所有动物实验均经上海中医药大学动物福利委员会批准。选用4周龄雄性BALB/c裸鼠，在进行骨肉瘤细胞原位注射手术前适应性饲养一周。小鼠饲养于独立通风&#160;每笼五只小鼠，饲养于12小时光照/12小时黑暗循环环境中，自由摄取SPF级饲料和无菌水。
1. 细胞的制备
<ol>
	<li>在注射成骨肉瘤细胞（143B-Luciferase）当天，用 PBS（pH 7.4）将培养于 10 cm 细胞培养皿中、融合度达 80%–90% 的细胞洗涤两次，加入 1.5 mL 0.25% 胰蛋白酶消化 3 分钟。随后加入 6 mL 含 10% 血清的 MEM 培养基以终止胰蛋白酶消化反应，并将细胞收集至 15 mL 离心管中。 
	注：143B-Luciferase 细胞系是通过将 pLV-luciferase 载体转染至 143B 细胞系获得23。</li>
	<li>吸取 20 µL 细胞悬液加入细胞计数板的计数室中，使用自动细胞计数仪（见材料表）计算细胞浓度。</li>
	<li>在室温下以 800 × g 离心 5 分钟。</li>
	<li>用移液器吸除上清液，将细胞沉淀重悬于 8.5 mg/mL 基底膜基质（见材料表）中，调整终浓度为 2 × 107 个细胞/mL。</li>
	<li>将细胞置于冰上并转移至手术室，细胞需在 2 小时内使用。 
	注：为避免注射剂量不准确（例如因注射器死腔所致），需额外准备细胞悬液（通常为所需体积的两倍）。基底膜基质需始终置于冰上，因其在室温以上具有凝固特性24。</li>
</ol>
2. 骨肉瘤细胞原位注射的手术操作
注意：手术工具如图1所示。
<ol>
	<li>小鼠在特定无病原体条件下饲养。所有操作均在无菌操作台中使用无菌器械进行。</li>
	<li>通过暴露于2%异氟烷和98%氧气（氧气流速为2 L/min）对小鼠进行麻醉。</li>
	<li>在小鼠眼睛上涂抹少量眼用软膏，以防止麻醉期间眼睛干燥。 
	注意：整个操作应在通风良好的环境中进行。在注射骨肉瘤细胞前，通过脚趾夹捏法确认每只小鼠已进入深度麻醉状态；若小鼠仍有抽搐或抖动等反应，需继续等待直至上述反应完全消失。</li>
	<li>将每只小鼠保持仰卧位。用拇指和食指夹住小鼠踝部，并用70%乙醇棉片对胫骨注射部位进行消毒。 
	注意：为牢固固定小鼠踝关节，拇指和食指的指尖在后续操作中至关重要。</li>
	<li>将每只小鼠的踝关节向外旋转，以移动胫骨和腓骨，并将膝关节弯曲至合适位置，直至通过皮肤清晰可见近端胫骨平台（胫骨顶部）（图2A）。</li>
	<li>将针头连接至1 mL注射器，并使针尖指向注射部位。确保注射器针头与胫骨长轴平行。
	<ol>
		<li>经皮将针头穿过或邻近髌韧带刺入皮肤/关节囊；然后旋转注射器（1/2至3/4圈），在胫骨平台上钻孔，朝向胫骨远端（骨髓腔），以便使用微量注射器注射骨肉瘤细胞（图2B、C）。 
		注意：若针尖位置准确，在钻孔过程中可同时感受到胫骨的旋转。确保针头随注射器旋转而前进，而非直接向前推挤，直至约一半针头进入胫骨。</li>
	</ol>
	</li>
	<li>检查注射器针头是否明显进入骨髓腔，以确认钻孔成功。 
	注意：进行X射线检查（见材料表）以确认针头位置正确，并采集图像。</li>
	<li>将143B骨肉瘤细胞悬液（来自步骤1.5）吸入微量注射器中，并替换胫骨处原有的1 mL注射器，换为装载143B细胞的微量注射器（图2D）。缓慢向每只无胸腺小鼠的胫骨内注射约10 µL（忽略针头内已存在的液体）143B细胞悬液（约2 × 105个细胞），避免施加高压。</li>
	<li>当取出微量注射器后，用棉签按压注射部位20–30秒。</li>
	<li>将每只小鼠放回清洁笼具中，并密切观察直至其完全从麻醉中恢复（约10分钟）。</li>
