培养方法必须始终考虑海藻叶状体的特性和整体形态。Gracilaria gracilis 是一种克隆型分类群，意味着其附着器官可产生多个营养轴。因此可通过碎片化（营养繁殖）实现繁殖，因为每个营养轴均能完全独立于母体叶状体而生存3。克隆型分类群可成功结合简单快速的单步培养方法，通过将叶状体分割成小片段获得大量生物量，这些片段能迅速再生并生长为新的、基因完全相同的个体。在此过程中，单倍体和二倍体叶状体均可使用。尽管该属具有复杂的单双倍体同形三相生活史，但除非需要通过遗传更新种群以获得更优良的品系，否则通常无需进行孢子形成。在此情况下，四分孢子（通过减数分裂形成的单倍体孢子）和果孢子（通过有丝分裂形成的二倍体孢子）均可发育为宏观叶状体，随后可通过营养繁殖进行培养和扩增4。生长周期受环境条件和个体生理状态等多种生物因素（如附生生物的出现及其他生物的附着）共同调控。因此，优化培养条件对于确保高生产力和获得高质量生物量至关重要5。
从海藻（包括G. gracilis）中提取生物活性化合物可通过多种方法实现6,7。提取方法的选择取决于目标化合物的种类、应用目的以及海藻的特性。在本研究中，我们采用溶剂提取法，即使用水或乙醇等绿色溶剂溶解并从海藻生物质中提取生物活性成分。该提取过程可通过浸渍法以一种通用且高效的方式进行，适用于多种化合物的提取。该方法操作简单，应用广泛，通常将生物质浸泡于溶剂中较长时间，一般在室温或略高的温度下进行，并通过搅拌以增强提取效率。在达到预定提取时间后，通过过滤或离心将溶剂与固体物质分离。
水因其安全性、易得性以及与多种食品的良好相容性，常被用作食品应用中的溶剂。水提法适用于提取多糖、肽类和某些酚类等极性化合物，但可能无法有效提取非极性化合物。乙醇也是食品应用中广泛使用的溶剂，可用于有效提取多种生物活性分子，包括酚类化合物、黄酮类以及某些色素。乙醇通常被认为在食品中使用是安全的，且易于蒸发，从而留下目标提取物。值得注意的是，选择提取方法时应综合考虑提取效率、选择性、成本效益以及环境影响等因素。优化提取参数（如溶剂浓度、提取时间、温度和压力）对于从G. gracilis或其他海藻中获得最佳生物活性成分产率至关重要。
研究表明，海藻对多种微生物（包括细菌、真菌和病毒）具有抗菌活性8。这种活性归因于其含有的生物活性成分，如酚类化合物、多糖、肽类和脂肪酸。多项研究已证实海藻对大肠杆菌（Escherichia coli）、金黄色葡萄球菌（Staphylococcus aureus）、沙门氏菌（Salmonella sp.）和铜绿假单胞菌（Pseudomonas aeruginosa）等病原体具有抑制作用9。海藻的抗菌作用机制主要与其所含的生物活性化合物有关，这些化合物可干扰微生物的细胞壁、细胞膜、酶系统及信号传导通路10。这些成分可能破坏微生物的生长、抑制生物膜的形成，并调节宿主的免疫反应。
红藻，也称为红藻门生物，是一类能够对多种微生物表现出抗菌活性的藻类。在这类藻中，G. gracilis 含有多种可能与其报道的抗菌活性相关的生物活性化合物。尽管具体的分子成分可能有所不同，但在 G. gracilis 中已有报道且可能具有抗菌特性的常见化合物类别包括多糖、酚类化合物、萜类化合物以及色素11。然而需要指出的是，这些成分的存在与否及其含量会受到多种因素影响，例如海藻采集地点、季节变化、藻体的生理状态以及环境条件。因此，G. gracilis 中抗菌化合物的具体类别和浓度也可能随之变化。
G. gracilis 还被发现具有抗氧化特性，含有多种酚类化合物，这些化合物已被证明能够清除自由基并减轻氧化应激12。 抗氧化剂有助于保护细胞免受活性氧物种造成的损伤，并具有潜在的健康益处。抗氧化能力可通过不同方法直接评估，包括2,2-二苯基-1-苦肼基（DPPH）自由基清除活性；也可通过总多酚含量（TPC）的定量进行间接评估13。
