D. antiqua 采自泰安范镇的田间。
1. 无菌饲料的制备
<ol>
	<li>剥去葱的外层，去除绿色叶子部分，保留葱的白色部分（图1A）并用无菌水清洗，重复冲洗三次。用剪刀将葱白部分切成1-2厘米见方的小块（参见 材料表用75% EtOH溶液（见步骤2.5）灭菌，以备后续使用。</li>
	<li>称取50 g切碎的葱段，放入食品加工机中（参见 材料表）。加入50 mL无菌水，研磨5次，每次2分钟。所有切碎的葱应研磨至呈糊状。图2A).</li>
	<li>使用镊子转移10片3rd 龄幼虫 D. antiqua 加入含有10 mL 1×PBS的10 mL离心管中，使用一次性研磨杵进行研磨（参见 材料表约5分钟，以获得幼虫研磨液。</li>
	<li>将葱糊状液体和幼虫研磨液倒入一个洁净的500 mL锥形瓶中（参见 材料表）。用两层透明封口膜包裹锥形瓶瓶口，然后将锥形瓶置于摇床培养箱中（参见 材料表在25 °C、180 rpm条件下发酵12小时（图2B).</li>
	<li>用剪刀剪出七层边长为10 cm的方形纱布（无需无菌），叠放在一起，制成一个简易过滤装置。将发酵后的葱液缓慢倒入纱布上，挤压收集滤过液至一个新的500 mL锥形瓶中。将纱布上残留的葱渣用锡箔纸包裹，留待后续使用。</li>
	<li>使用真空泵过滤上一步骤中获得的滤液（参见 材料表）和布氏漏斗。将经去离子水润湿的滤纸置于布氏漏斗中，然后倒入滤液，并使用不同的滤纸重复抽滤三次。将最终的滤液收集至500 mL锥形瓶中。</li>
	<li>使用0.22 µm滤膜瓶和真空泵进行再次过滤。在超净工作台中拧紧抽滤瓶的瓶盖（参见 材料表）以供进一步使用。此时，获得发酵后的葱渣及滤液（图2C). 
	注意：此步骤获得的滤液将为无菌状态。（0.22 µm 滤瓶是一种无菌真空过滤系统，用于滤液的除菌。）</li>
	<li>重复之前的步骤，以获得未发酵的葱渣和滤液。此处的不同之处在于无需进行发酵。</li>
	<li>称取0.24 g氯化胆碱和0.56 g L-抗坏血酸（参见 材料表) 于10 mL离心管中。加入3 mL去离子水，剧烈振荡直至完全溶解。在超净工作台中，使用一支新的5 mL注射器，并连接三个0.22 µm注射式滤器（参见 材料表）对溶液进行灭菌，并将其置于无菌的10 mL离心管中备用。 
	注意：氯化胆碱是昆虫生长所必需的神经递质，而L-抗坏血酸则起到防腐作用。</li>
	<li>称取 2 g 琼脂粉和 6 g TSB（见 材料表）倒入一个500 mL锥形瓶中，然后加入100 mL去离子水以配制培养基。用去离子水冲洗玻璃棒后，用其搅拌混合物至充分混匀。随后，用两层透明封口膜密封锥形瓶。</li>
	<li>将已发酵的葱和未发酵的葱（见步骤1.5和步骤1.8）用铝箔纸包裹，连同培养基一起在121 °C高压灭菌器中灭菌20 min。</li>
	<li>在超净工作台中，将灭菌的氯化胆碱、L-抗坏血酸和无菌滤液倒入培养基中，同时轻轻旋转混匀。最后加入发酵与未发酵的葱，手动剧烈振荡以获得无菌饲料。将饲料分装至50 mL离心管（无菌；带0.22 µm PVDF疏水膜通气盖）中（见 材料表）以一定角度（图 2D). 
