Cell Structure

Das Leben, von den größten Organismen bis zu den winzigsten, besteht aus Zellen. Zellen sind die Bausteine für das Leben und liefern die Materialien, die für die Existenz des Lebens benötigt werden. 
Während Zellen aus allen Lebensbereichen ähnliche Strukturen wie DNA und Proteine enthalten, gibt es zwei verschiedene Zelltypen, je nachdem, wie diese Strukturen intern organisiert sind. Prokaryotische Zellen, die von Bakterien und Archaeen, sind einfach organisiert. So ist ihr Genom zwar dicht gepackt, aber nicht von einer Membran umgeben. Eukaryotische Zellen, aus denen Tiere, Pflanzen und Pilze bestehen, sind komplexer. Sie unterteilen ihre inneren Strukturen in Organellen, membrangebundene Strukturen, die bestimmte Funktionen erfüllen. Im Gegensatz zu Prokaryoten ist die eukaryotische DNA in einer Kernmembran eingeschlossen. Weitere Organellen sind das endoplasmatische Retikulum, das Mitochondrium und das Lysosom. 
Die ersten Zellen auf der Erde tauchen im Fossilienbestand vor etwa 3,7 Milliarden Jahren auf und es wurde angenommen, dass sie prokaryotisch waren. Eukaryotische Zellen tauchten erstmals vor etwa 2,4 Milliarden Jahren auf, als eine amöbenähnliche Zelle eine Bakterienzelle verschlang und eine stabile Koexistenz bildete. Im Laufe der Zeit hat sich diese Zelle innerhalb der Zelle entwickelt, um Strukturen wie den Chloroplasten und das Mitochondrium zu bilden. 
Auch Pflanzen und Tiere haben vielfältige metabolische und strukturelle Bedürfnisse, was zu weiteren Unterschieden in ihren Zellen führt. Da Pflanzen ihre eigene Energie erzeugen müssen, enthalten Pflanzenzellen Chloroplasten, die die Photosynthese unterstützen. Zur Pflanzenzelle gehört auch eine große Vakuole, um Proteine, Stoffwechselprodukte und Wasser zu speichern. Schließlich haben Pflanzenzellen auch eine starre Zellwand, die für das Wachstum und die Wasserspeicherung notwendig ist. 
Da Zellen so klein sind, können sie nur mit dem Mikroskop gesehen werden. Eine der einfachsten Möglichkeiten, Zellen zu betrachten, ist das Hellfeldmikroskop. Dieses Mikroskop enthält eine Lichtquelle, eine Kondensorlinse zur Fokussierung des Lichts, eine Okularlinse zum Betrachten der Probe und eine Objektivlinse zur Vergrößerung. Ein Mikroskop ist nicht das Einzige, was benötigt wird, um das Unsichtbare sichtbar zu machen. Die Strukturen innerhalb der Zellen sind transparent, daher müssen die Zellen mit Farbstoffen angefärbt werden. Zum Beispiel wird Safranin verwendet, um Zellkerne rot zu färben. 
In dieser Laborübung verwenden Sie ein zusammengesetztes Mikroskop, um Wangen- und Pflanzenzellen sichtbar zu machen.

