1. 载玻片和盖玻片的准备
<ol>

	<li>将双面粘性成像间隔垫贴附至载玻片的FEP胶带涂层面，顶部保护衬层保持未剥离状态（图1A）。 
	&#8203;注意：载玻片和盖玻片应在实验前预先制备。可立即使用，或存放于盒中以防止表面灰尘积聚。成像间隔垫中的孔深度为120 µm，直径为9 mm。</li>
</ol>
2. 间期阻滞卵提取物的制备与活体成像
注意：以下实验方案改编自 Deming 和 Kornbluth19、Murray20 以及 Smythe 和 Newport25，并进行了修改。除非另有说明，所有步骤均应在室温下进行。
<ol>
	<li>在取卵前3至10天，将100 IU的孕马血清促性腺激素（PMSG）皮下注射至成熟的雌性Xenopus laevis青蛙的背侧淋巴囊中。</li>
	<li>在计划取卵前16至18小时，向步骤2.1中的青蛙注射500 IU的人绒毛膜促性腺激素（hCG）。将青蛙置于18 °C的产卵缓冲液（100 mM NaCl，2 mM KCl，1 mM MgSO4·7H2O，2.5 mM CaCl2·2H2O，0.5 mM HEPES，0.1 mM EDTA，配制成pH 7.4的20倍浓缩液，使用前用洁净的蛙养殖用水稀释至1倍）中，直至取卵。</li>
	<li>实验当天，在大号玻璃培养皿中收集卵子，并评估卵子质量。丢弃外观呈白色蓬松球状或成串出现的卵子（图1B）。在体视显微镜下检查卵子，保留外观正常的卵子（图1C），丢弃色素分布不规则或斑驳的卵子（图1D）。 
	注意：本方案适用于从一只青蛙收集的卵子，通常在注射hCG后16小时内可产约25 mL卵子。一般使用hCG诱导3至6只青蛙，选择卵子质量最高的个体用于提取物制备实验。</li>
	<li>将卵子转移至400 mL玻璃烧杯中，通过倾倒尽可能去除产卵缓冲液。</li>
	<li>将卵子在100 mL新配制的脱胶液（2% w/v L-半胱氨酸溶于水，用NaOH调节pH至8.0）中孵育，并定期轻轻摇动。约3分钟后，倒出溶液，加入100 mL新鲜脱胶液。继续孵育直至卵子紧密堆积（卵子间无空隙），但避免脱胶液处理总时间超过5分钟。</li>
	<li>通过倾倒尽可能去除脱胶液，并用0.25x MMR缓冲液（25 mM NaCl，0.5 mM KCl，0.25 mM MgCl2，0.5 mM CaCl2，0.025 mM EDTA，1.25 mM HEPES，配制成10倍浓缩液，用NaOH调节pH至7.8，使用前用Milli-Q水稀释）洗涤卵子：加入缓冲液，轻轻摇动卵子，然后倒出缓冲液。重复数次，直至总共使用1 L缓冲液完成洗涤。</li>
	<li>用总共400 mL卵裂解缓冲液（250 mM蔗糖，10 mM HEPES，50 mM KCl，2.5 mM MgCl2，1 mM DTT，现配并用KOH调节pH至7.7）洗涤卵子数次。在每次洗涤之间，用巴斯德移液管去除外观异常的卵子。 
	注意：外观异常的卵子指呈白色蓬松球状（图1B）、色素斑驳（图1D）、出现白色区域扩大的退化现象（图1E），或动物极色素区域变暗并收缩的卵子（图1F）。</li>
	<li>使用尖端剪宽的移液管，将卵子转移至含有1 mL卵裂解缓冲液的17 mL圆底离心管中。在临床离心机中以400 × g离心15秒，使卵子压紧。</li>
	<li>用巴斯德移液管尽可能从压紧的卵子上层去除卵裂解缓冲液。 
	注意：必须尽可能去除压紧卵子中的缓冲液，以最大限度减少卵提取物的稀释。有时需连同残留缓冲液一起去除部分松散的卵子以实现此目的。</li>