	&#8203;注意：灭菌后，培养基温度应保持在 65–80 °C 之间，以防止加入葱或其他成分时迅速冷却并凝固。此外，每支试管中加入约 10–15 mL 培养基。培养基凝固后，若不立即使用，应将试管置于 4 °C 冰箱中保存。</li>
</ol>
无菌卵的获取
<ol>
	<li>实验室养殖系统 D. antiqua
	<ol>
		<li>将饲养笼置于25 °C、内置LED光照的培养箱中（24小时为一个周期，光照与黑暗时间为16:8），用于饲养雄性和雌性成虫，使其交配并产卵。填充饮水装置（图1B用水润湿棉絮封口，为成虫提供水分。</li>
		<li>将四个 35 mm × 12 mm 的培养皿底置于一个 94 mm × 16 mm 的培养皿盖中。用去离子水润湿棉絮，然后将湿润的棉絮放置在其中两个培养皿底中，并在棉絮上方加入一汤匙蔗糖。</li>
		<li>用两汤匙蔗糖和酵母提取物填充另外两个培养皿底部（参见 材料表），这些是成虫的食物来源（图1C). 
		注意：步骤 2.1.2 和步骤 2.1.3 中提到的蔗糖和酵母提取物无需灭菌。</li>
		<li>将一个直径94 mm、高16 mm的培养皿底部放入湿润的沙子，并放置一块未生根且已去皮的大蒜瓣（图1D在果蝇培养笼内放置大蒜以收集卵。雌蝇会被大蒜的气味吸引，并在其周围产卵。</li>
	</ol>
	</li>
	<li>卵的收集
	<ol>
		<li>从笼中取出产卵装置（即步骤2.14中所述的含有沙子和大蒜的94 mm × 16 mm培养皿），将沙子与大蒜一同倒入500 mL烧杯中，用自来水冲洗大蒜，使残留的卵冲入烧杯。此时，卵漂浮在水中。</li>
		<li>取一个100目筛网（参见 材料表），并用自来水润湿。然后将含有漂浮卵的水从烧杯倒入筛子中，卵将留在筛子上。</li>
		<li>待筛网上的卵自然干燥后，用刷子轻轻将卵扫入培养皿中。</li>
	</ol>
	</li>
	<li>第一天，冲洗产卵装置以收集卵，无需进行消毒或灭菌。第二天下午四点，再次冲洗卵，并用清水过筛收集。 
	注意：第二天收集卵是为了确保后续各阶段发育速率的一致性。下午四点是产卵的高峰期 D. antiqua因此，此时收集卵子最为理想。</li>
	<li>将收集的卵置于无菌细胞筛上（参见 材料表）置于超净工作台中（图3A).</li>
	<li>制备卵子消毒溶液
	<ol>
		<li>取5 mL 5.2%的NaClO（参见 材料表）转移至一个已灭菌的250 mL锥形瓶中。加入95 mL无菌水，定容至100 mL，得到0.26 %的NaClO溶液。</li>
		<li>配制75%乙醇溶液时，取75 mL 99.7%乙醇，转移至已灭菌的250 mL锥形瓶中，加入25 mL灭菌水，总体积为100 mL，得到75%乙醇溶液。</li>
	</ol>
	</li>
	<li>将30 mL NaClO倒入一个培养皿中，另取30 mL EtOH倒入另一个培养皿中。将步骤2.4中提及的含有卵的细胞筛放入盛有NaClO的培养皿中，在0.26% NaClO溶液中浸泡0.5分钟。随后将筛子转移至盛有EtOH的培养皿中，在75% EtOH溶液中继续浸泡0.5分钟。</li>
	<li>重复此过程三次，每隔0.5分钟交替使用0.26% NaClO和75% EtOH。此后可获得无菌卵。此时，应将盛有卵的筛子浸入EtOH中。 
	注意：为确保彻底灭菌，使用1 mL移液器吸取次氯酸钠（NaClO）和乙醇（EtOH）并分散卵子。重复清洗过程多次。此步骤可进一步清除卵子表面残留的细菌和杂质，确保卵子表面清洁无菌。</li>
</ol>
3. 无菌幼虫的培育
<ol>
	<li>使用经酒精灯灭菌的剪刀，剪去1 mL移液器末端尖端，以转移已灭菌的卵图3B）。确保剪切后的移液器末端开口大于卵的直径（约2 mm）。然后用该移液器将卵连同乙醇溶液一起转移图3C)加入到含有无菌食物的离心管中，每管应包含约20-50个卵（图 3D）。重复此步骤，直至所有卵均已转移。 
