1. 昆虫饲养
<ol>
	<li>在水稻幼苗（Oryza sativa cv. Nipponbare）中饲养本实验所用的褐飞虱。将20株水稻幼苗种植于培养皿（65 mm × 200 mm）中，在25 °C、16小时光照/8小时黑暗光周期条件下培养。</li>
</ol>
2. 收集褐飞虱血淋巴的解剖步骤
<ol>
	<li>将SBPH放入离心管中，置于冰浴中10-30分钟。 
	注意：冰浴时间不得少于10分钟，否则昆虫可能复苏。</li>
	<li>在体视显微镜下（参见材料表），将一只冷冻的SBPH腹部朝上放置于载玻片（参见材料表）上。调节焦距，使昆虫的六条腿清晰可见。</li>
	<li>准备两把超细尖的高精度镊子（参见材料表）。用一把镊子压住昆虫身体以固定其位置，另一把镊子小心地将其中一条腿从昆虫体上拔下。 
	注意：对于外壳较硬的昆虫，可使用眼科手术刀切断其中一条腿。</li>
	<li>用镊子轻轻按压昆虫胸部，使血淋巴从伤口处流出。 
	&#8203;注意：血淋巴呈透明液滴状，无可见白色絮状物。若出现白色絮状物，表明存在脂肪体污染，应弃去脂质部分。</li>
</ol>
3. 使用微量移液器采集血淋巴
<ol>
	<li>准备一支微量移液器。将一支体积为 1 µL 的毛细管（参见材料表）插入移液球（参见材料表）中。为确保测量准确，应保证刻度线清晰可见。 
	注意：移液球顶部需有一个小孔。</li>
	<li>采集血淋巴时，用手握住微量移液器的移液球，将毛细管靠近昆虫伤口处。只需将毛细管尖端接触从伤口渗出的血淋巴，即可完成采集。</li>
	<li>当毛细管内的液体达到所需刻度线时，用手指轻轻堵住移液球顶部的小孔，以停止吸取过程。 
	注意：应连续且迅速地采集 1 µL 血淋巴样品，以避免脂质污染和毛细管堵塞。</li>
	<li>挤压移液球，将采集的血淋巴排入 100 µL 磷酸盐缓冲液（PBS 缓冲液，含 137 mmol/L NaCl、2.7 mmol/L KCl、4.3 mmol/L Na2HPO4、1.4 mmol/L KH2PO4，pH 7.2–7.4）中。 
	&#8203;注意：将毛细管插入 PBS 缓冲液中。</li>
	<li>采集另一份 1 µL 血淋巴样品时，需更换新的毛细管。</li>
</ol>
4. 考马斯亮蓝染色
<ol>
	<li>根据步骤3中所述方案，从三龄幼虫中采集3份等体积（1 µL）的血淋巴样品，并将收集的血淋巴排入PBS缓冲液中，使总体积达到100 µL。</li>
	<li>向100 µL稀释后的血淋巴样品中加入25 µL的5倍蛋白上样缓冲液（250 mmol/L Tris-HCl [pH 6.8]，10% W/V SDS，0.05% W/V 溴酚蓝，50% W/V 甘油，5% W/V β-巯基乙醇），加热以使蛋白质变性。</li>
	<li>取20 µL煮沸后的样品加入10% SDS-PAGE蛋白凝胶（见材料表），进行蛋白质分离。</li>
	<li>将凝胶置于考马斯亮蓝溶液中（将1 g考马斯亮蓝R-250与250 mL异丙醇、100 mL冰醋酸和650 mL ddH2O混合，用滤纸过滤除去颗粒）室温染色1小时，然后用脱色液（100 mL乙酸、400 mL甲醇、500 mL ddH2O）脱色1小时。</li>
</ol>
5. 蛋白质浓度测定
<ol>
	<li>用 PBS 缓冲液通过两倍稀释法将 10 mg/mL 牛血清白蛋白（BSA）稀释为 5 mg/mL、2.5 mg/mL、1.25 mg/mL、0.625 mg/mL 和 0.3125 mg/mL。</li>
	<li>吸取 5 µL PBS 缓冲液及第 5.1 步中各浓度的 BSA 溶液，分别加入 1 mL 1x Bradford 染料试剂（参见材料表），在室温下孵育 5 分钟。以 PBS 缓冲液加入 Bradford 染料试剂作为空白对照，在 OD = 595 nm 处调零。测定其余各蛋白浓度的吸光度值，并绘制标准曲线。</li>
	<li>取 1 µL 幼虫、雌性或雄性褐飞虱（SBPH）的血淋巴加入 100 µL PBS 缓冲液中，然后在 4 °C 条件下以 1,000 × g 离心 10 分钟，以去除血细胞。</li>
	<li>将 90 µL 上清液转移至新的离心管中，按照第 5.2 步的操作说明测定上清液的吸光度值。</li>
	<li>根据标准曲线的回归方程计算血淋巴样品的蛋白浓度。实验中包含三个生物学重复，每个生物学重复设三个技术重复。</li>
</ol>
6. 显微检测
<ol>
	<li>使用微量移液器从不同发育阶段的10-15只褐飞虱（SBPH）中采集血淋巴。将收集的血淋巴加入含有20 µL 4%多聚甲醛溶液的离心管中（参见材料表）。在室温下孵育30分钟，以固定血淋巴。</li>
	<li>将20 µL固定后的血细胞滴加至经硅烷处理的载玻片中央（参见材料表）。</li>
	<li>将液滴置于载玻片干燥架上干燥。 
	注意：避免过度干燥。</li>
	<li>用金抗荧光淬灭试剂4',6-二脒基-2-苯基吲哚（DAPI）覆盖样本（参见材料表），并在倒置显微镜下观察载玻片（参见材料表）。</li>
</ol>
7. 细胞定量
<ol>
	<li>用微量移液器收集1 µL的褐飞虱血淋巴，并将其溶解于20 µL的PBS缓冲液中。</li>
	<li>用PBS缓冲液将血淋巴样本稀释五倍。</li>
	<li>将20 µL血淋巴溶液加入细胞计数板中央（参见材料表），并用盖玻片覆盖液滴（参见材料表）。</li>
	<li>在倒置显微镜下（参见材料表）对细胞计数板上的五个方格进行细胞计数（参见补充图1）。每次实验包含三个生物学重复，每个生物学重复设三个技术重复。

