体型被认为是一种与新陈代谢、生长、呼吸和运动等其他重要性状相关的元性状4。此外，体型可以决定生物在食物网中的营养级位置及其相互作用5。在一个群落中，个体体型与按体型分级归一化的生物量（或丰度）之间的关系被定义为尺度谱6，其普遍模式表现为：在对数尺度上，随着个体体型的增加，归一化生物量呈线性下降7。这一线性关系的斜率已在理论上得到广泛研究，且跨生态系统的实证研究已将其用作群落体型结构的生态学指标4。另一个在生物多样性-生态系统功能研究中成功应用的群落体型结构综合指标是群落体型多样性，它以尺度谱中各体型类别的香农指数或其类似指标来表示，后者基于个体体型分布进行计算8。
在淡水生态系统中，不同动物类群的体型结构被用作一种非分类学指标，以评估生物群落对环境梯度9,10,11以及人为干扰12,13,14,15,16的响应。大型无脊椎动物也不例外，其体型结构同样会响应环境变化17,18以及采矿19、土地利用20或&#160;氮（N）和磷（P）富集20,21,22等人为干扰。然而，测量数百个个体以描述群落的体型结构是一项繁琐且耗时的工作，由于时间有限，实验室通常避免将其作为常规测量项目。因此，已开发出多种半自动或全自动成像方法用于分类和测量标本23,24,25,26。但这些方法大多侧重于分类学鉴定，而非生物个体的体型测量，且尚不能适用于所有类型的大型无脊椎动物。在海洋浮游生态学中，扫描图像分析系统已被广泛用于确定浮游动物群落的体型大小和分类组成27,28,29,30,31。该仪器在全球多个海洋研究机构中均有配备，可用于扫描保存的浮游动物样品，获得整个样品的高分辨率数字图像。本方案对这一仪器的使用进行了改进，使其能够快速、自动地估算河流中大型无脊椎动物群落的体型谱，而无需投入成本研发新设备。
该方案包括对样品进行扫描，并对整幅图像进行处理，以自动获取样品中各个对象的单个图像（即小图）。通过多种形态、大小和灰度特征参数来表征每个对象，并实现对象的自动分类，分类结果随后由专家进行验证。每个生物个体的大小通过椭球形生物体积（mm3）计算得出，该体积基于以像素为单位测量的生物体面积推算而来。这使得能够快速获得样品的粒径谱。据我们所知，该扫描成像系统目前仅被用于处理中型浮游动物样品，但该设备未来有望也可用于淡水底栖大型无脊椎动物的研究。
本研究的总体目标是通过改进先前用于海洋中型浮游动物的现有方案，建立一种快速获取保存状态下河流大型无脊椎动物个体大小的方法。27,32,33该步骤包括&#160;采用一种半自动方法，通过扫描设备对水样进行扫描，并利用三种开源软件处理扫描图像。本文介绍了一种经过改进的协议，用于扫描、检测和识别数字化的河流大型无脊椎动物，以自动获取群落体型结构及相关体型参数。基于从伊比利亚半岛东北部（NE）三个流域（Ter、Segre-Ebre 和 Besòs）采集的42幅河流大型无脊椎动物样本扫描图像，本文还对该方法的评估结果及提升效率的操作指南进行了阐述。
根据西班牙政府制定的可涉水河流底栖大型无脊椎动物野外采样与实验室分析方案34，在河流100米河段范围内进行样品采集。采用苏伯采样器（框架：0.3 m × 0.3 m，筛网孔径：250 µm）进行多生境调查法采集样品。在实验室中，样品经清洗后依次通过5 mm和500 µm筛网，获得两个子样品：粗颗粒子样品（5 mm筛网）和细颗粒子样品（500 µm筛网），分别储存于不同的样品瓶中，并用70%乙醇保存。将样品分为两个粒径组分有助于更准确地估算群落的个体大小结构，因为大型生物的数量通常比小型生物更稀少。否则，扫描所得样品将对大型个体组分产生偏差性表征。

Automatic Image Processing to Determine the Community Size Structure of Riverine Macroinvertebrates

在对大型无脊椎动物样本进行系统测试时，部分扫描图像质量较差。背景中出现深色过饱和区域，影响了图像的正常处理，并导致无法准确测量各个大型无脊椎动物的尺寸（图2）。背景中出现过饱和区域或图像像素化严重可能由多种原因造成：（1）扫描托盘上样本数量过多；（2）样本中含有过多杂质；（3）样本预览与正式扫描之间的时间间隔不足；或（4）在图像处理过程中所使用的背景图像质量不佳，原因可能包括冷凝、污渍或水质差33。在大型无脊椎动物群落样本中，若使用乙醇代替水，会在托盘上产生沉淀物；若未在每次扫描之间用清水充分冲洗，则会形成深色阴影。充分冲洗对于获得清晰图像以及最大限度减少扫描托盘玻璃的腐蚀至关重要。