<h4>Hintergrund</h4>
Zellen stellen die grundlegendsten biologischen Einheiten aller Organismen dar, seien es einfache, einzellige Organismen wie Bakterien oder große, mehrzellige Organismen wie Elefanten und riesige Mammutbäume. In der Mitte des 19. Jahrhunderts wurde die Zelltheorie vorgeschlagen, um eine Zelle zu definieren, die besagt:
<ul>
	<li>Jeder lebende Organismus besteht aus einer oder mehreren Zellen.</li>
	<li>Die Zellen sind die Funktionseinheiten aller Organismen.</li>
	<li>Alle Zellen entstehen aus bereits existierenden Zellen.</li>
</ul>
Alle Zellen haben gemeinsame Merkmale wie eine Plasmamembran, ein Zytoplasma, DNA und Ribosomen. Eine Plasmamembran ist eine Phospholipid-Doppelschicht, die die Zelle umgibt. Diese dünne und flüssige Schicht um die Zellen dient dazu, den Zellinhalt von seiner Umgebung zu isolieren und den Stoffaustausch mit seiner Umgebung zu regulieren und gleichzeitig die Interaktion mit anderen Zellen zu erleichtern. Im Inneren der Plasmamembran ist die Zelle mit einer gelartigen Flüssigkeit namens Zytoplasma gefüllt, die organische Moleküle, Salze und andere Materialien enthält, die für die Funktionen der Zelle lebenswichtig sind. Daher finden biochemische Reaktionen, die das Leben unterstützen, die sogenannten Stoffwechselprozesse, im Zytoplasma statt. Die Art der Stoffwechselprozesse, die eine Zelle ausführen kann, hängt von ihrer genetischen Information ab. Alle Zellen verwenden die DNA als genetisches Material, das der erbliche Bauplan für den Aufbau zellulärer Strukturen und Produkte ist. Schließlich verwenden alle Zellen Ribosomen, um ihre Proteinprodukte zu synthetisieren.
Es gibt zwei Arten von Zellen, die auf ihrer genetischen Materiallage basieren: prokaryotisch, was "vor dem Kern" bedeutet, und eukaryotisch, was "echter Kern" bedeutet. Während also beide Arten von Organismen DNA haben, haben Prokaryoten wie Bakterien Nukleoide oder "kernähnliche" Komponenten anstelle eines Kerns, während Eukaryoten echte, membrangebundene Kerne besitzen, um ihre DNA zu enthalten. Darüber hinaus sind Prokaryoten relativ klein, etwa 0,1 bis 5,0 Mikrometer (μm), im Vergleich zu Eukaryoten, die typischerweise zwischen 10 und 100 μm groß sein können. Die geringe Größe der Prokaryoten ermöglicht eine schnelle und mühelose Verteilung von Materialien in der Zelle und die Ausführung von Stoffwechselprozessen sowie die schnelle Entfernung von Abfallstoffen oder anderen Produkten aus der Zelle. Daher besitzen eukaryotische Zellen spezialisierte Strukturen, die als Organellen bekannt sind, wie Mitochondrien oder Golgi-Apparate, um die Ausführung lebenswichtiger Funktionen zu ermöglichen.
<h4>Die eukaryotische Zelle</h4>
Die eukaryotische Zelle ist ein gemeinsames abgeleitetes Merkmal aller Eukaryoten, was bedeutet, dass sie einen einzigen Ursprung hat, der seitdem von allen Eukaryoten vererbt wurde. Die frühesten eukaryotischen Zellen sind vor etwa 2,4 Milliarden Jahren in Fossilien zu sehen und sind daran zu erkennen, dass sie größer sind als prokaryotische Zellen1. Die Entstehung dieses Zelltyps resultierte aus einem endosymbiotischen Ereignis, bei dem eine amöbenähnliche Zelle ein Mikrokokken-Bakterium verschlang und eine stabile Koexistenz bildete2. Die verschlungenen Bakterien entwickelten sich zu den ersten energieproduzierenden Organellen, den Mitochondrien, den Organellen des aeroben Stoffwechsels in der Zelle. Mitochondrien haben ein eigenes Genom und sind ähnlich groß wie Prokaryoten. Sie bestehen aus zwei Membranschichten, die zwei unterschiedliche Kompartimente umschließen. Einige der Reaktionen, die hochenergetische Biomoleküle abbauen, finden im inneren Kompartiment statt, während im äußeren Kompartiment die Reaktionen stattfinden, die die von diesen Verbindungen freigesetzte Energie in Adenosintriphosphat (ATP)-Molekülen einfangen, die als Energiewährung der Zelle verwendet werden.