8. 统计分析

动物病毒和植物病毒均可通过节肢动物传播，这些病毒对人类健康构成严重威胁，并在农业领域造成巨大的经济损失1,2,3。重要的是，节肢动物的血淋巴作为其循环系统以及免疫系统的重要组成部分，在调控虫媒病毒传播过程中发挥着关键作用。病毒通过节肢动物肠道被获取后，必须成功逃逸不利的血淋巴环境，才能被运输至其他组织4,5,6,7。病毒在节肢动物血淋巴中的生命周期包括在血浆液体中存活、进入血细胞以及向其他组织运输，且多种病毒与媒介之间的相互作用机制均发生在血淋巴中8,9,10,11,12。例如，灰飞虱（SBPH）对水稻条纹病毒（RSV）的垂直传播依赖于灰飞虱卵黄原蛋白与RSV衣壳蛋白之间的分子相互作用13,14。一些病毒可通过结合特定的媒介因子来逃避免疫系统对血淋巴的免疫应答15,16,17,18。因此，研究节肢动物血淋巴中媒介与病毒之间的相互作用，对于深入理解虫媒病毒的传播机制具有重要意义。
由于体型微小，一些小型昆虫（如飞虱、叶蝉和某些蚊子）的血淋巴难以采集。为解决这一问题，已开发出多种血淋巴采集方法，包括将注射器针头直接插入昆虫体内以提取微量血淋巴、用尖头镊子收集伤口处渗出液以及直接离心法。这些方法使得研究人员能够测定血淋巴中的相对基因表达水平和病毒滴度19,20,21。然而，目前尚无有效方法可用于定量这些小型昆虫的血淋巴体积，而血淋巴体积的精确测定对于血细胞计数、蛋白质定量和酶活性分析至关重要。
SBPH（小褐飞虱）是一种小型昆虫媒介，体长约2-4 mm。SBPH能够传播多种植物病毒，包括水稻条纹病毒（RSV）、玉米粗缩病毒和水稻黑条矮缩病毒22,23,24。过去十年中，SBPH与RSV之间的相互作用已被深入研究。为了便于开展SBPH相关实验，我们开发了一种新颖且简便的血淋巴采集方法。该方法基于毛细作用原理，利用带有刻度标记的毛细管，以精确且可量化的方式获取昆虫血淋巴。此技术使我们能够高效地从小型昆虫中采集特定体积的血淋巴，从而更详细地研究小型媒介昆虫血淋巴环境。

