将磁性分离柱放置在磁铁上。向柱中加入0.5毫升洗涤缓冲液，使其完全通过。将一毫升的重悬细胞转移到柱上，让缓冲液一滴滴一滴通过。
清洗后，将柱转移到微型离心管中。然后将一毫升洗涤液移液移入色管。现在把活塞插入柱子顶部。
轻轻施加压力以洗脱细胞。然后取下活塞，将柱子放到新的微型离心管上。再往柱中加入一毫升洗涤缓冲液。
然后再次插入活塞，轻轻施加压力以洗出剩余细胞。将两根微型离心管分别用800克离心三分钟。将两管的细胞颗粒重新悬浮到一毫升的冷PBS中。
保持细胞悬浮液冷藏。将纯化细胞重新悬浮在由1%胎儿牛血清制成的标记缓冲液中，浸泡在PBS中。将250微升悬浮液转移到1.5毫升微离心管中。
将针对中性粒细胞膜分子的相应抗体加入试管。然后在四摄氏度下培养细胞30分钟，避免光线。现在用移液器吸一毫升PBS到管子里。
将管子放入微型离心机中，以800克旋转三分钟。抽取上清液。敲击管子以破坏颗粒。
并将细胞重新浸泡在0.5毫升的1%对甲醛中。然后将细胞保持在四摄氏度，保护光线，直到流式细胞术分析。利用流式细胞术分析样本，每个样本捕获10,000个事件。
健康个体中低密度中性粒细胞约占外周血单核细胞的5%，而描述的磁隔离方案中低密度中性粒细胞恢复率约为98%。中性粒细胞表达膜标记CD10、CD11b、CD15、CD62L和CD66b。磁纯化的低密度中性粒细胞表达与中性粒细胞相同的膜标记。
纯化的低密度中性粒细胞表现出多叶核，大小与中性粒细胞相似。中性粒细胞和低密度中性粒细胞均在PMA刺激下产生活性氧，低密度中性粒细胞产生的活性氧水平高于中性粒细胞。低密度中性粒细胞在PMA刺激下释放中性粒细胞外陷阱，这一点通过DNA与弹性蛋白酶和瓜氨酸共定位得到证明。
