1. 窗口钝化

</ol>
2. 手术准备
<ol>
	<li>在通风橱或层流柜中进行手术。为避免手术区域受到污染，应分别设置独立且分隔的准备区、手术区和恢复区。</li>
	<li>手术前，将所有手术器械在高压灭菌器中灭菌。若计划进行后续操作，可使用热珠灭菌器对器械重新灭菌。本手术操作采用仅使用尖端（tips-only）的技术。&#160;&#160;</li>
	<li>打开加热手术珠和热珠灭菌器的电源。</li>
	<li>在麻醉舱中使用5%异氟烷对小鼠进行麻醉。</li>
	<li>为去除毛发，将脱毛膏充分涂抹于左上胸部切口部位。不超过20秒后，用湿润的纸巾用力擦去毛发和脱毛膏。必要时重复操作，直至彻底清除手术部位的所有毛发。</li>
	<li>使用2-0丝线缝合线，在22 G导管末端打结，留出2英寸长的线尾（见图1A）。</li>
</ol>
3. 肺窗手术
<ol>
	<li>使用抗菌肥皂洗手。</li>
	<li>每次新手术前，更换新的无菌手套。</li>
	<li>为防止小鼠角膜干燥和眼睛损伤，将眼用软膏涂抹于双眼。</li>
	<li>将10 µL（0.1 mg/kg）的丁丙诺啡与90 µL无菌PBS混合稀释，然后皮下注射，以确保术前镇痛。</li>
	<li>用丝线绑扎的22 G导管对小鼠进行插管15使用气囊导管，通过挤压气囊并观察双侧胸部是否同步隆起，以确认插管成功。</li>
	<li>用2-0丝线在小鼠吻部绑扎固定插管导管（参见 图1B).</li>
	<li>将小鼠置于加热的手术台上，使其呈右侧卧位，以暴露左侧胸腔。</li>
	<li>将呼吸机连接至气管插管导管。</li>
	<li>确保呼吸机提供可控且稳定的通气，随后将异氟烷浓度降至3%。在手术开始时以及手术过程中定期通过脚趾夹捏试验评估麻醉深度。</li>
	<li>用纸胶带将小鼠的前肢向头侧、后肢向尾侧固定在加热的手术台上。再用一条胶带沿小鼠背部纵向贴附，以充分暴露手术区域（参见 图1C).</li>
	<li>在生物安全柜内打开所有外科器械，以保持无菌状态。</li>
	<li>通过大量涂抹消毒剂对小鼠皮肤手术部位进行消毒。</li>
	<li>使用镊子提起皮肤，在胸骨左侧约 7 mm、肋缘下约 7 mm 处做一个直径约 10 mm 的环形切口（图1D).</li>
	<li>仔细辨认任何主要血管。若需切断血管，应使用电凝笔在血管两端进行电凝止血。</li>
	<li>切除覆盖在肋骨上的软组织。</li>
	<li>提升 6th 或 7th 用镊子夹住肋骨。使用钝头显微解剖剪的一侧刀片，将圆头一侧朝向肺部，小心刺穿第6肋间肌。th 和 7th 肋骨以进入胸腔空间（图1E).</li>
	<li>使用压缩空气罐对缺损处进行轻柔排气，使肺组织塌陷并与胸壁分离。以短促脉冲方式喷射压缩空气，以防止医源性肺损伤。</li>
	<li>将活检打孔器置于切割工具上方（补充图1）并小心地将切割工具的底座穿过肋间切口（图1F).</li>
	<li>将切割工具的底部与胸壁平行放置。通过肋骨框架打一个5 mm的圆形孔图1G). 
	注意：确保暴露的肺组织呈粉红色，无损伤迹象。</li>
	<li>使用5-0丝线，在距离孔洞约1 mm处围绕其周围做一荷包缝合，缝合时与肋骨交叉穿插图1H).</li>
	<li>将窗框放置到位，使圆形缺损的边缘与窗框的凹槽对齐（参见 图1I).</li>
