1. 涡虫的饲养
<ol>
	<li>使用从生物供应公司购买的或必要时野外捕获的涡虫。本方案中使用的涡虫为供应清单中列出的 Dugesia tigrina。该物种也被称为 Girardia tigrina31。其他水生涡虫物种也可接受2,3。 
	注意：所述方案适用于从生物供应公司购买的动物。该检测系统尚未在野外捕获的涡虫中进行测试。然而，如果使用野外捕获的涡虫，建议在使用前至少将其适应实验用水及实验室环境1周。</li>
	<li>收到涡虫后，将其转移至含有洁净矿泉水的塑料食品储存容器中，并将容器盖略微打开。</li>
	<li>将涡虫维持在避光环境中。</li>
	<li>新到的涡虫在新环境中放置24–36小时后再进行喂食。</li>
	<li>在开始实验前，至少让涡虫在实验室环境中适应1周。</li>
	<li>按照每周两次的规律时间表喂养涡虫。
	<ol>
		<li>让涡虫自由摄食切碎的有机水煮蛋或匀浆的有机牛肝，持续1–2小时。</li>
		<li>喂食后，将涡虫转移至洁净容器中。
		<ol>
			<li>移除涡虫周围的脏水。</li>
			<li>使用小号平头水彩画笔（3–6号）将附着在容器壁及涡虫周围的残余食物和黏液转移到纸巾上。</li>
			<li>用新鲜的矿泉水轻轻旋转或搅动，使涡虫脱离容器，然后将其倒入洁净容器中。</li>

	</li>
	<li>清洁用于涡虫饲养的容器和器具时，不得使用肥皂或洗涤剂。应使用洁净水（自来水即可）充分冲洗，并用干净的布或纸巾擦干。</li>
</ol>
2. 实验用涡虫的准备
<ol>
	<li>新送达的涡虫在实验前至少应适应其环境1周。</li>
	<li>实验前对涡虫进行5–10天的饥饿处理。</li>
	<li>在饥饿期间至少更换1次培养用水。</li>
</ol>
3. 涡虫运动速度（pLmV）测试：兴奋行为
<ol>
	<li>实验前，确保饥饿的涡虫已完全发育，具有完整且具色素的头部和尾部。</li>
	<li>在开始实验前，准备好用于 pLmV 测试的玻璃或塑料 10 cm 培养皿以及适应容器。
	<ol>
		<li>将一个洁净的直径为 10 cm 的培养皿放置在预贴好的网格纸上（方格为 0.5 cm），用于 pLmV 测试。</li>
		<li>向直径为 10 cm 的培养皿中加入 20 mL 未添加任何物质的泉水（对照组），或含有待测天然产物适当浓度的泉水，用于 pLmV 测试。</li>
		<li>在准备好的直径为 10 cm 的培养皿上方放置一台相机（例如手机或高分辨率相机），以记录实验期间涡虫在网格纸上的运动情况。使用铁架台可方便地将相机固定在适当距离，以完整拍摄整个 10 cm 培养皿及网格线。</li>
		<li>准备一个适应容器，加入 5–10 mL 未添加任何物质的泉水（对照组），或含有待测天然产物适当浓度的泉水。类似闪烁计数小瓶或小型 5 cm 培养皿的容器均可使用。</li>
	</ol>
	</li>
	<li>使用洁净的小型扁头或圆头水彩画笔，轻轻地将一只涡虫从盛有泉水的储存容器中转移至含有 5–10 mL 未添加任何物质的泉水或含有待测天然产物的泉水的适应容器中。 
	注意：在进行 pLmV 检测操作涡虫时，应使用洁净的小型扁头或圆头水彩画笔（型号 3–6）。移动涡虫时，应将画笔置于其下方轻轻托起。为确保涡虫不受损伤，画笔刷毛不应在涡虫下方散开；若刷毛散开，涡虫可能被夹在刷毛之间而受到伤害。 
	注意：也可使用宽口移液管将涡虫转移至洁净干燥的适应容器中。
	<ol>
		<li>若使用移液管，需用该移液管将随涡虫带入适应容器中的多余水分吸出。</li>
		<li>小心地向含有涡虫的适应容器中加入适应液（即对照组用的泉水，或含有待测天然产物浓度的泉水）。</li>
	</ol>
	</li>
	<li>适应时间将取决于所测试天然产物的刺激动力学特性。本研究中，2 分钟的适应时间已足以检测到刺激效应1。</li>
	<li>在 2 分钟适应期结束后，使用水彩画笔将涡虫轻轻转移至已准备好的 10 cm 培养皿中央，进行 pLmV 刺激实验。</li>
	<li>启动相机以记录涡虫的运动，连续录制 10–11 分钟的视频。</li>
	<li>每次测试每只涡虫时，均需为 pLmV 实验准备新的适应容器和 10 cm 培养皿，并使用新鲜配制的溶液。</li>
	<li>为避免实验过程中意外将涡虫暴露于错误溶液中，应为每种天然产物的实验浓度配备专用的移液管、培养皿、容器和画笔。</li>
	<li>由于涡虫会表现出学习行为，每只涡虫（对照组或实验组）仅应使用一次21,22。</li>
</ol>
4. 涡虫运动速度（pLmV）测试：回避行为
<ol>
	<li>实验前，确保饥饿的涡虫已完全发育，具有完整且具色素的头部和尾部。</li>
	<li>准备一个10 cm的培养皿（玻璃或塑料）用于pLmV实验，一个5 cm的培养皿（玻璃或塑料）用于在适应后冲洗涡虫，并在实验开始前准备好适应容器。
	<ol>
		<li>将一个洁净的直径为10 cm的培养皿放置在预先贴好的网格纸上（方格为0.5 cm），用于pLmV实验。</li>

