在基础研究中，特别是在小鼠模型中，评估自发步态和运动活动是一种广泛使用的方法，用于表征运动表型，并评价生理干扰和治疗干预的影响。对运动参数进行客观量化，能够检测出仅靠肉眼观察难以可靠识别的细微运动变化，这一局限性在针对早期或轻度表型的临床前研究中尤为突出5。在此背景下，步态分析已成为研究运动行为改变、识别不同实验条件下（如神经系统疾病和神经退行性疾病）相关功能变化的重要工具6。
对小鼠模型运动行为和步态的分析传统上依赖于专门的技术平台，用于量化空间和时间运动学变量。常用系统包括 illuminated walkways、instrumented platforms 以及商业化的运动追踪软件；然而，这些系统对昂贵硬件和专用基础设施的依赖显著限制了许多实验室的可及性7,8,9,10。除经济因素外，这些方法还引入了额外的实验限制。一些系统要求动物穿越狭窄的通道或高度照明的路径，导致强制性的运动模式，从而减少自发行为；而另一些系统则依赖于物理标记、颜料斑点或附着在肢体上的粘性标签。此类干预可能引发应激反应，改变步态力学，并干扰对运动结果的生理学解释11,12,13,14。总体而言，这些局限性凸显了亟需发展能够在自然、自由活动条件下捕捉步态的方法，且无需依赖侵入性操作或昂贵的基础设施。
在无约束运动过程中评估步态能够观察到更为自然的运动模式，避免了人为调节速度、轨迹或姿势的外部限制。该方法有助于在最小约束条件下表征运动行为。DeepLabCut 是一个基于深度学习的开源姿态估计算法框架，利用卷积神经网络追踪用户自定义的解剖学标志点，无需使用物理标记物。通过从标准视频记录中实现解剖点的追踪，DeepLabCut 能够提取运动学参数，同时显著降低实验成本与复杂性15,16,17,18。重要的是，无标记追踪最大限度地减少了实验干扰，适用于自发行为的研究。这种单摄像头无标记方法最适合在自由活动条件下，对自发运动活动及二维步态相关特征进行简便、非侵入性的量化研究。然而，当需要获取直接的生物力学变量（如地面反作用力、足底压力、关节角度或完整的三维肢体运动学）时，则需使用仪器化步态平台或三维运动捕捉系统。

A Low-Cost Markerless DeepLabCut-Based Workflow for Spontaneous Gait and Locomotion Analysis in Freely Moving Mice
