Macromolecules

Zu wissen, was Makromoleküle sind, ist ein wichtiger Schritt, um zu verstehen, wie Nährstoffe von Zellen aufgenommen und verwendet werden. Der Begriff Makromoleküle bedeutet einfach große Moleküle und bezieht sich auf vier verschiedene Verbindungen, Kohlenhydrate, Lipide, Proteine und Nukleinsäuren. Jede von ihnen hat eine einzigartige Struktur und Funktion. Lassen Sie uns zuerst über Kohlenhydrate sprechen. 
Alle Kohlenhydrate setzen sich aus Kohlenstoff, Wasserstoff und Sauerstoff zusammen. Diejenigen mit einfacher Struktur werden Monosaccharide oder Zucker genannt. Monosaccharide können sich zu komplexeren Kohlenhydratketten verbinden, die als Polysaccharide oder Stärke bezeichnet werden. Die normale Form von Stärke ähnelt aufgrund der Bindungswinkel in der Polymerkette einer Spiralfeder. 
Die nächste Art von Makromolekülen, die wir besprechen werden, ist Protein. Proteine bestehen aus Ketten von Aminosäuren, organischen Verbindungen, die jeweils eine Aminogruppe oder N-Terminus an einem Ende und eine Carboxylgruppe oder C-Terminus am anderen Ende haben. Es gibt 20 Standardaminosäuren, und obwohl sie alle eine ähnliche Basenstruktur haben, hat jede eine einzigartige Seitenkette, die als R-Gruppe bekannt ist. Verschiedene Aminosäuren können sich zu einer Kette verbinden, die durch Peptidbindungen verbunden ist und ein Protein bildet. 
Die letzte Klasse von Makromolekülen, die wir untersuchen werden, sind Lipide. Die gebräuchlichsten Arten von Lipiden sind Fette, die auch Triglyceride genannt werden, da sie aus einem Glycerinmolekül bestehen, das mit drei Fettsäureketten verbunden ist. 
Lassen Sie uns nun über den Nachweis dieser Makromoleküle in einer Laborumgebung sprechen. Das Benedict-Reagenz wird häufig zum Nachweis von Monosacchariden verwendet. Das Benedikt-Reagenz ist normalerweise blau, weil es Kupferionen in Form von Kupfersulfat enthält. Dieses Metallsalz ist ein Beispiel für ein Oxidationsmittel. Monosaccharide, die eine Hemiacetalgruppe enthalten, wie z. B. Glukose, gelten als reduzierende Zucker. In einer wässrigen Lösung liegen Monosaccharide mit Halbacetalgruppen sowohl in der zyklischen als auch in der offenkettigen Form vor, die einen reaktiven Aldehyd enthält. Von diesem Aldehyd wird ein Elektron auf das Kupfer-II-Ion übertragen. Dadurch wird das Kupfer-II-Ion zu Kupfer-I-Ion reduziert und der reduzierende Zucker oxidiert. Da Kupfer-I-Ionen als rostrot angesehen werden, beginnt die Lösung, ihre Farbe zu ändern. In Gegenwart kleiner Mengen Monosaccharid wechselt die Lösung zu grün. Höhere Konzentrationen des Monosaccharids führen dazu, dass mehr Kupfer reduziert wird und sich die Farbe der Lösung dramatischer ändert und schließlich zu einem rötlichen Orange wird.
Stärke kann mit dem normalerweise gelb-braunen Jodindikator nachgewiesen werden. Dieser Indikator enthält verschiedene Polyiodid-Anionenspezies, wie z. B. I-3-negativ, I-5-negativ und I-7-negativ. Die negativ geladenen Jode in jedem Anion wirken als Ladungsdonatoren, während die neutralen Jode als Ladungsakzeptoren wirken. Dabei handelt es sich um einen Ladungstransferkomplex, und Elektronen in diesen Komplexen werden leicht mit Licht auf ein höheres Energieniveau angeregt. Das Licht wird dabei absorbiert und seine Komplementärfarbe kann vom menschlichen Auge gesehen werden. Die Absorptionen der verschiedenen Spezies in dieser Lösung verleihen ihr die gelb-braune Farbe. Wenn eine Stärke zugegeben wird, bildet sie mit den Polyiodid-Anionen einen neuen Ladungstransferkomplex. Dabei fungiert die Stärke als Ladungsdonator und das Polyiodid als Akzeptor. Obwohl die Einzelheiten der Reaktion nicht bekannt sind, wird angenommen, dass sich Jod und Stärke zu einem unendlichen Polyiodid-Homopolymer verbinden. Dieser Komplex absorbiert Licht einer anderen Wellenlänge und färbt die Farbe der Lösung dunkelblau.
