Cellular Respiration

Wie haben sich die Organismen auf der Erde diese Energie zunutze gemacht? Alles beginnt mit den Photosynthesizern. Diese Organismen sind in der Lage, Kohlendioxid und Wasser aufzunehmen und dann die von der Sonne eingefangene Energie in Form von Photonen zu nutzen, um diese Moleküle zusammenzubringen und Glukose und Sauerstoff zu produzieren. Die Glukose ist der Schlüssel zum nächsten kritischen Schritt, den fast alle Organismen in der einen oder anderen Form nutzen - der Zellatmung. Die Zellatmung erfolgt durch Enzyme, entweder in der Zellmembran bei Prokaryoten oder in den Mitochondrien bei Eukaryoten.

Die chemische Reaktion beginnt mit dem Abbau der Glukose unter Verwendung von Sauerstoff, um wieder Kohlendioxid und Wasser zu erzeugen, aber dabei geht die Energie, die ursprünglich in die Herstellung des Glukosemoleküls geflossen ist, an zwei neue Stellen. Eine davon ist die Synthese von ATP oder Adenosintriphosphat-Molekülen, einer Energiequelle, die Zellen leicht nutzen können. Der Rest geht als Wärme verloren. Bisher haben wir von einer Art der Zellatmung gesprochen, die Sauerstoff verwendet, und dies wird als aerobe Atmung bezeichnet, aber einige Organismen und Zellen sind in der Lage, in Abwesenheit von Sauerstoff zu atmen. Dies wird als anaerobe Atmung bezeichnet. Und anstatt CO2 und Wasser zu produzieren, entsteht bei diesem Prozess Ethanol als Nebenprodukt. Die Fermentation ist ein Beispiel für diese Art der Atmung. Auf diese Weise sind Hefen in der Lage, in verschlossenen Behältern Alkohol herzustellen. Wir können ein Werkzeug namens Respirometer verwenden, um die aerobe Atmung zu messen. Ganz einfach, dieses Gerät misst die Menge an Sauerstoff, die ein Organismus verbraucht, in diesem Fall die Pflanzensamen. Bei der Messung der Atmung in einem Respirometer wird eine clevere Methode verwendet, die auf dem idealen Gasgesetz basiert: P mal V ist n mal R mal T. P ist der Druck des Systems. V ist das Volumen des Gases. n ist die Anzahl der vorhandenen Mol Gas. R ist die ideale Gaskonstante... und T ist die absolute Temperatur. Vereinfacht ausgedrückt bedeutet dies, dass Sie durch Messung ihres Volumens herausfinden können, wie viele Gasmoleküle in einer Probe vorhanden sind, vorausgesetzt, Sie kennen den Druck und die Temperatur, denn R ist eine Konstante.

Respirometer enthalten Kaliumhydroxid, das Kohlendioxid in fester Form als Kaliumcarbonat einfängt. Aufgrund der Zellatmung wird der Sauerstoff in der verschlossenen Röhre von den Samen verbraucht, und sie setzen Kohlendioxid frei, das wiederum als Kaliumkarbonat eingeschlossen wird. Mit fortschreitender Zellatmung nimmt also das Gesamtvolumen des Gases im System ab. Wir können dies quantifizieren, indem wir das Respirometer an ein Gerät anschließen, das Manometer genannt wird. Hier fällt der Druck in der Respirometerkammer ab, wenn die Sauerstoffmoleküle verbraucht werden, und eine farbige Flüssigkeit in einem winzigen Kapillarröhrchen wird in Richtung des verringerten Drucks gezogen. Wir können dann die Menge des im Respirometerrohr verbleibenden Gases abschätzen, indem wir den Wert auf der Manometerebene ablesen. Dieses vielseitige System kann mit vielen verschiedenen Variablen eingerichtet werden, wie z. B. unterschiedlichen Temperaturen, und kann verwendet werden, um die Atmungsfrequenz in vielen verschiedenen Arten von Lebewesen zu testen.

In diesem Labor verwenden Sie ein Respirometer und ein Manometer, um die Atmungsrate von keimenden Samen zu messen.

