An Optimized Rhizobox Protocol to Visualize Root Growth and Responsiveness to Localized Nutrients

该方法有助于解答根系与根际科学领域中的关键问题，例如根系如何响应局部养分斑块。该技术的主要优势在于，它允许对土壤中的单个根系进行重复、非侵入式的测量，且无需使用专业设备，成本相对较低。收集根箱（Rhizobox）的组装部件。这些部件包括由透明亚克力制成的前后面板，尺寸约为 40 厘米 × 61 厘米。此时，在后面板上方已放置侧边和底部的间隔片。间隔片与面板上先前钻好的孔对齐。裁剪一段聚酯纤维垫料，使其沿根盒底部放置。将垫料铺在背板上，紧贴底部隔条上方，以帮助防止泄漏。将顶板放置在隔条上，以固定垫料位置。现在准备组装根盒所需的硬件。每个钻孔处使用一个螺钉、两个垫圈和一个六角螺母。将一个垫圈放在前面板上，然后将螺钉穿过垫圈并拧紧至后面板。另一个垫圈和六角螺母。为避免土壤流失，请确保所有螺丝都拧得非常紧。同时，制作两个间隔件，以形成处理区和对照区的区域。使用高密度聚乙烯板制作，并在顶部固定一个螺丝，以限制其仅部分插入。为实现遮光和降温，准备一个可将盒子完全包裹的保护罩。最后，构建一个框架，促使根系朝向背板生长。该PVC框架以约55度的角度固定盒子。准备好均质的田间土壤与沙子的混合物。每个根盒开始时使用一个装有基质混合物的大袋。为每个盒子的处理组和对照组区域分别标记两个小型自封袋。使用天平称取30克土壤-沙混合基质，并将其转移至对照组袋子中。同样，向处理袋中加入30克基质。然后称取1克氮15标记的氮源，并将其加入处理袋中。充分混合基质与氮源。每次操作一个根盒及其对应的垫片。将两个垫片插入根盒中，直至螺钉阻止其进一步移动。在根盒侧面标记垫片底边所处的深度位置。然后移除间隔物以进行后续步骤。使用漏斗，从大量基质中将Rhizobox填充至标记深度。缓慢而均匀地来回移动漏斗，以确保Rhizobox填充均匀。当基质达到标记深度时，将间隔块放入箱中，间隔块应距离箱体左右两侧各五厘米。继续填充基质，直至基质表面距离箱体顶部约五厘米处。接下来，使用滴灌头或手动方式，将150毫升水缓慢灌溉至盒中的基质内。静置数小时或过夜，使水分充分扩散，确保每个间隔物周围的区域完全湿润。此时，移除间隔物，留下用于放置样本的空腔。现在，将透明胶片贴在盒子的外侧。利用透明胶片将其中一个区域标记为处理组，另一个标记为对照组。然后使用漏斗和预先准备好的对照基质填充对照区域。这是填充后已标记的对照贴片。以相同方式，使用已准备好的处理底物填充处理贴片。完成后，用永久性记号笔在透明膜上描出每个贴片的边界。接下来，将大袋中剩余的基质均匀地填入根盒中。最后，用透明板描记基质表面的轮廓。待根盒准备就绪后，移栽一颗已萌发的种子。首先，使用窄 spatula 在 Rhizobox 中心挖一个孔，孔深应为 2.5 厘米。将已萌发的种子放入孔中，确保其胚根朝向正下方。在盒子上附着的透明纸上标记种子的位置。覆盖种子后，用最多50毫升的去离子水进行浇灌。接着，将盒子放入其配套外壳中，以防止光照在生长过程中影响根系。将盒子移至为其准备的支架上。将根盒放置在支架上，以促进植物向后部支架方向生长。每隔三到四天取回一次盒子。去掉外壳后，每次使用不同颜色的笔来描记植物可见的根系。如本示例所示，此步骤需保持一致性和系统性。将带盖的盒子放回支架，并确保其处于原始方向。此处将24个根箱背面追踪的根系总长度与使用扫描仪和计算机软件测量的根长进行对比作图。对根箱前面追踪的根系分布模式进行类似的作图，表明根系倾向于优先沿着根箱背面生长。该图中总根长对数值随生长时间（天数）变化曲线的斜率一致，表明不同根箱之间的根系生长速率相似。以下是一种玉米基因型的根长密度数据。对照组处理的根长密度低于氮素处理组的根长密度。该实验比较了六种不同玉米基因型的根长密度。在所有情况下，氮15标记处理区域的根长密度均高于对照区域。该方案可与其他方法（如测压计法或荧光原位杂交）结合使用，以可视化酶活性或微生物的空间分布。
