Genetics of Organisms

Ein Erbe ist etwas, das von einer Generation zur nächsten weitergegeben wird. In manchen Zusammenhängen bedeutet das Dinge wie Häuser und Geld. Im Kontext der Biologie wird die Erforschung von Genen und deren Vererbung jedoch als Genetik bezeichnet. Gregor Mendel gilt als Vater der modernen Genetik. Seine Arbeit ist verantwortlich für unser Verständnis davon, wie auffällige körperliche Merkmale oder Phänotypen von einer Generation an die nächste weitergegeben werden. Bekanntlich untersuchte er solche Eigenschaften bei Erbsenpflanzen. 
Die Informationen, die diese Phänotypen steuern, werden Gene genannt. Da es von jedem Gen zwei Kopien gibt, die als Allele bezeichnet werden, können wir diese als Buchstaben darstellen. Hier verwenden wir den Buchstaben P. Dies ist der erste Schritt von Mendels berühmtem Experiment zur Färbung von Erbsenblüten, das hier mit einem Werkzeug namens Punnett-Quadrat dargestellt wird. Mendel fand heraus, dass, wenn er violette Blüten mit weißen kreuzte, alle Nachkommen oder die Pflanzen der ersten Generation violette Blüten hatten. Dies liegt daran, dass die violette Farbe dominant ist, was durch den Großbuchstaben P dargestellt wird. Und wenn nur ein dominantes Allel vorhanden ist, bedeutet dies, dass der Phänotyp exprimiert wird. Aber interessanterweise, als diese violetten Blüten wieder gekreuzt wurden, waren 1/4 von ihnen weiß. Wo waren die weißen Blüten in der ersten Generation? Alle violett blühenden Pflanzen der ersten Generation waren heterozygot, was bedeutet, dass sie ein großes violettes P und ein kleinbuchstabiges oder weißes P hatten. Als sie ihre Allele in der F2-Generationenkreuzung weitergaben, bedeutete dies, dass 1/4 der Nachkommen zwei kleine P-Allele erhielten und somit den weißen Phänotyp exprimierten. Mendel hatte das Punnett-Quadrat-Werkzeug nicht zur Hand, er musste all dies herausfinden, indem er Tausende von Pflanzen im Auge behielt und dann Muster in ihrer Anzahl bemerkte. 
Aus diesen und noch viel mehr wissen wir jetzt, dass Gene auch in anderen Organismen, wie Menschen und Fliegen, in zwei Kopien vorhanden sind. Im Anschluss an Mendels Arbeit stellten mehrere Wissenschaftler fest, dass nicht alle Vererbungsmuster dem einfachen, grundlegenden Modell folgten, das Mendel vorschlug. Bei Hämophilie, einer genetischen Gerinnungsstörung, waren beispielsweise nicht betroffene Mütter in der Lage, die Krankheit auf ihre männlichen Kinder zu übertragen. Der Grund dafür liegt in den Chromosomen, die von Thomas Hunt Morgan mit seiner berühmten Fruchtfliege Drosophila untersucht wurden. Dank Morgan und anderen wissen wir heute, dass Chromosomen lange DNA-Stränge sind, die typischerweise paarweise existieren. Hier können wir sehen, dass Drosophila vier davon hat. Auf diesen Chromosomen befinden sich Gene für verschiedene Merkmale, ähnlich wie ein Kochbuch viele verschiedene Rezepte enthält. Heutzutage können wir mit Hilfe der modernen Mikroskopie diese Chromosomen tatsächlich sehen und sogar organisieren. Das Produkt dieses Prozesses wird als Karyotyp bezeichnet. Hier sehen Sie einen Menschen. Sowohl beim Menschen als auch bei der Fliege gibt es Autosomen und Geschlechtschromosomen. Der Mensch hat, wie die Fliegen, X- und Y-Chromosomen, die sein Geschlecht steuern. Die meisten Gene auf diesen Chromosomen steuern jedoch Dinge, die nichts mit Sex zu tun haben. Bei der seltenen Form der Hämophilie, die wir bereits erwähnt haben, liegt der Grund dafür, dass sie häufiger bei Männern auftritt, darin, dass der Phänotyp durch ein Gen gesteuert wird, das sich auf dem X-Chromosom befindet, in einem Abschnitt, der keinen Partner auf dem Y-Chromosom hat. Wenn eine Frau eine schlechte Kopie des Gens hat und ihr männliches Kind diese Kopie erbt, wird es die Störung haben, es hat keine Sicherungskopie des X-Chromosoms. Da eine Kopie des Gens ausreicht, damit eine Person normal gerinnt, muss eine Frau zwei schlechte Allele des Gens erben, eines von jedem Elternteil, um die Krankheit zu zeigen. Was in diesem Fall nicht möglich ist, da der Vater davon nicht betroffen ist. Infolgedessen betrifft diese Art von Hämophilie mehr Männer als Frauen. 
In diesem Labor werden wir uns die Vererbung bei Drosophila ansehen. Die Augenfarbe bei Fliegen wird durch eine Reihe von Genen gesteuert, von denen einige steuern, welche Arten von Pigmenten hergestellt werden, und ein besonders wichtiges Gen, der sogenannte ABC-Transporter, der den Transport von Pigmenten in die Granulate im Auge steuert. Wenn dieses Gen kaputt ist, ist dieses Pigment unsichtbar, selbst wenn die Fliege Pigmente produziert, und die Fliege hat weiße Augen. Die Laborübung besteht darin, eines der berühmtesten Experimente von Thomas Hunt Morgan nachzustellen und das genetische Vererbungsmuster des Gens zu erforschen, das für den Pigmenttransporter kodiert. Wird sie vererbt wie die violette Farbe von Mendels Erbsenblüten, oder ist sie geschlechtsgebunden, wie Hämophilie?