Zellkerne und Mitochondrien sind nicht die einzigen gemeinsamen Strukturen eukaryotischer Zellen. Andere ubiquitäre eukaryotische Organellen sind das glatte und raue endoplasmatische Retikulum (ER), der Golgi-Apparat, die Lysosomen und Vakuolen. Endoplasmatisches Retikulum bedeutet einfach "Netzwerk im Plasma" und ist, wie der Name schon sagt, ein großes Netzwerk von Membranen innerhalb der Zelle, insbesondere um den Zellkern herum. Teile des rauen ER erstrecken sich von der Kernmembran und unterscheiden sich vom glatten ER durch ihr raues Aussehen aufgrund zahlreicher Ribosomen auf ihrer Oberfläche. Rough ER ist der Ort für die Proteinsynthese, wie z. B. die Proteine, die in die Plasmamembran eingebettet sind, oder die Proteine, die von der Zelle ausgeschieden werden. Im Gegensatz dazu produziert glattes ER Produkte auf Lipidbasis, enthält aber auch Enzyme zur Entgiftung schädlicher Chemikalien. Daher enthalten Leberzellen reichlich glattes ER. Außerdem enthalten Muskelzellen aufgrund der kalziumspeichernden Funktion dieses Organells, das für die Muskelkontraktion unerlässlich ist, erhebliche Mengen an glattem ER. Der Golgi-Apparat sortiert, modifiziert und verpackt zelluläre Produkte in Vesikel, die mit der Plasmamembran verschmelzen, um die Produkte freizusetzen. Einige der Proteine, die im rauen ER produziert werden, sind intrazelluläre Verdauungsenzyme. Diese Enzyme werden im Golgi-Apparat in spezielle Vesikel, sogenannte Lysosomen, verpackt. Die Hauptfunktion der Lysosomen besteht darin, die von der Zelle verschlungenen Nahrungspartikel sowie alte Zellteile zu verdauen. Vakuolen sind Säcke der Zellmembran, die als Speichereinheiten innerhalb der Zellen dienen. Sie können sowohl zur Speicherung von Wasser zur Regulierung des Wassergehalts der Zelle als auch zur Speicherung von Stoffwechselprodukten oder sogar giftigen Molekülen dienen, je nach Zelltyp und Organismus.
<h4>Königreichsspezifische Organellen</h4>
Eukaryotische Zellen haben auch unterschiedliche Organellen entwickelt, die für jedes Reich spezifisch sind. Zum Beispiel sind die Plantae und Animalia des Königreichs beide eukaryotisch, aber die Organellen der pflanzlichen und tierischen Zellen unterscheiden sich in wichtigen Punkten, die es ihnen ermöglichen, ihr Leben als Produzenten bzw. Konsumenten zu führen. Landpflanzen müssen groß werden und starre Stängel haben, um Blätter zu halten, die sie für die Photosynthese verwenden. Sie müssen auch in der Lage sein, das von den Wurzeln aufgenommene Wasser zu speichern. Ihre Zellen spiegeln diese spezifischen Bedürfnisse wider. Im Gegensatz zu tierischen Zellen verfügen Pflanzenzellen über Chloroplasten, die für die Photosynthese verwendet werden und oft das grüne Pigment Chlorophyll enthalten. Darüber hinaus sind sie von Zellwänden umgeben, bei denen es sich um starre äußere Schichten aus Zellulose handelt, die das Wachstum und die Wasserspeicherung unterstützen. Da sie große Mengen Wasser speichern müssen, um den Wasserdruck in der Zelle aufrechtzuerhalten, haben sie größere Vakuolen als tierische Zellen. Darüber hinaus verfügen Pflanzenzellen auch über eine andere Art von spezialisierten Speicherorganellen, die Plastiden, die Pigmente sowie photosynthetische Produkte wie Stärke enthalten. Diese Unterschiede sind spürbar und unterscheiden pflanzliche Zellen von tierischen Zellen: Pflanzenzellen haben aufgrund ihrer starren Zellwände in der Regel eine regelmäßige, rechteckige Form, während tierische Zellen abgerundet und unregelmäßiger sind.
<h4>Mikroskopie</h4>
Einige Zellen, wie z. B. Frosch-Eizellen, sind groß genug, um mit bloßem Auge gesehen zu werden, aber die meisten Zellen können ohne visuelle Hilfe nicht gesehen werden. Daher nutzen Wissenschaftler Mikroskopietechniken, um Zellstrukturen zu untersuchen und Zelltypen voneinander zu unterscheiden. Während Mikroskope in der Lage sind, Objekte zu vergrößern, die mit dem menschlichen Auge nur schwer oder gar nicht zu erkennen sind, fehlt den meisten Geweben von Natur aus die Pigmentierung. Daher wurden Lösungen entwickelt, die Zellen anhand ihrer molekularen Zusammensetzung selektiv färben können. Damit können Forscher unter anderem zwischen Organellen in einer Zelle, Gewebetypen in einem