所有涉及人单核细胞来源巨噬细胞的操作均遵循《赫尔辛基宣言》（2013年）进行，并符合UNNOBA伦理委员会（COENOBA）的规定。在样本采集前已获得书面知情同意。男性/女性分组分布为13/6，中位年龄为32岁，四分位距（IQR）为18–75岁。既往存在病理状况、合并症或结核病阳性诊断被列为排除标准。试剂与设备的详细信息列于材料表中。
1. 单核细胞来源的巨噬细胞培养与刺激
<ol>
	<li>采血
	<ol>
		<li>使用肝素化注射器通过静脉穿刺从健康供者采集60 mL血液。 
		注意：血液提取必须由生物化学家或采血技术员进行。</li>
		<li>将针头丢弃至锐器处置容器中，所有接触过供者血液的材料均应丢弃至病理废物容器中。</li>
	</ol>
	</li>
	<li>单核细胞分离与巨噬细胞制备
	<ol>
		<li>将血液小心转移至50 mL离心管中，并用生理盐水等体积稀释。</li>
		<li>通过密度梯度介质离心分离获取外周血单个核细胞（PBMCs）。</li>
		<li>从白色晕环区域收集外周血单个核细胞（PBMCs）。</li>
		<li>用生理盐水以400 x g离心洗涤两次 g 10分钟，最后在200 x下洗涤一次 g 在20 °C下孵育15分钟以去除血小板。</li>
		<li>在细胞计数板中确定PBMC的数量，并使用CD14磁珠进行CD14阳性磁选。 
		注意：磁珠分选过程中，请严格遵循试剂盒生产商的说明书操作。此外，可使用流式细胞术检测所分离单核细胞的纯度。</li>
		<li>将分离的细胞重悬于RPMI 1640培养基中，使用细胞计数板测定单核细胞数量。</li>
		<li>接种 500 µL 的 1 × 106每孔加入1 mL经CD14阳性筛选的单核细胞，置于24孔细胞培养板中，无血清条件下培养以促进贴壁。</li>
		<li>2小时后，用预热的RPMI 1640培养基洗涤，去除未贴壁细胞。</li>
		<li>为了分化巨噬细胞，将贴壁的单核细胞在1 mL完全培养基（含L-谷氨酰胺的RPMI 1640培养基，添加10%胎牛血清（FBS）、100 U/mL青霉素和100 µg/mL链霉素）中继续培养16–18小时（过夜）。</li>
		<li>第二天更换培养基。移除原培养基，用1 mL 1×PBS洗涤，加入1 mL完全培养基。</li>
		<li>用超声处理的 M. tuberculosis (M. tuberculosis 抗原（Ags）孵育24小时。使用10 µL的 M. tuberculosis Ags（10 µL = 1 × 106 细菌）每 1 × 106 单核细胞来源的巨噬细胞 
		注意：10 µL的 M. tuberculosis Ags（Mtb Ags）= 1 × 106 细菌，通过OD600定量纳米米 在分光光度计中&#160;以光密度值（O.D.）为1时对应1 × 108 细菌/mL。使用BSL2生物安全柜，以及在含5% CO₂的 atmosphere 中37 °C的培养箱2 所有培养步骤。</li>
	</ol>
	</li>
	<li>SLAMF1 表达的验证
	<ol>
		<li>从单个孔中收集巨噬细胞。将培养板置于冷表面。加入1 mL冷FACS缓冲液（1× PBS + 2% FBS），重复两次，通过上下吹打使细胞脱落。使用流式细胞术专用圆底管收集细胞。</li>
		<li>以 500 x g 离心细胞 g 4 °C 下离心 5 分钟。弃去上清液。</li>
		<li>在4 °C避光条件下，用荧光染料偶联的抗人SLAMF1抗体对细胞染色30分钟。加入1.25 µL抗体后涡旋混匀。</li>
		<li>使用FACS缓冲液洗涤细胞以去除多余的抗体，将细胞沉淀重悬于FACS缓冲液中，然后使用流式细胞仪检测样本。 
		注意：1.25 µL 抗 SLAMF1 抗体相当于 0.0625 µg，用于标记 0.5 x 106 巨噬细胞。然而，强烈建议对抗体进行滴定，并设置相应的同型对照或荧光减一（FMO）对照。</li>
