In Vivo Calcium Imaging of Neuronal Ensembles in Networks of Primary Sensory Neurons in Intact Dorsal Root Ganglia

使用该方法监测的神经元数量使得能够收集利用传统或其他方法无法获得的神经网络数据。该技术的主要优势在于能够同时监测近 2,000 个初级感觉神经元，这些神经元可直接对施加于后肢足掌的刺激产生反应。该操作特别适用于研究针对周围性异样痛和疼痛过敏的治疗干预措施。该方法可适用于研究三叉神经节或膝状神经节，也可与编码电压或细胞信号分子（如环磷酸腺苷）的遗传编码传感器联合使用。该手术操作需要经过练习，可在单只动物身上对多达六个背根神经节进行练习。演示该操作的将由我本人完成，并由 John Shannonhouse 和 Mr. Ruben Gomez 协助。首先，将小鼠置于加热垫上，以维持其体温在 37 摄氏度。通过触摸小鼠的骨盆骨来定位腰膨大部位。然后，剃除小鼠腰膨大区域上方背部的毛发。使用剪刀在腰膨大上方做一三边矩形切口，并用镊子将皮肤掀开。使用13毫米弹簧解剖剪，在脊柱右侧剪开3至4毫米的切口。使用剪刀将皮肤和肌肉向两侧剪开，以暴露脊柱。然后使用8毫米剪刀清除右侧L5背根神经节（DRG）横突处的肌肉和结缔组织。尽量使用棉片或明胶海绵吸收血液，以减少出血。使用 Friedman-Pearson 咬骨钳或坚固的精细镊子切开右侧 L5 横突，注意不要触碰背根神经节（DRG）。接着，将小鼠和加热垫移至定制的载物台上。使用载物台胶带将动物和加热垫固定到位。将动物的鼻部置于鼻罩中以维持持续的异氟烷麻醉。将右侧后肢固定于平台外便于施加刺激的位置。使用平台夹具夹住L5背根神经节前后部位的椎骨或骨盆骨处皮肤，以固定脊柱。调整夹具和载物台，使背根神经节（DRG）表面尽可能水平。然后，将载物台置于显微镜下方，使物镜下降时正好位于DRG上方8毫米处。插入直肠温度计。将电源线连接至加热垫和直肠温度计。将鼻罩连接至异氟烷气体管路。使用配备10倍0.4微分干涉差（DIC）物镜及配套成像软件的直立式共聚焦显微镜进行成像。使用绿色 FITC 滤光片设置，激发波长为 495 纳米，发射波长为 519 纳米，检测波长范围为 500 至 580 纳米。在显微镜下找到 DRG 表面，调节载物台上的夹具，使 DRG 表面尽可能水平，并在焦平面上呈现最大表面积。在整个操作过程中监测动物，以维持异氟烷麻醉状态，避免过量。加载显微镜快速扫描程序时，使用以下典型设置：体素尺寸为 2.496 × 2.496 × 16 微米，图像分辨率为 512 × 512 像素，Z 轴堆栈包含 10 个光学切片，针孔大小为 1 个艾里单位（对应 32 微米），488 纳米激光功率为 1%（5 毫瓦），像素驻留时间为 1.52 微秒，扫描线时间为 0.91 毫秒，帧时间为 465 毫秒，LSM 扫描速度为 8，双向扫描，PMT 检测器增益为 650 伏，数字增益为 1。在“采集”选项卡下点击“开始实验”，对背根神经节（DRG）进行一次短
