本方案使用30天龄的 w1118 来自布卢明顿的果蝇 果蝇 保种中心 (BDSC_3605，参见 材料表).
1. 老年果蝇的制备
<ol>
	<li>食物制备
	<ol>
		<li>通过混合50 g/L玉米片、110 g/L蔗糖、5 g/L琼脂和25 g/L酵母制备标准玉米淀粉培养基。将玉米片和酵母加水加热至糊化，然后充分溶解所有成分。</li>
		<li>当培养基冷却至50–60 °C时，加入6 mL/L丙酸，并立即分装至培养瓶中。</li>
	</ol>
	</li>
	<li>果蝇饲养及老龄果蝇的制备
	<ol>
		<li>将果蝇品系w1118饲养于含标准玉米淀粉培养基的培养瓶中，并将培养瓶置于25 °C、相对湿度68%、光照强度500–1000 lux、光周期12 h光照:12 h黑暗的恒温培养箱中。</li>
		<li>根据果蝇生长周期，每7天将果蝇转移至新培养瓶一次，确保同一瓶内个体的年龄保持一致。</li>
		<li>在转移果蝇3天后，收集从原始培养瓶中新羽化的果蝇，并将其放入新的培养瓶中。按照每7天换瓶一次的原则，持续培养至果蝇约30日龄。</li>
	</ol>
	</li>
</ol>
2. 药用食物和用于监测的玻璃管的制备
注意：玻璃管的制备步骤参照 Jin 等人的研究并进行了修改34。
<ol>
	<li>玻璃管的清洗与干燥
	<ol>
		<li>将玻璃管（直径5 mm，长度65 mm，参见材料表）放入大烧杯中，用重蒸水浸泡并煮沸20分钟。重复3次。</li>
		<li>取出并捆扎玻璃管，用重蒸水冲洗管内壁3–5次，然后放入烘箱中干燥。</li>
	</ol>
	</li>
	<li>简单培养基的制备（100 mL）
	<ol>
		<li>将1.5 g琼脂和5 g蔗糖溶于重蒸水中，加热并定容至100 mL。</li>
		<li>当培养基冷却至约70 °C时，加入600 µL丙酸，并使用恒温水浴防止其凝固。</li>
		<li>取约4 mL简单培养基与利血平（参见材料表）加入10 mL小烧杯中，使药物终浓度达到20 µM或50 µM。在阴性对照组中加入二甲基亚砜（DMSO），使其终浓度为0.2%。</li>
	</ol>
	</li>
	<li>含药玻璃管的制备
	<ol>
		<li>为便于培养基流动，将适当长度的玻璃管小心插入小烧杯中，培养基将在大气压作用下自然进入玻璃管内。</li>
		<li>待培养基完全凝固后，取出玻璃管，擦干外壁，得到一端含有含药培养基的监测用玻璃管。</li>
		<li>将固体石蜡放入烧杯中加热至70 °C熔化，将玻璃管靠近食物的一端浸入石蜡液中约5 mm，迅速取出。待石蜡凝固后，即可密封玻璃管的食物端。</li>
	</ol>
	</li>
</ol>
3. 实验设计与果蝇处理
<ol>
	<li>根据表1设计果蝇处理实验。</li>
</ol>
4. 黑腹果蝇 组装与睡眠监测
注意：黑腹果蝇（Drosophila）基因组组装流程参考 Jin 等人的研究34，并进行了修改。
<ol>
	<li>用 CO2 气体麻醉果蝇，将果蝇放入石蜡密封的玻璃管中（每管一只），并在非食物端用棉球堵住，以防止果蝇逃逸并确保空气流通。</li>
	<li>将玻璃管装载到红外监测仪上进行监测。
	<ol>
		<li>将装有果蝇的玻璃管按同一方向组装到红外监测仪上，并记录每个药物对应的监测仪编号和孔位编号。</li>
		<li>调整每根玻璃管的位置，使红外线垂直穿过果蝇活动范围的中心。</li>
		<li>将监测仪放入位于果蝇睡眠暗室内的 25 °C 培养箱中，设置条件如下：温度 25 °C， Zeitgeber 12 (ZT12)（相当于当地时间晚上 08:00），ZT24（相当于当地时间早上 08:00）。该设置可确保果蝇经历 12 小时光照与 12 小时黑暗的交替周期。 
		注意：在数据采集完成前，尽量不要打开培养箱门，以维持监测期间培养箱内环境的稳定。</li>
		<li>使用 DAM2 系统开始监测（参见材料表）。</li>
