机械感觉系统的传入神经元在听觉和平衡过程中将信息从毛细胞传递到大脑。通过直接记录，电生理学可以揭示传入神经元的敏感性。虽然对毛细胞进行全细胞膜片钳记录具有挑战性，但对下游传入神经元进行记录则更为容易，并且能够在受控刺激条件下评估动作电位的反应1,2,3。刺激毛细胞会导致其发生偏转，从而改变机械感觉结构，进而引发传入神经元动作电位（尖峰）的增加4,5,6。在没有外部刺激的情况下，由于毛细胞向传入神经后突触末梢发生谷氨酸泄漏，传入神经元也会产生自发性放电7,8，并且已被证明有助于维持系统的敏感性9,10。对传入神经活动进行膜片钳记录，能够观察到毛细胞的敏感性和信号动态变化，而这些是使用时间分辨率较低的技术（如微音器电位11,12或功能性钙成像13,14,15）所无法实现的。以下实验方案可用于同步记录传入神经活动与运动指令，以揭示毛细胞敏感性的瞬时变化。
斑马鱼（Danio rerio）利用侧线系统中的感觉毛细胞来感知相对于其身体的水流，这种感知被转化为神经信号，对导航16,17,18、躲避捕食者、捕食19,20以及群游行为21至关重要。水流也可能由游泳22,23,24、呼吸22,25,26和摄食27等自身运动产生。这些行为包含重复性动作，可能导致毛细胞疲劳并削弱感知能力。因此，侧线系统必须能够区分外部（传入性）和自身产生的（再传入性）水流刺激。在斑马鱼运动过程中，伴随放电信号会抑制自身产生的水流信号。这种抑制性的预测性运动信号通过下行神经元传递至感觉受体，以调节输入或中断对再传入反馈的处理28,29。早期对这一前馈系统理解的关键研究依赖于体外制备模型，其中神经回路的连接性和内源性活动未能得以维持28,30,31,32,33,34,35。本实验方案描述了一种保持完整神经回路的方法，维持内源性反馈动态，从而更深入地在活体条件下理解伴随放电机制。

Activity of Posterior Lateral Line Afferent Neurons during Swimming in Zebrafish

感觉系统收集对指导行为至关重要的线索，但动物必须分辨出哪些信息具有生物学意义。运动会产生再感受性线索，动物必须将其与周围环境中相关的感官线索区分开来。例如，当鱼类游动时，身体波动产生的水流会被侧线系统中的机械感受性神经丘所检测，神经丘由毛细胞组成。随后，毛细胞通过感觉传入神经元将流体运动信息从感受器传递至大脑。与此同时，运动指令的伴随放电信号也会被传递至毛细胞，以防止感觉超载。因此，在评估侧线系统敏感性时，必须考虑运动信号预测性抑制效应的影响。我们开发了一种在体电生理方法，可同时记录斑马鱼幼鱼（受精后4-7天）的后侧线传入神经元和腹侧运动根活动，记录可持续数小时。传入神经元的胞外记录采用松贴式膜片钳技术实现，可检测单个或多个神经元的活动。腹侧运动根记录则通过皮肤使用玻璃电极检测运动神经元活动。本实验方案有望在完整且具有行为能力的脊椎动物中，监测不同运动行为过程中感觉输入的内源性或诱发性变化。

在斑马鱼幼鱼被适当固定并成功实现后侧线传入神经节和腹根（VR）记录后，可同时测量传入神经元和运动神经元的活动。使用无间隙记录程序（步骤1.4）显示记录通道，以持续监测传入神经和VR的活动。实时观察可见，自发性传入神经放电频率的下降与VR活动同步发生，后者提示存在拟态游泳发作活动。图1E）。我们发现，最佳结果和准确的峰电位检测依赖于达到至少0.5信噪比的记录。自行编写的预处理脚本可生成图表，以辅助传入神经和腹根（VR）峰电位检测的可视化。自发性传入峰电位通过结合多种峰电位参数进行识别，包括阈值、最小持续时间（0.01 ms）和最小峰间间隔（ISI；1 ms）。在建立记录过程中，随着负压的增加，常可同时检测到来自多个传入神经单位的信号。通过按振幅过滤，可区分独立传入神经信号的动力学特征。通过调整预处理脚本中的下限和上限检测变量，可实现对信号的分离。图2A）。通过强吸引力获取多单位记录可能导致记录不稳定、机械噪声、传入神经健康状况恶化，最终导致信号丢失。因此，在获得理想信号后，应缓慢减小吸力至大气压。腹根尖峰检测的参数与传入...