尽管某种成分被报道具有显著的生物活性，但在评估拟与活细胞或组织接触的天然和合成物质时，对其细胞毒性进行评价仍是不可或缺的。目前有多种测定细胞毒性的方法，每种方法均有其优势与局限性。总体而言，这些方法为评估多种物质对细胞的有害影响提供了丰富的选择，同时也有助于研究细胞损伤和死亡的机制14。
在本研究中，我们采用 Mosmann（1983）提出的 3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四氮唑溴盐（MTT）比色法15该方法通过检测代谢活跃细胞将四唑盐还原为紫色甲臜产物的过程来评估细胞活性。甲臜结晶生成量越多，表明活细胞数量越多，从而间接反映细胞毒性水平14由于在本研究中， G. gracilis 水提物和乙醇提物拟用于皮肤化妆品配方中， 体外 在角质形成细胞（HaCaT）细胞系中进行细胞毒性评估。
关于食品应用，海藻通常热量较低，且富含膳食纤维、必需元素和氨基酸、多糖、多不饱和脂肪酸、多酚以及维生素2,16. G. gracilis 也不例外，具有有趣的营养价值。Freitas 等（2021）4 发现培养的 G. gracilis 蛋白质和维生素C含量更高，且总脂质水平与野生海藻相当。这可能具有经济和环境优势，因为从营养角度来看，人工生产优于对野生资源的开发。此外，消费者日益关注所摄入食物的种类，因此引入新型成分用于食品强化，并利用新资源获取可提升产品附加值并宣称具有特定健康益处的提取物显得尤为重要 &#34;清洁标签&#34; 此外，当前市场竞争激烈，需要开发新产品并制定创新策略，以使制造商能够区别于竞争对手17.
通过海藻等海洋资源强化意大利面等营养价值较低的产品，是一种将此类资源作为新型食品引入市场，并通过具有独特营养价值的产品实现市场差异化的策略。另一方面，G. gracilis 是天然红色色素（如藻胆蛋白）的来源18，在食品工业中具有广阔的应用前景。这种海藻在多个领域均受到广泛关注，其应用形式可包括全海藻、提取物和/或残余生物质。在本研究中，我们展示了若干此类应用的实例。

在解读实验结果时，应注意抑制率越高，表明该提取物对特定菌株生长的抑制效果越强，因而作为抗菌剂的潜力也越大。通过该方法，我们能够快速鉴定出对某些细菌菌株具有较强活性的提取物，同时筛选出在后续应用方面更具研究价值的提取物，从而为针对该提取物的进一步研究提供起点。

我们的研究致力于从海藻中发掘具有潜在应用价值的新生物活性化合物，这些化合物可用于食品、化妆品和制药等多个领域。社会日益认识到海洋生物在提供生物活性化合物方面的潜力。这些化合物可用于开发可持续且更安全的功能性化妆品，提升食品产品的质量，并为新药研发提供新颖的化学结构来源。在开发新型食品时，使用海藻面临的一个挑战是避免使用过量，因为这可能导致食品感官特性不佳。使用海藻提取物是解决这一问题的一种方法。通过本方案，我们能够通过完全添加藻类来提升意大利面的营养价值。通常添加这些成分会带来较为少见的感官特性。然而，采用所开发的配方，仍实现了消费者的可接受性。首先，在气候室中，将江蓠属江蓠（Gracilaria gracilis）培养于250毫升的平底烧瓶中。将生物质扩大培养、干燥并研磨后，称量所得的干燥生物质。将其溶解于100毫升溶剂中，在避光容器中搅拌30分钟。分别在室温下用乙醇和水进行提取。将液体提取物通过滤纸过滤，以分离残余的生物量。在室温下以8,000G离心提取物10分钟。将水相提取物冷冻干燥，并在40摄氏度下使用旋转蒸发仪蒸发乙醇提取物。