本研究由国家关键研发计划资助&amp;中国国家重点研发计划（2017YFD0200400）、重大科技创新工程（2017CXGC0214）、山东省蜂产业创新团队、山东省农业科学院农业科技创新项目（CXGC2019G01）以及山东省农业科学院农业科技创新项目（CXGC2021B13）。

1. 喂食管的制备
<ol>
	<li>使用电热穿孔器（见材料表）在2 mL离心管管盖上打一个孔（直径约0.3 mm）。使用该离心管来饲养O. excavata幼虫及其食物团。</li>
</ol>
2. 农药的配制
<ol>
	<li>将工业级农药（参见材料表）溶解于丙酮中，配制成浓度为 1 × 104 µg a.i. mL-1 的储备液。随后对该溶液进行梯度稀释，获得至少五个不同浓度。 
	注意：本研究中使用的毒死蜱浓度为 0.1、0.2、0.4、0.8、1.6、3.2、6.4 µg a.i. mL-1。</li>
</ol>
3. 试剂的准备
<ol>
	<li>获取含有食物的塑料蜂管（参见 材料列表以及新孵化的幼虫 O. excavata 来自大规模饲养项目。 
	注意：从开花前20天至整个开花期间均未使用农药；化学分析结果显示，随机选取的五十份样品中常见农药含量均低于最低检测限。</li>
	<li>用软毛刷轻轻将食物团与幼虫分开。根据巢内食物团的大小及巢室位置选择雌性幼虫9然后，将大小均匀的饲料和选中的雌性幼虫放入培养皿（直径60 mm）中，备用。 
	注：随机选取五十份样品，分析其中常用农药氯吡啶、吡虫啉、芬普尼、辛硫磷、阿维菌素的含量。软毛刷参数为：（a）刷毛直径：0.3 mm，（b）刷毛长度：2 cm，（c）笔身长度：18 cm。</li>
</ol>
4. 施用农药处理
<ol>
	<li>使用笼子将选定的大小均匀的培养基（来自步骤 3.2）浸入稀释后的杀虫剂溶液（来自步骤 2.1；毒死蜱浓度分别为 0.1、0.2、0.4、0.8、1.6、3.2、6.4 µg a.i. mL-1）中 10 秒。对照组（CK）则浸入 0.2% 溶剂（本研究中为丙酮）中。 
	注意：每个浓度处理设三个重复，每个重复包含 60 个培养基。通过选择大小均匀的培养基可减少每个培养基间剂量的差异。</li>
	<li>在用杀虫剂处理培养基前后，测量杀虫剂溶液的体积。然后计算每次处理中昆虫接触的药剂浸入量，包括 60 个质量培养基（补充表 1）。将培养基置于无菌工作台晾干后，分别放入带有小孔的离心管（来自步骤 1.1）中。 
	注意：实验前，先将装有培养基的笼子浸入杀虫剂溶液中，随后测量浸泡前后的溶液体积，以消除误差。</li>
	<li>用软毛刷将雌性幼虫逐个转移至自然干燥的培养基表面。 
	注意：每根试管中放入一条幼虫。</li>
</ol>
5. 培养条件
<ol>
	<li>在黑暗条件下，于25 ± 2 °C、相对湿度65%–75%的培养室中饲养O. excavata幼虫16。</li>
</ol>
6. 结果的分析
<ol>
	<li>急性致死毒性试验
	<ol>
		<li>将幼虫置于处理组和对照组（CK）培养基上饲养48小时后，测定幼虫的死亡率。 
		注：死亡判定标准为：在黑光灯下用软毛刷轻触幼虫时，若其无任何反应，则判定为死亡22。使用黑光灯是为了模拟幼虫的黑暗生长环境，并避免检查生长指标时受光照影响。为消除人为误差，同时测定对照组在48小时后是否将幼虫从培养基上移除情况下的死亡率。</li>
		<li>在昆虫饲养试验开始前和48小时后分别称量60份培养基的重量，以确定每条幼虫所消耗的培养基量。</li>
		<li>根据每条幼虫摄食的培养基比例及各培养基中的农药含量，计算每条幼虫在各浓度下摄入的农药剂量。 
		注：剂量计算公式为23：

	<li>亚致死毒性试验
	<ol>
		<li>在饲养试验开始前和处理14天后分别称量幼虫体重，以测定幼虫体重增长量。</li>
		<li>在黑光灯下每日观察 O. excavata 结茧过程，记录幼虫发育历期。</li>
		<li>在处理组和对照组培养基上饲养14天后，称量剩余培养基重量，用于计算摄食量及食物转化效率（ECI）24。</li>
		<li>当对照组蜜蜂羽化为成虫时，使用小剪刀破茧检查，统计羽化个体数量。</li>

