1. 非侵入性前交叉韧带损伤
注意：通过外部施加的压缩载荷导致的前交叉韧带（ACL）损伤是一种简单且可重复的方法，能够很好地模拟人类前交叉韧带损伤的情况。本实验方案针对一种 commercially available load frame instrument（见材料表）编写，但也可根据类似系统进行调整。
<ol>
	<li>打开与载荷架仪器兼容的软件（参见材料表），选择一个现有的控制文件或创建一个新文件。</li>
	<li>打开作动器的电源。</li>
	<li>在校准菜单中，对载荷传感器的力读数进行去皮归零，并将作动器的位移设置为0。</li>
	<li>使用含1%–4%异氟烷的氧气麻醉小鼠，并通过脚趾夹捏和/或尾部夹捏确认动物已完全麻醉。</li>
	<li>将小鼠俯卧位放置在平台上。将下肢垂直置于两个加载夹具之间（图1）（参见材料表）。将足部放入上部夹具的切口内，膝关节放入下部夹具的凹槽内。</li>
	<li>手动调节下部夹具的高度，施加1–2 N的预载荷（在计算机屏幕上实时监测），并拧紧定位螺钉以固定位置。预载荷对于在施加损伤载荷前保持腿部处于正确位置是必要的。</li>
	<li>施加单次压缩载荷至目标力（约12–15 N）或目标位移（约1.5–2.0 mm）。 
	注意：以较慢的加载速率（约1 mm/s）施加载荷可提供更高水平的实时监测与控制，但可能导致前交叉韧带（ACL）发生撕脱性断裂。以较快的加载速率（约200 mm/s）施加载荷更可能产生ACL实质中部损伤22。如果确定发生了胫骨骨折或其他过度损伤，应在动物从麻醉中恢复之前，采用经IACUC批准的方法实施安乐死。
	<ol>
		<li>在软件控制文件中设置加载速率，并通过力-位移数据进行确认。 
		注意：由于胫骨压缩加载过程中骨骨折的力（约20 N）明显高于ACL损伤力，因此通常不必担心发生骨骨折。然而，仍应通过触诊进行监测，必要时可使用影像学检查（如X射线）确认是否发生胫骨骨折。</li>
	</ol>
	</li>
	<li>损伤通常表现为可听见的声音（&#34;咔哒声&#34;或&#34; crunch声&#34;），以及在力-位移曲线图上可识别的力值释放（图1C）。若使用较慢的加载速率，在损伤发生后应立即停止压缩载荷，以防止进一步加载并对其他关节组织造成可能的损伤。 
	注意：ACL损伤通常发生在8–15 N之间，具体取决于体重34。重要的是设定一个高于预期ACL损伤力的目标力。</li>
	<li>使用前后抽屉试验35,36或其他评估关节不稳定性的方法确认ACL损伤。</li>
	<li>在损伤后给予小鼠按体重计算的镇痛药物剂量（例如，0.05–0.1 mg/kg皮下或腹腔注射布托啡诺，参见材料表），给药持续时间和剂量应遵循所在机构IACUC推荐并批准的方案。 
	注意：非甾体抗炎药（NSAIDs）可能改变损伤后骨关节炎（PTOA）的发展进程，因此除非研究有特定目的，否则不建议在此损伤模型中使用NSAIDs进行疼痛管理。</li>
</ol>
2. FRI 成像的动物准备
注意：进行光学成像时，动物毛发（尤其是深色毛发）会强烈阻挡、吸收和散射光线，因此在成像前必须尽可能去除膝关节周围区域的毛发。脱毛膏通常比电推剪更有效地去除毛发。裸鼠或无毛鼠无需脱毛处理。然而，大多数常用小鼠品系（如 C57BL/6）需要进行脱毛。如果条件允许，成像前应喂食低荧光性纯化饲料。标准小鼠饲料含有叶绿素，其自发荧光波长约为 700 nm，可能影响近红外荧光成像系统（FRI）的数据采集。
<ol>
	<li>使用氧气中1%-4%的吸入用异氟烷对小鼠进行麻醉。尽可能将小鼠置于加热垫上，并涂抹眼膏以防止眼睛受到刺激。</li>
	<li>用棉签将脱毛膏（见材料表）涂抹于小鼠腿部膝关节周围前侧（头侧）区域。</li>
	<li>让脱毛膏停留约1分钟，然后用擦拭巾清除腿部的脱毛膏和毛发。如有必要，可重复操作。</li>
	<li>当膝关节完全暴露且无毛发覆盖时，使用酒精擦拭巾清洁腿部，以去除残留的脱毛膏。 
	注意：在一项研究过程中，同一小鼠可多次使用脱毛膏，但每次使用间隔应至少为一周，以避免对皮肤造成不必要的刺激。</li>
</ol>
3. 探针溶液的制备
<ol>
	<li>如有必要，根据生产商说明，用无菌 1× 磷酸盐缓冲液（PBS）稀释荧光可激活探针。市售探针一瓶（见材料表）通常含有 20 nmol 溶于 0.15 mL 的 1× PBS 中。为稀释瓶内溶液，加入 1.35 mL 的 1× PBS，使终体积为 1.5 mL 的 1× PBS 中含 20 nmol 探针。 
	注意：稀释后，按每只小鼠注射 0.15 mL 计算，一瓶溶液可用于十只小鼠的成像。</li>
	<li>使用涡旋振荡器以最低速度（约 2000 rpm）振荡 30 秒，确保探针充分溶解，随后短暂离心，使管盖内壁无液体残留。 
	&#8203;注意：溶液可在避光条件下于 2–8 °C 保存，最长可达 12 个月。</li>
</ol>
4. 眶后注射
注意：有关本实验步骤的详细信息，请参见 Yardeni 等人的文献37。
<ol>
	<li>使用含1%-4%异氟烷的氧气麻醉小鼠，并将小鼠侧放，鼻部置于鼻罩中。</li>