		<li>监测完成后，从系统中下载以 .txt 格式保存的采集数据。</li>
	</ol>
	</li>
</ol>
5. 数据处理
注意：使用 DAM 系统、DAMFileScan107 和 SCAMP 进行数据处理均按照其官方网站上的说明进行（参见材料表）。
<ol>
	<li>将上述 txt 文件导入 DAMFileScan107 软件进行扫描，并根据需要进行分割以获取睡眠数据。
	<ol>
		<li>将数据分段的起始时间设置为启动监测仪后第三天早晨的 8:01（1 分钟分段）或 8:00（30 分钟分段），终止时间为起始时间之后第三天的上午 8:00（图 2A1）。 
		注意：果蝇必须至少适应监测环境一天。因此，可将分段数据的起始时间设为监测开始后第三天上午 8:00。</li>
		<li>以 1 分钟和 30 分钟为间隔分割数据。将选项 &#34;Bin Length&#34; 更改为 1 分钟，将选项 &#34;Output File Type&#34; 更改为 Channel files，重命名并输出。30 分钟数据的分段方法与上述相同（图 2A2-5）。 
		注意：在以 1 分钟和 30 分钟为间隔进行数据分段时，两个文件的最终重命名应保持一致；否则，后续在 Matlab 处理过程中可能无法读取。如有需要，可在输出后更改文件名以便区分。</li>
	</ol>
	</li>
	<li>使用 SCAMP2020 进行数据处理
	<ol>
		<li>在 Matlab 中打开程序包 SCAMP2020，双击 Vecsey 睡眠与昼夜节律分析 MATLAB 程序（SCAMP）（图 2B）。</li>
		<li>将子文件夹 &#34;Vecsey SCAMP Scripts&#34; 添加至路径中，找到该文件夹内的文件 &#34;scamp.m&#34; 并运行。在随后弹出的窗口中，依次选择处理 1 分钟和 30 分钟的文件夹（图 2C、D）。</li>
		<li>选择一个监测仪，点击 Load individual See Plots 进行预览（图 3A1），并检查出现的图像。取消勾选死亡果蝇对应的通道（图 3A2、图 3B）。</li>
		<li>重复上述步骤，检查所有监测仪。</li>
		<li>根据各通道对应的待测药物对每个监测仪中的通道进行重命名（图 3A3），选择所有监测仪，然后点击 ANALYZE Selected Data 进行分析（图 3A4）。</li>
		<li>保留默认选中选项，点击 Analyze for Chosen Bin，勾选 Export Data，最后点击 GRAPH 30 min Data Types for All Days for Selected Groups and EXPORT All Data 输出结果（图 3C）。</li>
	</ol>
	</li>
	<li>从 CSV 文件中选择名为 s30 的文件，查找每个监测仪对应的均值和标准误数据，备份至 Excel 进行修改和调整，然后粘贴到 GraphPad Prism（见 材料表）中绘制睡眠状态图（图 4A、B）。</li>
	<li>找到名为 &#34;stdur&#34; 的文件，计算每只果蝇在三天内的白天、夜间及总睡眠时间的平均值（图 4A、C）。将数据粘贴至 Prism 软件中完成差异检验并绘制图表。</li>

睡眠是人类生存所必需的重要行为之一，其主要分为两种状态：快速眼动（REM）睡眠和非快速眼动（NREM）睡眠1。NREM睡眠包含三个阶段：N1期（清醒与睡眠之间的过渡阶段）、N2期（浅睡眠）和N3期（深睡眠，即慢波睡眠），代表了从清醒状态逐步进入深睡眠的过程1。睡眠在维持身体和心理健康方面发挥着关键作用2。然而，随着年龄增长，成年人的总睡眠时间、睡眠效率、慢波睡眠比例以及REM睡眠比例均会减少3。老年人相较于慢波睡眠，更多地处于浅睡眠状态，因此更容易在夜间觉醒。随着觉醒次数的增加，平均睡眠时间减少，导致老年人出现睡眠片段化现象，这可能与小鼠中Hcrt神经元过度兴奋有关4。此外，与年龄相关的昼夜节律机制衰退也会导致睡眠时间提前5,6。结合躯体疾病、心理压力、环境因素及药物使用等多种因素，这些变化使老年人更易罹患失眠、快速眼动睡眠行为障碍、发作性睡病、周期性肢体运动、不宁腿综合征以及睡眠呼吸障碍等睡眠障碍7,8。
