1. 肽溶液的制备
<ol>
	<li>从 -20 °C 冰箱中取出肽段（P2：CAAAAE，P4：CAAAAW，PKCδi：CSFNSYELGSL），在室温（RT）下解冻。 
	注意：保持瓶盖关闭，防止水分在肽段上冷凝。</li>
	<li>使用微量天平称取每种肽段各 0.01 g，分别放入不同的 50 mL 锥形管中。</li>
	<li>向 P2 管中加入 18.74 mL 去离子水（DI 水）。</li>

	<li>向 PKCδi 管中加入 8.21 mL 用去离子水稀释的 50% 乙腈。</li>
	<li>短暂涡旋混合肽段溶液，将 50 mL 锥形管置于超声仪（40 MHz）中处理 5 分钟。</li>
	<li>将肽段溶液转移至生物安全柜中。所配制的肽段溶液浓度均应为 1 mM。</li>
	<li>将每种肽段溶液各 1 mL 转移至新的 50 mL 锥形管中；向 P2 和 P4 管中各加入 19 mL 去离子水，向 PKCδi 管中加入 19 mL 50% 乙腈，使各溶液稀释至 50 µM，并分别储存于各自的管中。</li>
</ol>
2. GNP/PKCi 的配制
<ol>
	<li>应在生物安全柜内将多肽加入GNP溶液中</li>
	<li>向一个15 mL离心管中加入475 μL的P2、25 μL的P4和500 μL的δPKCi溶液。再向同一离心管中加入9 mL的20 nm GNP溶液（7.0×1011 粒子/mL）</li>
	<li>离开生物安全柜，用铝箔包裹15 mL离心管，并在室温下置于摇床上过夜</li>
	<li>将样品重新放回生物安全柜内，将1 mL的GNP/PKCi溶液分装至各1.5 mL微量离心管中</li>
	<li>在4 °C条件下，以15,294 × g离心30分钟</li>
	<li>在生物安全柜内小心吸除每根离心管中的上清液 
	注意：吸除上清液时需小心操作，确保GNP沉淀完整，避免吸出沉淀</li>
	<li>根据所需浓度，将沉淀重悬于适当的溶剂中。适用的溶剂包括去离子水、PBS和0.9% NaCl 
	注意：从1 mL的GNP/PKCi溶液开始，GNP沉淀含有6.3 × 1011 个粒子（基于制造商提供的GNP浓度）。若需在500 µL的0.9% NaCl溶液中给予1.3 × 1012 个粒子，则向三个沉淀中各加入232 µL的0.9% NaCl，合并后即可获得500 µL的GNP/PKCi溶液 
	注意：在稀释GNP/PKCi沉淀前，应充分混匀所用溶剂，否则GNP/PKCi可能发生聚集</li>

肺移植可挽救终末期肺病患者的生命1。然而，肺移植后严重的并发症仍然是一个障碍。在肺移植后的早期阶段，原发性移植物功能障碍是最严重的并发症1，其主要原因是缺血-再灌注（IR）引起的急性肺损伤2。
在低温保存期间，供体肺的代谢活动被限制在极低水平。然而，由于血流停止，活性氧和一氧化氮的合成被激活3。移植后，血液灌注恢复，冷缺血期间产生的活性氧和一氧化氮会加剧炎症反应和细胞死亡，导致组织损伤。
为预防缺血再灌注损伤（IR损伤），蛋白激酶Cδ抑制剂（PKCδi）已被应用于心脏、大脑和肺组织4,5,6,7,8。这些研究表明，PKCδi可在再灌注期间减轻炎症反应和细胞凋亡。此外，PKCδi还可在大鼠模型及肺移植模型中预防肺部IR损伤6。PKCδi通常与一种细胞穿透肽TAT偶联，以实现细胞内递送。然而，已有研究显示TAT肽本身具有非特异性的生物学效应，包括促进血管生成、诱导细胞凋亡以及抑制多种细胞因子9,10,11。纳米颗粒是直径在1至100 nm范围内的微小颗粒12，已被探索作为药物递送的候选载体13。特别是金纳米颗粒（GNPs）被认为具有无创性和低毒性。因此，我们已开发GNPs作为基于肽类药物的递送载体14,15。