A Precise and Quantifiable Method for Collecting Hemolymph from Small Arthropods

节肢动物可通过其血淋巴传播多种具有医学和农业重要性的病毒，血淋巴在病毒传播过程中起着关键作用。血淋巴采集是研究病毒与媒介相互作用的基础技术。本文以灰飞虱（Laodelphax striatellus，简称SBPH）为研究模型，介绍一种新型且简便的小型节肢动物血淋巴定量采集方法，该昆虫是水稻条纹病毒（RSV）的主要传播媒介。本实验方案中，首先将冷冻后的节肢动物用精细镊子轻轻掐断一条腿，从伤口处挤出血淋巴；然后利用由毛细管和吸头组成的简易微量移液器，依据毛细作用原理收集从伤口渗出的血淋巴；最后，所采集的血淋巴可溶解于特定缓冲液中用于后续研究。这种小型节肢动物血淋巴采集的新方法，为虫媒病毒及媒介-病毒相互作用的深入研究提供了一种有效且实用的工具。

微量移液器模型与血淋巴采集 
我们开发了一种基于毛细管毛细作用原理的简易微量移液器。该微量移液器由毛细管和移液球两部分组成（图1A）。毛细管有多种容积规格可供选择，范围从1 µL到20 µL，可根据实际需求选用合适的容积。不建议使用容积更小的毛细管，因为小容积毛细管的孔径过细，可能导致难以吸取血淋巴等液体。移液球顶部有一个小孔，在采集血淋巴过程中不应将其堵塞。该移液球便于在液体采集过程中握持微量移液器（图1B），并有助于将毛细管中采集的液体转移至收集缓冲液中。
为了便于收集血淋巴，本实验中首先将褐飞虱（SBPH）置于冰上或冰箱中冷冻。将冷冻后的褐飞虱置于载玻片上，在体视显微镜下定位，然后用尖头镊子逐个拔除其中一条腿（图1C）。为确保伤口足够大并实现最佳血淋巴收集效果，最好在腿的基部将其拔除（图1Ca、<st...

我们的研究聚焦于节肢动物病毒、昆虫与植物之间的相互作用。从小型节肢动物中获取血淋巴样本，对虫媒疾病研究而言是一项技术挑战。本实验方案展示了我们为解决这一问题所采用的精确且可量化的技术。关于节肢动物传播病毒的最新研究发现，病毒能够操控多种重要的媒介因子，以突破宿主的屏障。本实验方案是一种简单、经济且有价值的技術，可实现对血淋巴的精确定量。该方法还为研究病毒与媒介之间的相互作用提供了重要的技术手段。我们即将开展的研究将聚焦于了解媒介昆虫传播的各种分子机制。我们正在探索并改进新的操作技术。本研究中，请使用在水稻幼苗中饲养的小褐飞虱（SBPHs）。待培养物准备好后，将生长的SBPH转移至离心管中，置于冰浴中10至30分钟。然后将一只冷冻的SBPH置于体视显微镜下的载玻片上，使其腹部朝上，并将焦点对准其六条腿。准备两把超细尖的高精度镊子。用一把镊子按住昆虫的身体以固定其位置，再用另一把镊子小心地取下一条腿。然后轻压昆虫的胸部，使血淋巴从伤口流出。为收集血淋巴，可将一支1微升毛细管插入移液球中，制成微量移液器。 将毛细管靠近昆虫伤口，同时握住微量移液器的吸头。将毛细管尖端接触渗出的血淋巴并收集液体。当毛细管内液面达到所需刻度线时，用手指轻微堵住移液器吸头上的小孔，以停止吸收过程。然后将收集的血淋巴排入100 µL的PBS缓冲液中。通过SDS-PAGE验证血淋巴收集的准确性，结果显示三次独立收集的血淋巴样品中蛋白质含量相似。对于幼虫，总蛋白浓度约为3.7 mg/mL。在成年雌性和雄性褐飞虱中，蛋白质浓度分别为每毫升约3.5毫克和3.6毫克，三个样本之间无显著差异。幼虫血淋巴中可见大小为2至20纳米的血细胞。此外，所采集的三个血淋巴样本中的细胞浓度相近。