流行病学研究表明，睡眠障碍与中国老年人的慢性疾病密切相关9，包括抑郁症10、心血管疾病11和痴呆症12。解决睡眠障碍在改善和治疗慢性疾病、提高老年人生活质量方面发挥着关键作用。目前，患者主要依赖苯二氮䓬类药物、非苯二氮䓬类药物以及褪黑素受体激动剂来改善睡眠质量13。然而，长期使用苯二氮䓬类药物可导致受体下调和药物依赖，停药后会出现严重的戒断症状14,15。非苯二氮䓬类药物同样存在风险，包括痴呆16、骨折17和癌症18。常用的褪黑素受体激动剂雷美替胺可缩短入睡潜伏期，但无法延长睡眠时间，且因其广泛的首过消除效应，存在肝功能相关安全性问题19。阿戈美拉汀是一种褪黑素受体激动剂和5-羟色胺受体拮抗剂，可改善与抑郁症相关的失眠，但也存在肝损伤风险20。因此，迫切需要更安全的药物来治疗或缓解睡眠障碍。然而，当前基于分子和细胞实验结合自动化系统与计算机分析的药物筛选策略，成本高昂且耗时较长21。基于结构的药物设计策略依赖于受体的结构与特性，需要对受体三维结构有清晰的认识，且对药物效应缺乏预测能力22。
2000年，依据Campbell和Tobler于1984年提出的睡眠标准23，研究人员建立了简单的动物模型来研究睡眠24，包括 黑腹果蝇，表现出类似睡眠的状态25,26尽管在解剖结构上存在差异 果蝇 以及人类中，许多调节睡眠的神经化学成分和信号通路 果蝇 在哺乳动物睡眠中是保守的，有助于研究人类神经系统疾病27,28. 果蝇 尽管果蝇与哺乳动物的核心振荡器存在差异，该方法仍在昼夜节律研究中被广泛使用29,30,31因此， 果蝇 是研究睡眠行为和开展睡眠相关药物筛选的宝贵模式生物。
本研究提出了一种经济高效且简便的基于表型的方法，利用老龄果蝇筛选治疗睡眠障碍的小分子药物。睡眠调控在 黑腹果蝇 高度保守25，且随着年龄增长而出现的睡眠减少现象可能通过药物干预得以逆转。因此，这种基于睡眠表型的筛选方法能够直观反映药物疗效。我们将待研究的药物与食物混合后饲喂果蝇，利用该系统监测并记录其睡眠行为 果蝇 活动监测仪（DAM）32，并使用 MATLAB 中的开源 SCAMP2020 数据包分析所获取的数据（图1）。统计分析使用统计与绘图软件进行（参见 材料表）。作为示例，我们通过展示利血平的实验数据来证明本方案的有效性。利血平是一种小分子抑制剂，可抑制囊泡单胺转运体，据报道可增加睡眠33该方案为识别治疗衰老相关睡眠问题的药物提供了一种有价值的方法。

High-Throughput Small Molecule Drug Screening For Age-Related Sleep Disorders Using Drosophila melanogaster

睡眠是健康和整体福祉的重要组成部分，但老年人常常面临睡眠障碍的挑战，表现为睡眠时间缩短和睡眠模式碎片化。这些睡眠紊乱还与老年人多种疾病风险增加相关，包括糖尿病、心血管疾病和心理障碍。遗憾的是，目前治疗睡眠障碍的药物常伴随显著的副作用，如认知功能损害和成瘾性。因此，迫切需要开发更安全、更有效的新型睡眠障碍治疗药物。然而，现有药物筛选方法成本高昂且实验周期漫长，仍是制约因素。
本方案描述了一种经济高效且适用于高通量筛选的方法，该方法利用黑腹果蝇（Drosophila melanogaster）作为研究老年个体睡眠障碍的理想模型，因其睡眠调控机制与哺乳动物高度保守。通过向老年果蝇施用多种小分子化合物，可评估其对睡眠障碍的影响。果蝇的睡眠行为由红外监测装置记录，并使用开源数据分析包“2020版睡眠与昼夜节律分析MATLAB程序”（SCAMP2020）进行分析。本方案提供了一种低成本、可重复且高效的睡眠调控筛选方法。果蝇因其生命周期短、饲养成本低且易于操作，是该方法的理想实验对象。例如，所测试药物之一利血平（Reserpine）在老年果蝇中表现出延长睡眠时长的能力，凸显了本方案的有效性。