	<li>测定压紧卵子的近似体积，然后直接在压紧的卵子上层加入5 µg/mL抑蛋白酶肽（aprotinin）、5 µg/mL亮抑蛋白酶肽（leupeptin）、5 µg/mL细胞松弛素B（cytochalasin B）和50 µg/mL环己酰亚胺（cycloheximide）。 
	注意：抑蛋白酶肽和亮抑蛋白酶肽为蛋白酶抑制剂。细胞松弛素B抑制肌动蛋白聚合，防止提取物收缩和凝胶化26。环己酰亚胺抑制蛋白质合成，从而使提取物保持在细胞周期的间期。</li>
	<li>将离心管置于摆桶转子中，在4 °C下以12,000 × g离心15分钟，破碎卵子。 
	注意：离心结束后，卵子应已破裂，裂解物分为三层主要结构：顶部为黄色脂质层，中间为细胞质提取物（也称粗提物），底部为含有色素颗粒的深色致密层（图1G）。</li>
	<li>将18号针头连接至注射器。针尖斜面朝上，从离心管侧面刺入细胞质层底部，缓慢吸取提取物。 
	注意：应缓慢吸取细胞质提取物，以避免混入上层黄色脂质层的污染物。</li>
	<li>将收集的细胞质提取物转移至新的微量离心管中，置于冰上保存。提取物应在1小时内使用。</li>
	<li>准备成像时，向提取物中加入所需的试剂和荧光成像探针。 
	注意：荧光成像探针可标记特定的细胞质结构，使其能在荧光显微镜下被观察到。</li>
	<li>移除步骤1.2中制备的载玻片上成像间隔片的顶部保护膜，在孔中央滴加约7 µL提取物。立即盖上FEP膜涂层的盖玻片，使FEP面朝向提取物以密封孔位。迅速进行成像（图1H，I）。</li>
	<li>将载玻片置于配备电动载物台和数字相机的倒置或正置显微镜上。在明场和荧光通道中，按所需的空间位置和时间间隔对提取物进行成像。 
	注意：通常使用5倍物镜进行成像。电动载物台可实现多个预设空间位置的自动图像采集。明场显微镜可基于透明度差异观察细胞质结构。荧光显微镜可观察在步骤2.14中加入的荧光成像探针所特异性标记的细胞质结构。相机记录这些结构的时间序列图像，从而捕捉细胞质组织的动态变化。</li>

细胞质构成了细胞的主要体积，并具有独特的组织结构。真核细胞细胞质的组分能够自组装形成多种空间结构，例如微管星状体和高尔基体，这些结构又会根据细胞的身份和生理状态发生动态排列和更替。因此，理解细胞质的空间组织及其与细胞功能的关联，对于揭示细胞的工作机制至关重要。Xenopus laevis 卵提取物传统上被用于大规模生化分析1,2,3,4,5,6,7,8，但近年来的研究已将其确立为研究细胞质结构及其细胞功能机制的有力活体成像系统9,10,11,12,13,14,15,16,17,18。这些未稀释的提取物在保留细胞质多种结构和功能的同时，允许对细胞质内容进行直接操控，而这种操控在传统的基于细胞的模型中难以实现19,20。这使得它们成为表征细胞质现象并解析其机制基础的理想体系。
现有的提取物成像方法需要化学表面修饰，或微流控装置的制备。一种基于盖玻片的方法需要对玻璃盖玻片进行聚乙二醇（PEG）钝化处理21。一种基于微乳液的方法需要在玻璃表面进行三氯(1H,1H,2H,2H-全氟辛基)硅烷的气相沉积22,23。基于微流控的系统能够精确控制提取物液滴的体积、几何形状和组成，但需要专门的微加工设施11,12,24。
本文介绍一种用于成像卵提取物的替代方法，该方法易于实施，并使用 readily available、低成本的材料。该方法包括使用载玻片和盖玻片构建成像腔室，其中盖玻片表面涂有氟化乙烯丙烯（FEP）胶带。该腔室可用于多种显微镜系统对提取物进行成像，包括体视显微镜、直立显微镜和倒置显微镜。该方法无需对表面进行化学处理，即可实现与上述基于玻璃的现有方法相当的光学清晰度。该方法旨在对在二维视野内厚度均匀的提取物层进行成像，并可轻松扩展用于对三维体积的提取物进行成像。该方法特别适用于大视野下细胞质集体行为的延时成像。