	</ol>
	</li>
</ol>
2. 总细胞蛋白提取物的制备
<ol>
	<li>裂解缓冲液的制备
	<ol>
		<li>配制含有 150 mM NaCl、10 mM Tris、5 mM EDTA、1% SDS、1% Triton X-100 和 1% 脱氧胆酸钠的放射免疫沉淀分析（RIPA）缓冲液。</li>
		<li>在 RIPA 缓冲液中添加 1 mM 苯甲基磺酰氟（PMSF）以及蛋白酶抑制剂，如抑肽酶 A（Pepstatin A）和亮抑酶肽（Leupeptin）。</li>
	</ol>
	</li>
	<li>细胞收集与蛋白提取
	<ol>
		<li>使用 P1000 移液器弃去孔板中的完全 RPMI 培养基，并用预冷的 1× PBS 通过反复吹打收集巨噬细胞。</li>
		<li>将细胞转移至 1.5 mL 微量离心管中。每管收集一个孔的细胞。</li>
		<li>在 4 °C 条件下，以 500 × g 离心 5 分钟，弃去上清液。</li>
		<li>加入 1 mL 预冷的 PBS，重复步骤 2.2.2 和 2.2.3，将细胞收集至同一微量离心管中。</li>
		<li>将细胞沉淀重悬于 100 µL 1× PBS 中，并合并四管细胞。重复步骤 2.2.3。每管最终含有 2 × 106 个巨噬细胞。</li>
		<li>加入 100 µL 添加了抑制剂的 RIPA 缓冲液，冰上孵育 1 小时，每 10 分钟涡旋一次。</li>
		<li>在 14,000 × g 条件下离心 15 分钟。收集上清液，并于 -20 °C 保存直至使用。</li>
	</ol>
	</li>
</ol>
3. M. tuberculosis&#160;通过流式细胞术检测SLAMF1相互作用
<ol>
	<li>蛋白质-细菌交联
	<ol>
		<li>孵育 1 × 106 全灭活 M. tuberculosis 细胞（WCMtb， M. tuberculosis 经γ射线照射灭活）与1×10⁶个细胞的蛋白提取物混合6 在4 °C下，于旋转微量离心管架中孵育巨噬细胞（50 µL）过夜。 
		注意：10 µL WCMtb = 1 × 106 细菌，通过 O.D. 600 定量纳米米 在分光光度计中&#160;以光密度值（O.D.）1视为1 × 108 细菌/mL</li>
		<li>次日，加入500 µL用1×PBS稀释的1%甲醛溶液，在室温（RT）下振荡孵育15分钟。</li>
		<li>加入25 µL用水稀释的0.125 M甘氨酸。室温下振荡孵育5分钟。</li>
		<li>用1 mL 1×PBS在14,000 × g条件下洗涤两次 g 5 分钟。每次洗涤步骤之间弃去上清液。</li>
		<li>将沉淀重悬，并在500 µL含2 mM乙烯二醇双（琥珀酰亚胺基琥珀酸酯）（EGS）交联剂的溶液中孵育，该溶液用冰醋酸与水按1:1体积比配制，室温下孵育1小时。 
		注意：建议在使用时配制试剂。配制EGS时，需将冰醋酸和水按等比例混合，并加热至70 °C。若溶液温度不足，则EGS无法完全重悬。</li>
	</ol>
	</li>
	<li>SLAMF1 染色
	<ol>
		<li>使用500 µL的1x PBS进行第一次洗涤，1 mL进行第二次洗涤，重复步骤3.1.4。</li>
		<li>在4 °C避光孵育30分钟，向微管中加入抗人SLAMF1抗体（同1.3.3步骤），对蛋白-细菌复合物进行染色，随后涡旋混匀。使用适合1 x 10⁶细胞的抗体用量6 细胞（2.5 µL 或 0.125 µg）</li>
		<li>用FACS缓冲液洗涤细胞以去除多余的抗体，将细胞沉淀重悬于FACS缓冲液中，并在流式细胞仪中检测样本（同步骤1.3.4）。 
		注意：进行相应的同型对照或FMO对照。</li>
	</ol>
	</li>
</ol>
4. M. tuberculosis&#160; 抗原标记