首先，取自干燥的江蓠（Gracilaria gracilis）中获得的水相提取物和乙醇提取物。在96孔微孔板中，每孔加入2 µL样品和198 µL溶于无水乙醇的DPPH溶液。随后，使用微孔板分光光度计在517 nm处测定吸光度。取在96孔板中培养过夜的人角质形成细胞。将二甲基亚砜溶解的提取物2微升加入到微孔板孔中的198微升培养基中。移除培养基。向细胞中加入100 µL MTT，避光孵育30分钟。为溶解细胞内甲臜结晶，加入100 µL DMSO。使用酶标仪在570 nm处测定吸光度。DPPH 实验证明，40 摄氏度条件下获得的提取物对氧化活性的抑制值高于室温或 70 摄氏度条件下获得的提取物。在人角质形成细胞上未观察到细胞毒性效应，表明在最高酸浓度下，水提物和乙醇提取物均可安全用于皮肤。首先，将预定量的米粉、江蓠（Gracilaria gracilis）和小球藻（Chlorella vulgaris）加入设备中，并向混合物中加入约 30% 的水。混合几分钟后，使用设备挤出面条，然后手动切割。为了制作通心粉（chifferi），先在68摄氏度下干燥42分钟，再在76摄氏度下继续干燥5小时30分钟，以模拟工业化生产过程。最后将样品真空密封，并置于室温下的避光处保存。取一克试样置于带有滤底的玻璃坩埚中，并将其放入纤维分析仪内。向每个坩埚的柱管中加入150毫升预热的1.25%硫酸溶液和2毫升消泡剂。除去溶剂，并用去离子水冲洗坩埚三次，以制备用于碱性水解的样品。用 150 毫升丙酮将坩埚清洗三次后，小心转移至设定为 150 °C 的烘箱中，加热一小时。记录最终重量。将坩埚转移至设定为 500 °C 的马弗炉中，加热三小时，然后记录最终重量。为测定脂肪酸谱，向50毫克样品中加入2毫升甲醇中的2%硫酸溶液。将混合物在80°C下加热2小时。然后将混合物冷却至室温，再向每个样品中加入1毫升超纯水和2毫升正庚烷。将混合物涡旋震荡一分钟后，离心五分钟。随后收集含有脂肪酸甲酯的上层正庚烷相，并转移至气相色谱样品瓶中。使用配备TRFAME毛细管柱、自动进样器和火焰离子化检测器的玻璃色谱仪分析样品。通过将所得保留时间与标准品进行比较，确定脂肪酸谱型。将瓷坩埚在105摄氏度下孵育三小时。在干燥器中冷却后，称量其重量。按照前述方法将10克样品孵育后，使用所示公式计算其水分含量。然后将装有干燥样品的坩埚转移至已加热至525摄氏度的马弗炉中，灼烧4小时。待样品冷却后，记录其重量。为评估消费者接受度，使用蒸馏水煮8分钟的通心粉样品。在感官分析实验室的独立感官检测间内进行感官测试。评价样品的外观、颜色、质地、气味、海味、总体口感、总体评价及购买意愿。该意大利面在1至9级的享乐量表上，对于海洋风味和外观的评分分别为5分和6分。该意大利面的购买意愿评分为5分，总体喜爱度评分为6分。首先，将50毫升磷酸钠缓冲液加入1克江蓠（Gracilaria gracilis）中。匀浆后，使用研钵和研杵研磨样品 10 分钟。将溶液转移至离心管中，在 12,298×g 条件下离心 20 分钟。合并上清液后，缓慢加入 65% 硫酸铵。然后通过离心收集沉淀，并将其溶解于蒸馏水中。使用透析膜在水中对提取物进行透析，持续24小时，随后进行冷冻干燥。为将提取物加入酸奶中，以0.21%的浓度将其与酸奶混合。将样品分装于各自的玻璃瓶中，在4°C条件下保存直至分析。为检测提取物在酸奶中的颜色稳定性，使用白色陶瓷板校准色度计。取约28克样品放入样品池中，分析其颜色。添加色素的酸奶的颜色稳定性在负4摄氏度下评估了12天。将提取物添加至酸奶中后，在贮存12天后表现出良好的色泽保持性，色差为7.01 ± 2.36。
