1. 储备液的配制

2. 样本采集
<ol>
	<li>用玻璃容器采集约 0.5 kg 农田土壤表层 10 cm 的土样。本研究的土壤样品采自传统马铃薯种植区的农田。 
	注意：采用刮铲进行表层取样。然而，土壤深度可能影响其理化特性。因此，若有机碳含量随深度变化，则需在不同深度进行采样。</li>
	<li>将土壤样品带回实验室，用孔径为 1 mm 的筛子过筛，并在分析前于 4 °C 条件下保存在棕色玻璃容器中。 
	&#8203;注意：同一样品可在整个实验过程中重复使用，但每次使用后均需立即按上述条件保存。</li>
</ol>
3. 样品制备 通过 使用甲酸铵的改进型QuEChERS方法
注意：图1展示了改进的QuEChERS方法的示意图。
<ol>
	<li>称取10 g土壤样品于50 mL离心管中，加入50 µL浓度为20 mg/L的内标（IS）溶液，使终浓度为100 µg/kg。为进行回收率测定，另加入步骤1.2中配制的农药溶液，使终浓度分别为10 µg/kg、50 µg/kg和200 µg/kg（每种浓度设3个重复）。</li>
	<li>使用涡旋振荡器振荡30 s，使加标物充分混匀于样品中。</li>
	<li>加入10 mL水，使用自动振荡器在10 × g条件下振荡5 min。</li>
	<li>加入10 mL乙腈，再次在10 × g条件下振荡5 min。</li>
	<li>加入5 g甲酸铵（见材料表），手动剧烈振荡1 min，然后在1,800 × g条件下离心5 min。</li>
	<li>取1 mL乙腈提取液转移至含有150 mg无水MgSO4、50 mg初级-次级胺（PSA）和50 mg十八烷基硅烷（C18）的2 mL离心管中（见材料表），用于分散固相萃取（d-SPE）净化8，涡旋振荡30 s后，在1,800 × g条件下离心5 min。</li>
	<li>取200 µL提取液转移至带有300 µL熔融石英插管的适当标记的自动进样瓶中，使用气相色谱-质谱联用仪（GC-MS）进行仪器分析（步骤4）。 
	&#8203;注：基质匹配校准曲线的制备采用与上述相同步骤，但使用空白提取液，并在d-SPE步骤（步骤3.6）中将5 mL上清液于15 mL离心管中进行净化&#160;，内标和加标溶液直至步骤3.7才加入。将校准标准溶液加入自动进样瓶中，使终浓度分别为5 µg/kg、10 µg/kg、50 µg/kg、100 µg/kg、200 µg/kg和400 µg/kg，氮气吹干后加入200 µL基质提取液。</li>
</ol>
4. 采用气相色谱-质谱法进行仪器分析
<ol>
	<li>使用配备单四极杆质谱仪和电子轰击电离接口（−70 eV）的气相色谱-质谱联用系统进行GC-MS分析（参见材料表）。</li>
	<li>将质谱传输线温度设为280 °C，离子源温度设为230 °C。</li>
	<li>采用5%苯基-甲基聚硅氧烷色谱柱（30 m × 250 µm × 0.25 µm）（参见材料表），以超高纯度氦气作为载气，流速为1.2 mL/min，保持恒定。</li>
	<li>初始时将气相色谱柱温箱温度维持在60 °C，持续2 min，然后以25 °C/min的速率升温至160 °C，保持1 min；随后以15 °C/min的速率升至175 °C，保持3 min；再以40 °C/min的速率升至220 °C，保持3 min；接着以30 °C/min的速率升至250 °C，保持2 min；最后以30 °C/min的速率升至310 °C，保持2 min。总分析时间为22.125 min。</li>
	<li>在每一轮分析序列开始前，执行完整的自动调谐以及空气和水峰检查。
	<ol>
		<li>打开控制GC-MS系统所有参数的MassHunter采集软件。 
		注：仪器系统默认包含MassHunter采集软件。</li>
		<li>在工具栏中打开&#34;View&#34;选项，点击Vacuum control，再点击Tune，然后点击Autotune。自动调谐将在数分钟后完成。</li>
		<li>打开&#34;View&#34;选项，点击Instrument control。</li>
		<li>点击Yes，并保存此次自动调谐生成的新调谐文件。</li>
		<li>在工具栏中打开&#34;View&#34;选项，点击Vacuum control，再次点击Tune，然后点击Air &amp;#38; Water check。空气和水峰检查将在数秒后完成。</li>
		<li>打开&#34;View&#34;选项，点击Instrument control。</li>
		<li>点击Yes，并保存此次空气和水峰检查生成的新调谐文件。</li>
	</ol>
	</li>
	<li>使用自动进样器（参见材料表）在不分流模式下进样，进样口温度为280 °C，进样体积为1.5 µL。进样后0.75 min时，以40 mL/min的流速打开分流阀。 
	注：每次进样之间，10 µL进样针须用乙酸乙酯清洗三次，再用环己烷清洗三次。所有样品均进行双样重复进样。</li>
	<li>以选择离子监测（SIM）模式分析目标分析物。这是单四极杆质谱系统常用的标准检测模式。 
	&#8203;注：表1列出了有机氯农药（OCPs）和内标物（IS）的保留时间（min）及定量参数，其中每个目标物采用一个定量离子和两个定性离子进行分析。定量分析基于目标物定量离子峰面积与内标物离子峰面积的比值。</li>