	<li>使用X射线成像系统在活体（in vivo）监测肿瘤生长情况。每周使用游标卡尺测量肿瘤长径（a）和短径（b），并计算肿瘤体积（V）：V = 1/2 × a × b2。 
	&#8203;注意：通过暴露于2%异氟烷和98%氧气对小鼠进行麻醉。小鼠在X射线成像过程中处于麻醉状态。向胫骨内注射表达荧光素酶或荧光蛋白标记的骨肉瘤细胞，可实现对原发性和转移性骨肉瘤病灶的追踪。 
	注意：因膝关节肿瘤生长及肺转移导致的骨肉瘤小鼠人道终点判定标准如下：（1）体况评分；（2）体重减轻超过20%；（3）肿瘤平均最大直径达2 cm；或（4）动物行为严重受限。</li>
</ol>
3. 病理学检查（采集原发性和肺转移性骨肉瘤标本用于分析）
<ol>
	<li>在骨肉瘤细胞注射后六周，将小鼠暴露于 CO2 吸入后，通过颈椎脱位处死小鼠。</li>
	<li>将小鼠保持仰卧位，并拉伸双侧后肢。</li>
	<li>从腹股沟区域分离带有骨肉瘤的整个后肢。 
	注意：确保所有后肢均从相同的解剖位置分离。</li>
	<li>通过去除皮肤、肌肉和足部，制备带有骨肉瘤的后肢组织学标本，然后将每只小鼠的标本置于含 20 mL 甲醛溶液（10%）的 50 mL 管中固定 24 小时，随后在 10% EDTA 溶液中脱钙 14 天，期间定期更换缓冲液。</li>
	<li>将标本包埋于石蜡中，并按照先前发表的研究方法25制备切片用于组织学检查。</li>
	<li>轻柔分离肺脏，并将其放入盛有 20 mL 甲醛溶液（10%）的 50 mL 管中。24 小时后，将每只小鼠的肺脏转移至含 70% 乙醇的 15 mL 管中。将肺组织包埋于石蜡中，用于苏木精和伊红（H&amp;#38;E）染色及免疫组织化学检测25。</li>

骨肉瘤是儿童和青少年中最常见的原发性骨癌1,2，其主要侵犯周围组织，甚至在患者确诊时已发生肺转移。肺转移是骨肉瘤治疗面临的主要挑战，伴有肺转移的骨肉瘤患者五年生存率仍低至20%–30%3,4,5。然而，自20世纪70年代引入化疗以来，原发性骨肉瘤的五年生存率已提高至约70%6。因此，迫切需要深入了解骨肉瘤发生及肺转移的基本机制，以开发新的治疗方法。建立能够最好地模拟人类骨肉瘤进展的小鼠模型具有重要意义7。
骨肉瘤动物模型可通过自发形成、诱导型基因工程、移植及其他技术方法构建。由于肿瘤形成时间长、成瘤率不一致、发病率低且稳定性差，自发性骨肉瘤模型很少被使用8,9。尽管诱导型骨肉瘤模型比自发性模型更容易获得，但其应用仍受到限制，因为诱导因素会影响骨肉瘤的微环境、发病机制及病理特征10。转基因模型有助于理解肿瘤的发病机制，因其能更好地模拟人体生理和病理环境；然而，转基因动物模型也存在局限性，主要体现在转基因修饰操作难度大、耗时长且成本高。此外，即使是在通过p53和Rb基因修饰构建的、目前最广泛认可的转基因动物模型中，仅有13.6%的肉瘤发生于四肢骨11,12。
近年来，移植法因其操作简便、成瘤率稳定且均一性较好，已成为最常用的原发性和远端转移性癌症模型构建方法之一13。根据移植部位的不同，移植法可分为异位移植和原位移植。在骨肉瘤异位移植中，骨肉瘤细胞被注射到动物原发灶（骨骼）以外的部位，通常为皮下14。尽管异位移植操作直接，无需对动物进行手术，但注射部位并不能代表人类骨肉瘤的真实微环境。骨肉瘤原位移植则是将骨肉瘤细胞接种至动物骨骼内，例如胫骨15,16。与异位移植相比，原位移植的骨肉瘤移植物具有潜伏期短、生长迅速和侵袭性强等特点，因此是开展骨肉瘤相关研究的理想动物模型17。
最常用的动物是小鼠、犬和斑马鱼18,19。骨肉瘤的自发模型通常用于犬类，因为骨肉瘤是犬类中最常见的肿瘤之一。然而，由于肿瘤形成时间长、成瘤率低、均一性和稳定性差，该模型的应用受到限制。斑马鱼因繁殖速度快，常被用于构建转基因或基因敲除肿瘤模型20。但斑马鱼的基因与人类基因存在差异，因此其应用也受到限制。
