然后从磁铁上取下原来的管子。再次涡旋磁珠 30 秒。如前所述，在重悬和孵育之前，将 200 μL 磁珠添加到新转移的细胞悬液中。
将试管放回磁力架上，不带盖子。在室温下孵育 10 分钟后，将含有分离的中性粒细胞的透明细胞悬液转移到新试管中。将从全血中获得的分离的中性粒细胞悬液拉入单个 50 mL 试管中。
使用细胞分离缓冲液将试管加满至总体积为 50 毫升。在打开裂缝的情况下离心，然后弃去上清液，并将中性粒细胞沉淀重悬于 1 毫升细胞分离缓冲液中。再次离心悬浮液后，弃去上清液，将中性粒细胞沉淀重悬于 3 毫升 55% 密度梯度培养基中。
将含有细胞悬液的 3 毫升 55% 密度梯度培养基缓慢分层到预制的密度梯度管上，避免梯度干扰。将试管以 720 G 离心 30 分钟，不要使用隔断装置。离心后，小心处理试管，以免干扰各层。
使用移液管，小心分离含有低密度和正常密度嗜中性粒细胞的单独层，并将它们转移到单独的标记的 15 毫升试管中。要洗去任何残留密度梯度培养基，请将 PBS 添加到每个 15 mL 试管中，直至达到 15 mL。接下来，在室温下以 400 G 离心 5 分钟，并打开裂口。
在进一步实验之前，在血细胞计数器上计数每个级分中的细胞。总嗜中性粒细胞在密度梯度培养基上的分层导致成功形成两个不同的条带。低密度中性粒细胞形成上带，正常密度中性粒细胞形成下带。
中性粒细胞超过每毫升 5-600 万个细胞，导致条带呈弥散性，从而增加亚型混合的风险。低密度中性粒细胞的平均分离纯度为 93.8%，正常密度中性粒细胞的平均分离纯度为 96.3%，其他细胞类型的污染最小。