5. 数据采集

近几十年来，为满足日益增长的粮食生产需求，全球范围内已广泛且集约化地使用农药。农药被施用于农作物，以保护其免受病虫害侵袭并提高作物产量，但其残留物通常最终进入土壤环境，尤其是在农业地区1。此外，某些农药（如有机氯农药（OCPs））具有非常稳定的化学结构，因此其残留物难以降解，可在土壤中长期存留2。通常情况下，土壤具有较强的农药残留累积能力，特别是当土壤中有机质含量较高时，更易积累残留物3。因此，土壤是受农药残留污染最严重的环境介质之一。例如，迄今为止一项全面的研究发现，来自欧盟各地的317个农业土壤样本中，有83%受到一种或多种农药残留的污染4。
农药残留对土壤的污染可能因其残留物的高毒性，通过食物链影响非靶标物种、土壤功能以及消费者健康5,6。因此，评估土壤中农药残留对于判断其对环境和人类健康的潜在负面影响至关重要，尤其是在缺乏严格农药使用监管制度的发展中国家7。这使得农药多残留分析变得日益重要。然而，由于土壤中存在大量干扰物质，且这些分析物浓度低、理化性质多样，农药残留的快速准确分析仍是一项艰巨的挑战4。
在所有农药残留分析方法中，QuEChERS 方法已成为最快捷、简便、经济、高效、稳健且安全的首选方法8QuEChERS 方法包括两个步骤。第一步是基于分配原理的微型提取 通过 在第一步中，进行水相与乙腈相之间的盐析分离。第二步采用分散固相萃取（dSPE）进行净化；该技术利用少量多种多孔吸附剂的组合，去除基质干扰成分，克服了传统固相萃取的缺点。9因此，QuEChERS 是一种环境友好型方法，所用溶剂和化学试剂几乎无浪费，可提供非常准确的结果，并最大限度地减少随机误差和系统误差的潜在来源。事实上，该方法已成功应用于数百种农药的高通量常规分析，在几乎所有类型的环境、农业食品和生物样品中均具有很强的适用性。8,10本研究旨在应用并验证一种新改进的QuEChERS方法，该方法此前已开发并联用气相色谱-质谱法（GC-MS）用于分析农业土壤中的有机氯农药（OCPs）。

Analysis of Organochlorine Pesticides in a Soil Sample by a Modified QuEChERS Approach Using Ammonium Formate

采用在六种浓度水平（5 µg/kg、10 µg/kg、50 µg/kg、100 µg/kg、200 µg/kg 和 400 µg/kg）下添加空白样品的基质匹配校准曲线进行线性评估。所有有机氯农药（OCPs）的测定系数（R2）均大于或等于 0.99。最低校准水平（LCL）设定为 5 µg/kg，满足食品应用中监测目的所规定的最高允许限量 10 µg/kg11。

使用甲酸铵的QuEChERS方法是提取土壤样品中有机氯农药的一种成功替代方案，可替代原始方法及两种官方版本。每克样品仅需添加0.5克甲酸铵即可诱导水相与乙腈相之间的分相，并实现良好的分析性能。开始样品制备时，称取10克土壤样品于50毫升离心管中。向土壤样品中加入50微升浓度为20毫克/升的内标（IS）溶液，使土壤中内标含量达到100微克/千克。加入10毫升水，使用自动振荡器在10 ×g 条件下振荡5分钟。然后加入10毫升乙腈，在10 ×g 条件下振荡5分钟。接下来，加入五克甲酸铵，用手剧烈振荡离心管一分钟，然后在1800 ×g 条件下离心五分钟。进行净化时，采用分散固相萃取法，将一毫升乙腈提取液转移至含有无水硫酸镁、初级次级胺和十八烷基硅烷的离心管中。涡旋振荡该离心管30秒，然后在1800 ×g 条件下离心五分钟。离心后，将200 µL提取液转移至适当标记的自动进样瓶中，瓶内装有300 µL熔融石英衬管，并使用GC/MS系统进行仪器分析。在不分流模式下，通过自动进样器将1.5 µL样品注入配备单四极杆质谱仪的GC/MS系统，进样口温度为280 °C。所研究的有机氯农药（OCPs）的基质效应分布显示，既有表现为信号增强的正基质效应百分比，也有表现为信号抑制的负百分比。数值在负20%至20%之间表明存在较弱的基质效应，而负20%至负50%以及20%至50%之间的数值对应中等程度的基质效应。当百分比值高于50%或低于负50%时，则表明存在较强的基质效应。在不同加标水平下评估所有农药的总体回收率发现，大多数研究的有机氯农药（OCPs）平均回收率在70%至120%之间，相对标准偏差（RSD）低于20%。然而，七氯、艾氏剂和β-硫丹的平均回收率略高。该替代方法可用于农药的液相色谱-质谱分析，这将更加有意义，因为甲酸铵有助于正电喷雾离子化过程中的离子化。