	<li>用5-0丝线紧密结扎，牢固固定植入的窗口。</li>
	<li>将100 µL的氰基丙烯酸酯凝胶粘合剂装入1 mL注射器中。</li>
	<li>用压缩空气以稳定而轻柔的气流吹干肺组织，持续约10–20秒（图1J).</li>
	<li>用镊子夹住窗框的外缘，轻轻提起，以确保肺组织与窗框底面分离。</li>
	<li>沿光学窗口框架的底面均匀涂抹一层薄薄的氰基丙烯酸酯粘合剂图1K).</li>
	<li>增加呼吸机的呼气末正压（PEEP）以扩张肺部。</li>
	<li>保持10-20秒，施加轻柔但稳固的压力，将光学窗框固定在肺组织上（图1L).</li>
	<li>在矩形盖玻片上滴加一滴5 mm的剩余氰基丙烯酸酯凝胶粘合剂。</li>
	<li>使用真空镊子夹取5 mm盖玻片。将盖玻片底面浸入粘合剂中，然后在矩形盖玻片侧边反复刮擦三次以去除多余粘合剂，仅保留极薄一层（图1M).</li>
	<li>将盖玻片小心地置于光学窗口框架中央凹槽内，并使其倾斜悬于肺组织上方。短暂夹闭呼吸机以产生正压，使肺过度膨胀。通过旋转动作，将盖玻片调整至与肺组织平行，使肺表面与盖玻片底面直接贴合。保持轻柔压力，使氰基丙烯酸酯粘合剂固化（约25秒）。</li>
	<li>使用镊子将盖玻片从真空拾取头分离图1N).</li>
	<li>使用5-0丝线缝合，再次制作荷包缝合，这一次 &#60;从皮肤切口边缘向周围延伸1 mm。在用锁结扎紧之前，将多余的皮肤塞入窗框外缘下方。</li>
	<li>为确保盖玻片与窗框之间的气密性密封，需在金属-玻璃界面处滴加少量液态氰基丙烯酸酯（参见 图1O).</li>
	<li>将无菌针头连接至1 mL胰岛素注射器。将针头插入剑突下方，朝向左肩方向推进，穿过膈肌进入胸腔。轻柔回抽注射器，以清除胸腔内任何残留的空气（参见 图1P).</li>
	<li>移除小鼠身上的胶带。</li>
	<li>关闭异氟烷。</li>
	<li>继续用100%氧气通气，直至小鼠表现出即将苏醒的迹象。</li>
	<li>小心剪断小鼠吻部周围的2-0丝线缝合线，并拔除气管插管。</li>
	<li>将小鼠转移至清洁的笼子中，并监测直至完全恢复。若出现呼吸困难症状，实施安乐死。</li>

以往对肺部进行单细胞分辨率成像时，必须采用离体（ex vivo）或组织外植的制备方法10,11,12,13，这限制了分析仅能获取单一时间点的数据。尽管这些制备方法确实能提供有用的信息，但无法揭示肿瘤细胞在与完整循环系统相连的器官内的动态行为。
成像技术的最新进展使得在长达12小时的时间内以单细胞分辨率对完整肺组织进行活体可视化成为可能14,15,16。这一成果是在小鼠模型中实现的，所采用的方案包括机械通气、胸廓切除以及真空辅助的肺固定。然而，尽管该技术首次实现了对生理状态下完整肺组织的单细胞分辨率成像，但其具有高度侵入性且为终末性操作，因而无法在初次手术后进行进一步的成像观察。因此，这一局限性阻碍了该技术在研究持续时间超过12小时的转移过程（如肿瘤细胞休眠及重新启动生长）中的应用14,15,16。此外，由于真空诱导的压力梯度可能改变血流分布，因此对此成像方法所观察到的细胞行为模式需谨慎解读。

A Permanent Window for Investigating Cancer Metastasis to the Lung