我们使用处于间期阻滞的卵提取物来演示成像方法。提取物的制备遵循 Deming 和 Kornbluth19 的方案。简言之，天然停滞于减数分裂 II 期中期的卵通过低速离心破碎。该离心过程使细胞质脱离减数分裂阻滞状态，从而使提取物进入间期。通常在离心破碎前加入细胞松弛素 B，以抑制 F-肌动蛋白的形成。然而，如果需要保留 F-肌动蛋白，则可省略该步骤。此外，在离心破碎前还需加入环己酰亚胺，以防止间期提取物进入下一次有丝分裂。随后将提取物置于上述成像腔室中，并放置于显微镜上。最后，通过连接显微镜的相机在设定的时间间隔内连续采集图像，获得延时图像序列，从而记录提取物在二维视野中的动态行为。

该方法适用于利用蛙卵提取物对细胞质动态进行活体成像，能够研究细胞质中的空间模式信息。样品可被塑造成厚度均匀且可调的薄层，并可在二维视野或三维体积范围内进行成像。该方法成本低廉且易于实施。首先，使用滚筒涂布器将一层FEP胶带粘贴到玻璃载玻片上。用干净的剃刀片将边缘多余的胶带切除。接着，以相同方法制备涂有FEP胶带的盖玻片。然后将双面粘性成像垫贴在载玻片涂有FEP胶带的一侧，暂不揭去保护衬层。将卵转移至一个400毫升的玻璃烧杯中，并通过倾倒尽可能去除卵孵化缓冲液。随后将卵在100毫升新鲜配制的脱胶液中孵育，并轻轻摇动。大约三分钟后，倒掉溶液，加入 100 毫升新鲜的脱胶液。继续孵育，直至卵子紧密聚集，但避免卵子在脱胶液中总时间超过五分钟。孵育结束后，尽可能去除脱胶液，并用 0.25x MMR 缓冲液清洗卵子。轻轻旋转混匀卵子，然后倒掉缓冲液。重复数次，直至洗涤过程中总共使用一升缓冲液。接着，用总共400毫升的卵裂解缓冲液将卵子洗涤数次，在每次洗涤之间去除异常的卵子。然后使用带有宽口吸头的移液管将卵转移至含有1毫升卵裂解缓冲液的17毫升圆底离心管中。在临床离心机中以400 ×g离心15秒，使卵沉淀。随后用巴斯德吸管尽可能吸除卵裂解缓冲液。确定卵团的大致体积，然后在卵团上方直接加入抑蛋白酶肽、亮抑蛋白酶肽和细胞松弛素B，每种终浓度为每毫升5微克，同时加入放线菌酮至终浓度为每毫升50微克。将离心管置于水平转子中，在4°C、12,000 g条件下离心15分钟，压碎卵团。接下来，将一支18号针头连接到注射器上。针尖斜面朝上，从细胞质层底部的侧面刺入离心管，缓慢吸取以回收提取物。将回收的细胞质提取物转移至新的微量离心管中，并置于冰上保存。准备成像时，在提取物中加入所需的试剂和荧光成像探针。从已准备好的载玻片的成像间隔垫上移除顶部保护层，并在孔中央滴加约七微升提取物。立即盖上带有FEP膜的盖玻片，使FEP面朝向提取物以密封孔，随后迅速进行成像。将载玻片置于带有电动载物台和数字相机的倒置或正置显微镜上。在明场和荧光通道中，按所需的时间间隔和空间位置对提取物进行成像。此处展示的是利用非洲爪蟾（xenopus laevis）卵提取物研究间期胞质自组织的延时成像实验。在室温下孵育约20分钟后，明场和线粒体通道中均可观察到细胞质成分（如光散射物质）减少的区域。在室温下孵育约60分钟后，应已形成由类似细胞的区室构成的空间模式，微管形成中空的花环样结构，线粒体则被清晰地分配到各个区室中。此处展示了在玻璃表面有无FEP胶带的成像腔室中提取物性能的对比。在用FEP胶带固定玻璃制成的腔室中，提取物自组织形成了正常的类细胞结构。而在未用FEP胶带覆盖玻璃表面的腔室中，提取物显示出异常的明场和微管结构，且随时间推移逐渐变得紊乱。在GST-mCherry-NLS蛋白的核输入方面未观察到显著差异。需要牢记的重要事项是使用FEP胶带对玻璃表面进行钝化处理，并在提取物制备过程中去除所有不良样本。通过该系统，我们可以轻松控制提取物样品的厚度，为利用光片显微镜进行三维细胞质模式成像铺平了道路。