<h4>Autotrophe und Heterotrophe</h4>

Lebende Organismen benötigen einen kontinuierlichen Energiezufuhr, um zelluläre und organismische Funktionen wie Wachstum, Reparatur, Bewegung, Verteidigung und Fortpflanzung aufrechtzuerhalten. Zellen können chemische Energie nur nutzen, um ihre Funktionen zu betreiben, daher müssen sie Energie aus chemischen Bindungen von Biomolekülen wie Zuckern und Lipiden gewinnen. Autotrophe Organismen, nämlich Pflanzen, Algen sowie photosynthetische und chemosynthetische Bakterien, wandeln anorganische Materialien in solche Biomoleküle um, indem sie Energie aus der Umwelt nutzen, z. B. aus dem Sonnenlicht während der Photosynthese. Heterotrophe Organismen sind nicht in der Lage, hochenergetische Biomoleküle aus anorganischen Materialien zu synthetisieren, daher gewinnen sie Energie durch den Verzehr von Kohlenstoffverbindungen, die von anderen Organismen produziert werden, hauptsächlich aus autotrophen Stoffen. Wenn Energie benötigt wird, werden chemische Bindungen von Kohlenstoffverbindungen aufgebrochen, um die in diesen Bindungen gespeicherte Energie zu gewinnen. Die Prozesse zur Energiegewinnung aus Biomolekülen werden als Zellatmung bezeichnet.

Die Zellatmung findet sowohl in autotrophen als auch in heterotrophen Organismen statt, wobei die Energie dem Organismus am häufigsten durch die Umwandlung von Adenosindiphosphat (ADP) in Adenosintriphosphat (ATP) zur Verfügung steht. Es gibt zwei Haupttypen der Zellatmung – die aerobe Atmung und die anaerobe Atmung. Die aerobe Atmung ist eine spezielle Art der Zellatmung, bei der Sauerstoff (O2) benötigt wird, um ATP zu bilden. In diesem Fall kann Glukose (C6H12O6) in einer Reihe von enzymatischen Reaktionen vollständig oxidiert werden, um Kohlendioxid (CO2) und Wasser (H2O) zu erzeugen.

<h4>Aerobe Atmung</h4>

Die aerobe Atmung erfolgt in drei Stufen. Ein Prozess namens Glykolyse spaltet Glukose in zwei Drei-Kohlenstoff-Moleküle, die als Pyruvat bezeichnet werden. Bei diesem Prozess wird Energie freigesetzt, die zum Teil in ATP umgewandelt wird. Als nächstes gelangen Pyruvatmoleküle in die Mitochondrien, um an einer Reihe von Reaktionen teilzunehmen, die als Krebszyklus bezeichnet werden und auch als Zitronensäurezyklus bekannt sind. Dadurch wird der Abbau von Glukose abgeschlossen, ein Teil der Energie wird in ATP umgewandelt und Elektronen auf Trägermoleküle übertragen. In der letzten Phase, der sogenannten oxidativen Phosphorylierung, durchlaufen Elektronen ein Elektronentransportsystem in der mitochondrialen Innenmembran, das einen Gradienten von Wasserstoffionen aufrechterhält. Die Zellen nutzen die Energie dieses Protonengradienten, um während der aeroben Atmung den Großteil des ATP zu erzeugen.

<h4>Anaerobe Atmung</h4>

Für die aerobe Atmung wird Sauerstoff benötigt, aber es gibt viele Organismen, die an Orten leben, an denen Sauerstoff nicht ohne weiteres verfügbar ist oder an denen andere Chemikalien die Umwelt überwältigen. Extremophile sind Bakterien, die an Orten wie Hydrothermalquellen in der Tiefsee oder Unterwasserhöhlen leben können. Anstatt Sauerstoff für die Zellatmung zu verwenden, verwenden diese Organismen anorganische Akzeptoren wie Nitrat oder Schwefel, die in diesen rauen Umgebungen leichter zu gewinnen sind. Dieser Vorgang wird als anaerobe Atmung bezeichnet.

