一旦细胞分离，轻轻混合每一个井的内容，并转移100微升每个细胞悬浮液到适当的相应的井在新的24井板。然后将细胞返回细胞培养箱24小时。要用双荧光酶处理器结构转染细胞，请准备转染稀释混合物，如表中所示。
要准备记者稀释混合物，混合每个试剂的体积乘以转染的总数加上几个额外的体积。接下来，将转染稀释混合物与记者稀释混合物混合，在室温下孵育所得溶液15分钟，产生转染混合物。在孵育过程中，用250微升PBS冲洗细胞培养板的每口井，并在每口井中加入447微升的新鲜完整生长介质。
然后使用多通道移液器将53微升的转染混合物分发到细胞培养板的每个井中。将板在37摄氏度和5%的二氧化碳24小时。为了测量荧光素酶活性，用去电化水稀释百西解酶活性试剂盒中的5X无源解液缓冲液，并解冻试剂盒中所需的试剂A和试剂B缓冲液。
当试剂准备就绪后，用75微升无源解液将每井中的上流液更换，并在室温下孵育板30分钟，偶尔摇晃。在孵育过程中，用解冻试剂B缓冲液将试剂盒B基板稀释至1至50浓度。在孵化结束时，在四口控制井中加入30微升无源解液缓冲液，并将30微升的解盐从每口井转移到96孔平底白化检测板的重复井中。
当所有莱酸盐都镀上时，使用多通道移液器向每个井中添加 50 微升试剂 A，并使用板读卡器读取萤火虫荧光素酶信号。然后使用多通道移液器将 50 微升试剂 B 添加到每个井中，并使用板读卡器读取 Renilla 荧光素酶信号。正如预期的那样，YAP2SA 增加萤火虫荧光酶活性，但与控制载体相比，不会增加雷尼拉荧光酶活性。
相比之下，YAP2SA S94A不会增加萤火虫荧光素酶活性。重要的是，YAP2SA不会改变缺少 MCAT TEAD 绑定元素的最小启动子结构的活动。在与 YAP-TAZ-TEAD 记者结构相染的细胞中，串联的 YAP 和 TAZ 短发夹显著降低萤火虫荧光素酶水平，但对短发夹的非靶向控制不然。
相比之下，在与最小处理器结构相染的细胞中，YAP-TAZ 短发夹RNA不会显著改变萤火虫荧光素酶水平。每个井中的 Renilla 信号也未因短发夹非靶向控制或 YAP-TAZ 短发夹 RNA 而显著改变。正如预期的那样，针对 YAP 和 TAZ、SARC 或 PI3 激酶的短发夹RNA显著减少 YAP-TAZ-TEAD 活性。