传粉者在现代全球农业的生态系统服务中发挥着关键作用。虽然蜜蜂（Apis mellifera；膜翅目：蜜蜂科）传统上被认为是农作物最重要的经济传粉者，但近期研究表明，切叶蜂属（Osmia；膜翅目：切叶蜂科）在提高某些作物的授粉效果、增加果实大小和种子数量、以及降低世界各地商业果园中畸形果实比例方面也具有重要作用1。Osmia excavata 被认为是苹果授粉的理想物种，主要应用于亚洲地区，如中国西北和华北地区以及日本2,3,4。该物种可为某些作物提供与蜜蜂相似甚至更高效的授粉服务。在这方面，研究已表明其可替代蜜蜂，或与蜜蜂协同发挥作用4,5,6。
O. excavata 的生物学特性与社会性蜜蜂相比具有独特性。其一年一代、独居且筑巢活动主要发生在春季和初夏。在自然条件下，O. excavata 的巢穴通常位于预先存在的孔洞中，常见于枯木、中空植物、秸秆管和竹茎内3。成年 O. excavata 从茧中羽化后进行交配、采集花粉并筑巢产卵，卵约一周后开始孵化。受精卵发育为雌性，未受精卵则发育为雄性3。雌性个体多分布于蜂管底部，对应的营养供给量也较多；而雄性则靠近管口，营养供给相对较少7，因此雄性先羽化，雌性后羽化。雌蜂将花粉与少量花蜜混合成湿润的团状物，作为每个巢室内幼虫唯一的食物来源8。
多项研究报道了传粉昆虫种群数量的下降9,10。农药的广泛使用已被确定为导致传粉者丰度和多样性减少的主要因素之一，并可能威胁传粉服务11,12。为了减少并缓解农药的不利影响，有必要对农药进行传粉者风险评估。一些国家已建立监管框架，以确保所用农药对蜜蜂的安全性13,14。最近的研究表明，Osmia 属蜂类对农药的敏感性高于蜜蜂1,15。
有趣的是，大多数风险评估都集中在成年蜜蜂上11,12；针对O. excavata尤其是其幼虫的研究较少。此外，农药对Osmia的直接致死效应通常受到关注16。然而，慢性毒性（如幼虫体重增长、发育周期、取食模式、羽化能力、成虫后续行为及繁殖力）可能与急性致死毒性具有同等危害，但由于缺乏适用于独居蜂的有效实验方法，这些方面常常被忽视17。
目前，评估农药对独居蜂幼虫影响的方法主要有两种：（1）将适量农药施加于蜂粮的局部位置，而不移除独居蜂的卵1,18,19,20；（2）用含有特定剂量农药的人工花粉-花蜜混合物替代天然蜂粮21。然而，上述两种方法均存在一定局限性。前一种方法只能测定急性毒性，无法评估慢性毒性，因为幼虫在短时间内摄入了全部剂量；后一种方法由于人为操作干扰，可能导致幼虫死亡率显著升高1。本文介绍了一种浸渍法，通过模拟在真实环境中幼虫取食蜂粮中残留农药的行为，在高度可控的实验条件下研究农药对O. excavata的生态毒性。本研究所述方法克服了上述两种方法的缺点，适用于评估有害物质对独居蜂的急性与慢性毒性影响。

评估杀虫剂对独居蜂幼虫的影响

Evaluating the Effect of Pesticides on the Larvae of the Solitary Bees

目前对农药在传粉昆虫上的生态风险评估主要仅考虑实验室条件。对于独居蜂的幼虫而言，摄食被农药污染的食物可能会增加幼虫死亡率，并从种群动态的角度降低来年成年独居蜂的采集率和种群数量。然而，关于农药对独居蜂幼虫影响的研究仍十分有限。因此，理解农药如何影响独居蜂幼虫应被视为农药生态风险评估的重要组成部分。本研究提出了一种使独居蜂Osmia excavata幼虫暴露于致死或亚致死剂量农药的方法，用于监测幼虫体重增长、发育时长、羽化能力以及摄食效率转化率。为验证该方法的有效性，将O. excavata幼虫饲喂含有毒死蜱急性致死剂量和亚致死剂量的食物，随后检测处理组幼虫的上述指标。该技术有助于预测并减轻农药对传粉昆虫的风险。