Proteine können mit dem Biuret-Reagenz nachgewiesen werden, einer blauen Lösung, die Kupferionen enthält, aber an den in Peptidbindungen enthaltenen Stickstoff bindet und Chelatkomplexe bildet. Chelatkomplexe, an denen drei bis vier Peptidbindungen beteiligt sind, erzeugen eine violette Farbe, die die Farbe der Lösung verändert. Die Intensität der Farbe steht in direktem Zusammenhang mit der Konzentration der Peptidbindungen. Je höher die Konzentration, desto intensiver ist die violette Farbe. 
Schließlich können Lipide in einer Lösung mit dem Sudan IV-Test nachgewiesen werden. Sudan IV ist ein Farbstoff, der nicht wasserlöslich ist, aber er ist in Lipiden löslich, so dass er bei Zugabe zu einer Lösung, die Lipide enthält, diese rot färbt.

<h4>Biomoleküle</h4>
Organismen enthalten eine Vielzahl organischer Moleküle mit zahlreichen Funktionen, die von den chemischen Strukturen und Eigenschaften dieser Moleküle abhängen. Alle organischen Moleküle enthalten ein Kohlenstoffrückgrat und Wasserstoffatome. Das Kohlenstoffatom spielt eine zentrale Rolle bei der Bildung einer Vielzahl organischer Moleküle, die sich in Größe, Form und Komplexität unterscheiden. Anorganische Moleküle hingegen haben in der Regel einfachere Strukturen. Die äußerste Schale eines freien Kohlenstoffatoms kann acht Elektronen aufnehmen, ist aber nur von vier Elektronen besetzt, daher kann sie vier kovalente Bindungen bilden und mit bis zu vier Atomen binden. Alternativ kann es auch mit weniger Atomen binden, indem es Doppel- oder Dreifachbindungen bildet. Diese Vielseitigkeit von Kohlenstoffatomen ermöglicht es organischen Molekülen, komplizierte Strukturen wie Ketten, Äste und Ringe zu zeigen.
Organische Moleküle, die natürlicherweise in Organismen vorkommen, werden als Biomoleküle bezeichnet. Biomoleküle enthalten neben Kohlenstoff und Wasserstoff auch andere Elemente wie Sauerstoff, Stickstoff, Phosphor und Schwefel. Im Allgemeinen kommen kleinere Einheiten von Biomolekülen als sich wiederholende Sequenzen zusammen, um größere Biomoleküle zu bilden. Diese kleinen modularen Einheiten von Biomolekülen werden Monomere genannt. Zwei Monomere verbinden sich typischerweise zu einem Dimer durch einen Prozess, der als Dehydratisierungssynthese bekannt ist und bei dem einfach ein Wasserstoffatom aus einem Monomer und ein Hydroxyl (OH)-Ion aus dem anderen Monomer entfernt werden, um ein Wassermolekül zu erzeugen, das ausgestoßen wird, während die beiden Monomere mit einer kovalenten Bindung verbunden werden. Die Umkehrung dieses Prozesses wird als Hydrolyse bezeichnet, bei der sich das Molekül wieder in seine ursprünglichen Monomere aufspaltet, wobei ein Wassermolekül ein Wasserstoffatom für das eine Monomer und ein Hydroxyl-Ion für das andere Monomer bereitstellt. Viele Monomere können durch Dehydratisierung zu Polymeren zusammengebunden werden. Manchmal können verschiedene Polymere zusammenkommen, um noch größere und komplexere Moleküle zu bilden, die als biologische Makromoleküle bekannt sind.
