冷冻保存的精液样本由墨西哥相关兽医主管部门授权的商业供应商提供，已在液氮中保存超过19年，符合墨西哥官方标准NOM-027-ZOO-1995对家畜精液动物卫生处理的规定。所有精液样本，无论是否用于实验或不适合分析，均按照机构生物安全规程及适用法规进行灭活和处置，包括遵循墨西哥NOM-087-ECOL-SSA1-2002标准对生物感染性废物的管理要求。残留的氢氟酸（HF）在通风橱中安全中和：将其缓慢转移至塑料容器，并逐步加入少量CaCO₃3&#160;或 Ca(OH)2&#160;直至不再产生气泡，形成不溶性CaF2中和后悬液的pH值经确认在6至8之间，随后根据当地法规作为无机化学废弃物进行处理。所用试剂和设备列于下文 材料表.
1. 体细胞分离用微量移液针的制备
<ol>
	<li>选择硼硅酸盐玻璃毛细管（外径 1.0 mm × 内径 0.78 mm × 长度 100 mm）。</li>
	<li>使用微量移液器拉制仪制备尖端细小的毛细管。调节切割点以改变尖端直径。</li>
	<li>将毛细管插入拉制仪并牢固固定。</li>
	<li>拉制完成后小心取出微量移液器，并水平放置于显微操作仪上。</li>
	<li>在显微镜视野内对准加热器与微量移液器。</li>
	<li>使用目镜测微尺确定切割位置（图 1A、B）。</li>
	<li>使微量移液器尖端接触加热丝上的玻璃珠。</li>
	<li>逐渐升高温度，直至尖端熔融进入玻璃珠。
	<ol>
		<li>关闭加热器。</li>
		<li>使尖端附着于玻璃珠并自然分离。 
		注：无需弯曲微量移液器，可直接使用其直形状态。但若操作者需要，也可形成一定角度（图 1C）。</li>
	</ol>
	</li>
	<li>不要打磨尖端，此步骤在本实验中非必需。</li>
	<li>通过依次将每个尖端浸入以下溶液约 10 秒来清洗移液器尖端：
	<ol>
		<li>20% 氢氟酸（HF）。 
		&#8203;警告：氢氟酸具有高度毒性和腐蚀性。请使用供应商建议的个人防护装备，特别强调佩戴眼部/面部防护装置、手套和实验服，并始终在通风橱内操作。</li>

	</li>

	<li>将微量移液器储存于无菌容器中。</li>
</ol>
2. 微吸入装置
<ol>
	<li>检查微吸管系统是否处于正常工作状态，确保微吸管操纵杆位于控制面板中央。</li>
	<li>对于使用油的系统，需确认系统内部无气泡，因为气泡可能影响移动精度。</li>
	<li>将体细胞吸取用微吸管插入操作者惯用手一侧的持针器中。 
	注意：在体细胞采集过程中，若微吸管尖端发生堵塞，应更换为新的微吸管。</li>
</ol>
3. 冷冻保存的马精液的解冻与稀释
<ol>
	<li>从液氮中取出麦管，暴露于空气中10秒，然后在37 °C下解冻30秒。</li>
	<li>第一次精液稀释：取解冻后精液样品（250 µL），加入含10%胎牛血清、0.1&#8239;mg/mL牛血清白蛋白、12.5 µM β-巯基乙醇和1%抗生素-抗真菌剂的DMEM/F12培养基1.0 mL进行稀释。
	<ol>
		<li>将混合物在37 °C孵育30分钟。</li>
	</ol>
	</li>
	<li>第二次精液稀释：取第一次稀释液25 µL，加入含0.1 mg/mL聚乙烯醇（PVA）或聚乙烯吡咯烷酮（PVP）的1x DPBS溶液975 µL中，转移至1.5 mL微量离心管中，轻轻混匀。
	<ol>
		<li>在整个细胞收集过程中，将此混合液保持在37 °C。</li>
	</ol>

</ol>
4. 准备用于收集 SC 的培养皿
<ol>
	<li>使用两个直径为35 mm培养皿的皿盖收集精原细胞（SCs）。</li>
	<li>按以下步骤准备第一个皿盖（皿盖#1，图2A）。
	<ol>
		<li>取100 µL第二次精液稀释液，滴加于皿盖中央（见图2A）。</li>
		<li>在第一滴液滴上方再制备一个微滴（10 µL），该微滴为添加了0.1 mg/mL PVA或PVP的1x DPBS（见图2A）。</li>
		<li>用石蜡油完全覆盖这两个微滴，以防止蒸发，并在吸取精原细胞过程中维持温度稳定。</li>
	</ol>
	</li>
	<li>准备第二个皿盖用于去除细胞碎片及伴随的精子细胞（皿盖#2，图2B）。
	<ol>
		<li>在皿盖中央放置三滴30–50 µL的1x DPBS，每滴均添加0.1 mg/mL PVA或PVP；不要用矿物油覆盖。 
		&#8203;注意：仅在所有体细胞均已筛选完毕后才准备此培养皿，以避免DPBS溶液因暴露于空气中而发生理化性质改变。</li>
	</ol>
	</li>
	<li>所有显微注射操作均应在室温下倒置显微镜下进行。避免温度骤变。</li>
</ol>
5. 从冷冻保存的马精液中分离精原干细胞的微吸管操作步骤
<ol>
	<li>将第一个皿盖（皿盖 #1，图 2A）放置在显微镜载物台上，准备收集体细胞（SCs）。</li>
	<li>选择合适直径的显微操作毛细管用于收集体细胞，并将其连接至机械抽吸系统。</li>
	<li>小心地将毛细管插入微滴中，并使其与培养皿盖底部对齐。一旦毛细管吸入培养基，即可开始逐个捕获体细胞。</li>
	<li>在培养皿底部，应可观察到多种细胞形态（图 3）。</li>
	<li>将毛细管尖端置于每个体细胞旁，在负压下进行抽吸。无论细胞大小，均需吸取消除所有圆形和伸长形细胞。每次捕获体细胞时的抽吸体积可能有所不同。</li>
	<li>捕获10个细胞后，将其转移至上方的微滴（10µL）中（图 2A）。重复此步骤，直至获得克隆实验所需数量的体细胞。</li>
	<li>在整个收集过程中，将所有捕获的体细胞保留在该微滴中。</li>
	<li>当获得克隆所需的体细胞数量后，使用微滴进行简单的洗涤操作，以去除伴随的精子、细胞碎片及微生物污染物。</li>
	<li>取第二个培养皿盖，对体细胞进行三步连续洗涤（皿盖 #2，图 2B）：
	<ol>
		<li>用显微毛细管捕获体细胞，并小心地将其转移至第一滴液表面。静置3–5分钟（图 2B；滴液 #1）。</li>
		<li>重复步骤5.9.1，将细胞转移至第二滴液中（图 2B；滴液 #2）。</li>
		<li>重复步骤5.9.1，将体细胞转移至第三滴液中（图 2B；滴液 #3）。</li>
	</ol>
	</li>
	<li>最后一步，将分离后的体细胞转移至一个最终的微滴中，用于后续体细胞核移植操作（图 4D）。 
	注意：每次重新捕获体细胞时，应尽可能以最小体积的培养基进行抽吸，以最大限度减少精子、碎片及微生物污染物的残留。</li>

A Simple Microaspiration Technique for Isolating Somatic Cells from Cryopreserved Equine Semen as Nuclear Donors for Cloning
