1. 粪便样本的采集
注意：在兽医或动物主人的协助下对犬只进行操作。
<ol>
	<li>对于野狗（图1A）或紧张的动物，应在排便后立即或排便后不超过10分钟内从地面采集样本。</li>
	<li>用水或凡士林润滑外科手套或薄壁聚乙烯袋。若需从直肠采集粪便样本，应佩戴外科手套或薄壁聚乙烯袋。</li>
	<li>从每只狗采集至少2 g的粪便（图1B）。</li>
	<li>按以下方式标记个体粪便样本：日期、地点（使用Google地图获取全球定位系统[GPS]坐标）、为每只动物分配一个识别编号、狗的大致年龄、性别、品种、室内或室外（野狗）。</li>
	<li>用紧实的结扎方式封好装有粪便样本的袋子。若粪便样本在采集后3-4小时内无法进行分析，应将样本袋冷藏（4-8 °C）。</li>
</ol>
2. 用于现场诊断的饱和盐溶液的制备
注意：如果无法获得或难以使用天平、量具、炉具或燃气来加热水，可使用水、普通食盐、一个塑料制12盎司（355 mL）杯和一个1升装空塑料汽水瓶，轻松制备饱和盐水溶液。
<ol>
	<li>将一个空的1升汽水瓶彻底清洗干净，并用1升水将其装满。</li>
	<li>用常见的食用盐将一个12盎司的塑料杯装满。</li>
	<li>将盐转移到汽水瓶中。</li>
	<li>用螺旋瓶盖将汽水瓶紧密封好，剧烈摇晃溶液直至盐不再溶解。 
	注意：在汽水瓶中摇晃溶液至盐完全溶解可能需要长达90分钟。</li>
</ol>
3. 实验室诊断用饱和盐溶液的制备
<ol>
	<li>称取 420 g 普通食盐。</li>
	<li>将 420 g 食盐溶解于 1 L 水中。</li>
	<li>煮沸溶液直至食盐不再溶解。</li>
	<li>过滤溶液以去除未溶解的食盐。</li>
	<li>使用重液比重计或密度计检测溶液浓度。 
	注意：理想浓度的比重（SpG）应为 1.20，以获得更佳效果（图 2A）。鸡蛋在溶液中的浮力受其与溶液相互作用的影响，因此即使在相同比重的溶液中，不同鸡蛋的浮起能力也可能存在差异。因此，为了实现最佳的虫卵回收效果，必须考虑浮选溶液比重的上限，确保其高于目标寄生虫成分的比重6。</li>
</ol>
4. 漂浮法
注意：如果粪便样本过于干燥或坚硬，可在研钵中进行研磨处理。
<ol>
	<li>使用勺子将约 3 g 粪便放入一个塑料杯中（高度约 8.5 cm，直径约 5.5 cm）。</li>
	<li>加入 1 mL 饱和盐溶液，直至形成糊状物。</li>
	<li>搅拌 1 分钟，然后加入 100 mL 饱和盐溶液。</li>
	<li>将此悬浮液通过塑料滤网过滤至第二个塑料杯中，以去除粗颗粒（图 2B）。</li>
	<li>静置悬浮液 15–20 分钟。</li>

