演示程序时，将李玉亭和谭香玉，来自我实验室的研究助理。为了产生扁病毒颗粒，将400万293T扁病毒包装细胞播种到100毫米培养皿中，其中10毫升DMEM辅以10%FBS。在37摄氏度和5%的二氧化碳下孵育细胞，确保细胞在转染当天汇合50%至80%。

将适当数量的细胞播种到新的100毫米培养皿中，配以10毫升新鲜预热GCSC完整培养基和每毫升多氧环素2.5微克。然后，孵育48小时的菜。使用自动细胞计数器确定 10 微升细胞样本的密度。
然后，使用预热的GCSC完整培养培养介质调节体积，获得每毫升2万个活细胞的浓度。将100微升的电池悬浮液分配到每个井中，一个新的超低附着96井培养板。用5%的二氧化碳在37摄氏度下孵育细胞。
球体形成应在三到十天内发生。每两天对肿瘤球形成进行成像。原发性人类胃腺癌的胃癌干细胞在血清自由培养基培养中。
六天后，细胞从像表型一样的单细胞扩展到大球体。为了评估GCSC中聚类的功能，将针对聚类素的短发夹RNA序列克隆到Tet-GV307-RFP-Puro载体中。与表达载体转染的细胞用多氧环素治疗48小时。
然后，通过西方印迹验证聚类表达，通过密度测定进行量化。GCSC中多氧环素的存在和聚类素的敲落抑制了肿瘤球的形成。虽然在用炒 shRNA 对子转导的细胞中未观察到肿瘤球形成抑制，但表明多氧环素本身并不抑制肿瘤球的形成。
这些结果表明，在聚类沉默后，GCSC生长缓慢，不能形成肿瘤球体。基于这些数据，聚基在促进GCSC的自我更新活动方面发挥着关键作用，表明它可能是抑制胃癌患者癌症干细胞的有希望的药物靶点。肿瘤球体应是单个或聚合细胞上的固体圆形结构，不应被视为肿瘤球体。
