High-Throughput Identification of Resistance to Pseudomonas syringae pv. Tomato in Tomato using Seedling Flood Assay

番茄是一种具有重要农艺价值的作物，可被革兰氏阴性细菌Pseudomonas syringae侵染，导致细菌性斑点病。番茄与P. syringae pv. tomato的互作体系被广泛用于解析植物先天免疫反应和抗病性的遗传基础。尽管通过将Solanum pimpinellifolium中的Pto/Prf基因簇导入栽培番茄，已成功防控该病害数十年，但P. syringae的1号生理小种已进化出克服Pto/Prf基因簇所介导抗性的能力，并已在世界范围内出现。
野生番茄物种是病原体识别中天然多样性的重要资源库，因为它们在具有不同病原体压力的多样化环境中进化而来。在对野生番茄进行抗病性筛选时，通常使用成株植物，但由于成株生长期较长且需要更大的生长空间，限制了可筛选植株的数量。我们开发了一种针对10日龄番茄幼苗的抗性筛选方法，该方法可最大限度地缩短植株生长时间和生长室空间占用，实现植株的快速轮替，并支持大规模样本的测试。幼苗的存活或死亡结果可视为离散表型，也可根据淹水后存活幼苗的新组织生长量定义为一个抗性等级。该方法已优化用于10日龄番茄幼苗对两种P. syringae菌株的抗性筛选，并可轻松适用于其他P. syringae菌株。

检测 PtoR利用苗期抗性测定法评估品种和近等基因系中的介导免疫反应

该高通量实验方案可快速筛选来自野生种质资源的番茄幼苗对细菌性病原菌丁香假单胞菌（Pseudomonas syringae）的抗性。幼苗浸润法可最大限度缩短植物生长时间和减少对人工气候室的需求，实现植物的快速轮替，并支持大样本量的检测。该检测方法是一种强大的工具，可用于筛选具有复杂遗传背景的野生种质资源及其他品系中的抗病性状。本方案提供了针对两种丁香假单胞菌（Pseudomonas syringae）菌株进行喷雾接种的具体操作步骤。然而，该方法具有通用性，可经修改后用于检测植物对其他细菌病原体的抗性。本实验操作将由我实验室的研究专家 Yana Hasan 演示。首先培育番茄幼苗。将番茄种子放入一个2.2毫升的微量离心管中，加入2毫升50%的次氯酸钠溶液。轻轻摇动管子25分钟，然后用移液器吸除次氯酸钠溶液。加入两毫升超纯水，通过倒置离心管五次来清洗种子。吸去管中的液体，再重复清洗四次。最后一次清洗后，加入两毫升超纯水，将种子倒入无菌培养皿中。用乙醇火焰灼烧镊子进行灭菌，然后用其将五到七粒种子转移至含有0.5倍MS加0.8%琼脂培养基的100 mm × 25 mm培养皿中。用外科胶带密封培养皿边缘。确保培养皿平放且正面向上叠放。将灭菌后的种子置于4°C黑暗条件下层积至少三天，以同步萌发。三天后，将培养皿垂直放置，使根沿培养皿表面向下生长，种子排成水平线。将种子转移至生长培养箱中，设置温度为22°C，光照周期为16小时光照、8小时黑暗。在培养箱中培养幼苗10天，此时幼苗通常已完全长出并展开子叶，且第一片真叶开始出现。用无菌扁平牙签将新鲜细菌划线接种到适当的KB琼脂平板上。对于PstT1，将平板在28°C下培养48小时，然后用于喷雾接种实验。为制备接种物，需在无菌条件下将细菌重悬于无菌的 10 毫摩尔氯化镁溶液中，使其光密度达到 0.1。随后进行两次连续稀释，以获得 OD600 为 0.075 的工作浓度。同时，用 10 毫摩尔氯化镁溶液制备非离子型有机硅表面活性剂共聚物 1:10 的稀释液。涡旋混匀后加入细菌中，轻轻旋转离心管以充分混合。将盛有十日龄幼苗的培养皿从生长室转移至生物安全柜中，撕去外科胶带。然后向每个培养皿中加入6 mL的接种液。用移液枪枪头轻轻将幼苗推入接种液中，并启动计时器，持续三分钟。双手各持一个培养皿，将培养皿前端倾斜向下，使幼苗的子叶和叶片浸入接种液中。左右晃动五到七次，然后将培养皿向后倾斜，使接种液覆盖幼苗根部。再次倾斜培养皿，重复该循环操作，共计三分钟。将接种液从培养皿中倒出，将培养皿平放于水平表面，再次倾倒以去除残留的接种液。重新包好培养皿，放回培养箱中。将Moneymaker-PtoR和Moneymaker-PtoS品种用PSTDC3000菌液浸润，并在浸润后7至10天进行表型观察。Moneymaker-PtoR幼苗携带PtoPRF基因簇，对PSTDC3000具有抗性。而近等基因系的Moneymaker-PtoS幼苗无法识别PSTDC3000的效应子AVRPto或AVRPto-B，浸润后7天内迅速死亡。将10天大的幼苗用PstT1进行淹水处理，并在淹水后至少10天进行表型鉴定。感病材料植株死亡，顶端分生组织呈褐色，且无新组织生长；而抗病幼苗则表现出大量新生绿色组织，并能存活于PstT1感染。通过细菌生长实验定量验证了PstT1抗性及Solanum NeoRici LA1329的抗病性。在抗性与感病LA1329之间，细菌生长量相差1.7 log，在抗性LA1329与Moneymaker-PtoS之间相差1.6 log，该结果与表型数据一致。进行此实验时，需特别注意在整个操作过程中严格遵守无菌操作技术，并确保平板上所有幼苗均被充分浸润。务必对受感染的植物组织和细菌进行高压灭菌，并根据相关规定处理废弃物。