		<li>向注射后的眼部滴加生理盐水或涂抹眼膏。 
		&#8203;注意：根据成像探针产品说明书，最佳成像时间通常在注射探针溶液后1至2天。如条件允许，建议先进行初步的时间点筛选，以确定每种具体应用的最佳成像时间。小鼠通常在约7天内代谢掉注射的探针，若需后续时间点成像，需重新注射探针溶液。</li>

5. 荧光反射成像
注意：本节中的操作步骤专用于一种市售的光学成像系统（参见材料表）。使用类似的成像系统也可进行相应的成像操作。
<ol>
	<li>使用1%-4%的异氟烷氧气混合气体对小鼠进行吸入麻醉，并将动物仰卧位放置于成像系统中，鼻部置于鼻罩内。</li>
	<li>将小鼠后肢伸展定位，使膝关节略微朝上（必要时可用胶带固定足部）。所有动物必须采用一致的体位摆放，这一点至关重要。</li>
	<li>在成像系统计算机上打开配套软件（见材料表），&#34;采集控制面板&#34;将出现。</li>
	<li>为预热系统，点击初始化，并等待温度指示灯变为绿色。</li>
	<li>点击成像向导，确保弹出&#34;成像向导&#34;窗口。</li>

	<li>成像参数可在&#34;采集控制面板&#34;右侧面板查看和修改。确认所有设置无误后，点击采集序列按钮。图像显示后，确认曝光是否充分；若不足，调整曝光时间设置后再次点击采集序列。</li>
	<li>为分析图像，在黑白图像上每个膝关节处放置大小一致的兴趣区域（ROI）圆形（此举可避免因荧光信号区域差异而导致的定位偏差）。</li>
	<li>计算每个膝关节的总辐射效率和/或平均辐射效率。若同时对对侧肢体进行了测量，则通过将损伤侧肢体的辐射效率值除以对侧未损伤肢体的辐射效率值进行数据归一化处理。 
	注：若所有膝关节均使用面积一致的兴趣区域，则总辐射效率与平均辐射效率结果相近。当使用不同大小的兴趣区域时，建议采用平均辐射效率。将损伤关节的辐射效率数据与未受伤的对侧膝关节数据进行归一化处理，可作为内部对照，以消除不同动物间探针注射量及递送效率差异的影响。</li>

非侵入性膝关节损伤小鼠模型为研究损伤后早期的生物学和生物力学变化提供了机会，可能获得更具临床相关性的结果20。我们实验室已建立一种非侵入性损伤模型，通过单次外部施加的胫骨压缩过载来诱导小鼠前交叉韧带（ACL）断裂21,22,23,24。这种非侵入性损伤方法能够在不破坏皮肤或关节囊的情况下产生无菌性关节损伤。
荧光反射成像（FRI）是一种光学成像方法，通过特定波长的红外光激发目标，并定量检测在另一波长处反射的光信号。 commercially available protease-specific probes 可注射入动物模型体内，随后利用 FRI 定量检测特定部位（如膝关节）的蛋白酶活性。该方法已广泛用于体内（in vivo）生物学活动（如炎症）的检测。用于此应用的探针在遇到相应的蛋白酶之前处于荧光淬灭状态；当这些蛋白酶作用于探针上的酶切位点并将其断裂后，探针即释放出近红外荧光信号。这些探针及该成像方法已在癌症25,26,27,28和动脉粥样硬化29,30,31,32研究中得到广泛验证和应用，本课题组也已将其用于肌肉骨骼系统的研究，以检测炎症和基质降解的标志物23,24,33。

在3月龄雄性C57BL/6J小鼠的下肢施加单次压缩力（以1 mm/s的速度加压直至损伤）后，所有小鼠均稳定诱导出前交叉韧带（ACL）损伤。膝关节损伤时的平均压缩力约为10 N（图1）。
