1. 乙醇提取物的制备

	<li>用10 mL 95%乙醇重新溶解干燥物。</li>

	<li>使用孔径为0.22 µm的无菌注射器滤膜再次过滤乙醇提取物，收集滤液，可直接用于HPLC分析。</li>

3. 使用高效液相色谱法分析蜂胶素含量

	<li>蜂胶素标准曲线的建立

		<li>配制1 L流动相，甲醇:水（v/v）为88.8:11.2的溶液。</li>
		<li>使用上述流动相作为溶剂，对蜂胶素标准品（C、D、F和G）进行系列稀释，浓度分别为15.625 mg/mL、31.25 mg/mL、62.5 mg/mL和125 mg/mL。</li>
		<li>依次从低浓度到高浓度，将20 µL各浓度蜂胶素标准品注入反相色谱柱中。</li>

		<li>将细菌菌株Staphylococcus aureus和Escherichia coli解冻，随后分别在胰蛋白胨大豆肉汤和营养肉汤中于37 °C培养24 h。</li>

蜂胶是由蜜蜂（Apis mellifera）产生的一种天然树脂混合物。自古以来，蜂胶在民间医学中被广泛使用。最近一项研究表明，蜂胶有助于预防微生物感染和炎症1。大量研究证实，蜂胶中的主要生物活性成分包括黄酮类化合物、酚酸酯类、 prenylated p-香豆酸和二萜酸2,3。迄今为止，已通过高效液相色谱法（HPLC）从中国台湾绿蜂胶中鉴定出10种 prenylated 黄烷酮衍生物4,5,6,7。其中含量最丰富的为蜂胶素C、D、F和G5,7。中国台湾绿蜂胶显著的生物学效应与其高含量的蜂胶素密切相关8。
蜂胶中生物活性成分的浓度因采集季节和地理来源的不同而存在显著差异。欧洲蜂胶主要含有黄酮类苷元和酚酸9。巴西蜂胶中的主要生物活性成分是带有异戊烯基取代的p-香豆酸，例如阿替匹林C（artepillin C）10。一项研究表明，季节是决定中国台湾绿蜂胶中总原黄酮（propolin）含量的关键因素11。中国台湾绿蜂胶中的原黄酮含量在夏季（5月至7月）最高，在冬季最低11。蜂胶的抗菌特性通常被视为其生物活性的重要指标。总体而言，来自不同地区的蜂胶样品表现出相似的抗菌特性；例如，蜂胶通常对几乎所有革兰氏阳性菌有效，而对革兰氏阴性菌的抗菌作用则较为有限10,12,13。研究证实，蜂胶中的黄酮类化合物之间存在协同作用，从而产生抗菌效果14。类似地，中国台湾绿蜂胶也被报道对革兰氏阳性菌具有抗菌作用15。此外，有研究还发现，中国台湾绿蜂胶中原黄酮成分之间的相互作用也具有抗菌活性8。
蜂胶中生物活性化合物的表征较为困难，因其化学成分会随来源不同而变化。因此，有必要建立一种可行且可重复的方法，用于评估中国台湾绿蜂胶的质量。然而，目前尚未建立针对中国台湾绿蜂胶提取及其后续功能分析的标准操作流程。已有多种使用有机或无机溶剂的提取方法被提出用于蜂胶提取16,17,18,19,20。由于蜂胶是一种亲脂性混合物，研究表明，有机溶剂提取效果优于无机溶剂提取8,18,19。中国台湾绿蜂胶中总propolin含量及其抑菌特性是评价其质量的关键指标。因此，本研究旨在建立一种以乙醇为溶剂提取并表征中国台湾绿蜂胶抑菌特性的实验方案。