Protein Kinase C-delta Inhibitor Peptide Formulation using Gold Nanoparticles

应仔细评估 GNP/PKCi 杂交体的生物物理特性，因为 GNP 在溶剂中容易发生聚集。当 GNP 聚集时，溶液颜色会从粉红色变为紫色（图 1a）。紫外-可见分光光度计能够更灵敏地检测到这种变化。若 GNP/PKCi 未发生聚集，则吸收峰应位于 525 nm 处（图 1b）。若 GNP 发生聚集，吸收峰将向长波方向移动。作为另一种分析方法，当形成聚集体时，Δ光密度（ΔOD = 525 nm 处的 OD 减去 440 nm 处的 OD）会降低。

我们独特的药物递送系统采用20纳米的金颗粒与两种特定短肽结合，可在生理溶液中稳定金纳米颗粒，并增强其细胞摄取效率，从而有效地将基于肽类或其他药物递送至细胞内。我们的金纳米颗粒-肽杂合物易于合成且性质稳定。通过将金纳米颗粒-肽杂合物作为药物载体，我们能够高效且大量地将肽类药物转入细胞内。我们使用GNP/PKCi杂合体治疗肺部缺血-再灌注损伤。GNP可将PKC抑制剂肽递送至细胞质中。该递送方法能够以低于其他递送方法的PKCi剂量，减轻更为严重的肺损伤。开始此操作前，从零下20摄氏度的冰箱中取出多肽，并在室温下解冻。使用微量天平称取每种解冻后多肽0.01克，并将每种多肽分别转移至单独的50毫升锥形管中。向P2管中加入18.74毫升去离子水，向P4管中加入16.93毫升去离子水。向 PKC delta 抑制剂管中加入 8.21 毫升用去离子水稀释的 50% 乙腈。短暂涡旋振荡肽溶液。然后将管子置于 40 兆赫的超声仪中处理 5 分钟。此后，将溶液转移至生物安全柜中。将50 mL锥形管放入超声仪中处理五分钟至关重要。如果跳过此步骤，GNP/PKCi将形成聚集体。将每种肽溶液各一毫升转移至各自新的50毫升锥形管中。向含有P2和P4的管中加入19毫升去离子水。向含有PKC delta抑制剂的管中加入19毫升50%乙腈。将475毫升P2溶液、25微升P4溶液、500微升PKC delta抑制剂溶液加入一个15毫升离心管中。然后向同一管中加入九毫升20纳米金纳米颗粒（GNP）溶液。将混合溶液从生物安全柜中取出，用铝箔包裹试管，并将试管置于摇床中过夜。第二天，将样品放回生物安全柜中。将1毫升的GNP/PKCi溶液分装至1.5毫升的微量离心管中。在4°C条件下，以15294 ×g的离心力在微量离心机中离心30分钟。将微量离心管重新转移回生物安全柜中，小心移除每管中的上清液，确保不扰动金纳米颗粒（GNP）沉淀。根据所需浓度，用适当的溶剂重悬每份沉淀。为评估GNP/PKCi杂合物的溶解度，将0.5毫升GNP/PKCi溶液倒入比色皿中。将比色皿置于紫外-可见分光光度计上，检测其最大吸收峰。在本研究中，蛋白激酶 C-delta 抑制剂与金纳米颗粒偶联形成 GNP/PKCi 杂合体。在评估该杂合体的生物物理特性时必须谨慎，因为金纳米颗粒在溶剂中容易发生聚集。当纳米颗粒发生聚集时，溶液的颜色会从粉红色变为紫色。使用紫外-可见分光光度计对样品进行分析，该仪器具有高度的检测灵敏度。如果杂交体未发生聚集，吸收峰应位于525纳米处。如果金纳米颗粒（GNP）发生聚集，其吸收峰将向右偏移。可以看出，当形成聚集体时，光密度差值会降低。在制备GNP-肽杂合物时，需注意肽与GNP溶液的比例应为1:9。这种GNP/PKCi配方也可用于肺以外器官系统的缺血-再灌注损伤。只要肽段在N端含有半胱氨酸残基，即可与GNP结合。这意味着GNP-肽杂合体可用于将其他肽类药物递送入细胞内。经过合理设计的多肽是一种新技术，可在细胞内实现特定的蛋白质-蛋白质相互作用。本研究所开发的这一技术可用于递送新型多肽药物，或对现有药物的改良版本进行递送。