	注意：移液器吸头的直径应尽可能大，以避免在转移过程中损坏卵子，否则可能导致卵子死亡。</li>
	<li>使用移液器从离心管中移除多余的乙醇。将盖好盖子的离心管放入自封袋中，并在袋口放置一小块棉花以利于空气流通。最后，将自封袋置于25 °C培养箱中（相对湿度：50%–70%；光周期：16小时光照：8小时黑暗），用于观察与培养。</li>
	<li>大约三天后，饲料中的无菌卵开始孵化（图4A孵化后的幼虫将变得活跃，无菌饲料表面将不再光滑，幼虫将开始取食饲料。</li>
	<li>继续观察幼虫，直至其发育至第2阶段nd 龄幼虫。在此期间持续观察其生长和发育情况。</li>
	<li>9至12天后，超过80%的卵已发育为3rd 龄幼虫（图4B这表明幼虫成功生长和发育。</li>
</ol>
4. 通过依赖培养的检测方法验证无菌幼虫
<ol>
	<li>随机选取三个无菌的3rd 离心管中的龄期幼虫。</li>
	<li>将幼虫放入含有75%酒精的94 mm × 16 mm培养皿中进行清洗。随后，将其转移至盛有无菌水的容器中进一步冲洗。</li>
	<li>使用经75%乙醇溶液消毒的解剖镊夹取幼虫（图中显示头部和尾部） 图5A）。使用解剖剪剪除头部和尾部末端（图5B）。用镊子夹住幼虫的尾部，用另一把镊子从尾部向头部轻轻挤压，以挤出内脏器官（结果如图所示 图 5C）。将内脏置于去离子水中，用镊子轻轻按压脂肪组织以剥离肠管，并将提取的内脏（图 5D) 于无菌水中，以备后续使用。</li>
	<li>将肠道放入无菌的1.5 mL离心管中，加入500 µL无菌1x PBS缓冲液，使用一次性研磨棒将肠组织研磨成匀浆。</li>
	<li>取100 µL匀浆液，接种于营养琼脂培养基中。使用L型涂布棒（参见 材料表用于划线接种琼脂平板。</li>
	<li>将培养皿置于28 °C的生化培养箱中，培养72 h以观察菌落生长情况。</li>
</ol>
5. 采用非培养依赖性检测方法验证无菌幼虫
<ol>
	<li>使用DNA提取试剂盒（参见材料表），按照制造商说明书从无菌幼虫的肠道组织中提取总DNA。</li>
	<li>使用紫外分光光度计测定DNA浓度。</li>
	<li>使用通用细菌16S rRNA引物（1492R: 5'- GGTTACCTTGTTACGACTT -3'，27F: 5'- AGAGTTTGATCATGGCTCAG -3'）进行PCR反应。总反应体积为25 µL，包含1 µL DNA模板、0.5 µL正向引物、0.5 µL反向引物、10.5 µL ddH2O和12.5 µL 2x Taq PCR Master Mix（参见材料表）。PCR反应条件设置如下：94 °C预变性3 min；进行35个循环，每个循环包括94 °C变性30 s、55 °C退火30 s、72 °C延伸90 s；最后72 °C延伸10 min。将PCR产物储存于4 °C，以备后续分析。</li>
	<li>使用通用真菌ITS引物（ITS1: 5'- TCCGTAGGTGAACCTGCGG -3'，ITS4: 5'- TCCTCCGCTTATTGATATGC -3'）进行PCR反应。总反应体积为25 µL，包含1 µL DNA模板、0.5 µL正向引物、0.5 µL反向引物、10.5 µL ddH2O和12.5 µL 2x Taq PCR Master Mix。PCR反应条件设置如下：95 °C预变性5 min；进行35个循环，每个循环包括95 °C变性30 s、55 °C退火1 min、72 °C延伸90 s；最后72 °C延伸10 min。将PCR产物储存于4 °C，以备后续分析。</li>
	<li>使用Super Green核酸染料（参见材料表）将上述两种PCR产物分别均匀混合，并在1%琼脂糖凝胶中以1x TAE缓冲液（由50x TAE稀释而成，参见&#160;材料表）进行电泳分析。使用3 µL DNA Marker（参见材料表）作为参照。</li>