<h4>Mendelsche Genetik</h4>
Die Evolution wird durch Veränderungen in der genetischen Zusammensetzung von Populationen verursacht. In der Populationsgenetik modellieren Wissenschaftler diesen Prozess als Veränderungen in der Häufigkeit von Allelen an einzelnen genetischen Orten. Diese einfache Darstellung des Ablaufs der Evolution geht auf Gregor Mendels Analyse der Merkmalsvererbungsmuster bei Erbsenpflanzen zurück, die erstmals 1865 vorgestellt wurde. Mendel stellte anhand einer rigorosen Datensammlung fest, dass auffällige Merkmale durch zwei Allele jedes Gens (er nannte sie Faktoren) gesteuert werden, eines von jedem Elternteil. Darüber hinaus kam er zu dem Schluss, dass einige Allele gegenüber anderen dominant sind (als rezessiv gelten). Interessanterweise wurden Mendels Daten erst um 1900 gewürdigt, wahrscheinlich zum Teil, weil sein Ansatz zur Erforschung der Vererbung so unterschiedlich von seinen Vorgängern war. Selbst nachdem seine brillanten Experimente wiederentdeckt worden waren, dauerte es weitere 30 Jahre, bis die Mendelsche Vererbung in die Evolutionstheorie integriert wurde und universelle Akzeptanz fand.
Warum schlummerte ein so scheinbar klarer und unkomplizierter Prozess so lange und stand dann jahrzehntelang im Mittelpunkt von Kontroversen? Ein Schlüssel zu der Kontroverse ist, dass wir die von Mendel beschriebenen klaren Vererbungsmuster nur selten sehen. Tier- und Pflanzenzüchter sehen am häufigsten Beweise für das, was als Vermischung von Vererbung oder Nachkommen beschrieben wurde, deren Phänotypen zwischen denen ihrer Eltern liegen, und nicht für die diskrete Trennung von Merkmalen in klar definierte Kategorien, wie glatte versus runzelige Erbsen oder rote versus weiße Blüten. Die Mendelsche Vererbung stimmte also nicht gut mit der praktischen Erfahrung überein, und es dauerte Jahrzehnte, um beides in Einklang zu bringen. Darüber hinaus ist inzwischen bekannt, dass es Vererbungsformen gibt, die auf den ersten Blick gegen die Mendelschen Gesetze zu verstoßen scheinen. Tiefergehende Untersuchungen der Genetik mit Hilfe der Molekularbiologie haben diese neuartigen Vererbungsformen effektiv charakterisiert. Die neuen Informationen entkräften Mendels Ergebnisse nicht, sondern verbessern lediglich sein Modell.
<h4>Phänomene, die gegen Mendelsche Regeln verstoßen</h4>
Es gibt drei Hauptphänomene: Epistase, Pleiotropie und Geschlechtsbindung, die die grundlegenden Dominanz- und Rezessivitätsregeln der Mendelschen Vererbung zu verletzen scheinen. Epistase tritt auf, wenn zwei oder mehr Gene oder Allele interagieren, um einen Phänotyp zu beeinflussen. Die Funktion eines Genprodukts kann erforderlich sein, um die Expression eines anderen Gens zu ermöglichen oder normal zu funktionieren. Auf der anderen Seite tritt Pleiotropie auf, wenn ein Gen die Expression mehrerer Phänotypen in einem Individuum steuert. Zum Beispiel ist ein großer Teil der Katzen mit weißem Fell und blauen Augen taub 1. Pleiotropie ist oft antagonistisch, was bedeutet, dass dasselbe Gen vorteilhafte Veränderungen in einer Facette des Phänotyps eines Individuums verursacht, während es gleichzeitig schädliche Veränderungen in einem anderen Aspekt des Phänotyps eines Individuums verursacht. Antagonistische Pleiotropie wird als Kompromiss oder Einschränkung der Evolution angesehen, wie von George C. Williams vorgeschlagen. Zum Beispiel wird angenommen, dass der altersspezifische Leistungsabfall oder die Seneszenz ein pleiotropes Merkmal ist, das von denselben Genen gesteuert wird, die die Fruchtbarkeit im frühen Leben erhöhen2,3.