<ol>
	<li>将罗丹明B用乙醇重悬，配制成5 mg/mL（1000倍浓缩液）的储存液。保存于-20 °C，待用。</li>
	<li>在1.5 mL微量离心管中，每100 µL结核分枝杆菌抗原（M. tuberculosis Ags）加入1 µL罗丹明B。</li>
	<li>室温下持续振荡（涡旋）孵育45分钟。</li>
	<li>用1 mL 1× PBS在24 °C、14,000 × g 条件下洗涤至少3次，每次5分钟，直至上清液透明。沉淀物（Mtb-R Ags）呈淡粉色。</li>
	<li>用100 µL 1× PBS重悬Mtb-R Ags，恢复至初始体积。 
	注意：建议在无菌条件下操作。确保染色在实验前立即进行。</li>
</ol>
5. M. tuberculosis&#160;通过荧光显微镜检测SLAMF1相互作用
<ol>
	<li>细菌-蛋白交联
	<ol>
		<li>使用圆头手术镊将直径12 mm的圆形盖玻片放入24孔细胞培养板中（每孔1片）。 
		注意：用70%乙醇彻底清洗盖玻片。如需要，可进行高压灭菌或在紫外灯下照射30分钟以灭菌。</li>
		<li>在4 °C条件下，用400 µL含10 µg/mL多聚-L-赖氨酸（Poly-D-Lysine）的溶液孵育盖玻片过夜。用铝箔包裹培养板以避光。</li>
		<li>次日，从冰箱中取出培养板，用1 mL去离子水洗涤盖玻片两次。</li>
		<li>让盖玻片自然干燥。</li>
		<li>将20 µL（2 × 106）Mtb-R Ags加入180 µL 1× PBS中稀释。最终体积200 µL足以完全覆盖一片盖玻片。</li>
		<li>在细胞培养箱中，37 °C条件下用Mtb-R Ags孵育盖玻片1小时。</li>
		<li>弃去剩余液体，用1 mL 1× PBS洗涤两次。 
		注意：在此步骤中，可通过荧光显微镜使用对应红色通道的滤光片检查Mtb-R是否正确黏附于盖玻片上。</li>
		<li>加入400 µL封闭缓冲液（含10%胎牛血清的1× PBS），在室温下振荡（摇床）孵育30分钟。</li>
		<li>用400 µL封闭缓冲液洗涤两次。</li>
		<li>加入100 µL蛋白提取物与300 µL 1× PBS的混合液，在室温下振荡孵育2小时。</li>
		<li>加入400 µL用1× PBS稀释的1%甲醛溶液，在室温下振荡孵育15分钟。</li>
		<li>加入500 µL用去离子水稀释的0.125 M甘氨酸溶液，在室温下振荡孵育5分钟。</li>
		<li>用1 mL 1× PBS洗涤两次。</li>
		<li>加入400 µL 2 mM EGS溶液，在室温下振荡孵育1小时。</li>
		<li>重复步骤5.1.13 
		注意：建议在使用前现配试剂。</li>
	</ol>
	</li>
	<li>SLAMF1染色
	<ol>
		<li>用石蜡膜包裹细胞培养板的盖子。</li>
		<li>在覆盖有石蜡膜的盖子上滴加60 µL预先滴定好的抗SLAMF1一抗（1:75稀释）。</li>
		<li>使用弯头精细手术镊和针小心地从培养板中取出盖玻片。 
		注意：为便于取出盖玻片，可将针尖弯曲成约45°角。</li>
		<li>将盖玻片倒置覆盖在含有抗体的液滴上，在室温、避光条件下孵育30分钟。</li>
		<li>抬起盖玻片，用封闭缓冲液洗涤两次。</li>
		<li>将盖玻片倒置覆盖在新的含有二抗（1:200稀释）的液滴上，在室温、避光条件下孵育30分钟。</li>
		<li>重复步骤5.2.5。</li>
		<li>去除多余液体，将盖玻片置于载玻片上的封片液滴中进行封片。</li>
		<li>使用合适的滤光片在荧光显微镜下观察。 
		注意：建议在无菌条件下操作。抗体应使用封闭缓冲液稀释。操作盖玻片时需小心，避免产生划痕影响显微镜观察。强烈推荐使用适用于荧光成像的封片液。</li>
