心血管疾病（CVD）是全球主要的死亡原因1。预防和治疗心血管疾病需要深入理解组织对生物力学力的分子适应机制，以及由此引起的力学性质变化。在整个心血管系统中，生物力学力在组织的功能和结构中起着重要作用2。心血管（CV）组织的力学性质受到这些力的影响，因此可作为健康与疾病的指标3,4,5,6。为了预防、诊断和治疗心血管疾病，开发能够理解并观察疾病起始与进展过程的方法至关重要。生物医学成像在获取生理学和机制性认识方面发挥了关键作用，新的成像技术和分析方法也正在不断涌现。本实验方案展示了一种将两种心血管成像与分析技术相结合的方法，以验证这些成像模式在缺血性心脏病和血管老化研究中的潜力。

本实验室采用临床前超声成像技术，研究多种疾病和生理状态下心血管生物力学。我们还利用质谱成像技术，研究心血管和脑组织中脂质的空间分布。我们的目标是明确组织中哪些区域发生了功能和分子层面的变化。尽管常用的分子成像技术（如组织学和免疫组织化学）能够提供分子数据，但其应用受限于当前可用的染料和抗体。相比之下，质谱成像是一种用于多组学研究的非靶向方法，同时能够保持组织的空间完整性。通过我们的技术，可将多组学扩展至包括脂质、糖链和肽类，并可与超声等功能成像技术相结合。通过这些技术，研究人员现在可以将功能成像与分子成像技术相结合。在我们的实验室中，心血管疾病研究主要有两个重点领域，其中之一是研究心肌梗死或癌症治疗引起的心脏重塑，即心脏毒性；第二个重要的心血管研究方向是探讨衰老及其对血管系统的影响。进行心脏四维超声检查时，将小鼠仰卧放置在成像板上。然后将换能器置于支架上的半固定位置，使换能器上的凸起点与屏幕上的蓝点对齐，并朝向小鼠的右侧。将换能器旋转至与小鼠矢状面一致，使凸起的缺口朝向尾侧。在胸腔腹侧表面涂抹足量超声耦合凝胶，以实现换能器与组织间的声学耦合。缓慢下放换能器，直至其接触超声凝胶。使用载物台底部的 XY 调节旋钮进行精确调整，以实现精细对焦。接下来，确保屏幕上的胸骨旁长轴切面包含心尖、左心室流出道和主动脉，并呈水平对齐，以获得准确的成像效果。点击屏幕底部角落的“命名图像”选项，将该图像保存至当前序列中。将探头顺时针旋转90度，以获得胸骨旁短轴切面。确保图像中左心室边界清晰，并可见乳头肌。点击屏幕左上角的立方体图标，以设置四维图像。重置换能器后，将起始位置调整至心尖稍下方，终止位置调整至主动脉弓。将步长设置为 0.08 至 0.13 毫米，帧率设置为 200 至 300 赫兹。在开始扫描前，确保生命体征和心电图信号保持稳定。完成扫描和处理后，开启“保存EKV/4D数据”和“呼吸门控”以进行后期处理。在右下角选择“命名图像”，并在名称中包含小鼠编号。为可视化心脏周期中每个视角的切面，选择“更多控制”，然后选择“载入四维图像”。逐一检查心脏的各个切面，确认心脏中心在整个心动周期中保持稳定。首先，准备用于小鼠组织速冻的铝箔船。使用镊子提起已处死小鼠的皮肤，在颈部上方用剪刀剪开所提起的皮肤以进行血管通路操作，或在胸骨下方剪开以进行心脏通路操作。继续切开皮肤和肌肉层，以暴露血管系统。如需摘取心脏，应切开骨骼以暴露心脏。使用棉签进行钝性分离，将心脏或血管系统与周围组织（包括脂肪）分离。仔细将颈动脉与神经分离。然后使用手术工具取出心脏或血管系统。将颈动脉、心脏和血管系统置于预先标记的铝箔舟中。然后，将样本盒放入液氮中进行速冻。首先，取一个烧杯装入HPLC水，并将其与5毫升和1毫升的注射器一起放置备用。接着，将冷冻切片机的温度设置为零下25摄氏度，并安装刀片。将一对镊子放入冷冻切片机腔室内，冷却，以备安装组织前使用。用注射器抽取5毫升HPLC水，放入冷冻切片机中，使水部分冻结。在注射器内的水完全冻结之前，将其挤出至金属样品头表面，并使其完全冻结。现在，用1 mL注射器吸取HPLC水，并将其放入冷冻切片机中。30至60秒后，取少量部分凝固的水滴加到持样头中央。用镊子迅速将取出的小鼠心脏放入水滴中，并固定位置，直至周围水完全冻结。首先，将载有小鼠心脏的玻片从冷冻箱中取出，放入干燥器中干燥。打开 HTX M3+Sprayer 仪器电源，启动笔记本电脑上的 HTX 软件，在“方法”设置中将喷嘴温度设为 75 摄氏度，流速设为 100 微升每分钟，压力设为 10 psi。在实验记录本中记录样品名称、电离模式、基质、溶剂和浓度。然后，计算达到目标浓度所需的基质量。接着，称取适量的2,5-二羟基苯甲酸用于正离子模式基质。在15毫升离心管中用70%甲醇溶解基质。将基质溶液超声处理10分钟。超声过程中，从干燥器中取出载玻片。打开喷雾器托盘。然后将载玻片放置在左下角，并用胶带固定边缘。选择样品喷雾区域，然后关闭托盘。使用注射器和滤器，将基质溶液吸入注射器中。通过注射器将基质溶液过滤至喷雾仪左侧带有黑色瓶盖的小瓶中。将小瓶放回喷雾仪上指定的位置，并将 D 管路牢固地插入小瓶中。接下来，打开惰性气体并确认喷雾器上的压力表读数为 10 psi。按下“开始”按钮，当喷雾器达到设定温度后，选择闪烁的“开始”按钮以开始喷雾。喷雾完成后，将样品从喷雾器中取出，并放入 MALDI 载玻片架中。使用扫描仪扫描MALDI载玻片架和载玻片。将图像保存至U盘，用于质谱成像。在喷雾器上选择“Wash”选项，并将D管路从基质小瓶移至废液杯。最后，将甲醇喷洒到喷雾器托盘上并擦拭干净。关闭氮气。对梗死心肌组织进行的基质辅助激光解吸电离质谱成像鉴定出质荷比为577.52的分子离子峰，该峰很可能对应于C或D区域中的COHb，提示其参与心肌重塑过程。四维超声图像显示心肌区域的应变幅度小于20%表面积，表现为黄绿色组织，提示梗死区域。在心肌梗死组织的长轴视图中，可见脂质异位分布，这使得组织生物力学与分子组成之间的关联更加复杂。
