔板进行培养。46小时后，向培养基中加入氚标记的胸腺嘧啶核苷，并在37°C下孵育2小时。然后，吸除培养基后，加入300微升预热的0.25%胰蛋白酶EDTA，于37摄氏度孵育20分钟，以解离细胞并释放DNA。接着，开启细胞收集器和泵，将滤纸放入细胞收集器中。将细胞收集器的收集管持续放入一个空的空白托盘中。然后，按下预洗键以润湿滤纸。将收集管放入样品孔中，然后开始运行。使用热光源烘干滤纸，并将相应的样品瓶排列在托盘上。将滤纸片打孔至样品瓶中，然后向每个样品瓶中加入 2 毫升液体闪烁液。在使用闪烁计数仪前，将样品瓶在仪器中孵育 1 小时，以获取每分钟计数（CPM），该数值反映 DNA 的活跃合成水平。在35毫米培养皿中接种500,000个细胞。然后，将培养皿向各个方向轻轻晃动，使细胞均匀分布。接着，在37°C条件下孵育细胞46小时。然后，向培养物中加入终浓度为5 mM的EdU，每毫升培养物加2 µL，孵育2小时。接着，将细胞解离并离心成团。向细胞沉淀中加入3 mL含或不含生长因子或药物的30%扩增培养基。将1毫升细胞悬液接种到预先包被的培养皿中，在37摄氏度下孵育2小时。为研究培养中的神经前体细胞数量，在培养两天后，标记并准备好微量离心管。然后吸出孔中的培养基，加入200微升1X细胞解离溶液，在37摄氏度下孵育15分钟。细胞脱离后，向每个孔中加入300微升1×磷酸盐缓冲液。使用P1000移液器反复吹打细胞溶液数次，使细胞从培养板上完全脱离。然后将细胞悬液转移至预先准备好的1.5毫升离心管中，并将离心管上下颠倒2至3次。用移液器从管中点位置吸取50微升细胞，加入至0.5毫升离心管中的台盼蓝溶液中。然后使用血细胞计数板进行细胞计数。为形成神经球，将1毫升100%扩增培养基加入无任何包被基质的35毫米培养皿中。然后，向每个培养板中加入一百万个神经前体细胞，并在37摄氏度下孵育48至96小时。为准备用于神经球迁移的培养板，将ECM模拟凝胶等分试样溶解于6毫升30%扩增培养基中。在六孔板的单个孔中加入1毫升含30%扩增培养基的ECM模拟凝胶。然后，将培养板在37摄氏度下孵育30分钟。接种神经球时，使用移液器从35毫米培养皿中收集含有细胞的培养基，并转移至一个锥形管中。随后，在100 ×g 条件下离心锥形管，使神经球沉淀。然后，将沉淀重悬于1至3毫升预热的30%扩增培养基中。接着，用移液器将200微升重悬的神经球加入六孔板中，六孔板内含有ECM模拟凝胶和30%扩增培养基，轻轻摇动培养板。随后，将培养板置于37摄氏度孵育48小时。用含30%扩张培养基的ECM模拟凝胶重悬细胞，然后使用4%多聚甲醛固定细胞30分钟。为分析神经球，采用10倍放大倍数的相差显微镜设置采集图像。使用NIH ImageJ软件测量神经球的平均迁移距离。在 ImageJ 中，使用手绘线条工具勾勒神经球的外轮廓。然后，使用测量功能计算所勾勒区域的面积，并勾勒球体内内部细胞团的区域。用神经球总面积减去内部细胞团面积，以量化平均迁移距离。通过免疫细胞化学染色研究第3代神经前体细胞中细胞骨架蛋白以及SOX2、PAX6和巢蛋白（nestin）等转录因子的表达情况。随后，采集相差显微图像以识别具有神经突的细胞。根据图像，将突起长度等于或长于两个细胞体的细胞视为具有神经突的细胞，而突起长度短于两个细胞体的细胞则不视为具有神经突的细胞。为确认神经球的形成，采集了相差显微图像，结果显示在72小时时间点收集的神经球大小介于80微米至120微米之间，因此可用于迁移实验。为明确增殖能力，将神经前体细胞在对照或成纤维细胞生长因子条件下培养，并对EdU的存在进行染色。相差荧光图像显示，经成纤维细胞生长因子处理的细胞中处于S期的细胞比例更高，这通过EdU染色得以证实。接下来，为了检测神经突的生长情况，将细胞在对照条件下进行检测，并与PACAP处理组进行比较。采集了相差图像，显示在PACAP处理下具有神经突的细胞数量增加。随后，为了研究神经球的迁移，用PACAP神经肽处理细胞。获得的相差图像显示，与对照组相比，神经球的迁移增加。本方案包含多种技术和检测方法，每种方法所需时间各不相同。掌握之后，平均每项检测的建立需要两到三小时，分析则需要30分钟至三小时。在进行这些操作时，至关重要的是确保使用高质量的神经前体细胞，并且这些细胞在除神经球形成实验以外的所有条件下均被完全解离并均匀分布。完成这些操作后，可进一步开展其他分析，包括蛋白质印迹（Western blotting）、定量逆转录 PCR（QRT-PCR）、转录组学、蛋白质组学和代谢组学，以阐明其潜在的作用机制。感谢观看，祝您的实验顺利成功。