Biomoleküle werden auf der Grundlage der Elemente, aus denen sie bestehen, sowie ihrer Struktur und Funktion in lebenden Organismen klassifiziert. Fast alle Biomoleküle können in eine der vier allgemeinen Kategorien eingeteilt werden: Kohlenhydrate, Lipide, Proteine und Nukleinsäuren.
<h4>Kohlenhydrate</h4>
Kohlenhydrat bedeutet einfach "Kohlenstoffwasser", da diese Moleküle aus Kohlenstoff-, Wasserstoff- und Sauerstoffatomen in etwa im Verhältnis 1:2:1 bestehen. Kohlenhydratmonomere sind als Monosaccharide bekannt, die auch als Einfachzucker bezeichnet werden. Glukose (C6, H,12,O6) ist das häufigste Monosaccharid in lebenden Organismen und eine Untereinheit vieler Polysaccharide. Zahlreiche Organismen synthetisieren auch andere Monosaccharide mit sechs Kohlenstoffatomen mit der gleichen chemischen Formel wie Glukose, aber leicht unterschiedlichen Strukturen, wie Fruktose und Galaktose. Wenn zwei Monosaccharide miteinander verbunden werden, bilden sie Disaccharide. Zum Beispiel setzt sich Saccharose aus Glukose und Fruktose zusammen, während Laktose Glukose und Galaktose enthält. Diese Monosaccharide und Disaccharide werden zur kurzfristigen Energiespeicherung in lebenden Organismen eingesetzt. Maltose ist ein weiteres Disaccharid, das aus zwei Glukosemolekülen besteht und normalerweise gebildet wird, wenn Polysaccharidketten wie Stärke und Glykogen während der Verdauung abgebaut werden. Stärke ist ein Polysaccharid, das in Pflanzen als Energiespeichermolekül dient und aus zwei Arten von Glukosepolymeren besteht: Amylose und Amylopektin. Amylose macht 10-20% der Stärke aus und ist ein helikales Polymer von Glukose. Amylopektin macht den Großteil der Stärke aus und ist ein verzweigtes Polymer der Glukose. Glykogen ist praktisch das gleiche wie Stärke, wird jedoch in tierischem Leber- und Muskelgewebe synthetisiert, gespeichert und verwendet.
Kohlenhydrate dienen nicht nur als Energiespeicher, sondern haben auch andere Funktionen im Organismus. Die fünf Kohlenstoffatom-Monosaccharide, Ribose und Desoxyribose, sind in die Nukleinsäurestruktur integriert und in jeder lebenden Zelle vorhanden. Darüber hinaus dient das Polysaccharid Cellulose, ein langes Polymer aus Glukose, in Pflanzen als starres Strukturmaterial. Der Mensch verfügt nicht über Verdauungsenzyme, um die in der Nahrung enthaltene Zellulose, die auch als Ballaststoffe bezeichnet wird, abzubauen. Der Verzehr von Ballaststoffen trägt jedoch zur Erhaltung einer gesunden Darmflora bei, was wiederum zur Gesundheit des Verdauungs- und Immunsystems beiträgt1. Ähnlich wie Pflanzen verwenden einige Tiere und Pilze ein weiteres Polysaccharid, Chitin, als Strukturmolekül. Gliederfüßer verwenden Chitin, um ihre Exoskelette aufzubauen und zu erhalten, während Pilze es in ihre Zellwände einbauen, um die Steifigkeit zu erhalten.
<h4>Lipide</h4>
Die zweite Klasse biologischer Makromoleküle sind Lipide, zu denen Fette, Öle und Wachse gehören. Lipide sind hydrophobe Moleküle, die fast ausschließlich aus Kohlenstoff- und Wasserstoffatomen bestehen. Oft werden Lipide in drei Hauptkategorien eingeteilt; Triglyceride, Phospholipide und Steroide.
