Culturing and Genetically Manipulating Entomopathogenic Nematodes

为了评估经过无菌化处理的 H. bacteriophora 线虫的状态，需确定其感染性幼虫（IJs）中是否存在 P. luminescens 细菌菌落。为此，收集约500条经表面消毒并用PBS缓冲液匀浆处理的IJs，制成沉淀。阳性对照组为含有共生 P. luminescens 细菌的线虫培养物中约500条IJs的沉淀。将无菌化处理组和阳性对照组的线虫沉淀在1×PBS中进行匀浆处理，随后将匀浆液用移液器接种至琼脂平板上，并涂布均匀以促进单个菌落的生长。琼脂平板在28 °C条件下培养24小时。次日观察 P. luminescens 菌落（CFU）的出现情况。结果如预期所示，来自保种培养物的 H. bacteriophora 携带共生的 P. luminescens 细胞；而缺乏相关 P. luminescens 细菌的线虫则为无菌状态，并被单独保存以供后续实验使用（图6）。

维持和扩增昆虫病原性线虫的高效方案对于理解线虫致病性的分子基础以及宿主抗线虫免疫机制至关重要。本方案可利用昆虫宿主大蜡螟（Galleria mellonella）大量生产共生状态及无菌状态的小杆线虫（Heterorhabditis bacteriophora）和斯氏线虫（Steinernema carpocapsae）感染性幼虫。首先，在培养皿中铺上一张滤纸，并加入约10至15只大蜡螟幼虫。使用移液器，将含有约每 10 微升悬浮液 25 至 50 条感染性幼虫的 2 毫升水溶液滴加到蜡虫上。将培养皿置于室温柜中保存。根据滤纸的湿润程度，每隔两天添加 1 至 2 毫升水。感染侵染性幼虫的蜡虫通常在48小时内死亡。在蜡虫感染侵染性幼虫约10天后，制备White氏水诱捕器。小心将死亡的昆虫转移到滤纸未浸湿的部分。使用自来水填充底部的培养皿，并将一个15毫升离心管的管盖倒置作为通风隔片。应使用自来水而非去矿物质水或去离子水，以避免线虫聚集。确保水陷阱中的水位达到培养皿高度的约一半。当小培养皿中的水因线虫而变得浑浊时，使用移液器将新一代的感染性幼虫转移至T25或T75细胞培养瓶中。随后加入自来水，直至总体积达到40%或达到适宜密度。将细胞培养瓶水平放置保存，以避免线虫聚集。重复将感染性幼虫转移至细胞培养瓶并加水，直至大约三到五天后昆虫尸体中不再有感染性幼虫孵出为止。使用接种环从 Photorhabdus temperata Ret16 的冷冻培养物中刮取数小块，接种于麦康凯平板上，并划线分离单菌落。将平板在 28 摄氏度下培养 2 至 3 天。培养结束后，用麦康凯琼脂平板上的单菌落接种 10 毫升 LB 液体培养基，置于 50 毫升试管中，在 28 摄氏度摇床培养箱中过夜培养。孵育后，从50毫升离心管中转移100微升过夜培养物至新的微量离心管中，然后在1.5毫升微量离心管中，用900微升单倍浓度的PBS通过离心（17,900 × g）洗涤过夜培养物。弃去上清液。接下来，用单倍浓度的PBS将菌液稀释10倍，并将离心管置于冰上。现在，将蜡虫浸入70%乙醇溶液中。用纸巾将昆虫干燥后，放入一个50毫升的离心管中。将离心管置于冰上20分钟，以固定蜡虫。使用滤纸覆盖培养皿的顶部和底部。将覆盖有滤纸的培养皿盖用作注射时的底部支撑。润湿培养皿底部的滤纸，并将其置于冰上，以帮助注射后的蜡虫恢复。用移液器吸取50微升冰上预冷的细菌，置于一块Parafilm上，然后准备一支22号精制针头注射器，轻轻推动活塞以排出针尖处的空气。在体视显微镜下，将蜡虫靠近其后端固定，将细菌培养物注射到胸部的背侧。最好在角质层下方、与蜡虫平行的两个节段交界处进行注射，以尽量减少内部损伤，并注意观察幼虫身体开始膨胀的情况。接下来，将注射后的昆虫转移至恢复用培养皿中。待所有昆虫注射完毕并放入恢复皿后，将培养皿置于黑暗环境中。蜡虫在注射后两天内死亡，大约三到四天后呈现砖红色。然而，呈棕色的昆虫表明细菌感染未成功。感染后七天，将具有典型砖红色的昆虫转移至铺有新鲜滤纸的培养皿中，并重复共生线虫感染性幼虫的生产。开始表面灭菌时，通过离心收集足量的共生型异小杆线虫（Heterorhabditis bacteriophora）及候选无菌感染期幼虫。在1.5毫升离心管中形成沉淀后，向每根微量离心管中的沉淀加入500微升5%次氯酸钠溶液，并颠倒混匀试管。在室温下孵育10分钟后，于17,900 × g离心1分钟，弃去上清液。然后用一毫升无菌水洗涤沉淀物。离心后弃去上清液，并重复此步骤四次。该图显示了经过无菌化处理的嗜菌异小杆线虫（Heterorhabditis bacteriophora）状态评估结果。库存培养的嗜菌异小杆线虫携带共生的发光杆菌细胞。然而，不带有共生发光杆菌的线虫为无菌状态。为了保证实验的可重复性，应始终从细菌库存培养物的新鲜菌落上划线接种，并在细菌培养物进入稳定期之前取出过夜培养物。仅使用砖红色的蜂蜡虫（Galleria wax worm）进行怀特氏水分陷阱实验，其余颜色的应弃去。经过该步骤后，线虫可用于研究其与昆虫免疫系统之间的相互作用。该技术为研究人员探索线虫寄生的分子机制以及宿主抗线虫免疫反应奠定了基础。