<h4>Geschlechtliche Verknüpfung</h4>
Die Geschlechterbindung ist ein weiteres Phänomen, das als Gegenbeispiel zur grundlegenden Mendelschen Vererbung entdeckt wurde. In vielen Organismen bestimmen Paare von Geschlechtschromosomen das Geschlecht. Beim Menschen und vielen Säugetieren sind XX-Individuen weiblich, während XY-Individuen männlich sind. Männliche Nachkommen erben ein X von ihrer Mutter und ein Y von ihrem Vater; Weibchen erben ein X von ihrer Mutter und ein X von ihrem Vater. Bei den meisten Arten beträgt das Verhältnis von Männchen zu Weibchen bei der Geburt unter "normalen" Umweltbedingungen 1:1. Y-Chromosomen rekombinieren sich während der Meiose nur wenig mit X-Chromosomen, da sie im Laufe der Evolution eine beträchtliche Anzahl von Genen verloren haben. Interessanterweise gibt es nur 16 Gene, die von den menschlichen X- und Y-Chromosomengeteilt werden 4.
Bei X-chromosomalen Merkmalen (Merkmale, die von Genen auf dem X-Chromosom kodiert werden) können Frauen homozygot (mit zwei Kopien desselben Allels) oder heterozygot (mit zwei verschiedenen Allelen) sein. Wenn das Merkmal rezessiv ist, zeigen Weibchen den Phänotyp für das Merkmal nur, wenn sie homozygot sind. Heterozygote Frauen "tragen" das rezessive Allel, exprimieren aber nicht den Phänotyp dieses Allels. Statistisch gesehen wird die Hälfte ihrer Söhne dieses rezessive Allel erben und alle werden den Phänotyp exprimieren, da Männer nur ein X-Chromosom haben.
Ein häufig verwendetes Beispiel für einen X-chromosomalen Phänotyp ist die Hämophilie, eine Krankheit, die die Synthese von Blutgerinnungsproteinen verhindert. Wenn sich ein Weibchen heterozygot (Träger, Genotyp XH Xh) mit einem normalen Rüden (Genotyp XH Y) paart, sind 50% der Söhne betroffen. Wenn das Weibchen stattdessen homozygot für das Merkmal wäre (Genotyp Xh Xh), wäre keine ihrer Töchter bluter, aber 100% ihrer Söhne wären es. Das unterschiedliche phänotypische Verhältnis, das bei Söhnen und Töchtern beobachtet wird, scheint gegen die Mendelschen Gesetze zu verstoßen, aber Mendels erstes Gesetz gilt immer noch; Jeder Nachkommen hat eine 50%ige Chance, ein X-Chromosom zu erben, das das Hämophilie-Allel von einer heterozygoten Mutterträgt 5.
Die Fruchtfliege Drosophila melanogaster verfügt wie der Mensch über ein XY-Geschlechtsbestimmungssystem. D. melanogaster eignen sich hervorragend als Labororganismen, da sie leicht zu halten, zu züchten und zu manipulieren sind. Wildtyp-Fliegen haben eine normale Körpermorphologie und rote Augen. Viele Linien sind leicht käuflich zu erwerben, darunter auch einige mit Mutationen, die zu Fliegen ohne Augen, mit unterschiedlich farbigen Augen oder fehlenden Flügeln führen. Diese Organismen können verwendet werden, um sowohl Mendelsche als auch nicht-Mendelsche Vererbungsmuster zu untersuchen oder zu demonstrieren. Der Nobelpreisträger von 1933, Thomas Hunt Morgan, bemerkte als erster, dass einige Gene mit diesem System geschlechtsgebunden sind6.