	<li>使用紫外透射仪观察凝胶并定位目标片段。</li>

无菌动物是指无法检测到任何可存活微生物或寄生虫的动物，是研究宿主与微生物相互作用的宝贵实验模型1,2。昆虫作为最大的无脊椎动物类群，能够与微生物形成共生关系3。无菌昆虫可用于研究共生系统中的宿主-共生体相互作用4。例如，Nishide 等人5建立了一种针对臭虫Plautia stali的实用无菌饲养方法，从而能够在模式共生系统中对宿主-共生体相互作用进行可靠且严谨的分析。无菌昆虫可通过对卵阶段进行消毒，并为幼虫和成虫提供无菌食物来获得6,7。无菌昆虫在生物学研究中具有重要意义并被广泛应用。例如，Somerville 等人8开展的一项研究表明，接种了Enterobacter cloacae的小菜蛾可提高转基因雄性个体的适应能力。
Delia antiqua Meigen 是一种在全球范围内对洋葱及其他百合科作物具有重要经济影响的害虫，其幼虫会危害洋葱和其他百合科作物的鳞茎9。D. antiqua 主要分布于温带气候地区，在美洲、欧洲和亚洲的洋葱种植区广泛存在。若未得到适当控制，该害虫可导致洋葱（Allium cepa L.）、大蒜（Allium sativum L.）、小葱（Allium fistulosum L.）和韭菜（Allium schoenoprasum L.）等作物减产50%至100%10,11。幼虫取食植物地下部分，导致幼苗萎蔫并最终死亡。此外，受害植株易受病原菌侵染，引发鳞茎腐烂12。即使植株未被幼虫完全取食，所造成的损伤也会使洋葱失去商品价值，从而造成经济损失。
昆虫与肠道微生物群关系密切，大多数昆虫肠道内含有多种共生细菌，这些细菌依赖宿主提供的营养物质生长繁殖13,14。Jing 等人15研究表明，肠道共生菌群的主要功能是提供必需营养物质，其次为参与消化和解毒相关功能。在某些情况下，肠道细菌可作为害虫管理的微生物资源。因此，研究特定肠道细菌在D. antiqua体内的作用及其具体功能具有重要意义。为此，制备无菌幼虫对于研究特定菌株与昆虫之间的相互作用尤为关键16。目前，消除昆虫肠道细菌的常用方法是使用抗生素组合来清除相关微生物17,18,19。与仅使用抗生素（只能减少微生物数量）不同，无菌饲养昆虫可实现对微生物组成和数量的精确控制，从而更准确地验证肠道微生物群的功能。
因此，本文介绍了一种制备和培育无菌D. antiqua的实验方案。通过高温灭菌天然食物并结合半发酵食物，获得无菌幼虫饲料。卵经实验方案进行表面灭菌以获得无菌卵，最终由无菌卵培养出无菌幼虫。本实验仅对该无菌培养体系维持了一代。该体系将为研究昆虫与肠道微生物群之间的相互作用提供便利。

无菌昆虫是通过使用无菌培养基的无菌人工饲养系统获得的。这类昆虫具有个体小、生长周期短、饲料需求低等特点，非常适合用于研究微生物与宿主之间的关系。肠道微生物群显著影响昆虫宿主的生理特征，将特定菌株引入无菌昆虫体内，可为验证肠道微生物功能提供有效方法。Delia antiqua 是双翅目（Diptera）、花蝇科（Anthomyiidae）、葱蝇属（Delia）的一种重要害虫，主要取食洋葱、大蒜、韭菜等百合科（Liliaceae）植物。其幼虫取食植物鳞茎，导致鳞茎腐烂、植株萎蔫，甚至整株死亡。通过饲养无菌幼虫，可进一步开展研究，观察肠道微生物群对D. antiqua生长发育的影响。与使用抗生素清除共生微生物的方法不同，本文介绍了一种低成本、高效率的无菌D. antiqua饲养方法。在对D. antiqua卵进行表面消毒后，采用半发酵的无菌饲料饲养幼虫，并通过依赖培养和非依赖培养的方法验证了D. antiqua的无菌状态。综上所述，结合昆虫卵的表面灭菌与无菌饲料制备技术，已成功建立了一种高效且简便的获得无菌D. antiqua的方法。该方法为研究昆虫与微生物群落的相互作用提供了有力工具。