Wenn kein Sauerstoff vorhanden ist und keine Zellatmung stattfinden kann, findet eine spezielle anaerobe Atmung statt, die als Fermentation bezeichnet wird. Die Fermentation beginnt mit der Glykolyse, um einen Teil der in Glukose gespeicherten Energie in ATP einzufangen. Da jedoch keine oxidative Phosphorylierung stattfindet, werden bei der Fermentation weniger ATP-Moleküle produziert als bei der aeroben Atmung. Beim Menschen findet die Fermentation in den roten Blutkörperchen statt, denen die Mitochondrien fehlen, sowie in den Muskeln während der anstrengenden Aktivität, bei der Milchsäure als Nebenprodukt entsteht, daher wird sie als Milchsäurefermentation bezeichnet. Einige Bakterien führen eine Milchsäuregärung durch und werden zur Herstellung von Produkten wie Joghurt verwendet. In Hefe erzeugt ein Prozess, der als alkoholische Gärung bekannt ist, Ethanol und Kohlendioxid als Nebenprodukte und wurde vom Menschen zur Gärung von Getränken oder Sauerteig verwendet.

<h4>Photosynthese</h4>

Die Zellatmung zusammen mit der Photosynthese ist ein Merkmal der Übertragung von Energie und Materie und unterstreicht die Interaktion von Organismen mit ihrer Umwelt und anderen Organismen in der Gemeinschaft. Die Zellatmung findet in einzelnen Zellen statt, aber auf der Ebene der Ökosysteme beeinflusst der Austausch von Sauerstoff und Kohlendioxid durch Photosynthese und Zellatmung den Sauerstoff- und Kohlendioxidgehalt in der Atmosphäre.

Interessanterweise sind die Prozesse der Zellatmung und der Photosynthese genau gegensätzlich, wobei die Produkte der einen Reaktion die Reaktanten der anderen sind. Bei der Photosynthese wird die Glukose produziert, die bei der Zellatmung zur Herstellung von ATP verwendet wird. Diese Glukose wird dann während der Atmung wieder in CO2 umgewandelt, das ein Reaktant ist, der bei der Photosynthese verwendet wird. Genauer gesagt baut die Photosynthese ein Glukosemolekül aus sechs CO2 - und sechs H2O-Molekülen auf, indem sie Energie aus dem Sonnenlicht einfängt und sechs O2 -Moleküle als Nebenprodukt freisetzt. Bei der Zellatmung werden sechsO2-Moleküle verwendet, um ein Glukosemolekül in sechs CO2 - und sechs H2O-Moleküle umzuwandeln und dabei Energie in Form von ATP und Wärme zu nutzen.

<h4>Messung der Atmung</h4>

Wissenschaftler können die Rate der Zellatmung mit einem Respirometer messen, indem sie die Rate des Sauerstoffaustauschs bewerten. Das Verständnis des idealen Gasgesetzes ist von grundlegender Bedeutung, um zu wissen, wie das Respirometer funktioniert. Das ideale Gasgesetz besagt, dass die Anzahl der Gasmoleküle in einem Behälter aus dem Druck, dem Volumen und der Temperatur bestimmt werden kann. Genauer gesagt entspricht das Produkt aus dem Volumen und dem Druck eines Gases dem Produkt aus der Anzahl der Gasmoleküle, der idealen Gaskonstante und der Temperatur des Gases. Respirometer enthalten Kaliumhydroxid, das Kohlendioxid einfängt, das durch die Atmung in fester Form als Kaliumcarbonat produziert wird. Wenn Zellen Sauerstoff verbrauchen, nimmt das Gasvolumen im Respirometersystem ab, ohne dass Kohlendioxid es wieder erhöht, was es den Wissenschaftlern ermöglicht, die Menge an verbrauchtem Sauerstoff anhand der idealen Gasgleichung zu berechnen.

