Chromosomal Theory of Inheritance

1866 veröffentlichte Gregor Mendel die Ergebnisse seiner Experimente zur Erbsenpflanzenzüchtung. Damit lieferte er den Beweis für vorhersehbare Muster bei der Vererbung von physischen Merkmalen. Die Bedeutung seiner Erkenntnisse wurde jedoch nicht sofort erkannt. Tatsächlich war die Existenz von Genen zu dieser Zeit unbekannt. Mendel bezeichnete die Erbanlagen allgemein als Faktoren.
 
 Die Mechanismen, die Mendels Beobachtungen, wie die Spaltungsregel und die Unabhängigkeitsregel zugrunde liegen, blieben schwer verständlich. In den späten 1800er Jahren ermöglichten Fortschritte in der Mikroskopie und der Färbetechniken den Wissenschaftlern zum ersten Mal, Mitose und Meiose zu visualisieren.
 
 Anfang des 20. Jahrhunderts schlugen die Wissenschaftler Theodor Boveri, Walter Sutton und andere unabhängig voneinander vor, dass die Chromosomen den Mendelschen Regeln der chromosomalen Vererbung folgen könnten. Bei der Erforschung von Seeigeln bzw. Heuschrecken stellten Boveri und Sutton auffällige Ähnlichkeiten zwischen den Chromosomen während der Meiose und den Mendel-Faktoren fest.
 
 Wie die Mendel-Faktoren kommen die Chromosomen paarweise vor. In Anlehnung an die Mendelsche Spaltungsregel werden diese Paare während der Meiose so getrennt, dass jede Gamete (z.B. Spermium oder Eizelle) ein Chromosom von jedem Paar erhält. Die Chromosomenpaare werden unabhängig voneinander getrennt, was der Mendelschen Unabhängigkeitsregel entspricht.
 
 Der erste konkrete Beweis für die chromosomale Vererbung kam von einem ihrer Kritiker, Thomas Hunt Morgan. Morgan fand heraus, dass eine Mutation, die die Augenfarbe von Fruchtfliegen beeinflusst, von männlichen und weiblichen Fliegen unterschiedlich vererbt wird und er zeigte, dass dieses Merkmal durch das X-Chromosom bestimmt wird.
 
 Wir wissen jetzt, dass die Mendel-Faktoren DNA-Abschnitte, die Gene, an bestimmten Orten der Chromosome sind. Die unabhängige Anordnung von Genen auf verschiedenen Chromosomen ist eine Folge der zufälligen Anordnung der Chromosomen an der Mittellinie einer Zelle während der Metaphase I. Diese bestimmt, welche Gene in dieselben Tochterzellen segregieren. Jedes homologe Chromosomenpaar wandert unabhängig von den anderen. Die Spaltungsregel entspricht der Bewegung der Chromosomen während der Anaphase I, die unter normalen Bedingungen sicherstellt, dass jede Keimzelle nur ein Exemplar jedes zufällig verteilten Chromosoms erhält.

Genes map to chromosomes, and this idea explains how Mendel’s inheritance patterns work. Gregor Mendel published his pea plant breeding results in 1866. He showed that physical traits follow predictable patterns. At the time, scientists did not yet know that genes existed. Mendel used the word &quot;factors&quot; for the hereditary units he studied.

The details behind Mendel’s laws of segregation and independent assortment were still unknown for many years. In the late 1800s, better microscopes and staining methods let scientists see mitosis and meiosis for the first time. These advances gave researchers a way to study how hereditary material behaves in cells. The chromosomal theory of inheritance grew from this work in the early 1900s.

Theodor Boveri and Walter Sutton independently suggested that chromosomes could explain Mendel’s laws. Boveri studied sea urchins, and Sutton studied grasshoppers. They saw that chromosomes during meiosis looked much like Mendel’s factors. Chromosomes also come in pairs, just as Mendel’s factors do. During meiosis, paired chromosomes separate so each gamete, such as a sperm or egg, receives one chromosome from each pair. Chromosome pairs also separate independently from one another, matching Mendel’s law of independent assortment.

Strong evidence came from Thomas Hunt Morgan, who had been a critic of the theory. He studied a fruit fly eye color mutation and found that it was inherited differently in males and females. This showed that the trait was linked to the X chromosome. We now know that Mendel’s factors are DNA segments called genes, and each gene sits at a specific place on a chromosome. The random arrangement of chromosome pairs at metaphase I affects which genes end up in the same daughter cells, and anaphase I helps ensure that each gamete gets one copy of each chromosome under normal conditions.
