1. 体外正常浆细胞分化模型
注意：浆细胞通过四步培养法获得11,12,13。
<ol>
	<li>B 细胞扩增与分化
	<ol>
		<li>使用健康志愿者的外周血细胞进行记忆 B 细胞的纯化。</li>
		<li>将 7.5 mL 血液用 24.5 mL RPMI 1640 培养基稀释。</li>
		<li>在无菌的 50 mL 锥形管中加入 12.5 mL 室温密度梯度液（例如 Ficoll），然后将 32 mL 稀释后的血液轻轻叠加在密度梯度液上方，尽量减少两相之间的混合。</li>
		<li>以 500 × g 离心 20 分钟，关闭刹车。收集稀释血浆与密度梯度液界面处的外周血单个核细胞（PBMCs），转移至无菌 50 mL 管中，并用 Hank's 平衡盐溶液补至 45 mL。</li>
		<li>以 500 × g 离心 5 分钟，小心弃去上清液，保留沉淀细胞。</li>

			<li>利用磁铁分离装置将结合磁珠的细胞与未结合细胞分离，收集未结合细胞，并将细胞沉淀与抗 CD19 APC 和抗 CD27 PE 抗体在 4 °C 孵育 15 分钟（每 106 个细胞加入 2 µL 抗体）。</li>
			<li>在 PBS/10% 小牛血清中洗涤两次。</li>
			<li>在 FSC 和 SSC 图中去除杂质事件。通过 FSC-A 与 FSC-H、SSC-A 与 SSC-H 图绘制单细胞群，以去除碎片并选择总白细胞群体。在 CD19/CD27 图上选择记忆 B 细胞，即 CD19+CD27+ 细胞。</li>
			<li>使用细胞分选仪纯化记忆 B 细胞（MBCs），纯度达到 95%。</li>
		</ol>
		</li>
		<li>所有培养步骤均使用 IMDM 培养基和 10% FCS，并添加 50 mg/mL 人转铁蛋白和 5 mg/mL 人胰岛素。</li>

	<li>长寿命浆细胞的生成
	<ol>
		<li>通过改变培养条件，将早期浆细胞分化为长寿命浆细胞。</li>
		<li>将细胞接种于 12 孔培养板（每孔 2 mL，密度为 5 × 105/mL）或 24 孔培养板（每孔 1 mL），在 37 °C 下使用含 IL-6、基质细胞条件培养基和 APRIL（200 ng/mL）的培养体系进行培养。</li>
		<li>通过将基质细胞在培养基中培养 5 天获得基质细胞条件培养基，收集上清液经 0.2 µm 滤膜过滤后冷冻保存。</li>
		<li>在浆细胞培养体系中加入 50% 的基质细胞条件培养基，并每周更换一次。所生成的长寿命浆细胞可维持数月13。已有报道表明，仅使用 IL-6 和 APRIL 即可实现浆细胞的长期存活13。</li>

B细胞和浆细胞（PC）是体液免疫中的关键参与者。了解调控正常浆细胞生成与存活的生物学过程，对于需要确保有效免疫应答并预防自身免疫或免疫缺陷的治疗干预至关重要。浆细胞在体内属于稀有细胞，其早期分化阶段发生在解剖学上难以完全进行生物学表征的部位，尤其是在人体中。我们已报道了一种通过多步骤培养系统建立的体外B细胞向浆细胞分化的模型。该模型可重现体内不同器官中发生的连续性细胞分化与成熟过程11,12,13.&#160;第一步，记忆B细胞通过CD40配体、寡脱氧核苷酸及细胞因子组合刺激激活四天，分化为前浆母细胞（PrePBs）。第二步，通过去除CD40L和寡脱氧核苷酸的刺激，并更换细胞因子组合，诱导前浆母细胞进一步分化为浆母细胞（PBs）。第三步，再次更换细胞因子组合，诱导浆母细胞分化为早期浆细胞（early PCs）11,12。第四步，通过将早期浆细胞培养于骨髓基质细胞条件培养基或特定生长因子中，获得完全成熟的浆细胞13。这些成熟浆细胞可在体外存活数月，并分泌大量免疫球蛋白（图1）。值得注意的是，我们的体外模型能够重现体内可检测到的B细胞向浆细胞各阶段分化过程中协调发生的转录变化及表型特征11,12,13,14,15。由于浆细胞本身数量稀少，本体外分化模型为研究人类B细胞向浆细胞的分化过程提供了有效手段。

