NIR-II成像研究的动物实验在武汉大学动物实验中心进行，该中心已获得国际实验动物护理评估和认可委员会（AALAC）认证。所有动物实验均遵循中国实验动物福利委员会《实验动物护理与使用指南》，并经武汉大学动物实验中心动物护理与使用委员会（IACUC）批准。
本研究采用6周龄的雌性BALB/c裸鼠（约20 g）。
1. 近红外二区成像准备
<ol>
	<li>将市售黑色卡纸（参见材料表）放置于载物台中央，然后将样品置于黑色卡纸上，使样品位于载物台中心（成像设备中的成像平台位置）。 
	注意：与白色卡纸相比，黑色卡纸在近红外二区（NIR-II）成像过程中产生的背景干扰更少。</li>
	<li>根据NIR-II探针的波长选择合适的滤光片。当系统将滤光片移入光学成像光路时，在屏幕界面上长按（&#62;2秒）以控制对应滤光片型号的区域（例如900 LP）。</li>
	<li>在载物台控制区域的触摸屏界面上长按平台上升，使载物台上升；长按平台下降，使载物台下降。</li>
	<li>将平台高度调节至&#34;0 mm&#34;（高度调节），并使用自动聚焦功能使NIR-II图像清晰。</li>
</ol>
NIR-II 染料（HLY1）的合成
<ol>
	<li>称取合成实验所需的原料，确保其不发生降解。</li>
	<li>将化合物1（200 mg，0.18 mmol）、PdCl2(dppf)2CH2Cl2 （28 mg，0.04 mmol）、N-苯基-N-(4-(4,4,5,5-四甲基-1,3,2-二氧杂硼烷-2-基)苯基)萘-2-胺&#160;（170 mg，0.4 mmol）和K2CO3 （46 mg，0.34 mmol）加入25 mL圆底烧瓶中的四氢呋喃（THF）溶液中。在N2气氛下，75 °C搅拌反应混合物4小时（图1A）。 
	注意：化合物1和N-苯基-N-(4-(4,4,5,5-四甲基-1,3,2-二氧杂硼烷-2-基)苯基)萘-2-胺的合成步骤参见Li等21。化学结构见图1A。</li>
	<li>冷却至室温后，用去离子水（DI水）（80 mL）淬灭反应，并用DCM（二氯甲烷）/H2O（30 mL）萃取混合物（三次）。通过柱层析16纯化粗产物（石油醚:DCM = 10:1），得到绿色固体HLY1（78 mg，产率30%）。</li>
	<li>将染料HLY1置于氮气保护下，存于冰箱中以备后续使用。可保存长达6个月。</li>
</ol>
3. 水悬浮纳米探针的制备
<ol>
	<li>称取HLY1（1 mg）和两亲性包封材料1,2-二硬脂酰基-sn-甘油-3-磷酸乙醇胺-N-[氨基（聚乙二醇）-2k]（DSPE-PEG2k，10 mg；参见材料表）。</li>
	<li>采用DSPE-PEG2k作为包封基质（纳米沉淀法12），制备HLY1纳米点20（图1C）。将HLY1溶于四氢呋喃（THF，1 mL）中，随后在25 °C条件下，缓慢加入含有DSPE-PEG2k水溶液（9 mL）的烧杯中，并同时进行超声处理。接着通过透析20去除混合物中的THF。</li>
	<li>使用超滤法18对上述溶液进行离心浓缩（7100 × g，10分钟），然后置于4 °C冰箱中保存以备后续使用。该溶液可保存长达1个月。 
	注意：由DSPE-PEG2k负载的纳米探针水溶液应保存在0 °C以上，并尽快使用。</li>
</ol>
4. 构建荷瘤小鼠模型
<ol>
	<li>在添加了10%（v/v）胎牛血清（FBS）和1%（v/v）青霉素-链霉素的杜氏改良伊格尔培养基（DMEM）中培养4T1小鼠乳腺癌细胞（4T1），并在37 °C、含5% CO2的湿化培养箱中维持培养（参见材料表）。</li>
	<li>进行近红外二区（NIR-II）荧光成像实验时，将4T1细胞（5 × 107）培养24小时，用胰蛋白酶（1 mL）消化，并用无血清DMEM洗涤两次（每次4 mL）。</li>
	<li>使用异氟烷（2%）对小鼠进行麻醉。通过刺激小鼠的脚趾或足底，观察其是否产生反应，以确认麻醉是否充分。若无反应，则表明麻醉已达到足够深度32。</li>
	<li>随后，使用胰岛素注射针头将4T1细胞悬液通过皮下注射方式注入小鼠体内（100 µL）。 
	&#8203;注意：NIR-II成像研究在接种后约2周进行，此时肿瘤体积已生长至约100 mm3。在进行NIR-II肿瘤成像前，请确认肿瘤大小。本研究中肿瘤尺寸采用电子数显卡尺进行估算11。</li>
</ol>
5. 体内 近红外二区荧光成像
<ol>
	<li>使用异氟烷（2%）麻醉小鼠，并利用近红外二区（NIR-II）光学成像系统对小鼠全身进行NIR-II成像（参见材料表）。
	 
