Energy-requiring Steps of Glycolysis

Die Glykolyse beginnt mit einem einzelnen Glukosemolekül.
 Unter Verwendung von ATP überträgt das Enzym Hexokinase eine Phosphatgruppe auf den Sechs-Kohlenstoff-Zucker, um Glucose-6-phosphat zu produzieren, das aufgrund seiner negativen Ladung in der Zelle eingeschlossen wird.
 Als nächstes katalysiert das Enzym Phosphoglucose-Isomerase die Umwandlung der Phosphoglucose in eines ihrer Isomere, Fructose-6-phosphat.
 Die Phosphofructose kann nun durch ein geschwindigkeitsbegrenzendes Enzym, Phosphofructokinase, phosphoryliert werden, um Fructose-1,6-bisphosphat herzustellen.
 Schließlich wird mit zwei gebundenen Phosphatgruppen das Zuckermolekül durch Aldolase in zwei 3-Kohlenstoff-Isomere gespalten, Glycerinaldehyd-3-phosphat oder G3P und Dihydroxyacetonphosphat, DHAP.
 Ein anderes katalysierendes Enzym Triosephosphat-Isomerase wandelt das DHAP in G3P um, um zwei Moleküle zu erhalten.
 Während dieser Energieinvestitionsphase werden also insgesamt zwei ATPs verwendet, um ein anfängliches Glucosemolekül in zwei kleinere Zucker aufzuspalten.

<h4>Überblick</h4>
 
 Glukose ist die Quelle für fast die gesamte verbrauchte Energie in Organismen. Den ersten Schritt der Umwandlung von Glukose in nutzbare Energie nennt man Glykolyse. Die Glykolyse findet im Zytosol der Zelle in zwei Phasen statt. Die erste Phase ist die energieaufnehmende Phase und die anschließende wird als energieabgebende Phase bezeichnet. In den ersten drei Reaktionsschritten wird die Glucose in verschiedene Formen umgewandelt und bindet sich an zwei Phosphatgruppen, die von zwei ATP-Molekülen abgegeben werden. So entsteht ein instabiler Zucker. In den nächsten beiden Schritten spaltet sich der instabile Zucker in zwei Zuckerisomere, die entweder umgewandelt oder direkt in der nächsten Phase der Glykolyse verwendet werden.
 
 <h4>Der Prozess</h4>
 
 Zuerst erhält Glukose eine Phosphatgruppe von ATP, die sie in eine reaktionsfähigere Form (Glukose-6-Phosphat) umwandelt. Da die an das negativ geladene Phosphat gebundene Glucose die hydrophobe Zellmembran nicht mehr durchqueren kann, führt das Anhängen der Phosphatgruppe dazu, dass die Glukose im Inneren der Zelle eingeschlossen ist.
 
 Weiter wird die reaktionsfreudigere Form der Glucose in eines ihrer Isomere, Fruktose-6-Phosphat, umgewandelt. Es wird für die nachfolgenden energieaufwendigen Schritte der Glykolyse benötigt.
 
 Fructose-6-Phosphat erhält dann eine Phosphatgruppe von einem zweiten ATP-Molekül. Dieses wandelt Fructose-6-phosphat in Fructose-1,6-bisphosphat, einen instabilen Zucker, um.
 
 Dieser instabile Zucker spaltet sich in zwei unterschiedliche Drei-Kohlenstoff-Zucker-Isomere, Glycerinaldehyd-3-Phosphat und DHAP. Glycerinaldehyd-3-Phosphat kann direkt in der nächsten Stufe der Glykolyse verwendet werden, während DHAP auch in Glycerinaldehyd-3-Phosphat umgewandelt werden muss.

Glycolysis begins in the cytosol, where glucose is prepared for use as energy. This process has two phases: an energy-requiring phase and an energy-releasing phase. The first phase changes glucose into a more reactive, unstable form before it can move into later steps.

In the first step, glucose is phosphorylated, which means a phosphate group is added from ATP. This produces glucose 6-phosphate. The added phosphate is negatively charged, so it cannot cross the hydrophobic cell membrane. As a result, glucose is trapped inside the cell.

Next, glucose 6-phosphate is converted into its isomer, fructose 6-phosphate. An isomer is a molecule with the same atoms but a different arrangement. Fructose 6-phosphate is needed for the remaining energy-requiring steps of glycolysis. It is then phosphorylated by a second ATP molecule, forming fructose 1,6-bisphosphate.

Fructose 1,6-bisphosphate is unstable and splits into two three-carbon sugar isomers. These are glyceraldehyde 3-phosphate and DHAP. Glyceraldehyde 3-phosphate can go straight into the next phase of glycolysis. DHAP is converted into glyceraldehyde 3-phosphate before continuing.