对照组中常见农药（毒死蜱、吡虫啉、芬普尼、辛硫磷、阿维菌素）在饲料中的残留含量均低于定量限（0.01–0.02 mg kg-1），排除了农药残留对各处理组的影响。评估了对照组在48小时后是否移除幼虫对死亡率的影响，结果表明无显著差异（表1），说明人为误差较小。
在急性致死毒性试验（表2）中，将供试材料浸入七种不同稀释浓度的农药溶液（0.1、0.2、0.4、0.8、1.6、3.2 和 6.4 µg a.i. mL-1 毒死蜱）以及 0.2% 丙酮（作为对照组）中。采用对数-概率回归分析，根据幼虫在处理 48 小时后摄入的农药剂量（范围为 0.0001–0.005 µg a.i. mL-1）及其相应的死亡率，评估毒死蜱对O. excavata的半数致死剂量（LD50 值）。结果显示，毒死蜱对O. excavata幼虫的 LD50 值为 0.001（0.001–0.002）µg...

本方案可用于预测并减轻农药对独居蜂的不利影响。该技术的主要优势在于能够模拟幼虫取食储粮中残留农药的过程。对该方法进行可视化演示至关重要，因为与社会性蜜蜂相比，掘地切叶蜂（Osmia excavata）的生物学特性具有独特性，且目前针对O. excavata尤其是其幼虫阶段的研究十分有限。为准备饲喂管，使用电热螺旋钻在2毫升离心管盖上打一个直径为0.3毫米的孔。为配制农药溶液，将技术级农药溶解于丙酮中，配制成10毫克/毫升的母液，然后对该溶液进行梯度稀释，得到至少五个不同浓度。接下来，从大规模繁育项目中获取含有饲料和新孵化的Osmia excavata幼虫的塑料蜂管。使用软毛刷轻轻将幼虫与食物分开，根据食物团的大小和巢室内幼虫的位置挑选雌性幼虫，然后将大小均一的食物团与选好的雌性幼虫一同放入直径为60毫米的培养皿中，备用。在进行农药处理前，称量食物团的质量。为施加农药处理，首先测量农药溶液的体积。然后将选定的大小均匀的样本在稀释后的杀虫剂溶液中浸泡10秒。用杀虫剂处理样本后，再次测量杀虫剂溶液的体积。在无菌工作台上将样本风干后，将其加入预先准备好的带孔离心管中。随后，称取60只雌性幼虫，用软毛刷将幼虫转移至自然干燥的饲料表面，每管接种一只幼虫。将幼虫置于培养箱中，在黑暗条件下、25 ± 2 °C、相对湿度65%～75%的环境中饲养。进行急性致死毒性试验时，将幼虫置于处理组和对照组饲料上，48小时后测定幼虫死亡率。接着，称量昆虫饲养48小时后剩余的饲料，以确定每条幼虫消耗的饲料量，然后根据每条幼虫摄食饲料的百分比及每份饲料中的农药含量，利用此公式计算每条幼虫在各浓度下摄入的农药剂量。对于亚致死毒性试验，在幼虫结茧期间每日在黑光灯下观察O.excavata，以测定幼虫发育持续时间。处理14天后称量幼虫体重，以确定幼虫的体重增长情况。接下来，称量饲喂14天后剩余的饲料，以计算食物消耗量和食物转化效率。当对照组蜜蜂羽化为成虫时，使用小剪刀剪开茧，检查羽化数量。急性致死毒性试验显示，毒死蜱对O.excavata幼虫的LD50值为每只蜜蜂0.001微克。在亚致死毒性试验中，随着剂量增加，处理组幼虫的体重增长、摄食量及食物转化效率的指数值均下降，其中每只蜜蜂摄入0.013微克毒死蜱时，这些指标的数值相对于对照组最低。相反，每只蜜蜂摄入0.016微克毒死蜱时，幼虫发育历期最长，与对照组相比差异明显。对毒死蜱对O.excavata羽化率影响的评估显示，摄入的毒死蜱剂量与O.excavata的羽化率之间存在显著的负线性关系。当每只蜜蜂摄入的剂量超过0.02微克时，羽化率明显低于对照处理组。在本实验方案中，筛选大小一致的储备食物至关重要，以最大限度地减少实验误差。该技术有助于探究杀虫剂对独居蜂成虫飞行能力及繁殖力的影响，从而解决回收率低和种群数量下降的问题。
