Energy-releasing Steps of Glycolysis

In der zweiten Hälfte der Glykolyse werden die beiden Moleküle Glycerinaldehyd 3-phosphat G3P mit der Reaktion oxidiert, durch das Enzym Glycerinaldehydphosphatdehydrogenase katalysiert und eine Phosphatgruppe an den instabilen Zucker gebunden, wobei 1,3-Bisphosphoglycerat gebildet wird.
 Infolgedessen werden zwei energiereiche Elektronen und zwei Protonen freigesetzt und vom Träger NAD+ aufgenommen, wodurch zwei NADHs und Wasserstoffionen gebildet werden.
 Die Phosphoglyceratkinase überträgt dann eine Phosphatgruppe von jedem 1,3-Bisphosphoglycerat auf ADP, wodurch zwei Moleküle ATP und 3-Phosphoglycerat erzeugt werden.
 Als nächstes wandelt das Enzym Phosphoglyceratmutase dieses Molekül in sein Isomer 2-Phosphoglycerat um, wodurch das Enzym Enolase ein Wassermolekül freisetzen und eine neue Doppelbindungsstruktur, Phosphoenolpyruvat oder PEP, bilden kann.
 Mit Hilfe der Pyruvatkinase werden die Phosphatgruppen von PEP entfernt und an ADP abgegeben, wobei zwei weitere ATP-Moleküle zusammen mit dem Endprodukt Pyruvat erzeugt werden.
 Somit werden am Ende der Glykolyse insgesamt zwei ATPs zusammen mit zwei NADHs und zwei Pyruvatmolekülen erzeugt.
 In Gegenwart von Sauerstoff kann Pyruvat weiter abgebaut werden, während NADH seine Elektronen in die Elektronentransportkette leiten kann, um NAD+ zu regenerieren.

Während die erste Phase der Glykolyse Energie verbraucht, um Glucose in Glycerinaldehyd-3-Phosphat (GAP) umzuwandeln, produziert die zweite Phase Energie. Die Energie wird in eine Reihe von Reaktionen freigesetzt. Dabei wird GAP in Pyruvat umgewandelt. Die energiefreisetzende Phase (Schritte 6-10 der Glykolyse) erfolgt zweimal. Sie läuft nämlich für jeden der beiden 3-Kohlenstoffzucker ab, die während der Schritte 1-5 produziert worden sind.
 
 Der erste energiefreisetzende Schritt (der 6. Schritt der Glykolyse) besteht aus zwei gleichzeitigen Ereignissen: Die Oxidation und Phosphorylierung von GAP. Der Elektronenträger NAD+ entfernt einen Wasserstoff aus GAP, oxidiert den 3-Kohlenstoffzucker und wandelt (reduziert) NAD+ in NADH und H+ um. Die freigesetzte Energie wird genutzt, um GAP zu phosphorylieren und in 1,3-Bisphosphoglycerat umzuwandeln.
 
 Im nächsten Schritt wandelt 1,3-Bisphosphoglycerat ADP in ATP, durch die Abgabe einer Phosphatgruppe, um. Dabei wird 3-Phosphoglycerat gebildet. Das 3-Phosphoglycerat wird dann in ein Isomer, 2-Phosphoglycerat, umgewandelt.
 
 Nachfolgend verliert 2-Phosphoglycerat ein Wassermolekül und wird zu dem instabilen Molekül 2-Phosphoenolpyruvat oder PEP. PEP gibt seine Phosphatgruppe leicht an ADP ab, wandelt es in ein zweites ATP-Molekül um, wobei Pyruvat gebildet wird.
 
 In der Energiefreisetzungsphase werden pro umgewandelten Zucker zwei Moleküle ATP und ein Molekül NADH freigesetzt. Da während der energieverbrauchenden Phase der Glykolyse zwei 3-Kohlenstoff-Zucker produziert wurden, findet dies doppelt statt. Es werden also vier ATP-Moleküle und zwei NADH-Moleküle freigesetzt. So ergibt die Glykolyse für jedes Glukosemolekül eine Nettoproduktion von zwei ATP-Molekülen (4 produziert minus 2 verbraucht, während der energieaufwendigen Phase) und zwei NADH-Molekülen.
 
 Glykolyse produziert zwei 3-Kohlenstoff-Pyruvat-Moleküle aus einem 6-Kohlenstoff-Glucosemolekül. In Gegenwart von Sauerstoff kann Pyruvat im Krebszyklus in Kohlenstoffdioxid zerlegt werden, wobei viele ATP-Moleküle freigesetzt werden. NADH sammelt sich in der Zelle an, wo es wieder in NAD+ umgewandelt werden kann und für weitere Kreislaufe der Glykolyse verwendet werden kann.

Glycolysis produces ATP and NADH during its energy-releasing phase. These reactions are the last steps of glycolysis and convert glyceraldehyde-3-phosphate, or G3P, into pyruvate. The phase is often described as steps 6 to 10, and it happens twice because two 3-carbon sugars are made earlier in glycolysis.

The sixth step begins with two linked events. G3P is oxidized, which means it loses hydrogen, and it is phosphorylated, which means a phosphate group is added. NAD+ acts as the electron carrier and removes one hydrogen atom from G3P, forming NADH and H+. The energy released in this step helps turn G3P into 1,3-bisphosphoglycerate.

Next, 1,3-bisphosphoglycerate loses a phosphate group and becomes 3-phosphoglycerate. That phosphate group is transferred to ADP, which makes ATP. Then 3-phosphoglycerate changes into its isomer, 2-phosphoglycerate. After that, 2-phosphoglycerate loses water and becomes the unstable compound 2-phosphoenolpyruvate, or PEP.

PEP quickly loses its phosphate group to ADP. This creates a second ATP molecule and leaves pyruvate. For each converted 3-carbon sugar, the energy-releasing phase makes two ATP molecules and one NADH molecule. Because the phase runs twice, glycolysis produces four ATP molecules and two NADH molecules in this stage.

Overall, one glucose molecule yields two pyruvate molecules. Glycolysis has a net gain of two ATP molecules because four ATP are produced and two ATP are used in the energy-requiring phase. It also leaves two NADH molecules in the cell. In the presence of oxygen, pyruvate can continue into the Krebs cycle, where it is broken down into carbon dioxide and more ATP is released. NADH can later be converted back into NAD+ so glycolysis can continue.