Die häufigste Art von Lipiden sind Triglyceride, zu denen Fette von Tieren und Öle von Pflanzen gehören. Triglyceride dienen in der Regel als langfristige Energiespeichermoleküle, mit Ausnahme von unverdaulichen Wachsen, die stattdessen sowohl bei Pflanzen als auch bei Tieren als Imprägniermittel verwendet werden. Triglyceride enthalten drei Fettsäureketten, die entweder gesättigt oder ungesättigt sein können und mit einem Glycerinmolekül verbunden sind. Gesättigte Fettsäureketten sind lineare Moleküle mit einer maximalen Anzahl von Wasserstoffatomen, bei denen jeder Kohlenstoff in der Kette über eine einzige Bindung verbunden ist. Auf der anderen Seite haben ungesättigte Fettsäureketten Knicke, die auf das Vorhandensein von mindestens einer Doppelbindung zurückzuführen sind. Darüber hinaus können ungesättigte Fette "Trans"-Fette sein, wenn sich die Wasserstoffatome um die Doppelbindung gegenüberstehen. Transfette kommen zwar natürlich vor, entstehen aber bei der industriellen Herstellung von gesättigten Pflanzenölen mit Wasserstoff. Ähnlich wie gesättigte Fettsäuren lassen sich Transfette aufgrund ihrer relativen Linearität sehr gut stapeln. Transfette verursachen jedoch Probleme für die Gesundheit des menschlichen Herzens, wie z. B. die Schädigung der Arterienschleimhaut und Entzündungen bei der Verdauung 2.
Phospholipide ähneln den Triglyceriden, jedoch wird eine der Fettsäureketten durch eine phosphathaltige polare Gruppe ersetzt. Daher haben Phospholipide einen hydrophilen Kopf und zwei hydrophobe Fettsäureschwänze. Diese Eigenschaften von Phospholipiden sind entscheidend für die Struktur und Funktion der Zellmembran.
Steroide sind Lipide, die aus geschmolzenen Kohlenstoffringen mit unterschiedlichen funktionellen Gruppen bestehen. Cholesterin ist ein Steroid, das auch Bestandteil der Zellmembran ist. Darüber hinaus wird Cholesterin zur Synthese anderer Steroide verwendet, einschließlich Sexualhormonen wie Östrogen und Testosteron. Obwohl Cholesterin für die Zellmembranstruktur und die Hormonsynthese unerlässlich ist, sind hohe Plasmacholesterinspiegel an der Ansammlung von Plaque in den Blutgefäßen beteiligt und verursachen koronare Erkrankungen3.
<h4>Proteine</h4>
Die dritte Klasse biologischer Makromoleküle sind Proteine, die aus Ketten von Aminosäuren bestehen. Es gibt 20 verschiedene Aminosäuren, die alle eine ähnliche Basenstruktur haben, aber jeweils eine einzigartige Seitenkette haben, die als "R-Gruppe" bezeichnet wird. Eine einzelne Aminosäure hat ein N-terminales Ende, das eine Aminogruppe (NH3+) ist, und ein C-terminales Ende, das eine Carboxylgruppe (-COOH) ist. Diese Gruppen verbinden sich miteinander, N-Terminus zu C-Terminus, in einer Kette, die durch Peptidbindungen verbunden ist. Proteine sind wichtig für die Aufrechterhaltung der Körperfunktionen als Enzyme, Hormone, Strukturkomponenten und Transportmoleküle und spielen eine wichtige Rolle bei der Kontraktionsfähigkeit der Muskeln, der Immunität und der Blutgerinnung. Es können jedoch Probleme in der Proteinstruktur und -funktion auftreten, und diese Probleme sind oft genetisch bedingt. Zum Beispiel sind normale rote Blutkörperchen rund, aber bei Menschen, die von Sichelzellenanämie betroffen sind, haben die Zellen eine gekrümmte Form mit einer freiliegenden hydrophoben Region, die durch eine Mutation in einem Protein namens Hämoglobin S verursacht wird. Diese Form verringert die Fähigkeit, Sauerstoff zu transportieren, und führt dazu, dass die Zellen in den Blutgefäßen stecken bleiben. Dies führt zu vielen nachteiligen Symptomen für die Person, die die Mutation trägt, und Menschen, die zwei Kopien des Sichelzellen-Gens erben, erleiden oft negative Auswirkungen oder sterben möglicherweise sogar aufgrund der verminderten Fähigkeit der Sichelzellen, Sauerstoff zu transportieren. Anders ausgedrückt sind diejenigen, die nur eine Kopie des Gens tragen, resistent gegen eine Infektion durch Malaria, so dass die Krankheit in Ländern mit erhöhten Malariainfektionen weitergegeben werden konnte und fortbesteht4.