In Vitro Differentiation Model of Human Normal Memory B Cells to Long-lived Plasma Cells

浆细胞（PCs）分泌大量抗体，由已活化的B细胞发育而来。浆细胞是稀有细胞，主要位于骨髓或黏膜组织中，负责介导体液免疫。由于其频率低且定位特殊，在人体中研究浆细胞具有较大难度。我们报道了一种从B细胞向浆细胞 利用特定细胞因子和活化分子的组合构建体外分化模型，可重现体内发生的连续性细胞分化过程。在此体外模型中，记忆B细胞（MBCs）将依次分化为前浆母细胞（prePBs）、浆母细胞（PBs）、早期浆细胞（early PCs），最终形成长寿浆细胞（long-lived PCs），其表型接近健康个体中的相应细胞.&#160;我们还开发了一个开放获取的生物信息学工具，用于分析与浆细胞分化相关的基因表达谱（GEP）数据中最显著的信息。这些资源可用于研究人类B细胞向浆细胞的分化过程；在本研究中，我们探讨了人类B细胞向浆细胞分化过程中表观遗传因子的基因表达调控。

体外正常浆细胞分化的整体流程如图1所示。通过本方案，我们能够生成足够数量的细胞，而这些细胞无法通过人源原代样本直接获得。尽管参与浆细胞分化的复杂转录因子网络的作用已被研究，但调控关键浆细胞分化转录网络的机制仍知之甚少。细胞分化主要由表观遗传学和转录水平的变化驱动。利用我们的体外模型以及B细胞至浆细胞的基因表达谱图谱（GEP atlas），我们研究了正常浆细胞分化过程中表观遗传调控因子的表达变化。

