1. 材料与设备
注意：Bacillus altitudinis LZP02 (CP075052) 在本研究中分离自中国东北水稻的根际12,13。
<ol>
	<li>在LB培养基（胰蛋白胨 10 g L-1；NaCl 8 g L-1；酵母提取物 5 g L-1）中培养 B. altitudinis LZP02 菌株 10 小时。于 4 °C 条件下，以 9,569 × g 离心 2 分钟收集菌体，并用含 15% 甘油的溶液于 -80 °C 保存。 
	注：本实验所用水稻种子（Oryza sativa Longgeng 46）由黑龙江省农业科学院水稻研究所提供。</li>
</ol>
2. 根系分泌物的收集
<ol>
	<li>将水稻种子随机放置于人工气候室中。 
	注意：水稻种子使用30% H2O2 消毒30分钟，并在水中浸泡过夜。培养条件如下：控制光照（16/8小时光暗循环）、温度（22 ± 2 °C）和相对湿度（῀70%）。</li>
	<li>培养水稻种子一周，并添加无菌水两次。</li>
	<li>选取大小相近的水稻幼苗，种植于50 mL的Murashige和Skoog（MS）液体培养基中。在22 °C下，于无菌条件下培养48小时。 
	注意：水稻根系分泌物将释放到MS培养基中14,15,16。</li>
</ol>
3. 根系分泌物的液相色谱-质谱分析
<ol>
	<li>收集 100 µL 样品（含根系分泌物的MS培养基）置于1.5 mL离心管中。加入 20 µL 加入提取溶剂（乙腈-甲醇-水，2:2:1，含内标），在45 Hz条件下均质化样品4分钟，随后在冰浴中超声处理5分钟。</li>
	<li>重复匀浆和超声处理循环三次。将样品在 -20 °C 1小时，随后在133,778 x g 和 4 °C 15分钟。</li>
	<li>将所得上清液转移至液相色谱-质谱小瓶中，并于 -80 °C 直至进行UHPLC-QE分析。通过混合所有样品上清液的等量部分制备质量控制（QC）样品。 
	注意：每个样品的体积为 600 µL （每次实验设六个重复）</li>
	<li>使用超高效液相色谱系统（UHPLC）和UPLC HSS T3色谱柱（2.1 mm × 100 mm）进行LC-MS/MS分析， 1.8 µm）和 Q Exactive12.
	<ol>
		<li>使用0.1%甲酸水溶液和5 mmol/L乙酸铵水溶液作为流动相A，乙腈作为流动相B。分别使用甲酸和乙酸铵作为正离子和负离子模式。</li>
		<li>洗脱梯度设置如下：0 分钟，1% B；1 分钟，1% B；8 分钟，99% B；10 分钟，99% B；10.1 分钟，1% B；12 分钟，1% B。流速和进样量分别设置为 0.5 mL/min 和 2 µL分别。 
		注意：大分子（&gt;1,000 道尔顿）无法被检测到。</li>
	</ol>

</ol>
4. 趋化性实验
<ol>
	<li>制备趋化剂水溶液，确保其无菌。使用0.22 µm细菌滤器过滤趋化剂溶液。 
	注：趋化剂水溶液为通过液相色谱-质谱（LC-MS）研究获得的单一物质溶于水所得。浓度和体积可根据不同实验适当调整。此处以柠檬酸溶液为例。所有操作均需在灯旁进行。</li>
	<li>在玻璃载片上以1 cm间隔标记中心位置。确保玻璃载片已在火焰上多次灭菌。</li>
	<li>在玻璃载片左侧加入30 µL趋化剂溶液。确保细菌已在LB培养基中培养至对数生长期（2 × 108 CFU/mL）。在玻璃载片右侧加入30 µL菌液。 
	注：设置阴性对照组，加入等体积无菌水。使用生理盐水（0.9% NaCl）作为阳性对照，以排除分子间作用力对实验结果的影响。</li>
	<li>将接种环在火焰上多次灭菌。用接种环连接趋化剂水溶液与菌液，并在洁净台中室温静置20分钟。 
	注：实验必须在无风环境中进行。根据不同菌株的运动能力，可适当调整断开连接线前的静置时间。</li>
	<li>20分钟后，用滤纸断开连接线。</li>
	<li>确保1.5 mL离心管无菌。收集玻璃载片左侧的趋化剂水溶液，并转移至1.5 mL无菌离心管中。</li>
	<li>向离心管中加入适量体积的沙黄染液。2分钟后，取混匀液体，使用血细胞计数板进行细菌计数和显微镜观察。</li>
</ol>
5. 结果分析

	<li>使用以下公式计算被吸引的可存活微生物数量：

已有多种方法用于分析细菌趋化性。泳动平板法是先前描述的方法之一9。该方法使用半固体培养基制备平板，将含琼脂（1.0%，w/v）的趋化缓冲液加入平板中，加热后与趋化吸引物混合。然后将8 µL细菌悬液滴加至平板中央，将平板置于28 °C培养箱中。定期观察并拍照记录。然而，泳动平板法的实验周期非常长。在类毛细管法10中，移液器吸头用作盛装100 µL细菌悬液的腔室，1 mL注射器针头作为毛细管。将含有不同浓度梯度趋化吸引物的注射器针头插入100 µL移液器吸头中。室温孵育3小时后，取出注射器针头，内容物经稀释后接种于培养基上。通过平板上的菌落形成单位（CFUs）表示注射器内细菌的聚集情况。然而，类毛细管法在重复实验间的实验误差较大。另一种方法采用了微流控SlipChip装置11。简言之，将牛血清白蛋白（BSA）溶液注入所有通道，并用真空泵抽除。随后分别向顶部、中部和底部微孔中加入含不同趋化吸引物的溶液（仅用于定性检测时浓度为1 mM）、磷酸盐缓冲液悬浮的细菌细胞以及磷酸盐缓冲液（阴性对照）。然后在室温避光条件下孵育30分钟，随后检测微孔中的细菌细胞。然而，微流控SlipChip装置成本较高。因此，上述每种方法均各有优缺点。
我们建立了一种改进的趋化性检测方法，利用无菌载玻片快速鉴定根系分泌物中的根际细菌趋化物，无需复杂步骤。本研究通过多次比较实验结果，证明该方法克服了传统细菌趋化性实验方法的多种缺陷。该方法降低了平板计数的误差，并缩短了实验周期。因此，若用于鉴定趋化性物质，此新方法可节省2-3天时间，并降低实验材料成本。