本文介绍了使用乙醇作为溶剂提取和表征具有抑制细菌活性的中国台湾绿蜂胶的实验方案。该方案是一种可靠且可重复的方法，用于评估中国台湾绿蜂胶的质量。实验操作演示由本实验室的博士研究生陈俊廷（Chun'Ting Chen）和简翊轩（Yi'Hsuan Chien）完成。首先，称取 100 克中国台湾绿蜂胶。使用香料研磨机将蜂胶研磨成细粉，确保无大颗粒残留。准备五个锥形瓶，分别加入 100 毫升不同浓度的乙醇水溶液。将10克粉碎的蜂胶加入每个烧瓶的乙醇溶液中，在25摄氏度下以250转/分钟的转速振荡孵育48小时。之后，使用孔径为25微米的滤纸过滤乙醇提取物。使用容量瓶，用95%乙醇将滤液重新定容至原来的100毫升体积。将重组的提取物在零下20摄氏度保存，直至使用。当准备进行HPLC分析时，使用真空蒸发法在40摄氏度下浓缩每种提取物10毫升，持续10分钟。将干燥物在45摄氏度下烘烤24小时。然后，用10毫升95%乙醇重新溶解每份干燥样品。使用孔径为0.22微米的注射器滤器，对每份提取物进行再次过滤，并收集滤液，用于直接进行高效液相色谱（HPLC）分析。为开始建立标准曲线，配制1升流动相甲醇-水溶液，其中甲醇与水的比例为88.8:11.2。以流动相溶液为溶剂，按照文本方案中所述制备黄芩素标准品的系列稀释浓度。将每种标准浓度溶液各取 20 微升，按浓度从低到高的顺序依次注入反相色谱柱。然后，将高效液相色谱柱温度设置为 30 摄氏度，紫外检测器波长设为 280 纳米，流速设为每分钟 1 毫升，记录时间设为 20 分钟。至少对标准品进行三次分析。随后，使用计算表格软件将测得的响应值与浓度对应，绘制标准曲线，并得到相应的方程和R²值。开始分析提取物时，将每种提取物各20 µL注入反相色谱柱中。将高效液相色谱柱温度设置为 30 摄氏度，流速设置为每分钟 1 毫升。将紫外检测器的波长设置为 280 纳米，记录时间设置为 20 分钟。至少重复分析提取物三次。然后，利用标准曲线确定每种乙醇提取物中丙泊林的浓度。在制备好测试生物体和乙醇提取物后，向96孔板的各孔中加入10微升稀释后的乙醇提取物，其浓度范围为每毫升0.156至640微克。使用肉汤将每孔的体积调整至100微升，并确保所有稀释液中二甲基亚砜（DMSO）的浓度保持在5%。接下来，将100 µL细菌培养物接种至96孔板的每个孔中。在37 °C下培养48小时。使用酶标仪在590 nm波长下根据浊度分析细菌生长情况，并确定最低抑菌浓度。此后，从每个显示有生长的孔中取 10 微升液体培养物接种到琼脂平板上。在 37 摄氏度下培养 24 小时。确定未见明显细菌生长的最低浓度，以判断细菌活性。完全抑制细胞生长的浓度被视为最低抑菌浓度。本研究中，使用乙醇提取中国台湾绿蜂胶。当使用高浓度乙醇时，干物质得率似乎最高；而使用水时，干物质得率最低。这些结果表明，在提取过程中使用有机溶剂效果最佳，且提取产率与乙醇浓度呈正相关。同样，黄酮类化合物（propolins）的浓度在提取过程中也似乎与乙醇浓度呈正相关，其中95%和99.5%乙醇提取物中黄酮类化合物的得率最高。随后研究了乙醇提取物对金黄色葡萄球菌（S aureus）和大肠杆菌（E coli）的抑菌作用。金黄色葡萄球菌的平均最小抑菌浓度似乎在每毫升10至20微克之间，平均细菌浓度似乎为每毫升20微克。然而，水提取物对金黄色葡萄球菌无任何抑菌作用。使用水提取物或乙醇提取物时，均未观察到对大肠杆菌的抑菌作用。一个关键的实验步骤是在研磨过程中避免台湾绿蜂胶的均匀化。