Die Zellatmung ist ein wichtiger Prozess, der nutzbare Energie für Organismen erzeugt, daher ist es nicht nur interessant, sondern auch notwendig, die Kontexte zu untersuchen, in denen sie verbessert oder behindert wird. Insbesondere sind die Mitochondrien für die Zellatmung unerlässlich und alle Erkrankungen, die die Gesundheit der Mitochondrien beeinträchtigen, haben immense Folgen für die Gesundheit des Organismus. Zum Beispiel sind mitochondriale Myopathien eine Gruppe von neuromuskulären Erkrankungen, die durch mitochondriale Schäden verursacht werden und hauptsächlich Nerven- und Muskelzellen betreffen, die ein hohes Maß an Energie benötigen, um zu funktionieren1. Darüber hinaus wirken viele Gifte, indem sie die Zellatmung hemmen. Zum Beispiel hemmt Cyanid die Produktion von ATP durch oxidative Phosphorylierung, so dass das Verständnis der Mechanismen von Cyanid oder anderen Stoffwechselgiften die Behandlung von Personen ermöglicht, die ihnen ausgesetzt waren2. In ähnlicher Weise können einige Medikamente wie bestimmte Antibiotika, Chemotherapeutika, Statine und Anästhetika die mitochondriale Funktion beeinträchtigen und sind möglicherweise nicht zur Behandlung von Patienten mit mitochondrialen Störungen geeignet3.

Cellular respiration is the process cells use to release usable energy from biomolecules such as sugars and lipids. Living things need a steady energy supply for growth, repair, movement, defense, and reproduction. Cells use that energy mainly by converting adenosine diphosphate (ADP) to adenosine triphosphate (ATP).

Autotrophs and heterotrophs get the molecules for respiration in different ways. Autotrophs, such as plants, algae, and photosynthetic and chemosynthetic bacteria, build high-energy biomolecules from inorganic materials by using energy from the environment, such as sunlight. Heterotrophs cannot make those molecules from inorganic materials, so they obtain carbon compounds by eating other organisms, mainly autotrophs.

Aerobic respiration needs oxygen to make ATP. In this form of respiration, glucose can be fully broken down through a series of enzyme-driven reactions into carbon dioxide and water. Aerobic respiration has three stages: glycolysis, the Krebs cycle, and oxidative phosphorylation.

Glycolysis splits one glucose molecule into two three-carbon pyruvate molecules. It releases energy, and some of that energy is captured in ATP. The pyruvate then enters the mitochondria, where the Krebs cycle, also called the citric acid cycle, finishes breaking down the glucose and loads electrons onto carrier molecules.

The last stage is oxidative phosphorylation. In this step, electrons move through an electron transport system in the inner mitochondrial membrane. This helps build a hydrogen ion gradient, and cells use that gradient to make most of the ATP in aerobic respiration.

Some organisms live where oxygen is scarce or where other chemicals dominate the environment. Extremophiles, which are bacteria found in places such as deep ocean hydrothermal vents or underwater caves, may use inorganic acceptors like nitrate or sulfur instead of oxygen. This is called anaerobic respiration.

When oxygen is absent and cellular respiration cannot continue, cells can use fermentation. Fermentation begins with glycolysis and makes some ATP, but it does not include oxidative phosphorylation, so it produces fewer ATP molecules than aerobic respiration. In humans, lactic acid fermentation occurs in red blood cells, which lack mitochondria, and in muscles during strenuous activity. Some bacteria also use lactic acid fermentation, which is why they are used to make yogurt.

Yeast carry out alcoholic fermentation. This pathway produces ethanol and carbon dioxide as byproducts. People use it to ferment beverages and to help dough rise.

Cellular respiration and photosynthesis are linked processes that move energy and matter through ecosystems. They are direct opposites in many ways because the products of one are the reactants of the other. Photosynthesis uses sunlight to build one glucose molecule from six carbon dioxide and six water molecules and releases six oxygen molecules. Cellular respiration uses six oxygen molecules to break one glucose molecule into six carbon dioxide and six water molecules while capturing energy as ATP and heat.

Scientists can measure respiration rates with a respirometer by tracking oxygen exchange. This method depends on the Ideal Gas Law, which relates pressure, volume, temperature, and the number of gas molecules. Respirometers contain potassium hydroxide, which traps carbon dioxide as potassium carbonate. As cells use oxygen, gas volume in the system drops, and scientists can use the ideal gas equation to calculate oxygen use.

Cellular respiration matters for health because mitochondria are essential to the process. Mitochondrial myopathies are neuromuscular diseases caused by mitochondrial damage, and they mainly affect nerve and muscle cells that need a lot of energy. Many poisons also block respiration. Cyanide, for example, inhibits ATP production through oxidative phosphorylation, and some antibiotics, chemotherapeutics, statins, and anesthetics can also interfere with mitochondrial function.