An Improved Chemotaxis Assay for the Rapid Identification of Rhizobacterial Chemoattractants in Root Exudates

趋化性鉴定对于根际促生细菌的研究与应用具有重要意义。我们建立了一种简便的方法，通过简单的步骤在无菌载玻片上快速鉴定能够诱导根际促生细菌趋化运动的化学引诱物。via 将细菌溶液（OD600 = 0.5）和无菌化学引诱物水溶液分别以相隔1 cm的距离滴加到载玻片上，再用接种环将化学引诱物水溶液与细菌溶液连接。将载玻片置于洁净工作台中室温静置20分钟，最后收集化学引诱物水溶液进行细菌计数和显微观察。本研究通过多次实验结果的比较，表明该方法克服了传统细菌趋化性实验方法的多种不足，减少了平板计数的误差，缩短了实验周期。在化学引诱物物质的鉴定方面，与传统方法相比，该新方法可节省2-3天时间。此外，该方法使任何研究人员均可在1-2天内系统完成一次细菌趋化性实验。该实验方案可作为研究植物-微生物互作的重要工具。

在正离子模式和负离子模式下分别检测到584种和937种已知代谢物。既往研究表明，趋化因子通常是有机酸、氨基酸和碳水化合物17,18。
本研究从水稻根际分泌物的液相色谱-质谱（LC-MS）分析中筛选出16种化学引诱物用于后续实验（表1）。采用泳动平板法，筛选水稻根部分泌的低分子量化合物中对根际B. altitudinis LZP02具有趋化吸引作用的物质。为比较不同化学引诱物的趋化能力，引入趋化指数（RCI），即处理组细菌数量与相应对照组的比值19。RCI值越高，表明对引诱物的趋化反应越强。结果表明，100 µM柠檬酸（RCI = 2.34）和100 µM咖啡酸（RCI = 1.85）的趋化指数最高（图2）。代表性图像显示，与对照组（CK）相比，柠檬酸和咖啡酸周围的菌群聚集区域更大（<strong class=...

该论文报道了利用改进的趋化性检测方法鉴定根际细菌在根系分泌物中的化学引诱物。根际细菌的趋化性对根系定殖、促进植物生长以及介导有益的植物-微生物相互作用具有重要意义。目前已采用多种方法分析细菌趋化性。例如，泳动平板法、毛细管样方法、微流控滑动芯片装置。泳动平板法的实验周期较长，毛细管样方法的实验误差较大，微流控滑动芯片装置的实验成本较高。我们建立了一种简便的方法，利用无菌玻璃载玻片快速鉴定能够诱导根际促生细菌趋化运动的趋化物质，且无需复杂步骤。该方法降低了平板计数的误差，并缩短了实验周期。在鉴定趋化物质方面，新方法可节省2至3天，同时减少实验材料的成本。将水稻种子随机分布在培养室的培养基中，培养一周，并添加无菌水两次。选取大小相似的水稻幼苗，种植于50毫升MS液体培养基中。在22摄氏度及无菌条件下培养48小时。水稻根系分泌物将释放到Ms培养基中。使用超高效液相色谱系统进行液相色谱-质谱分析。配制趋化物水溶液，并确保其无菌。使用0.22微米的细菌滤器过滤趋化剂溶液。趋化剂水溶液是将LCMS研究中获得的单一物质溶解于水中制备而成。以柠檬酸溶液为例。所有操作均须在灯旁进行。以1厘米为间隔，在载玻片上标记中点位置。确保载玻片在火焰上多次灭菌。在载玻片左侧加入30微升趋化引诱剂溶液，确保细菌已培养至对数生长期。在载玻片右侧加入30微升细菌溶液。将接种环在火焰上反复灼烧灭菌数次，用该接种环将趋化引诱剂水溶液与细菌溶液连接，然后在洁净工作台上室温静置20分钟。20 分钟后，断开带有滤纸的连接管。确保 1.5 毫升离心管无菌。收集玻璃片左侧的趋化引诱物水溶液。将溶液转移至1.5毫升无菌离心管中。向离心管中加入适量体积的细胞前体染料。2分钟后，用血细胞计数板收集混悬液，用于细菌计数和显微镜观察。使用以下公式计算被吸引的可存活微生物数量。RCI 值越高，表明对趋化物的反应越强。结果表明，柠檬酸和咖啡酸的趋化指数最高。新的趋化性检测方法结果显示，细菌在柠檬酸、咖啡酸和半乳糖实验中的浓度显著高于对照组和阳性对照组。其中柠檬酸引起的趋化作用最强。实验结果与传统方法的结果一致。本视频方案可作为阐明植物-微生物相互作用的宝贵资源。