浆细胞是稀有细胞，其在解剖学位置上经历30个分化阶段，这些位置阻碍了对细胞生物学特性的全面表征，尤其是在人体中。我们的体外B2浆细胞分化模型可重现体内不同器官中发生的有序细胞分化与成熟过程。该体外模型能够再现体内可检测到的不同B2浆细胞阶段的协调性转录变化及其表型特征。我们还构建了一个开放获取的生物信息学工具，用于分析与浆细胞分化相关的探索性数据中最显著的信息。演示该实验步骤的是我实验室的博士后 Hugues de Boussac。首先，从健康志愿者中获取外周血细胞，用于记忆B细胞的纯化。将7.5毫升血液与24.5毫升RPMI 1640培养基混合稀释。接着，向一个无菌的50毫升锥形管中加入12.5毫升室温密度梯度液。轻轻将32毫升稀释后的血液覆盖于密度梯度液上方，注意尽量减少两相之间的混合。在无刹车条件下，以500 × g离心20分钟。从稀释的血浆与密度梯度界面处收集外周血单个核细胞。将细胞转移至无菌的50毫升离心管中，并加入汉克平衡盐溶液至45毫升。在500 × g条件下离心5分钟。小心弃去上清液，保留沉淀。随后加入45毫升RPMI 1640培养基，添加10% FCS。在500 × g条件下离心5分钟。小心弃去上清液，保留沉淀。加入30 mL PBS，其中添加2% FCS。使用抗CD磁珠去除CD2阳性细胞，每四个磁珠对应一个靶细胞。在4°C下孵育30分钟。利用磁力架进行细胞分离，将与磁珠结合的细胞与未结合的细胞分离开。收集未结合的细胞，并在4°C下与抗CD19 APC抗体和抗CD27 PE抗体孵育15分钟。用含有10%小牛血清的PBS洗涤细胞两次。然后，使用细胞分选仪将记忆B细胞纯化至95%纯度。此后，加入可溶性 CD40 配体和抗多聚组氨酸单克隆抗体以激活记忆 B 细胞。将纯化的记忆 B 细胞接种于六孔培养板中，用白细胞介素-2、白细胞介素-10 和白细胞介素-15 培养四天。进行基因表达谱分析时，使用细胞分选仪在第4天纯化 CD38 阴性 CD20 阴性的前浆母细胞。第四天，计数细胞，并将细胞悬液以每毫升250,000个细胞的密度接种至12孔培养板中，每孔加入两毫升细胞悬液。为诱导分化，去除CPG寡核苷酸和可溶性CD40配体，加入含有新细胞因子组合的新鲜培养基，该组合包括白细胞介素-2、白细胞介素-6、白细胞介素-10和白细胞介素-15。将细胞在37摄氏度条件下培养三天。进行基因表达谱分析时，使用细胞分选仪在第7天纯化CD38阳性、CD20阴性的浆母细胞。在第7天对细胞进行计数。将细胞以每孔500,000个细胞的密度接种于12孔培养板中，每孔加入2毫升细胞悬液。使用含有白细胞介素-6、白细胞介素-15和干扰素-α的新鲜培养基，在37摄氏度下将浆母细胞分化为早期浆细胞，持续三天。为了进行基因表达谱分析，在第10天使用细胞分选仪纯化CD20阴性、CD38阳性、CD138阳性的早期浆细胞。首先，用培养基将基质细胞培养五天。使用0.2微米滤器过滤基质细胞培养上清液，以获得基质细胞条件培养基，并冷冻保存至使用前。接下来，将早期浆细胞接种于12孔培养板中，每孔加入2毫升细胞悬液，细胞密度为每孔500,000个细胞，使用先前获得的含有白细胞介素-6和APRIL的基质细胞条件培养基进行培养。于37°C孵育，每周更换一次基质细胞条件培养基。然后，通过流式细胞术分选的浆细胞检测其免疫球蛋白分泌情况。将浆细胞以每毫升一百万个细胞的密度培养24小时，收集培养上清液。使用ELISA试剂盒检测免疫球蛋白G（IgG）和免疫球蛋白A（IgA）。首先，启动GenomicScape开放获取生物信息学网络工具。在浏览数据菜单中，选择名为 human B cells to plasma cells 的数据集。可使用分析工具中的“表达/共表达报告”工具来可视化单个基因或基因列表的基因表达谱。接下来，选择分析工具 webSAM 以加载 SAM 工具。选择人B细胞向浆细胞分化的数据集，并选定要比较的样本组。使用可用的各种筛选条件，应用感兴趣的筛选选项。若要使用SAM工具比较基因表达谱，请调整包括倍数变化、置换次数、错误发现率以及比较类型在内的筛选参数。该软件将进行分析，并提供在所选组别之间差异表达的基因。本研究探讨了正常浆细胞分化过程中表观遗传因子的表达变化。通过多类别SAM分析发现，在记忆B细胞、前浆母细胞、浆母细胞、早期浆细胞和骨髓浆细胞之间，共有71个基因显著差异表达，错误发现率低于5%。记忆B细胞阶段的特征是23个表观遗传相关基因的过表达，其中包括39.13%的组蛋白乙酰转移酶、30.43%的组蛋白甲基转移酶、21.74%的组蛋白去甲基化酶、4.35%的甲基结合蛋白以及4.35%的组蛋白去乙酰化酶。在前浆母细胞阶段，有14个表观遗传调控基因显著过表达，其中包括50%的组蛋白甲基转移酶、14.28%的DNA甲基转移酶、21.43%的组蛋白去乙酰化酶、7.14%的组蛋白去甲基化酶以及17.14%的组蛋白乙酰转移酶。通过该实验流程，可进一步开展多项分析，以鉴定并理解B2浆细胞分化过程中染色质结构与构象的改变。该体外分化模型能够产生足够数量的细胞，用于完成上述分子分析，从而揭示参与整个建模过程的相关信号通路及其在B2浆细胞分化期间的下游转录影响。本研究所获得的知识可为浆细胞疾病（如多发性骨髓瘤）的诊断和治疗策略提供依据和指导。多发性骨髓瘤是一种尚无明确治愈手段的癌症，迫切需要更佳的预后评估和更优的治疗策略。其中，特别值得关注的是B2浆细胞分化过程中影响染色质的表观遗传重塑、结构及构象变化的特征与机制。这些研究可通过基因测序、ERRBS、ATAC测序、IC以及RNA测序等技术实现。根据用于记忆B细胞纯化的健康供体血液的不同，活化B细胞、前浆母细胞、浆母细胞和浆细胞的比例可能存在一定程度的异质性。