体型大小一直被视为监测的关键性状。然而，测量个体大小十分耗时。我们的方法可实现大型无脊椎动物大小谱的常规应用，以评估对淡水生态系统的影响。本方案提供了一种标准化的方法，可在约一小时内自动测定样本中河流大型无脊椎动物的个体大小分布。由维克大学的Rosa Guri演示该操作流程。首先，打开扫描仪。然后将双位置的灯打开，以从顶部和底部投射白光。接下来，用自来水清洁并冲洗扫描托盘。为制备空白样品，将110毫升室温自来水倒入扫描托盘中，直至覆盖玻璃表面。将大号框架放置在扫描托盘上，使其角落位于左上角，并继续加入自来水，直至水位覆盖框架的台阶部分，以避免弯月面效应影响扫描图像。接下来，打开图像处理软件，选择工作项目，然后单击扫描转换背景图像。接着，打开扫描软件并单击预览。检查图像是否存在线条或斑点，并在开始扫描前至少等待30秒。然后在第二次扫描前按下说明窗口中的“确定”按钮，以将扫描软件中的数据发送至图像处理软件。将110毫升70%乙醇倒入扫描托盘中，直至覆盖玻璃表面，并放置大框架。然后将大型无脊椎动物样品倒入由边框固定的扫描托盘中，如有需要可加入更多乙醇。接着使用木针将样品在边框区域内充分混匀，并将较大的个体置于托盘中央，以利于后续图像处理。并使漂浮的生物沉降。同时将聚集的生物分散开。将接触视野边缘的生物拉至中央。接下来，点击“使用Zoo Scan扫描样本”以进行扫描，选择样本并按照说明操作，归档无需处理。在扫描软件中预览图像，确保无条纹或斑点。30 秒后，点击扫描软件中的扫描按钮，并确认原始扫描图像正确无误。取下框架，使用装有 70% 乙醇的挤压瓶在扫描托盘上方冲洗，以回收任何附着的大型无脊椎动物。抬起扫描仪的上部，使所有生物体和乙醇通过扫描回收漏斗进入烧杯中。保持扫描仪上部处于抬起状态，使用洗瓶冲洗托盘，将残留的生物体全部冲入漏斗。在回收完所有样本后，用自来水清洗托盘。要预测样本身份，请在自动识别软件中点击“数据分析”。在“选择学习文件”中，找到 PID_Process，然后选择 learning_set 以选用相应的学习集文件。接下来，在“选择样本文件”中，从 PID_results 文件夹中选择待预测的样本。在选择方法时，选择随机森林方法，然后勾选“保存每个样本的详细结果”按钮。在原始变量中，取消勾选位置变量。最后，在自定义变量中，仅勾选 ESD。单击“开始分析”，并将结果保存为 analysis_name.txt，存储在 PID_process 预测文件夹中。为进行手动验证，将 PID_process 预测文件夹中的 analysis sample_dat_1.txt 文件复制到 PID_process、PID_results 文件夹中。根据图像处理软件中的预测或验证结果，在文件夹中选择提取的片段。然后从 PID_results 文件夹中选择已使用的预测文件。使用默认设置，点击“确定”将创建一个新文件夹。现在转到 PID_process 排序后的小插图文件夹，复制新创建的以样本名称、日期和时间命名的文件夹以进行验证。将该文件夹重命名为“样本名称、日期和时间_validated”。为手动验证自动分类结果，请打开重命名后的文件夹“样本名称、日期和时间_validated”。检查每个子文件夹中的所有小图像，以识别是否存在分类错误的对象。若发现对象分类错误，将该小图像拖动至正确的文件夹。然后，选择“从已分类的小图像加载鉴定结果”。保持默认设置，选择名为“date, time, name validated”的文件以进行处理。然后进入 PID_process、PID_results，再进入 dat1_validated。打开名为“ID_from_sorted_vignettes date and time txt”的文件，确认最后一列“prediction, validated ID, date and time”（该列指定了每个对象的专家分类）已成功生成。在对大型无脊椎动物系统进行测试时，部分处理后图像的扫描质量较差。尽管如此，原始图像和处理后图像中仍存在一批扫描质量良好的优质子样本，其效果如图所示。在分析的 vignette 集合中，86.1% 对应为碎屑。包括碎屑、纤维、身体部分或扫描伪影。其余13.9%的检测对象对应于无脊椎动物生物体。自动识别后接人工验证的方法在所有类别中均表现出高召回率。尽管其他无脊椎动物的污染程度相对较低。自动识别与人工验证的大型无脊椎动物丰度比较显示高度相关，但由于碎屑污染，自动识别结果存在轻微高估。自动预测的个体大小分布概率密度函数与人工验证预测结果在细颗粒和粗颗粒子样本中高度一致。从选定的天然样品中分离出接触颗粒后进行验证的子样品，其丰度有所增加，但平均椭球体积与已验证样品相近。经校正样品的粒径分布与已验证样品相比略有差异。但观察到了显著的相关性。不同处理之间的归一化生物体积大小谱相似，仅在少数几个谱图的个别尺寸类别中存在差异。花必要的时间将相互接触的器官分开以获得精细的扫描结果，并在每次扫描后用乙醇清洗扫描托盘，防止沉淀产生。我们已将原本为浮游生物设计的这一方法应用于河流大型无脊椎动物。因此，该方法有望被进一步调整，用于获取其他更精细类群个体的体长数据。例如陆生无脊椎动物。该方法将实现对大范围空间和时间梯度下大型无脊椎动物群落体型结构的系统性估算，并通过评估探讨体型在河流生态系统功能中的作用。
