1. 在lrECM中接种MCF10A细胞
<ol>
	<li>在培养基中培养 MCF10A 细胞（贴壁型乳腺上皮细胞），每 4 天传代一次。 
	注意：培养基成分：高糖 DMEM（不含丙酮酸钠），添加马血清（5%）、胰岛素（10 µg/mL）、氢化可的松（0.5 µg/mL）、表皮生长因子 EGF（20 ng/mL）、霍乱毒素（100 ng/mL）以及青霉素-链霉素（100 单位/mL）（参见 材料表）。</li>
	<li>实验开始前 20 分钟将 lrECM（参见 材料表）置于冰上解冻。通常将细胞接种当天记为第 0 天。</li>
	<li>消化细胞时，吸除培养基，用 2 mL PBS 洗涤一次，加入 900 µL 0.05% 胰蛋白酶-EDTA，于 37 °C 孵育约 15 分钟，或直至细胞完全消化脱落。</li>
	<li>在八孔腔载玻片（参见 材料表）的八个孔中制备 lrECM 基质层。
	<ol>
		<li>在细胞消化过程中，每孔加入 60 µL lrECM 进行包被，并置于 37 °C CO2 培养箱中孵育，时间不超过 15 分钟。</li>
	</ol>
	</li>
	<li>按以下步骤制备细胞悬液。
	<ol>
		<li>待细胞完全脱落之后，加入 5 mL 重悬培养基以终止胰蛋白酶活性，于 25 °C、112 × g 离心 10 分钟。 
		注意：重悬培养基成分：高糖 DMEM（不含丙酮酸钠），添加马血清（20%）和青霉素-链霉素（100 单位/mL）。</li>
		<li>吸除上清液，将细胞重悬于 2 mL 检测培养基中，充分混匀以确保形成单细胞悬液。 
		注意：检测培养基成分：高糖 DMEM（不含丙酮酸钠），添加马血清（2%）、氢化可的松（0.5 µg/mL）、霍乱毒素（100 ng/mL）、胰岛素（10 µg/mL）以及青霉素-链霉素（100 单位/mL）。</li>
		<li>使用血细胞计数板对细胞计数，并计算每孔接种 6 × 103 个细胞所需的细胞悬液体积。 
		注意：通常建议在计算时额外多计一孔，以补偿可能的移液误差。</li>
		<li>根据所需孔数，用覆盖培养基稀释细胞悬液。 
		注意：单孔覆盖培养基的组成为：400 µL 检测培养基、8 µL lrECM（终浓度 2%）和 0.02 µL 100 µg/mL EGF（终浓度 5 ng/mL）。</li>
	</ol>
	</li>
	<li>进行细胞接种。
	<ol>
		<li>将 400 µL 稀释后的细胞悬液加入已制备好的 lrECM 基质层（步骤 1.4）中，注意避免扰动 lrECM 层。置于 37 °C、5% CO2 恒湿培养箱中孵育。</li>

2. PAF 处理
<ol>
	<li>在细胞接种后3小时加入PAF。用PBS配制100 µM的PAF储备液，每孔加入0.2 µL（相当于终浓度为200 nM）。</li>
	<li>每次更换培养基时均加入相同浓度的PAF。</li>
</ol>
3. 用新鲜培养基重新喂养
<ol>
	<li>每4天更换一次新鲜培养基（即第4天、第8天、第12天和第16天）。</li>
</ol>
4. 免疫荧光研究以检测长期PAF暴露诱导的表型变化
<ol>
	<li>培养 20 天后，小心地用移液器吸出每孔中的培养基，并用 400 µL 预热的 PBS 洗涤各孔。</li>
	<li>通过向每孔加入 400 µL 4% 多聚甲醛（用 1× PBS 稀释 16% 多聚甲醛新鲜配制）并在室温下孵育 20 分钟，固定腺泡结构。</li>
	<li>用冰冷的 PBS 洗涤孔板一次，然后在 4 °C 下用含 0.5% Triton X-100 的 PBS 透化处理 10 分钟。</li>
	<li>10 分钟后，立即但小心地吸出 Triton X-100 溶液，并用 400 µL PBS-甘氨酸（在 1× PBS 中加入少量甘氨酸新鲜配制）冲洗。重复此步骤三次，每次 15 分钟。</li>
	<li>加入 400 µL 主要封闭液（含 10% 山羊血清（见材料表）的免疫荧光（IF）缓冲液），在室温下孵育 60 分钟。 
	注：IF 缓冲液的组成：0.05% 叠氮化钠、0.1% BSA、0.2% Triton X-100 和 0.05% Tween-20（溶于 1× PBS）。</li>
	<li>移除主要封闭液，加入 200 µL 用主要封闭液配制的 2% 二抗封闭液（山羊抗小鼠抗原的 F(ab')2 抗体片段，见材料表），在室温下孵育 45–60 分钟。</li>
	<li>将一抗（见材料表）用 2% 二抗封闭液按 1:100 稀释。吸除二抗封闭液后，加入新鲜配制的一抗溶液，在 4 °C 下过夜孵育。</li>
	<li>上一步可能导致基底膜液化。在继续实验前，需等待载玻片恢复至室温。小心吸出一抗溶液，并用 400 µL IF 缓冲液洗涤三次。</li>
	<li>在最后一次 IF 缓冲液洗涤过程中，将荧光标记的二抗（见材料表）用主要封闭液按 1:200 稀释。将载玻片置于二抗溶液中，在室温下孵育 40–60 分钟。</li>
	<li>用 400 µL IF 缓冲液冲洗载玻片 20 分钟，随后用 PBS 洗涤两次，每次 10 分钟。</li>
	<li>在室温下用含 0.5 ng/mL 核染料（见材料表）的 PBS 对细胞核进行复染 5–6 分钟。用 400 µL PBS 洗涤三次，以去除多余的染料。</li>
	<li>小心吸净全部 PBS，确保彻底去除残留液体。向每孔中加入一滴封片试剂（见材料表），并在室温下静置过夜。</li>
	<li>将载玻片在室温下保存，并尽快进行成像。</li>
	<li>在共聚焦显微镜（见材料表）下以 40× 或 63× 放大倍数进行成像，设置光学 Z 轴步长为 0.6 mm（NA = 1.4）。</li>
	<li>使用图像处理软件（见材料表）打开获取的图像，并通过三维投影展示形态学差异。 
	注：最中心的光学 Z 切片最适合展示极性标志物定位的差异。可通过手动定量分析，表示呈现特定染色模式的球状体所占百分比。</li>
	<li>为说明蛋白表达水平的差异，可进行半定量分析，测量平均灰度值或计算校正总细胞荧光强度（CTCF）23。数据以小提琴图或点图形式呈现。</li>
