1. 轮子设置
<ol>
	<li>在每个转轮的外侧中周位置粘贴一块磁铁。该磁铁将被速度计的传感器检测，以监测转轮转速（VWR）（图 1A）。</li>
	<li>在转轮的对侧位置，粘贴一个与磁铁重量相近的物体（如硬币），以平衡磁铁重量，防止因磁铁重量导致转轮发生不必要的旋转。</li>
	<li>为速度计和转轮制作一个支架。使用白色泡沫板，裁剪出与转轮底座形状相匹配的一块板，并用胶带固定在底座上（图 1B）。</li>
	<li>将新电池装入速度计后，在初始设置过程中通过测量转轮的外周长来设定轮径大小。</li>
	<li>将速度计的传感器粘附在支架平台上。确保速度计的传感器与转轮上的磁铁保持平行，以保证对运动的准确记录（图 1C）。每次磁铁经过传感器时，即计为一次转轮转动。</li>
	<li>用胶带将传感器及其导线包裹起来，以防止小鼠啃咬导线（图 1D）。</li>

2. 仪器设置
<ol>
	<li>使用行为学实验装置，将 40 cm × 40 cm × 35 cm（高）的箱体划分为四个象限，以同时独立测试四只小鼠。
	<ol>
		<li>根据以下尺寸裁剪两块白色泡沫板：长 30 cm，宽 40 cm（图 2A）。</li>
		<li>在泡沫板 40 cm 宽的一侧中间位置进行 20 cm 的切口，以便两块泡沫板相互交叉。</li>

3. VWR
注意：除了速度计指标外，我们还使用了行为追踪软件——ANY-maze行为追踪系统，以提供额外的行为追踪信息。
<ol>
	<li>每天在光照期的同一时间进行VWR实验（上午9点至11点）。将不运动的小鼠放置在相同的实验区域内2小时，但轮子处于锁定状态。使用热熔胶固定跑轮的旋转中心，以限制其转动。</li>
	<li>将小鼠放入VWR实验房间，适应环境30–60分钟。</li>
	<li>重置所有计速器。</li>
	<li>将每只小鼠放置在象限的角落，使其可自由奔跑2小时。</li>
	<li>2小时后，将小鼠放回其原饲养笼，并记录计速器数据。</li>
	<li>在两组实验之间，使用70%乙醇清洁实验区域和跑轮。</li>
</ol>
4. 排序
注意：在自由转轮跑（VWR）5周结束后，根据已发表的方案21进行SORT实验，以检测运动是否改善了海马功能。
<ol>
	<li>在环境光照的房间内，于光照期进行适应性训练和物体位置识别测试（SORT）。在SORT前一天进行适应性训练，将小鼠移入单独的笼子，并将其带入测试房间至少适应30分钟。
	<ol>
		<li>将测试 arena 的一个角落指定为“释放角落”，确保每只小鼠均面朝该释放角落放置，使其自由探索 arena 6分钟。之后将小鼠放回各自的单独笼子，并用70%乙醇清洁 arena。</li>
	</ol>
	</li>
	<li>重复步骤4.1.1两次，使每只小鼠共经历三次适应性训练。完成所有适应性训练后，将所有小鼠放回其原居住笼中。</li>
	<li>24小时后，将小鼠重新带回测试房间，在测试前让其在单独笼子中适应30–60分钟。</li>
	<li>将两个不同的物体以交叉平衡的方式固定在 arena 内，距离两个非释放角落各6 × 6 cm处。</li>
	<li>将每只小鼠面朝释放角落放置，使其自由探索物体10分钟。之后将小鼠放回各自的单独笼子中静置20分钟，并用70%乙醇清洁物体和 arena。</li>
	<li>移动两个物体中的一个，将其重新固定在距离新的非释放角落6 × 6 cm的位置，另一个物体位置保持不变。</li>

Tracking Individual Running Metrics in Mice Using a Voluntary Wheel Running Protocol that Minimizes Social Isolation

啮齿类动物的自主转轮跑动（VWR）用于评估运动表现和耐力，并模拟运动状态。与其他需要厌恶刺激来诱导主动运动的运动范式相比，转轮跑动是一种自主行为。VWR 通常是在小鼠的饲养笼内放置一个跑动转轮。然而，对运动参数的分析通常只能在单只饲养的小鼠中进行，而群体饲养的小鼠则可用于报告整个笼内群体的效应。因此，现有的模型迫使研究者必须在单只饲养条件下测量个体运动参数，或在群体饲养条件下测量群体效应之间做出选择；根据具体的研究问题和实验模型，这种限制可能会影响实验设计。

我们的目标是开发一种高效且经济的自主转轮跑动方案，能够在最小化社会隔离影响的同时，测量小鼠个体的跑动参数。目前自主转轮跑动（VWR）方案存在的不足在于，无法在避免隔离诱导应激或不依赖笼内整体表现数据的情况下，准确测量小鼠个体的跑动指标。本方案的优势在于提供了一种适用于多种研究场景的高效自主跑动模型，无需单独饲养，同时能够精确追踪小鼠个体的跑动数据。该方案可使应激实验范式在探索运动对群养小鼠行为影响的同时，精确测量个体指标，而不会引入单独隔离饲养所带来的应激反应。首先，将胶水涂抹在每个转轮外侧的中周位置，并固定一个磁铁。该磁铁将被速度计的传感器检测，以监测小鼠的自发转轮跑动。在轮子的另一侧，粘贴一个重量相似的物体（例如硬币），以平衡磁铁重量带来的影响，防止不必要的旋转。裁剪一块与轮子底部形状相配的白色泡沫板，用防水胶带纸将其固定，作为速度计和轮子的安装平台。将一组新电池插入速度计中。然后在初始编程期间，通过测量车轮的外周长来设置车轮尺寸。将速度计传感器粘贴到平台表面。确保将传感器与轮子的磁铁平行放置，以实现对运动的准确记录。使用胶带将传感器和导线包裹起来，防止小鼠啃咬导线。设置包含盒子的行为学装置，以便同时对小鼠进行测试。然后将白色泡沫板裁切成两块，每块长30厘米、宽40厘米。在泡沫板宽度方向的中点处切开一条20厘米长的切口，以便两块泡沫板能够交叉连接。随后用防水胶粘纸包裹泡沫板表面，便于在不同实验组之间进行清洁。使用两块相交的泡沫板将装置箱划分为四个象限。现在将一个准备好的轮子放置在每个象限的角落。用胶带将导线固定在箱壁上，并将速度计安装在透明有机玻璃装置的侧面。对于不进行运动的对照小鼠，使用热熔胶固定跑轮，使跑轮的旋转中心无法转动，从而禁用其旋转功能。对于自主跑轮运动实验，将小鼠放入自主跑轮运动房间内适应环境30至60分钟，然后在开始实验前重置所有速度计。将每只小鼠放置在其指定象限的角落，使其能够自由奔跑。两小时后，将小鼠放回其原饲养笼，并记录速度计数据。每组实验之间使用70%乙醇清洁活动区域和跑轮，以防止气味污染。小鼠在第二周表现出最高的自主跑轮活动水平，平均在跑轮上活动2,135.87秒，移动距离达1.16公里，跑速为2.25公里/小时。在第六周记录到最低的自主转轮跑动活动，小鼠在转轮上平均花费 1,136.48 秒，移动距离为 0.56 公里，平均速度维持在 2.21 公里/小时。经过五周的自主转轮跑动后，小鼠在空间物体识别测试中的探索时间增加了 26.87%，表明海马功能得到改善。
