通过引入人源造血干细胞构建具有人类免疫系统的小鼠模型，需要使用不会排斥异种移植物的免疫缺陷型小鼠作为受体。目前的人源化免疫系统（HIS）小鼠模型均源于30多年前报道的免疫缺陷小鼠品系。最早报道的免疫缺陷小鼠品系为SCID小鼠，其缺乏T细胞和B细胞11；随后出现了NOD-SCID杂交品系，该品系具有SIRPα多态性，可导致小鼠巨噬细胞对人源细胞产生耐受，原因是NOD型SIRPα等位基因与人源CD47分子之间的结合能力增强12,13。在21世纪初，将IL-2受体（IL-2Rγc）的共同γ链基因在BALB/c和NOD免疫缺陷小鼠品系中进行敲除，显著提高了人源细胞的植入效率，因为该基因缺失可阻断宿主自然杀伤（NK）细胞的发育14,15,16,17。其他替代模型如BRG和NRG小鼠，则通过敲除Rag1或Rag2基因实现T细胞和B细胞缺陷，这些基因对T细胞和B细胞受体基因的重排至关重要，从而影响淋巴细胞的成熟与存活18,19。本研究中使用的BRGS（BALB/c -Rag2nullIl2RγCnullSirpαNOD）小鼠，是在Rag2-/-遗传背景下，同时结合了IL-2Rγ链缺失和NOD型SIRPα等位基因，从而获得一种高度免疫缺陷的小鼠模型，其缺乏T细胞、B细胞和NK细胞，但仍具备足够的活力和健康状态，可支持超过30周的长期移植13。
可通过多种方式构建人源化免疫系统（HIS）小鼠，其中最直接的方法是注射人外周血单个核细胞（PBMC）15,18,20。然而，这类小鼠体内会显著扩增活化的人T细胞，导致在12周龄左右出现移植物抗宿主病（GVHD），从而限制了长期研究的开展。另一种替代方法是使用来自脐带血（CB）、骨髓或胎儿肝脏的人造血干细胞进行移植，以从头重建人类免疫系统de novo。在此模型中，造血干细胞可发育为多谱系的人类免疫系统，生成T细胞、B细胞及天然免疫细胞，并且重要的是，这些细胞对小鼠宿主具有耐受性，而PBMC模型小鼠则主要发育出T细胞。因此，GVHD在该系统中不存在或显著延迟，研究可延长至小鼠10月龄。脐带血提供了一种简便、易获取且无创的CD34+人造血干细胞来源，有助于建立多个具有遗传一致免疫系统的人源化小鼠模型17,18,20,21。近年来，HIS小鼠模型已被广泛用于免疫治疗和肿瘤微环境（TME）的研究3,4,5,6。尽管这些小鼠体内已发育出人类来源的免疫系统，人源异种移植肿瘤的生长速度仍与对照组免疫缺陷小鼠相似，且允许癌细胞与免疫细胞之间发生复杂的相互作用，这对于维持移植的患者来源异种移植瘤（PDX）的微环境至关重要3,7,8。本实验方案已用于开展超过50项研究，评估HIS-BRGS小鼠中PDX和CDX模型的治疗效果。一个重要结论是，HIS小鼠中的人类肿瘤保持了其独特的肿瘤微环境（TME），这一特性通过肿瘤组织相对于原始患者样本的分子特征以及免疫细胞浸润特征得以确认3,22,23。我们课题组专注于利用多参数流式细胞术，深入评估HIS小鼠免疫器官及肿瘤组织中的人源免疫系统。本文中，我们描述了一套完整的实验流程，包括BRGS小鼠的人源化、嵌合率评估、人源肿瘤移植、肿瘤生长监测、抗癌治疗给药以及通过流式细胞术对人源免疫细胞进行分析。

Testing Cancer Immunotherapeutics in a Humanized Mouse Model Bearing Human Tumors