	<li>为每只涡虫的pLmV实验，均需使用新鲜溶液重新准备适应容器、冲洗容器和10 cm培养皿。</li>

5. 数据分析
<ol>
	<li>准备一个数据收集表格，用于记录在 pLmV 实验期间每分钟内涡虫的行为和运动性，包括每分钟穿过的网格线数量。该表格还应能够记录涡虫每分钟累计穿过的网格线总数。表格中应包含用于记录备注的行，以及一个行为定义表，用于统计实验期间观察到的各种行为（如“游走”和“停止”）的发生情况（见讨论部分）。</li>
	<li>通过视频回放，记录涡虫在10分钟内每分钟穿过的完整网格线数量，并将该数值记录在数据表中。典型的涡虫行为表现为持续的、向前的、水平方向的匀速运动，期间伴有周期性的转向，但无停顿。
	<ol>
		<li>实验计时应从涡虫完全离开用于将其转移至10 cm培养皿的毛刷时开始，并记录起始时间。</li>
		<li>判断动物是否穿过一个完整的网格时，应以头部为参考标准：当头部完全越过一条网格线时，计为穿过一条线。</li>
		<li>当涡虫沿培养皿边缘移动时，可通过观察从皿边缘延伸出的线条来估算0.5 cm的距离。若涡虫穿过方格的角落，则以第二条被穿过的线计为一条完整的网格线。在进行此类判断时，仍应以头部位置为准。</li>
		<li>每分钟结束后暂停视频，以便记录数据。</li>
		<li>重新播放视频以记录下一分钟的数据时，若暂停时虫体头部位于两条网格线之间，则首次穿过下一条线时应计为一个完整的网格。</li>
		<li>持续记录10分钟内穿过的网格线总数。</li>
	</ol>
	</li>
	<li>若在 pLmV 测试过程中，动物停止移动，在10分钟记录期内不再穿过任何网格线，则应在行为记录表中注明其行为状态（例如“游走”或“停止”）。在 pLmV 检测中停止前进运动的个体应单独记录，并以暴露于该试剂浓度下的总动物数中的频率形式呈现数据。若在适应期出现卷曲或痉挛性行为（称为C型行为），导致无法进行任何前向运动，则表明该天然产物的浓度不适用于 pLmV 检测，因为 pLmV 检测是基于运动能力的。C型行为可采用其他类型的分析方法进行评估（见讨论部分）。</li>
	<li>若条件允许，为评估试剂对涡虫生理的整体影响，应在不同日期和不同时间段使用至少9–12只涡虫测试多个实验浓度。然而，若研究人员希望减少昼夜节律引起的变异性，可在每天固定时间、恒定光照条件下进行实验，并使用经过定时明暗循环培养和定时喂食的涡虫。建议至少安排两名实验人员参与项目，其中一人负责记录数据，另一人则在不知晓实验分组条件的情况下“盲法”计数网格线穿过次数。在数据收集、统计计算与分析过程中由不同人员参与，也有助于降低潜在的偏倚。</li>
	<li>将每一天每种天然产物浓度下的网格线穿行次数，相对于对照组每分钟的穿行次数进行归一化处理，以便整合来自不同日期、不同时间和不同实验者的數據。这些数据可进行平均后采用学生t检验（Student’s T-test）进行分析。每个时间点的p值可与对照组以及不同试剂浓度之间进行比较。使用来自不同实验浓度的数据集进行方差分析（ANOVA）可提供另一种分析方法。</li>

使用涡虫作为此类研究的动物模型相较于其他动物具有多项优势，实验者可轻松控制实验环境。具体而言，在实验前对涡虫进行饥饿处理，可避免其接触其他可能干扰实验结果的营养或药理物质；而待研究的特定生物调节剂只需直接加入培养水中即可引入涡虫体内，从而实现暴露条件的标准化。由于涡虫拥有与“高等动物”相似的神经系统和神经递质，其对神经肌肉刺激的生理反应和实验响应被认为对其他生物具有生物学相关性12,13,14,15,16。此外，由于涡虫在实验室中相对廉价且易于维持，因此为众多研究者提供了一种可及的生物模型。
作为一种实验动物，涡虫适用于多种研究。例如，我们课题组及其他研究者利用涡虫研究肿瘤发生过程17,18,19。涡虫还表现出对化学、热力、重力、电、光以及磁场刺激的一系列反应行为，这些反应构成了其他检测系统的基础。其中一些效应已被用于研究这些动物的学习与记忆能力20,21,22,23,24,25,26,27。目前文献中涡虫模型的主要用途集中在一类称为新胚细胞（neoblasts）的多能干细胞的活性及其在再生过程中的作用28,29,30。因此，采用本文所述模型可进一步结合其他基于涡虫的检测方法，从而更全面地理解天然产物及其他生物调节剂对生物体的影响。