本方案使研究人员能够在完整且具有行为活动的脊椎动物中，监测单个感觉神经元对受控刺激的活动与反应。这种膜片钳电生理技术是一种快速、直接的方法，可实时记录单个神经元的动作电位活动，与大多数光学成像技术相比，具有更高的时间分辨率和灵敏度，且成本更低。侧线毛细胞与人类内耳中的毛细胞同源，因此该技术有望揭示与人类耳聋及听力障碍相关的毛细胞回路特性。电生理学是一门依赖精细运动技能的技术，需要通过观察并进行实际模仿来掌握。仅提供文字说明容易产生误解。首先，在底部带有盖玻片的组织培养皿中加入一层薄薄的自混合硅胶弹性体（如Sylgard），制成硅胶弹性体底部记录皿。为制备解剖针，使用直流电源对盛有蚀刻剂的100毫升烧杯施加5伏的负电荷，并将一根钨丝连接至正电荷输出端。将金属丝尖端反复浸入蚀刻液中，直至尖端变细成锐利的尖点。在体视显微镜下，使用直刃剃须刀片从尖端起约一毫米处切断金属丝。重复此步骤三次，然后使用精细镊子将针插入已固化的记录皿中。制备记录电极时，使用配备盒式灯丝的微电极拉制仪将硼硅酸盐玻璃毛细管拉制成电极。电极尖端直径应为 30 微米，具有适当的锥形，用于记录后侧线系统的传入神经元。另取一根硼硅酸盐玻璃毛细管，拉制一对尖端直径为 1 至 5 微米的较小电极。每只手各持一个电极，轻轻将电极尖端相互摩擦以打断尖端。使用微电极拉制仪将斜面尖端打磨至光滑。最终尖端直径应在 30 至 50 微米之间。用大口径吸头将斑马鱼幼鱼固定并转移至35毫米的培养皿中，尽可能移除周围的溶液。将幼鱼浸入10微升0.1% α-银环蛇毒素中，孵育约5分钟。用细胞外溶液冲洗瘫痪的幼虫10分钟。然后，使用移液管将幼虫从细胞外溶液浴中转移至硅胶底部的记录皿中。在体视显微镜下，用尖头镊子轻轻将幼虫置于硅胶垫中央上方，使幼虫侧面向上，身体的前端与后端呈从左至右排列。使用尖头镊子，将边缘针垂直插入幼虫肛门正上方背侧脊索的硅胶中。将第二根针插入尾部末端附近的脊索，第三根针插入鳔背侧的脊索。第四根针插入耳囊时，在针进入包埋剂的过程中应轻微旋转。 当施加轻微旋转时，观察肩胛骨与耳囊之间的组织，以显露传入神经元胞体的聚集区域。将固定好的幼虫置于微分干涉相差（DIC）显微镜的10倍物镜下，并调整肌节的肌裂方向与头部左侧记录装置的矢量方向平行。将接地导线插入浴液中，并确保其连接至左侧头部记录装置。使用柔性凝胶上样移液器吸头将30微升细胞外液注入VR记录电极中，并将其插入左侧头部阶段的移液器支架。将放大倍数增加至40倍，并在施加由气动换能器产生的正压的同时，将记录移液器缓慢下降至培养皿溶液中。将电极尖端移至侧线腹侧两个肌节之间的肌隔上方，直至裂隙位于VR电极尖端孔口的中心。继续缓慢下降移液器，直至尖端孔口的后缘轻轻接触上皮层。初次接触后，将移液管以对角线方向移动，确保其前缘接触并形成封接。使用气动换能器施加负压并保持。在膜片钳软件中，单击工具栏上的“播放”按钮以监测 VR 信号。当观察到运动神经元活动呈现威尔斯（Wells）典型的爆发信号动态时，确保开始进行VR记录。将30微升细胞外液注入传入神经记录电极，并将其插入右侧头部支架的移液管固定器中。然后在施加由气动换能器产生的正压的同时，将电极缓慢下放至培养皿的溶液中。找到电极并将电极尖端移至标本上方。使用显微操作器，降低传入电极的尖端，直至其位于肩带骨（cleithrum）上方并保持位置。将放大倍数增加至40倍浸油镜，找到后侧线神经与肩带骨的交汇处。将电极尖端移至传入神经节上方，缓慢下降移液管，直至尖端接触上皮层。轻柔地调整电极位置，使整个尖端边缘与传入神经节充分接触。使用气动换能器施加负压，并保持该压力。在补丁钳制软件中点击“播放”后，当传入神经元的全细胞松散封接记录出现大约每100至200毫秒自发产生一次的动作电位时，确认记录已成功建立。一旦检测到传入神经元和运动神经元均有活动，立即点击峰钳10工具栏上的“记录”按钮，以同时在两个通道中获取连续无间隙的记录数据。按照文本稿件中的描述进行数据预处理。通过使用这种无间隙记录方案，可同时测量传入神经元和腹根神经元的实时活动。由自定义编写的预处理脚本生成图表，以帮助可视化动作电位的检测过程，所用参数包括阈值、最小持续时间和最小峰电位间隔。通过调整预处理脚本中的下限和上限检测变量，获得分离的信号。腹根尖峰检测采用相同的参数，并增加额外输入。运动指令中的簇发由至少在0.1毫秒内出现两次尖峰、持续至少五毫秒来定义，并由至少三个簇发组成，簇发间间隔小于200毫秒。预处理脚本将叠加在明确定义的时间段内输入的传入活动片段。在此情况下，平均自发活动在运动活动开始时表现出显著变化，运动活动的起始在X轴上表示为时间等于零。在游泳前、游泳期间及游泳后的传入峰电位频率之间检测到显著差异。传入峰电位频率与游泳持续时间呈负相关。相对峰电位频率与游泳频率之间未检测到相关性。在尝试进行该操作时，动物健康是最重要的因素。必须密切监测快速的血液流动，这不仅是为了确保动物福利，也是为了提高神经记录成功的可能性。通过利用斑马鱼中可用的遗传工具，该电生理学方案可结合转基因品系，从而有力地研究毛细胞系统及其更广泛范围内的解剖与功能环路连接性。这种对膜片钳技术的简化改进使我们能够在活体中监测感觉系统的功能。