<h4>Nukleinsäuren</h4>
Die vierte Klasse biologischer Makromoleküle sind die Nukleinsäuren, die aus Monomeren bestehen, die als Nukleotide bekannt sind. Diese Monomere bestehen aus drei Teilen: einer Phosphatgruppe, einem Ribosezucker und einer Stickstoffbase. Nukleotide unterscheiden sich voneinander durch ihre Stickstoffbasen und die Art der Ribose, die sie enthalten. Einzelnukleotide fungieren in der Regel als Energieträger in Zellen sowie als Botenmoleküle. Bei den Nukleotidpolymeren oder Nukleinsäuren wie Desoxyribonukleinsäure (DNA) oder Ribonukleinsäure (RNA) handelt es sich jedoch um erbliche Moleküle, die die genetische Information enthalten, um zelluläre Produkte zu konstruieren.
<h4>Nachweis von Makromolekülen</h4>
Biologische Makromoleküle in Lebensmitteln oder andere Substanzen können anhand ihrer spezifischen chemischen Eigenschaften nachgewiesen werden. Zum Beispiel haben Monosaccharide freie Aldehyd- (-CHO) oder Ketongruppen (-C=O), die andere Verbindungen reduzieren können, was bedeutet, dass sie Substanzen sind, die dazu führen, dass andere Moleküle Elektronen verlieren, daher sind Monosaccharide auch als reduzierende Zucker bekannt. Diese Eigenschaft wird genutzt, um das Vorhandensein von Monosacchariden mit einem Indikator namens Benedict-Reagenz nachzuweisen. Dieser Indikator enthält Kupferionen (Cu2+), die durch Monosaccharide reduziert werden, wie bei einer Änderung der Lösungsfarbe von blau zu rötlichem Orange beobachtet wird, obwohl die Intensität der Farbe je nach der ursprünglichen Konzentration des reduzierenden Zuckers variiert. Die Beobachtung einer grünen Farbe bedeutet also, dass nur eine geringe Menge an reduzierenden Zuckern in der Lösung vorhanden ist. Amylose in Stärke liegt aufgrund der Bindungswinkel in der Polymerkette in einer spiralförmigen Struktur vor. Der Jodindikator, Jod-Kaliumiodid (IKI), reagiert mit diesen gewickelten Molekülen und färbt die Lösung in ein dunkles Blauschwarz, um das Vorhandensein von Amylose anzuzeigen. Wenn jedoch keine Stärkeamylose vorhanden ist, findet die Reaktion nicht statt und die Lösung behält eine gelblich-braune Farbe. Ein Sudan IV-Test wird durchgeführt, um auf das Vorhandensein von Lipiden zu testen. Dieser Farbstoff ist lipophil und löst sich auf, wenn Lipide vorhanden sind, so dass eine rote Farbe in Gegenwart von Lipiden erhalten bleibt. Das Biuret-Reagenz, ein Indikator für das Vorhandensein von Proteinen, enthält Kupferionen (Cu2+), die mit den Peptidbindungen reagieren und die Lösung von blauer zu dunkelvioletter Farbe färben. Dieses Reagenz muss mit einer ausreichenden Anzahl dieser Peptidbindungen reagieren, um die erwartete violette Farbe zu erzeugen, so dass eine rosafarbene Farbe entsteht, wenn die Aminosäureketten nicht lang genug sind.

Biomolecules are the organic molecules that living things use for structure, energy, and heredity. They all have a carbon backbone and hydrogen atoms, and carbon can form four covalent bonds. That bonding flexibility lets carbon build chains, branches, rings, and other complex shapes. Biomolecules also commonly include oxygen, nitrogen, phosphorus, and sulfur.