Mendelian genetics explains how traits move from parents to offspring. Gregor Mendel studied pea plants in 1865 and showed that many visible traits are controlled by two alleles, or gene versions, with one inherited from each parent. He also found that some alleles are dominant and others are recessive.

Mendel’s work was not widely accepted until 1900, and it took about 30 more years for Mendelian inheritance to fit into evolutionary theory. One reason for the delay was that real breeding results often looked like blending inheritance, where offspring have intermediate traits instead of clear categories. That pattern seemed different from Mendel’s smooth-versus-wrinkled peas or red-versus-white flowers.

Molecular genetics has since shown that some inheritance patterns can seem to break Mendel’s rules without making his model wrong. The transcript highlights three major examples: epistasis, pleiotropy, and sex linkage. Epistasis happens when two or more genes interact to shape a trait. Pleiotropy happens when one gene affects more than one phenotype, such as white cats with blue eyes that are often deaf.

Pleiotropy can also create tradeoffs. The same gene may help in one part of life and cause harm in another. This kind of antagonistic pleiotropy has been linked to senescence, or age-related decline, where genes that improve early-life reproduction may also contribute to later-life aging effects.

Sex linkage is another case that looks non-Mendelian at first. In humans and many mammals, XX individuals are female and XY individuals are male. Males inherit an X from their mother and a Y from their father, while females inherit an X from each parent. Because Y chromosomes recombine very little with X chromosomes and have lost many genes over time, only 16 genes are shared by the human X and Y chromosomes.

X-linked traits are encoded on the X chromosome. Females can be homozygous or heterozygous for these traits. If a trait is recessive, a female shows it only when she has two copies. A heterozygous female can carry the allele without showing the phenotype, but about half of her sons may inherit it and express the trait because males have only one X chromosome. Hemophilia is a common example. If a carrier female with genotype XH Xh mates with a normal male XH Y, 50% of the sons will be affected. If the female is Xh Xh, none of her daughters will be hemophiliacs, but 100% of her sons will be.

Fruit flies, Drosophila melanogaster, also use an XY system and are useful lab organisms because they are easy to keep, breed, and manipulate. Wild-type flies have normal body shape and red eyes. Researchers can also study flies with no eyes, different eye colors, or missing wings. Thomas Hunt Morgan used this system to notice sex-linked genes, and it remains a useful model for both Mendelian and non-Mendelian inheritance.