	注意：注意麻醉剂的剂量，避免小鼠死亡。通常麻醉持续5–10分钟。刺激小鼠的脚趾或足底，观察其是否产生反应。若无反应，则表明麻醉已充分。</li>
	<li>取HLY1量子点溶液（0.8 mg/mL，200 µL），将HLY1量子点经尾静脉注射至麻醉后的小鼠体内。注射3分钟后，使用NIR-II成像系统对小鼠全身血管进行NIR-II荧光成像，并进一步聚焦于小鼠头部，采集脑部血管成像。
	 
	注意：成像过程中应佩戴洁净的实验手套，有助于获得清晰的NIR-II图像。</li>
	<li>在向小鼠注射HLY1量子点5分钟后采集图像，并使用ImageJ软件处理数据。光学NIR-II成像系统的仪器参数为90 mW/cm2（808 nm激光）。</li>
	<li>实验结束后，按照机构批准的安乐死程序处理动物。
	 
	注意：本研究中，动物通过暴露于过量异氟烷实施安乐死32。</li>

荧光成像是一种在基础研究中常用的分子成像工具，也常用于临床手术中肿瘤切除的引导1。荧光成像的基本原理是利用相机接收激光照射样品（组织、器官等）后所发出的荧光信号2。该过程在数毫秒内即可完成3。荧光成像的波长范围可分为紫外区（200–400 nm）、可见光区（400–700 nm）、近红外一区（NIR-I，700–900 nm）和近红外二区（NIR-II，1000–1700 nm）4,5,6。由于生物组织中的内源性分子（如血红蛋白、黑色素、脱氧血红蛋白和胆红素）在可见光区域具有强烈的吸收和散射效应，导致光的穿透深度和灵敏度显著降低，从而对可见光波长范围内的荧光成像产生不利影响7,8,9。
NIR-II 荧光成像具有光子吸收和散射低、成像速度快以及图像对比度（或灵敏度）高的优点10,11。随着荧光波长的增加，荧光在生物组织中的吸收和散射逐渐减少，且 NIR-II 区域的自发荧光极低12。因此，NIR-II 窗口显著提高了组织穿透深度，并获得更高的分辨率和信噪比13,14,15。NIR-II 窗口可进一步细分为 NIR-IIa（1300–1400 nm）和 NIR-IIb（1500–1700 nm）窗口16。迄今为止，已有多种具有里程碑意义的 NIR-II 材料被报道，包括无机材料单壁碳纳米管、稀土纳米颗粒、量子点，以及有机材料半导体聚合物纳米颗粒、小分子染料、聚集诱导发光材料等1,17,18,19,20,21,22。无机纳米材料容易在肝脏、脾脏等器官中积累，可能存在长期生物毒性23。有机小分子荧光团具有代谢快、毒性低、易于修饰和结构明确等优势，是最有希望用于临床的探针24。

A Bright NIR-II Fluorescence Probe for Vascular and Tumor Imaging

通过近红外荧光纳米探针实现了高分辨率的血管和肿瘤成像，为血管疾病和癌症的诊疗提供了准确有效的方法。在近红外二区（NIR-II）窗口进行的活体成像使我们能够深入观察活体对象，为生物医学研究和临床应用带来了新的机遇。进行近红外二区（NIR-II）成像前，需将市售黑色卡纸放置于成像设备载物台的中心位置。然后，将样品放置在黑色纸板上方，使其位于载具的中心位置。选择合适的滤光片后，进行平台高度调节。长按载具控制台触摸屏界面上的“平台上升”选项，使平台向上移动。长按平台向下移动。合成近红外二区染料HLY1后，使用DSPE-PEG-2K作为包封基质，通过纳米沉淀法配制HLY1纳米点。具体操作为：将含有10毫克DSPE-PEG-2K溶于9毫升水中的烧杯置于25°C条件下进行超声处理。然后，将一毫克HLY1溶解于一毫升四氢呋喃（THF）中，并缓慢加入正在超声处理的DSPE-PEG-2K水溶液中。随后，通过透析法从混合物中去除THF。将上述溶液在7,100 G条件下通过超滤离心浓缩10分钟后，置于4摄氏度冰箱中保存备用。进行活体成像时，将HLY1量子点经尾静脉注射至麻醉小鼠体内，3分钟后使用光学NIR-II成像系统对小鼠全身血管进行NIR-II荧光成像。进一步聚焦小鼠头部以采集脑血管成像。将光学近红外二区（NIR-II）成像系统的仪器参数设置为90毫瓦每平方厘米和808纳米激光。在小鼠注射HLY1量子点后5分钟采集图像，并使用ImageJ软件处理数据。HLY1在含有90%水的90% THF溶液中的荧光强度是纯THF溶液中的五倍，表明HLY1具有显著的聚集诱导发光（AIE）特性。HLY1纳米点在1,500纳米低通（LP）滤光片下发出强烈的荧光信号，证明其适用于NIR-IIb成像。HLY1纳米点的最大吸收波长和发射波长分别为740纳米和1,040纳米。此外，通过动态光散射测得HLY1量子点的流体动力学尺寸为145纳米。在给予HLY1量子点的小鼠中，使用1,500纳米长通滤光片进行近红外二区（NIR-II）成像，可清晰识别脑血管及后肢微血管。此外，荷瘤小鼠的四号T1肿瘤亦可通过NIR-II成像清晰可见，表明HLY1量子点具有增强的通透性和滞留效应（EPR效应）。上述所有结果表明，HLY1 纳米点是一种可用于血管和肿瘤成像的高亮度近红外二区荧光探针。制备适用于体内近红外二区荧光成像的水悬浮纳米探针是该实验流程中最关键的步骤。