5. 漂浮法的解读
注意：阴性结果无法得出明确结论。

在粪便样本中可发现肠道寄生虫的生命周期阶段，例如卵、包囊、卵囊或幼虫。因此，对粪便样本的检查可为了解动物体内的寄生虫提供重要信息。由于需要一种检测人类粪便中钩口科（Ancylostomidae）虫卵的方法，1878年首次采用了简单的粪便涂片法，该方法曾长期用于检测胃肠道寄生虫，但被认为敏感性较低。因此，开发更优的粪便显微镜检方法的需求应运而生14。自首次描述用于从粪便样本中回收和计数寄生虫卵的漂浮技术以来，已过去一百多年15。此后，多种漂浮技术及其改良方法已被视为检测某些寄生虫在宿主体内存在的标准方法。
例如，Lane 在 1924 年描述了一种直接离心漂浮法，该方法结合了离心步骤，随后将沉淀物置于比重（SpG）为 1.2 的饱和氯化钠溶液中进行漂浮，使用 1 g（Lane 法）或 10 g（Stoll 改良法）的氯化钠。此后，该漂浮法通过使用不同比重的溶液进行了改进14。1939 年，Gordon 和 Whitlock 报道了 Stoll 方法的缺点，即杂质会干扰寄生虫卵的观察，并由此开发了被称为 McMaster 的定量方法16。1979 年，O'Grady 和 Slocombe 证明，漂浮液的比重、操作时间以及过滤筛网的孔径大小均会影响漂浮法检测虫卵的准确性17。近几十年来，由于漂浮技术已出现多种改良方法，因此迫切需要对漂浮法进行标准化。目前，在防控人畜共患寄生虫的背景下，检测犬类蠕虫感染对于合理使用驱虫药物、减少环境被人畜共患线虫感染阶段污染具有重要意义18。
在定性检测方法中，粪便漂浮法因所需设备少、操作简便、成本低廉且可重复性强而被广泛应用和认可；但其主要缺点在于当感染强度较低时，该方法的灵敏度不足19。检出更多寄生虫成分（如虫卵、卵囊、包囊或线虫幼虫）的能力通常取决于漂浮液的密度20。
既往研究已对检测犬类线虫卵的粪便寄生虫学技术进行了比较。针对活动性原虫的检测，通常采用直接粪涂片法；而沉淀法则适用于诊断吸虫等具有较重虫卵的寄生虫21。其中应用最广泛的现场诊断检测方法之一是粪涂片法。然而，该技术灵敏度较低，可能与其含有干扰寄生虫卵检出的粪便杂质有关。通过引入筛滤步骤，并结合具有合适比重（SpG）的漂浮液，漂浮法可更清晰、更少干扰地观察蛔虫和钩虫卵，从而实现更精确、高效的显微镜筛查过程22。同样，简易漂浮法和直接离心漂浮法也常用于回收寄生虫卵和卵囊14。经典的漂浮法可根据是否使用计数室（如麦克马斯特法）而被视为定性或定量方法15。然而，由于漂浮法灵敏度较低，且主要针对排虫期寄生虫的检测，阴性结果不应视为最终结论。此外，检测准确性不仅取决于粪便样本的保存方法或漂浮液的比重（SpG），还依赖于操作人员进行粪便检查的技术熟练程度和经验。
因此，人们已探索了其他用于检测犬类粪便寄生虫的方法。普遍认为，诊断犬肠道线虫感染最广泛使用的方法之一是FLOTAC技术，这是一种多价、灵敏且准确的方法，相较于试管漂浮法和麦克马斯特（McMaster）技术，其在诊断犬钩虫（A. caninum）感染时可提供更准确可靠的结果19,23。沉淀法适用于回收吸虫卵、已胚胎化的线虫卵以及大多数绦虫卵，因为这些结构无法在漂浮液表面被检出，其本身不具备漂浮特性24。其中，改良双离心漂浮法已被证实优于传统的漂浮/沉淀技术，该方法能够有效检测粪便中的绦虫卵，操作更省时，可将无盘属（Anoplocephala ）虫卵与粪便碎片分离，并减少结晶干扰25。此外，该技术已被成功用于高灵敏度地检出蛔虫卵26。然而，上述部分技术以及诸如Ovassay等离心方法，与本研究提出的漂浮方案不同，需要使用福尔马林等试剂保存样本、依赖商业试剂盒、在实验室条件下进行样本处理，并使用硫酸锌等试剂27&#160;，这些试剂价格昂贵，且需特殊处置程序以避免对环境造成毒性危害。
近年来，人们倾向于使用通过添加高比重（SpG）溶液来提高浮聚法灵敏度的技术。然而，必须考虑到这些溶液的缺点是会增加最终样本中的杂质，从而导致寄生虫卵的误检。此外，材料的商业可获得性、试剂成本、环境影响问题以及离心方法操作的复杂性，都会影响浮聚技术的选择14，在野外条件下这种选择可能具有挑战性，相比之下，本文所介绍的方案则更具优势。使用食盐配制浮聚溶液优于使用糖类，因为在野外条件下，糖容易吸引黄蜂、蜜蜂等昆虫，且样本容易变黏。此外，为防止糖溶液黏附而添加的苯酚，或硫酸锌（ZnSO4）等溶液，其废弃处理需遵循环境保护指南，操作复杂，无法在野外直接丢弃；而食盐溶液则无此问题。
本文旨在演示在野外和实验室条件下，通过改良简易漂浮法检测粪便样本中T. canis和Ancylostoma spp. 虫卵的操作步骤。按照本文所述方案，并使用配备备用电池的显微镜，即使在缺乏实验室设备和基础设施的农村及城乡结合地区，也可实现对这些犬类人畜共患寄生虫的诊断。本研究所描述的简易漂浮法可快速获得结果，是一种无创、经济高效的常规筛查技术。

A Simple Fecal Flotation Method for Diagnosing Zoonotic Nematodes Under Field and Laboratory Conditions

在野外条件下，诊断具有人畜共患潜力的犬类寄生虫（如Toxocara canis和Ancylostoma caninum）通常较为困难，原因是在墨西哥的农村和郊区难以获得实验室资源。本研究旨在检测2017年至2021年间在墨西哥野外条件下采集的犬类粪便样本中的T. canis和Ancylostoma spp.。样本量计算结果显示，需在全国范围内纳入534只犬作为研究目标。

达到均匀状态后，将粪便样本通过筛网倒入新的塑料杯中，静置10-15分钟。使用灭菌的接种环从混合物上层取三滴样本。将载玻片置于显微镜下，由经过培训的寄生虫学家鉴定寄生虫。共对1,055只犬的粪便样本进行了显微镜检查。Ancylostoma spp.&#160;阳性样本数为833例（检出率为78.95%），T. canis阳性样本数为222例（检出率为21.04%）。这些结果表明，在实验室和现场条件下，采用粪便寄生虫学检测技术识别墨西哥城乡地区犬类体内的动物源性线虫具有重要意义。
