本研究及实验程序已获得第二军医大学（中国上海）动物实验伦理委员会批准，符合《实验动物护理和使用指南》（美国国家研究委员会，1996年）的规定。
1. 动物
<ol>
	<li>使用两个月大（200–250 g）的Sprague-Dawley（SD）大鼠。每次行为学检测均需使用独立的大鼠组别。对照组与实验组始终应分开使用。 
	注：本研究包含两项自主神经功能检测：EVM 和 FWR。EVM 包含对照组在内的四个组别（= 4），每个组别进行三项行为学检测（平衡木、排便计数和旷场实验 = 3），每组8只大鼠，共计96只大鼠（4 × 3 × 8）。FWR 包含对照组在内共两个组别（= 2），每个组别进行三项行为学检测（平衡木、排便计数和旷场实验 = 3），每组8只大鼠，共计48只大鼠（2 × 3 × 8）。总计报告使用144只大鼠。</li>
	<li>将啮齿类动物饲养于恒定25 °C温度、60%–70%湿度的环境中。</li>
	<li>将啮齿类动物置于12 h光照/12 h黑暗的循环条件下，并可自由获取食物和饮水。 
	注：由于以下实验均为行为学实验，操作大鼠时应轻柔。应以双手抓握，支撑其身体和后肢，避免引发焦虑。</li>
	<li>在黑暗环境中进行实验（EVM 和 FWR）及评估检测（平衡木和旷场实验评估），以尽量减少视觉线索的干扰。</li>
</ol>
2. 电梯垂直运动装置
<ol>
	<li>在完全黑暗的环境中进行升降机垂直运动操作，以尽量减少视觉提示。</li>
	<li>将啮齿类动物放入聚甲基丙烯酸甲酯（Plexiglas）盒中（22.5 cm × 26 cm × 20 cm）。该聚甲基丙烯酸甲酯盒可容纳四只啮齿类动物（定制装置）。</li>
	<li>确保盒子已牢固关闭，防止动物掉落。将聚甲基丙烯酸甲酯盒放置在升降机垂直运动装置的升降平台垫上（定制装置）。</li>
	<li>启动升降机垂直运动装置，并调至最低设置以进行适应性训练。</li>
	<li>设定振幅为从中性位置向上22 cm、向下22 cm。按以下方式逐步调整升降机垂直运动参数：
	<ol>
		<li>初始周期设定为：2500 ms 持续5分钟，2000 ms 持续5分钟，1500 ms 持续5分钟。</li>
		<li>测试周期使用1000 ms，持续2小时。</li>
		<li>反向逐步减慢设备，依次使用1500 ms 持续5分钟、2000 ms 持续5分钟、2500 ms 持续5分钟。</li>
	</ol>

3. 摩天轮旋转装置
<ol>
	<li>摩天轮旋转装置设置
	<ol>
		<li>将有机玻璃容器（22.5 cm × 26 cm × 20 cm）放置在木制工作台（定制装置）上。</li>
		<li>将啮齿类动物放入有机玻璃容器中，使其身体长轴与摩天轮的水平旋转杆垂直（定制装置）。 
		注：将身体长轴垂直于水平杆放置，可确保在旋转过程中刺激耳石器官（前后方向和垂直方向）。</li>
		<li>牢固关闭有机玻璃箱。</li>
		<li>将另一组啮齿类动物放置在摩天轮旋转装置第二臂的有机玻璃容器中，其身体长轴同样垂直于水平旋转杆。使用质量相近的另一组啮齿类动物以平衡摩天轮。</li>
		<li>牢固关闭有机玻璃箱，并将其安装到摩天轮旋转装置上。</li>
	</ol>
	</li>
	<li>摩天轮旋转操作流程
	<ol>
		<li>在完全黑暗的环境中进行摩天轮旋转操作，以尽量减少视觉线索的干扰。</li>
		<li>启动摩天轮，以16°/s2的加速度顺时针旋转，直至角速度达到120°/s，随后以48°/s2的减速度减速至0°/s。暂停1秒后，容器以相同方式逆时针继续旋转（以16°/s2加速度达到120°/s角速度，再以48°/s2减速度减速至0°/s）。顺时针—暂停—逆时针循环约需10秒回到初始位置。</li>
		<li>每次实验持续进行顺时针—逆时针旋转2小时，共计约720次旋转。</li>
	</ol>

