本文提供了一个使用最新版寿命机器（Lifespan Machine）进行实验的详细且易于操作的实验方案。我们已证明，获得分辨清晰的生存曲线的关键步骤是在图像采集后的手动排除非线虫物体。手动标注死亡时间对生存曲线的整体形态影响较小，表明即使不进行人工标注，全自动死亡时间估计也具有较高效率（图8）。相反，若要获取高质量的行为衰老数据，则需要更精确地标注死亡时间，尤其是在长寿个体中（图9）。因此，手动分镜标注所需的时间最终取决于所测量的具体指标。总体而言，我们发现使用LSM时，研究人员的主要精力应集中于实验设计阶段，其次为图像采集后的分析阶段。最后，我们强调高通量、自动化检测在生成高分辨率的生存数据和行为衰老数据方面的价值，同时可提高研究人员的工作效率，并支持实验的可重复性。

衰老是一个复杂且多方面的过程，其特征是生物体生理功能的衰退，从而导致随着时间推移疾病风险和死亡概率增加1。寿命通常被定义为从出生或成年期开始到死亡的时间，是衰老的一个明确结局指标2，也是衡量不同群体间相对衰老速率的一种间接但严格量化的代理指标3。衰老研究通常依赖于对寿命的精确测量，类似于临床试验，通过比较接受干预的群体与未接受干预的对照群体之间的结局来进行评估。然而，目前衰老研究普遍存在可重复性问题，有时是由于统计效力不足的实验设计4，而更多情况下则是因为寿命测定本身对环境中的细微变化具有高度敏感性5。可靠的实验需要对大规模群体进行多次重复，而这一过程尤其受益于自动化所提供的实验可扩展性6。
寿命测定实验的严格要求源于衰老过程本身的不可预测性。在相同环境中饲养的同基因型个体表现出不同的死亡时间和生理衰退速率7，这表明寿命具有高度的随机性7,8。因此，需要使用大样本群体来定量测量衰老过程中的变化，例如平均寿命或最大寿命的变化，并克服个体差异带来的偏差。此外，高通量寿命测定能力对于研究生存曲线的形态以及衰老动态模型至关重要9。
线虫 Caenorhabditis elegans 是衰老研究中不可或缺的模式生物，因其寿命短、遗传背景清晰且世代周期快，非常适合开展高通量的衰老与寿命测定实验。传统上，C. elegans 的寿命测定方法是通过在固体培养基上同步培养约50–100条线虫的小群体，随时间推移记录每条个体的死亡时间。随着线虫衰老并逐渐丧失运动能力，人工判定死亡时间需要逐条用工具轻触线虫并观察其头部或尾部是否仍有微小活动。这一过程通常十分繁琐且耗费人力，尽管已有研究尝试对其进行加速10,11,12。重要的是，实验流程的缓慢会阻碍人们对衰老机制的理解以及对干预措施有效性的评估进展。
为了满足衰老研究对定量数据的需求，已开发出多种技术以实现数据采集的自动化，包括从微流控腔室到平板扫描仪在内的多种方法，这些技术在文献中已有广泛报道13,14,15,16,17,18。LSM 与其他方法的不同之处在于其对高精度和高准确度寿命数据采集的全面优化，这一目标通过精心设计的设备校准流程以及一套功能完善的软件系统得以实现，该软件系统允许用户验证、修正并优化自动化分析结果13。尽管该软件在理论上可应用于多种成像方式，但在实际应用中，大多数用户采用经过改装的平板扫描仪，以实现对环境温度和湿度的精细调控——这两个因素对寿命具有显著影响，因此至关重要19。LSM 会以每20分钟一次的频率对线虫进行成像，持续时间可从数天至数月不等，具体取决于环境条件和基因型。相比人工检测所获得的数据，LSM 产生的数据具有更高的时间分辨率，所采集的图像还提供了线虫在整个生命周期中位置变化的永久可视化记录。通过机器学习方法，系统可自动判定每个个体的死亡时间。这些结果可通过名为 &#34;Worm Browser&#34; 的客户端软件快速进行人工验证。得益于其硬件与软件的协同设计，LSM 生成的生存曲线在统计学上与经验丰富的研究人员手动评分的结果无显著差异，同时具备工作量更小、可扩展性更高的优势13。