Small building blocks join together to make larger molecules. These units are called monomers. Two monomers can form a dimer through dehydration synthesis, which removes a hydrogen atom from one monomer and a hydroxyl group from the other. This releases water and creates a covalent bond. The reverse reaction is hydrolysis, where water helps split the molecule back into its original monomers. Many monomers can link into polymers, and different polymers can combine into biological macromolecules. Most biomolecules fall into four main groups: carbohydrates, lipids, proteins, and nucleic acids.

Carbohydrates are often called carbon water because they contain carbon, hydrogen, and oxygen in about a 1:2:1 ratio. Their monomers are monosaccharides, or simple sugars. Glucose is the most common monosaccharide in living organisms, and fructose and galactose are other six-carbon sugars with the same formula but different structures. When two monosaccharides join, they form disaccharides such as sucrose, lactose, and maltose. These sugars support short-term energy storage.

Larger carbohydrate polymers also have important jobs. Starch stores energy in plants and includes amylose and amylopectin. Amylose makes up about 10-20% of starch and forms a helical chain, while amylopectin is the main branched form. Glycogen is very similar to starch, but animals store and use it in liver and muscle tissue. Ribose and deoxyribose are five-carbon sugars found in nucleic acids. Cellulose is a rigid plant polymer made of glucose, and humans cannot digest it because they do not have the right enzymes. Even so, dietary fiber helps support healthy gut flora. Chitin is another structural polysaccharide. Arthropods use it in exoskeletons, and fungi use it in cell walls.

Lipids include fats, oils, and waxes. They are hydrophobic, which means they do not mix well with water, and they are made mostly of carbon and hydrogen. The main lipid groups are triglycerides, phospholipids, and steroids. Triglycerides contain glycerol and three fatty acid chains. They usually serve as long-term energy storage, while waxes act as waterproofing substances in plants and animals. Fatty acid chains can be saturated or unsaturated. Saturated chains are straight and have only single bonds. Unsaturated chains contain at least one double bond and have kinks. Some unsaturated fats are trans fats, which can form during industrial hydrogenation of vegetable oils. These fats stack well, but they can damage artery linings and promote inflammation. Phospholipids have a polar phosphate head and two hydrophobic tails. This structure helps form cell membranes. Steroids are fused carbon-ring lipids. Cholesterol is one steroid found in cell membranes, and it is also used to make sex hormones such as estrogen and testosterone. However, high plasma cholesterol can contribute to plaque buildup and coronary disease.

Proteins are chains of amino acids. There are 20 common amino acids, and each has the same basic structure plus a unique R-group, or side chain. Each amino acid has an N-terminal amino group and a C-terminal carboxyl group. Amino acids join from N-terminal to C-terminal through peptide bonds. Proteins work as enzymes, hormones, structural materials, and transport molecules. They also help with muscle contraction, immunity, and blood clotting. Protein changes can cause disease. In sickle cell anemia, a mutation in hemoglobin S changes red blood cell shape and exposes a hydrophobic region. This lowers oxygen transport and can make cells stick in blood vessels. People with two copies of the sickle cell gene can be severely affected. People with one copy are more resistant to malaria, which helps explain why the gene remains common in areas with malaria.

Nucleic acids store and pass on genetic information. Their monomers are nucleotides, each made of a phosphate group, a ribose sugar, and a nitrogen base. Nucleotides differ by their bases and by the type of sugar they contain. Single nucleotides can act as energy carriers and messenger molecules inside cells. Polymer forms of nucleotides include DNA and RNA, which contain the instructions used to build cellular products.

Biomolecules can be detected by using their chemical properties. Benedict’s reagent tests for reducing sugars, which are monosaccharides with free aldehyde or ketone groups that can reduce other compounds. Copper ions in the reagent change from blue to reddish orange, and a green color shows only a small amount of reducing sugar. IKI, or iodine-potassium iodide, tests for amylose in starch and turns dark bluish-black when amylose is present. If amylose is absent, the solution stays yellowish-brown. Sudan IV tests for lipids and leaves a red color when lipids are present. Biuret’s reagent tests for proteins by reacting with peptide bonds and changing from blue to dark violet. If the amino acid chain is too short, the color may appear pink instead of violet.