4. EVM 和 FWR 的评估
注意：摩天轮旋转装置和电梯垂直运动的评估通过以下三个步骤进行：平衡木测试、排便计数和开放旷场实验。评估电梯垂直运动时采用相同的步骤。这些评估步骤应在摩天轮旋转或电梯垂直运动结束后尽快进行。

	<li>平衡木实验

		<li>平衡木装置设置
		<ol>
			<li>在实验区域内放置两张木凳（高度约为 0.75 m），间距约 110 cm，以搭建平衡木装置10,11,12。</li>
			<li>在终点木凳上放置一个黑色塑料盒（15 cm × 15 cm × 8 cm）。</li>
			<li>将一根窄木条（2.5 cm × 130 cm）架在两张木凳之间，使凳边之间的距离为 100 cm，从起点凳延伸至终点凳。 
			注：黑色塑料盒的入口应位于 100 cm 距离的终点线上。</li>
			<li>在起点木凳处放置一盏灯，并打开光源。</li>
			<li>关闭室内灯光，确保房间尽可能黑暗。这可促使啮齿类动物从明亮区域沿平衡木向阴暗区域移动。</li>
		</ol>
		</li>
		<li>平衡木实验步骤 
		注：通过测量动物穿越架高木条所需时间，评估其运动协调能力。
		<ol>
			<li>在正式检测前，连续 3 天每天对每只啮齿类动物进行训练，以使其在平衡木上的表现趋于稳定10。训练时将大鼠置于光源处的角落，引导其穿越木条，最终大鼠会自主完成穿越。本实验方案中大鼠的平均穿越时间为 3.6 ± 0.9 秒。 
			注：部分啮齿类动物在训练期间无法达到稳定表现，应予以剔除。有些动物不执行任务，或缺乏穿越木条的动机。稳定表现定义为连续两次测试的穿越时间均少于 4 秒。若大鼠在训练或评估过程中掉落，应记录为“跌落”，并停止进一步测试。</li>
			<li>正式实验时，将训练好的动物置于靠近光源的起点木凳上，同时启动秒表。动物应迅速穿越平衡木并进入终点木凳上的黑色盒子中。</li>
			<li>当动物就位后启动秒表，当其鼻尖进入终点木凳的暗盒时停止计时。穿越时间指从起点木凳到终点木凳的总耗时。 
			注：动物完成训练后，可在评估前实施干预或处理（如诱发运动病）。也可利用最后一次训练的穿越时间获取基线数据，用于后续比较。</li>
		</ol>

	<li>排便计数
	<ol>
		<li>在旋转轮测试结束后，将装有四只啮齿类动物的有机玻璃容器放置于实验台上。</li>
		<li>取出动物，并分别放入独立的旷场箱（见下文）。</li>
		<li>统计每只动物在有机玻璃箱内排出的粪便颗粒数量。 
		注：可通过在电梯垂直运动前统计粪便颗粒数量，获得基线值，以便与运动后的评估结果进行比较。</li>
	</ol>
	</li>
	<li>旷场实验
	<ol>
		<li>将啮齿类动物放入旷场箱（40 cm × 40 cm × 45 cm）中。</li>
		<li>使用红外视频摄像机记录动物 3 分钟内的旷场行为28,29,30。</li>
		<li>计算动物的总移动距离。 
		注：在电梯垂直运动前，切勿将动物提前放入旷场箱。环境对动物而言必须是全新的，因此不应为旷场实验采集基线数据。</li>