LSM 的最新版本还可通过在整个线虫生命周期中收集形态学和行为学数据，并将其与每个个体的寿命一并报告，从而实现对行为衰老的研究。特别是，LSM 能够记录每只动物剧烈运动停止（VMC）的时间，这一标志常被用来量化个体的&#34;健康寿命&#34;，以区别于其寿命。通过同时收集寿命和行为衰老数据，LSM 支持研究那些可能对衰老的不同表型结果产生差异性影响的干预措施20。多种肉眼可观察的表型可用于研究行为衰老，例如身体运动或咽部泵动21、组织完整性22，以及运动速度或刺激诱导的转向行为17。不同衰老表型之间的比较有助于分析衰老过程的因果结构。例如，最近通过对 VMC 与寿命的比较，揭示了 C. elegans 中两种不同的衰老过程23。
尽管最初是为测量秀丽隐杆线虫（C. elegans）的寿命而开发，LSM 技术也支持从多种线虫物种中收集生存和行为数据，包括C. briggsae、C. tropicalis、C. japonica、C. brenneri和P. pacificus23。该技术有助于研究生物和环境干预对寿命、抗应激能力及抗病原体能力的影响，并可与靶向 RNA 干扰检测或生长素诱导的蛋白质降解系统等实验工具联用。迄今为止，该技术已在科学文献中被广泛应用于多种研究场景6,24,25,26,27,28,29,30。

在我们的实验室中，我们以定量的方式研究衰老和寿命的生理学与分子决定因素。为此，我们采用线虫秀丽隐杆线虫（C. elegans）作为遗传操作便捷且衰老迅速的模式生物，并结合寿命分析仪等工具开展研究。可重复性是衰老研究中的一个重要问题。为克服这一问题，通常需要大量动物种群以测量寿命的显著变化，因此该过程极大地受益于自动化的高通量实验方案。许多研究人员已将寿命检测仪用于多种不同目的，并对此表示满意。在我们自身的应用中，两项最重要的发现分别是：首先，2016年我们发现时间缩放是多种衰老干预措施的共同结果。其次，在2022年，我们发现健康寿命和寿命是由不同的基础生理衰退过程所决定的。寿命检测仪通过自动化和高频图像采集，将常用的琼脂平板实验方案进行改造，从而从大量线虫群体中收集存活率和行为衰老数据。该设备消除了传统手工存活率检测每日繁琐的操作，并获取了以往无法获得的线虫行为数据集。首先，确保扫描板已准备好并接种了线虫。通常，每个平板接种约40只动物和200微升细菌菌苔，是寿命菌苔实验的最佳条件。开始实验设置时，在将用于支撑扫描板的玻璃片两面均涂抹防雾玻璃清洁剂。然后，在将与扫描仪平板接触的玻璃面涂抹保护性疏水玻璃涂层。静置5到10分钟，清除玻璃板上的任何残留物。使用70%乙醇对将与扫描仪平板接触的玻璃面进行消毒。静置一到两分钟，然后用布或纸巾除去乙醇。将平板装载到扫描仪上时，先将经高压灭菌的橡胶垫放置在处理过的平板支撑玻璃上方。取用于成像的、已加载线虫的扫描仪平板，打开并移除盖子。将培养皿放置在面向玻璃表面的橡胶垫位置上。在加载培养皿时，拔掉扫描仪风扇的电源，以保护实验人员的手指。轻轻将培养皿及其支撑的玻璃片滑动到扫描仪表面。重新启动扫描仪风扇，并确认前面和侧面的风扇已通电。此时打开扫描仪。在开始实验之前，执行必要的图像采集前步骤，包括获取整个扫描仪表面的预览图像。通过网页界面，找到主页面上的图像采集部分，然后点击“采集设备和图像服务器”链接。在打开的窗口中，点击“搜索新设备”，并确保连接到服务器的每台扫描仪均出现在图像采集设备列表中。选中对应于已加载新培养皿的每台扫描仪的复选框，然后点击“请求预览采集”按钮。接下来在蠕虫浏览器中，选择“文件”、“打开图像”，然后选择要打开的每张预览捕获图像。对于每张图像，单击以选择对应于包含培养板区域的列。定义完所有图像后，选择“图像采集”、“定义扫描区域”，将选定的扫描区域保存到磁盘，然后选择所需位置将区域规格导出至磁盘。创建一个文件，其中包含实验名称、扫描仪上每列的物理位置、实验总时长以及图像采集频率。将该文件保存为文本文件和 XML 文件。在蠕虫浏览器中，点击“图像采集”，选择“提交实验计划”，然后选择生成的 XML 文件。线虫浏览器将提示是否输出实验计划的摘要或直接运行实验。请生成一个摘要文件，并核对摘要中计划拍摄的日期、位置