Pflanzenstamm und Fettschichten bei Tieren unterscheiden. Der Farbstoff Methylenblau färbt Nukleinsäuren toter Zellen und bindet an negativ geladene DNA. Safranin-Lösung ist ein weiterer biologischer Farbstoff, der Zellkerne rot färbt. Die Zellen müssen sich nur für kurze Zeit in den Färbelösungen befinden und können sofort nach dem Färbeschritt montiert werden. Die gebräuchlichsten Montagetechniken sind Nassmontage und Ölimmersion. Eine Nasshalterung wird erstellt, indem eine Probe entnommen und auf einen Objektträger mit Flüssigkeit zwischen Objektträger und Deckglas gelegt wird. Zellproben werden in Flüssigkeiten wie Wasser oder Glycerin suspendiert. Glycerin ist besser mit lebenden Kulturen zu verwenden, da es die Vermehrung von Bakterien verhindert3. Immersionsöl kann auf das Deckglas aufgetragen werden, um die Betrachtung der Probe bei hoher Vergrößerung zu verbessern. Dies wird erreicht, weil das Öl den gleichen Brechungsindex wie Glas hat, was bedeutet, dass es das Licht genauso gut durchlässt wie Glas. Die Glas-Luft-Grenzfläche streut Licht stärker als Öl oder Glas, so dass die Klarheit des Bildes beeinträchtigt wird, wenn Proben "trocken" oder ohne Öl montiert werden. Sobald die Zellen gefärbt und montiert sind, sind sie bereit, unter dem Mikroskop untersucht zu werden.
Es gibt verschiedene Mikroskopietechniken, von der Elektronenscanning-Technologie, die es Forschern ermöglicht hat, Objekte auf atomarer Ebene zu betrachten, bis hin zur fluoreszierenden Bildgebung lebender Zellen, die eine Echtzeitüberwachung der Bewegung von Molekülen in einzelnen Zellen ermöglicht4. Die Hellfeldmikroskopie ist die einfachste Mikroskopietechnik, die nur eine Halogenlichtquelle, eine Kondensorlinse zur Fokussierung des Lichts, eine Okularlinse zum Betrachten des Bildes und eine Objektivlinse zur Vergrößerung des Bildes benötigt. Bei jeder Mikroskopietechnik ist es wichtig, die Teile eines Mikroskops zu verstehen, bevor man es verwendet. Im Allgemeinen haben Verbundmikroskope, die für die Hellfeldbildgebung verwendet werden, ein Okular an der Oberseite des Zielfernrohrs, das am Kopf und an den Objektiven befestigt ist. Das Okular hat eine 10-fache Vergrößerung, und die Objektive sind auf eine bestimmte Vergrößerung in einem Bereich von 4-100x eingestellt. Bei einem Standardmikroskop gibt es zwischen drei und fünf Objektive. Die Objektive zeigen auf die Bühne, auf der ein Exemplar zur Betrachtung platziert wird. Die Bühne verfügt oft über mechanische Teile und Bühnenclips, um eine Folie zu halten und sie während des Betrachtens zu bewegen. Eine Blende ist ein Loch in der Bühne, durch das Licht dringt. Dieses Licht wird durch eine einstellbare Kondensorlinse über einer Beleuchtung oder Lichtquelle gesteuert. Um den Zoom des Tisches für das zu betrachtende Objekt zu steuern, verfügen die Mikroskope über Einstellknöpfe für den Grob- und Feinfokus. Der Grobfokus-Knopf bewegt sich auf einer größeren Skala als der Feinfokus, aber sie befinden sich auf der gleichen Achse. Die Feinfokussierung ist nützlich, wenn das Objekt auf dem Tisch in die Nähe der Objektive gebracht wurde. Es ist wichtig, dass die Objektivlinse das Objekt auf dem Tisch nicht berührt, da sie die Linse zerkratzen kann. Objekte sollten immer zuerst auf dem Objektiv mit der niedrigsten Vergrößerung betrachtet und klar fokussiert werden, bevor auf Objektive mit höherer Vergrößerung umgeschaltet wird.
Die Mikroskopie ist ein wichtiges Werkzeug für viele Aspekte des medizinischen Bereichs, einschließlich Forschung, Diagnose und Behandlung. Dies hat die Anwendung des Einsatzes der Nanotechnologie in der Medizin als neue Behandlungsmethode anstelle einer invasiveren Operation5. Chirurgen verwenden auch Mikroskope, von denen einige so modifiziert wurden, dass sie am Kopf des Chirurgen montiert und mit Fußpedalen bedient werden. Diese haben eine viel geringere Vergrößerung als selbst die heute verwendeten Lichtmikroskope, erleichtern aber die sichere Durchführung heikler Eingriffe, wie z. B. der optischen und neurochirurgischen Chirurgie.

Cells are the basic units of life, and their structures help explain how living things work. Cell theory says that every living organism is made of one or more cells, cells are the functional units of organisms, and new cells come from preexisting cells. Cells also share key features, including a plasma membrane, cytoplasm, DNA, and ribosomes.

The plasma membrane is a phospholipid bilayer that surrounds the cell. It separates the cell from its environment and controls what enters and leaves. Inside it, the cytoplasm is a gel-like fluid that contains organic molecules, salts, and other materials needed for cell activity. Many metabolic processes, or life-supporting chemical reactions, take place there.

Cells use DNA as their genetic material, which acts as the blueprint for cellular structures and products. Ribosomes make proteins. Cells are often grouped as prokaryotic or eukaryotic based on where their genetic material is found. Prokaryotes, such as bacteria, have nucleoids instead of a true nucleus, while eukaryotes have membrane-bound nuclei.

Prokaryotic cells are usually small, about 0.1 to 5.0 micrometers, while eukaryotic cells are often 10 to 100 micrometers. Their small size helps prokaryotes move materials quickly and remove waste efficiently. Eukaryotic cells use organelles, such as mitochondria and the Golgi apparatus, to carry out important functions.

Eukaryotic cells are thought to have a shared origin and appear in fossils about 2.4 billion years ago. The earliest eukaryotic cells were larger than prokaryotic cells and likely formed through an endosymbiotic event. In this event, an amoeba-like cell engulfed a micrococcal bacterium, and that bacterium later became the mitochondrion. Mitochondria perform aerobic metabolism and have their own genome.

Mitochondria have two membranes that enclose two compartments. Some reactions that break down high-energy biomolecules happen in the inner compartment. Other reactions in the outer compartment capture released energy and convert it into ATP, the cell’s energy currency. Eukaryotic cells also contain the smooth and rough endoplasmic reticulum, the Golgi apparatus, lysosomes, and vacuoles.

The endoplasmic reticulum means “network inside the plasma,” and it forms a membrane network around the nucleus. Rough ER has ribosomes on its surface and makes proteins, including proteins for the plasma membrane and proteins that are secreted from the cell. Smooth ER makes lipid-based products and helps detoxify harmful chemicals, which is why liver cells contain much of it. Muscle cells also have a lot of smooth ER because it stores calcium needed for contraction.

The Golgi apparatus sorts, modifies, and packages cellular products into vesicles. Some proteins made in the rough ER are digestive enzymes. The Golgi packages these enzymes into lysosomes, which digest food particles taken into the cell and break down old cell parts. Vacuoles are membrane sacs used for storage, including water, metabolic products, or even poisonous molecules, depending on the cell.

Plant and animal cells are both eukaryotic, but they do not have the same organelles. Plant cells have chloroplasts for photosynthesis and often contain chlorophyll, the green pigment. They also have cell walls made of cellulose, which give support and help with water retention. Their larger vacuoles help maintain water pressure, and they also contain plastids that store pigments and products such as starch.

These differences make plant cells and animal cells easy to tell apart. Plant cells usually have a regular, rectangular shape because of their rigid cell walls. Animal cells are more rounded and irregular. Cell walls, chloroplasts, and large vacuoles are key features that distinguish plant cells from animal cells.

Many cells are too small to see without help, so scientists use microscopy to study them. Some cells, such as frog oocytes, are large enough to see with the naked eye, but most samples need magnification. Because many tissues lack color, stains are used to highlight cell parts and tissue types. Methylene blue stains nucleic acids in dead cells by binding to DNA, and safranin stains cell nuclei red.

After staining, cells can be mounted as a wet mount or with immersion oil. In a wet mount, the sample is placed on a slide with liquid between the slide and coverslip. Water or glycerol can be used as the liquid, and glycerol is better for live cultures because it prevents bacterial growth. Immersion oil improves high-magnification viewing because it has the same refractive index as glass and reduces light scattering.

Once stained and mounted, the sample is ready for the microscope. Microscopy methods range from electron scanning technology, which can view objects at atomic scale, to fluorescent live-cell imaging, which tracks molecules in real time. Bright-field microscopy is the simplest method and uses a halogen light source, condenser lens, ocular lens, and objective lens.

Compound microscopes have an eyepiece, a head with objectives, and a stage for the specimen. The eyepiece usually has 10X magnification, and objective lenses commonly range from 4X to 100X. The stage may have clips and mechanical parts to move the slide, and light passes through an aperture in the stage. Coarse and fine focus knobs control sharpness, and the slide should be viewed first at low magnification before moving higher.

Microscopy is important in medicine for research, diagnosis, and treatment. It also supports newer approaches such as nanotechnology in medicine, which can reduce the need for more invasive surgery. Surgeons use modified microscopes as well, including head-mounted models operated with foot pedals. These lower-magnification tools help doctors perform delicate procedures such as optical and neurosurgery safely.
